Adrenergic receptor subtype activation by (+)-, (-)- and (+/-)-norephedrine in the pithed rat.

The ability of (+/-)-norephedrine (phenylpropanolamine) and its component isomers, (+)-and (-)-norephedrine, to activate adrenergic receptor subtypes in the cardiovascular system of the urethane/chloralose-anaesthetized pithed rat has been investigated. At all adrenoceptor subtypes, (-)-norephedrine was the most potent agonist followed by (+/-)- then (+)-norephedrine. The greatest activity was observed at the alpha 1-receptor, with little activity observed at either beta 1 or beta 2-adrenoceptors. Reserpinization shifted the (-)-norephedrine dose-response curve slightly to the right, indicatin

Cardiovascular effects of two potential beta-hydroxylase inhibitors.

The purpose of this study was to evaluate the cardiovascular effects of two potential beta-hydroxylase inhibitors, a dithiocarbamate and a thiourea derivative of d-amphetamine, 1 and 2, respectively. These compounds when given intravenously elicited a significant dose-dependent depressor effect in normotensive rats and dogs anesthetized with chloralose-urethane. The hypotensive effect of these compounds appears to be the result of a combination of two action components: (a) the major component, which can be blocked by atropine, exerted through the parasympathetic division and (b) a decreased a

Effect of phenytoin on cardiac slowing induced by cholinergic stimulation.

Phenytoin was tested for its ability to counteract cholinergic-induced slowing in sinus rate in chloralose-anesthetized cats subjected to bilateral cervical vagotomy and spinal cord transection. The cholinergic stimuli consisted of electrical stimulation of the cervical vagus nerves and injected acetylcholine. Phenytoin administered as a continuous infusion of 0.5 and 1.0 mg/kg/min was found to significantly reduce the cardiac slowing produced by both modes of cholinergic stimulation. Blockade of the acetylcholine response suggested that the inhibitory effect of phenytoin was on the postsynapt

General anesthetics hyperpolarize neurons in the vertebrate central nervous system.

The effect of general anesthetics on frog motoneurons and rat hippocampus pyramidal cells was examined with sucrose gap and intracellular recording, respectively. A number of volatile and intravenous anesthetics directly hyperpolarized the motoneurons. The potency of these agents in hyperpolarizing motoneurons was strongly correlated with their anesthetic potency. While the responses to barbiturates and alpha-chloralose were blocked by gamma-aminobutyric acid antagonists and were dependent on the chloride gradient, the responses to all the other anesthetics tested were generated by a separate

The influence of naloxone on the C fiber response of dorsal horn neurons and their inhibitory control by raphe magnus stimulation.

In intact rats anesthetized with chloralose, the effects of naloxone were studied on the responses of spinal cord dorsal horn neurons to C fiber stimulation and upon the inhibition induced on these responses by stimulation of the nucleus raphé magnus (NRM). (1) A mean 44% facilitatory effect on responses to C fibers was observed for 12/19 units. (2) A mean 30% reduction of the inhibitory effects of NRM was found for 14/29 units. (3) However there is no clear relationship between these facilitatory effects and the diminution of the efficiency of NRM stimulation. These results demonstrate a faci

Interaction of visceral and somatic afferent systems in the cerebral cortex.

Using the method of evoked potentials on chloralose-anesthetized and suxamethonium-immobilized cats, a comparative study was made of the effects of interaction of somatic and visceral afferent systems in the second somatosensory area and in the parietal cortex of the brain. It was found that a common property of the interaction of the two afferent systems is an inhibition of the second response irrespective of which of the systems is activated first. Fairly prolonged inhibition was noted in the association region of the parietal cortex. Preliminary stimulation of visceral nerves caused less pr

The effects of hyperventilation on the reflex cardiac response from the carotid bodies in the cat.

1. Cats were anaesthetized with chloralose and urethane, and ventilated by an artificial intermittent negative pressure applied to the thorax. The carotid body chemoreceptors were isolated and perfused with oxygenated blood. They were stimulated by substituting hypoxic blood obtained from a donor animal.2. Stimulation of the carotid bodies during constant ventilation caused a bradycardia. When an artificial hyperventilation was induced during carotid body stimulation the heart rate increased.3. The increase in heart rate during hyperventilation, and while the carotid bodies were being stimulat

Morphine-induced modulation of sciatic nerve stimulation evoked blood pressure responses.

Effect of i.v. morphine (1-2 mg/kg) on blood pressure changes evoked by sciatic nerve stimulation (SNS) were studied in chloralose anaesthetised cats. SNS gave a depressor, pressor or a biphasic BP response generally linked to the parameters of stimulation. Morphine produced marked attenuation of depressor and some facilitation of pressor response, suggesting a possible reciprocal relationship between depressor and pressor responses. Depressor response has been correlated with deep tissue or visceral pain mediated through A delta fibres and pressor response to cutaneous nociception involving C

Stimulation of caudal and cranial ends of vagi in vagotomised chloralose-urethane anaesthetised dogs with 7.5V, 30Cps pulses of 2 msec width after 0, 25, 50, 75 and 100 ml/kg body weight infusion dose of Ringer-Locke solution caused well known effects on heart rate, BP, amplitude of peripheral blood pressure waves, amplitude and frequency of respiration and amplitude of ECG waves. The vagal stimulation effects did not appear to be related to volume of fluid infused into circulation except that caudal end stimulation effect on QRS amplitude was prominent during higher infusion doses while its e

[Assessment of the influence of pharmacologic substances on pressor vasomotor reflexes in anesthetized animals].

Causes accounting for differing results of experiments during studies on the effects of morphine and chlordiazepoxide upon the intensity of pressor vasomotor reflexes provoked by an electric stimulation of afferent fibers in the tibial nerve were investigated in tests set up on urethan and chloralose anesthetized animals. With this purpose in view the relationship between the amplitude of the vasomotor reflexes and the intensity of electric stimulation, as well as the nature of the drugs action upon the vasomotor reflexes, depending on the stimulation parameters adopted during their registrati

Stimulation of carotid chemoreceptors and ventilation by doxapram in the cat.

Small doses of doxapram (less than 0.5 mg . kg-1 iv) were used to study its effect on carotid chemoreceptor activity and ventilation in cats anesthesized with alpha-chloralose. The effects were studied at several levels of partial pressures of O2 and CO2 in arterial blood (PaO2 and PaCO2). It was found that doxapram stimulated discharge rate of the carotid chemoreceptor afferents by the same magnitude at all levels of PaO2 and PaCO2 studied. However, the ventilatory effect of doxapram was more than additive to the concomitant stimulation by hypoxia or hypercapnia. This ventilatory effect was e

Tachyphylaxis to the hypotensive effect of clonidine and the possible mechanism of this action.

In cats under chloralose-urethan anaesthesia, tachyphylaxis developed to the hypotensive effect of clonidine. Development of this tachyphylaxis was slow in the case of intravenous low doses (10--20 micrograms/kg) and rapid after high doses (50 micrograms/kg). No tachyphylaxis developed to contractions of the nictitating membrane. The phenomenon resembles the tachyphyaxis seen after the application of indirect sympathomimetics. A tachyphylaxis developed to the latter agents inhibited the hypotensive action of clonidine which, after such an intervention, invariably turned into a vasopressor effe

Digoxin-induced decrease in intraocular pressure in the cat.

A constant intravenous infusion of digoxin (2 micrograms/kg/min) to alpha-chloralose-anesthetized cats produced a progressive decrease in intraocular pressure with increasing doses of digoxin between 60 micrograms/kg and drug-induced ventricular arrhythmia which occurred at a mean dose of 172 micrograms/kg. Digoxin elicited a 40% decrease in intraocular pressure just prior to ventricular arrhythmia compared to a decrease of only 10% with a control infusion of diluent in the same animal (P less than 0.05). There was no significant difference (P less than 0.1) between changes in blood pressure a

Vesico-somatic reflexes in the chronic spinal cat.

1. In cats anaesthetized with chloralose whose spinal cords were transected 6-7 weeks previously, spontaneous contractions of the bladder had no effect on mono- or polysynaptic reflexes recorded electrically from the cut lumbar ventral spinal roots.2. On the direct and crossed components of reflex responses to unilateral stimulation of the carotid body chemoreceptors in the dog.

1. In dogs under chloralose-urethane anaesthesia the chemoreceptors of the two carotid bodies were separately stimulated.2. The distribution of three primary reflex responses to carotid body stimulation was studied: parasympathetic bradycardia, sympathetic vasoconstriction, and increase in somatic phrenic nerve activity.3. The reflex bradycardia evoked by either carotid body was mediated by both vagus nerves, but when either vagus was blocked a greater response could be obtained from the contralateral than from the ipsilateral carotid body.4.
