Chloroplast genomes of nine Primula species: comparative genomics and phylogenetic implications.

INTRODUCTION: Primula is the largest genus in Primulaceae and a classic model for studying heterostyly and plant evolution. METHODS: In this study, we sequenced, assembled, and annotated the complete chloroplast genomes of seven Primula species from China. Two additional plastomes of Primula dejuniana and Primula meishanensis, previously generated and published by our research group, were retrieved from GenBank and integrated into our dataset. We further combined these nine plastome resources with 48 published Primula plastomes and three outgroup plastomes downloaded from GenBank, forming a t

Characterization of the complete chloroplast genome of Bupleurum komarovianum Lincz. 1950 (Apiaceae).

Bupleurum komarovianum is distributed in northeastern China and has occasionally been used as a substitute for the official Radix Bupleuri due to its morphological similarity and comparable pharmacologically active constituents. To facilitate its accurate identification, the complete chloroplast genome of B. komarovianum was assembled and annotated, followed by phylogenetic analysis. The chloroplast genome is 155,835 bp in length, with a GC content of 37.70%, and exhibits a typical quadripartite structure. A total of 128 genes were annotated, including 84 protein-coding genes, 36 tRNA genes, a

Complete chloroplast genomes of endemic Astragalus and Oxytropis species from Uzbekistan.

Chloroplast genomes provide important insights into plant phylogeny, genome evolution, and molecular marker development. In this study, we sequenced, assembled, and analyzed the complete chloroplast genomes of two endemic species from Uzbekistan, Astragalus nuratensis and Oxytropis pseudorosea. Genome skimming generated high-quality paired-end reads, enabling the recovery of complete plastomes with mean sequencing depths of 638× and 1,725×, respectively. The chloroplast genomes were 122,316 bp in A. nuratensis and 122,708 bp in O. pseudorosea. Both genomes encoded 110 unique genes, including 7

Disequilibrium between chloroplast proton motive force and ATP levels in Arabidopsis.

Current dogma holds that CO2 fixation by photosynthesis requires additional ATP production via cyclic electron transfer, mediated by two distinct pathways involving the NADH dehydrogenase-like complex (NDH) and proton gradient regulation 5 protein (PGR5), to augment the ATP relative to NADPH produced by linear electron transfer. Here we use the MgATP2- fluorescent biosensor ATeam1.03-nD/nA and find that, while proton motive force in the Arabidopsis pgr5CAS and pgr5 ndho mutants is only 50-75% that of the wild type, chloroplast MgATP2- levels are unchanged. These data demonstrate that disequili

Iron allocation to chloroplast proteins depends on the DNA-binding protein WHIRLY1.

The DNA-binding protein WHIRLY1, sharing structural similarities with ferritin, plays a role in the formation of iron cofactor proteins within chloroplasts. Previous studies indicated that barley plants with a knockdown of HvWHIRLY1 containing a minimal amount of the protein are compromised in chloroplast development and photosynthesis, and get chlorotic leaves when grown at high irradiance. Thereby, the leaves display signs of iron deficiency. Metal determination revealed, however, that leaves of WHIRLY1-deficient plants had a regular iron content. Nevertheless, WHIRLY1-deficiency affected th

Dihydroxyacetone phosphate generated in the chloroplast mediates the activation of TOR by CO2 and light.

Light and CO2 assimilation activate the target of rapamycin (TOR) kinase in photosynthetic cells, but how these signals are transmitted to TOR is unknown. Using the green alga Chlamydomonas reinhardtii as a model system, we identified dihydroxyacetone phosphate (DHAP) as the key metabolite regulating TOR in response to carbon and light cues. Metabolomic analyses of synchronized cells revealed that DHAP levels change more than any other metabolite between dark- and light-grown cells and that the addition of the DHAP precursor, dihydroxyacetone (DHA), was sufficient to activate TOR in the dark.

The complete chloroplast genome sequence of Primula medogensis (Primulaceae) and its phylogeny.

Primula medogensis W.B Ju, B. Xu & X.F. Gao 2023, a new species categorized under P. sect. Cordifoliae, was officially described in 2023. Given its recent classification, the genetic resources for this species are currently very limited. Here, we sequenced and assembled the first complete chloroplast genome of P. medogensis using Illumina sequencing technology. The complete chloroplast genome of P. medogensis is 151,486 bp in length, exhibiting a typical quadripartite structure. It consists of a large single-copy region (LSC; 83,407 bp) and a small single-copy region (SSC;17675 bp), separated

Dual roles for CND1 in maintenance of nuclear and chloroplast genome stability in plants.

The coordination of chloroplast and nuclear genome status is critical for plant cell function. Here, we report that Arabidopsis CHLOROPLAST AND NUCLEUS DUAL-LOCALIZED PROTEIN 1 (CND1) maintains genome stability in the chloroplast and the nucleus. CND1 localizes to both compartments, and complete loss of CND1 results in embryo lethality. Partial loss of CND1 disturbs nuclear cell-cycle progression and photosynthetic activity. CND1 binds to nuclear pre-replication complexes and DNA replication origins and regulates nuclear genome stability. In chloroplasts, CND1 interacts with and facilitates bi

Expression of a mosquito larvicidal gene in chloroplast and nuclear compartments of Chlamydomonas reinhardtii.

As a part of the search for environment-friendly biocontrol of mosquito-borne diseases, mosquito larvicidal potential of Bacillus thuringiensis subsp. jegathesan (Btj) Cry toxins is explored for toxins with increased toxicity. Safe delivery of the Cry toxins to mosquito larvae in aquatic habitats is a major concern. This is because in water bodies Bacillus thuringiensis (Bt) protein formulations degrade by sunlight, can sink down and get adsorbed by the silt. So, because of its short persistence the toxin requires repeated applications at the given site. Therefore, an upcoming approach is inco

Multiple regulatory mechanisms in the chloroplast of green algae: relation to hydrogen production.

A complex regulatory network in the chloroplast of green algae provides an efficient tool for maintenance of energy and redox balance in the cell under aerobic and anaerobic conditions. In this review, we discuss the structural and functional organizations of electron transport pathways in the chloroplast, and regulation of photosynthesis in the green microalga Chlamydomonas reinhardtii. The focus is on the regulatory mechanisms induced in response to nutrient deficiency stress and anoxia and especially on the role of a hydrogenase-mediated reaction in adaptation to highly reducing conditions

EMB1211 is required for normal embryo development and influences chloroplast biogenesis in Arabidopsis.

Chloroplast biogenesis is tightly linked with embryogenesis and seedling development. A growing body of work has been done on the molecular mechanisms underlying chloroplast development; however, the molecular components involved in chloroplast biogenesis during embryogenesis remain largely uncharacterized. In this paper, we show that an Arabidopsis mutant carrying a T-DNA insertion in a gene encoding a multiple membrane occupation and recognition nexus (MORN)-containing protein exhibits severe defects during embryogenesis, producing abnormal embryos and thereby leading to a lethality of young

N-myristoylation-mediated shuttling of TaMP from plasma membrane to chloroplasts increases wheat susceptibility to rust fungi.

Chloroplasts are significant generators of reactive oxygen species and are crucial for plant immunity. However, how these subcellular organelles detect external stimuli and initiate specific immune responses remains largely unknown. Here we identified a plasma membrane (PM)-anchored N-myristoylated protein, TaMP, which detaches from the PM and relocalizes to chloroplasts upon exposure to pathogen-associated molecular patterns. We also uncovered a secreted protease from the rust fungus, Puccinia striiformis f. sp. tritici, that hijacks and promotes TaMP relocalization, increasing wheat suscepti

Expression of endochitinase and exochitinase in lettuce chloroplasts increases plant biomass and kills fungal pathogen Candida albicans.

Lettuce (Lactuca sativa) is a popular leafy vegetable with global production of ~28 million Mt, cultivated >1 million hectares, with a market value of US$ 4 billion in 2022. However, lettuce is highly susceptible to fungal pathogens that drastically reduce biomass and quality due to spoilage/rot. Therefore, in this study, we investigated the expression of chitinase genes via the lettuce chloroplast genome to enhance biomass and disease resistance. Site-specific integration of the expression cassette into chloroplast genomes was confirmed using two sets of PCR primers. Homoplasmy in transplasto

Functional relationship of atypical thioredoxins with NADPH-thioredoxin reductase C and 2-Cys peroxiredoxins in Arabidopsis chloroplasts.

In addition to typical thioredoxins (TRXs) of the types m, f, x, y, and z, Arabidopsis (Arabidopsis thaliana) chloroplasts contain atypical TRXs such as cysteine- and histidine-rich TRXs (ACHTs) and TRXs-like 2 (TRXs L2), but their contribution to the redox regulation of the organelle and their functional relationship with the NADPH-TRX reductase C (NTRC)-2-Cys peroxiredoxin (PRX) redox system are poorly understood. To address these issues, we have focused on atypical TRXs ACHT3, ACHT4, and TRX L2.2. Either single or multiple Arabidopsis mutants combining deficiencies in these atypical TRXs sh

[Attachment of horse cecum Ciliata to plant fragments. Degradation of chloroplasts. Attachment of bacteria to cecal Ciliata].

Cecum microfauna association with different plant tissues was examined by scanning and transmission electron microscopy. The ciliates were attached to the damaged areas of the leaves and their highest concentrations were found on the epidermis and mesophyll tissues. The degradation of plant tissue was due to protozoal ingestion of the plant fragments. The morphology of ingested envacuolized chloroplasts changed rapidly, showing different stages of digestion inside the ciliate endoplasm. Intact chloroplasts were rarely observed but the grana of fragmented thylakoid membranes was often seen. The

Far-red light induces ultrastructural reorganization of chloroplasts in Marchantia polymorpha to compensate for reduced photosynthetic efficiency.

Far-red light is stressful to Marchantia polymorpha and reduces the efficiency of photosystem II. In response, Marchantia reorganizes the chloroplast ultrastructure, forming Rubisco condensate regions to physicochemically concentrate CO₂. The light spectral composition is one of the key photomorphogenesis factors. Far-red light (FRL) resulted in marked changes in the morphology of Marchantia polymorpha chloroplasts. However, most aspects of this photomorphogenesis remain unexplored, and this work is devoted to elucidating their nature. We studied in vitro cultured M. polymorpha under FRL or wi

Antibiotic use and abuse: a threat to mitochondria and chloroplasts with impact on research, health, and environment.

Recently, several studies have demonstrated that tetracyclines, the antibiotics most intensively used in livestock and that are also widely applied in biomedical research, interrupt mitochondrial proteostasis and physiology in animals ranging from round worms, fruit flies, and mice to human cell lines. Importantly, plant chloroplasts, like their mitochondria, are also under certain conditions vulnerable to these and other antibiotics that are leached into our environment. Together these endosymbiotic organelles are not only essential for cellular and organismal homeostasis stricto sensu, but a

Chemical cross-linking and its effect on fatty acid synthetase activity in intact chloroplasts from Euglena gracilis.

Intact chloroplasts were isolated from Euglena gracilis variety bacillaris, aliquots were exposed to several different chemical cross-linking reagents. The reagents penetrated the triple membrane of Euglena chloroplasts. This was shown by gradient acrylamide gel electrophoresis under denaturing conditions. The activity of the nonaggregated fatty acid synthetase of Euglena was located within the chloroplast stroma, and the effects of dimethylsuberimidate cross-linking on the activity of the enzyme system were examined. The acyl-carrier protein concentration in the chloroplast was measured at ab

The effects of carbonic anhydrase inhibitors formate, bicarbonate, acetazolamide, and imidazole on photosystem II in maize chloroplasts.

Inhibitors of carbonic anhydrase were tested for their effects on Photosystem II (PS II) activity in chloroplasts. We find that formate inhibition of PS II turnover rates increases as the pH of the reaction medium is lowered. Bicarbonate ions can inhibit PS II turnover rates. The relative potency of the anionic inhibitors N-3, I-, OA-c, and Cl- is the same for both carbonic anhydrase and PS II. The inhibitory effect of acetazolamide on PS II increases as light intensity decreases, indicating a lowering of quantum yields in the presence of the inhibitor. Imidazole inhibition of PS II increases
