OBJECTIVE: To explore the efficacy and feasibility of interstitial chemotherapy using thermosensitive gel-coated ricin in hepatoma H22-bearing mice.

A nitrocellulose-based assay was developed using a dot-blot apparatus to detect phenoloxidase activity in column fractions. Using this assay, plasma phenoloxidase was partially purified from Aedes aegypti larvae using hydrophobic interaction chromatography, gel filtration, and ion-exchange chromatography. The molecular weight (M(r)) native enzyme was 130,000, and it contained subunits of 76,000, 62,000, and 58,000. Two phenoloxidase peaks were observed by ion exchange chromatography, and these fractions had distinct polypeptide profiles as detected by SDS-PAGE.

Two allergenic components, termed J1 and J2, were isolated from a soluble egg antigen preparation (SEA) of Schistosoma japonicum by anion-exchange chromatography on DE52 and gel chromatography on Sephacryl S-200. This report concerns the isolation of bullous pemphigoid antigen from the nondialyzable urinary components of a patient with the disease. The isolation was accomplished by ion exchange chromatography and gel filtration. Pemphigoid antigen was found to be a basic glycoprotein that on SDS gel electrophoresis showed two major bands, one in the 18,000 m. w. region and the second with a m. w. of 74,000. Cryptorchid testes of testicular feminization rats are very low in zinc in spite of normal zinc status of the animals. Analysis of the cytosol of the cryptorchid testes by gel permeation chromatography showed decreased zinc binding by proteins eluted at fractions corresponding to 30,000 dalton. Further analysis by sodium dodecylsulphate polyacrylamide gel electrophoresis indicated the absence of a protein with molecular weight of 23,000.

By means of DEAE-Sepharose CL-6B ion exchange chromatography and TSK-GEL G2000 SW high-performance gel filtration, a purified protein with fibrinolytic activity was obtained from the venom of Agkistrodon halys brevicaudus (Korean mamushi). The hyphomycete Chalara paradoxa CH32 produced an extracellular beta-glucosidase during the trophophase. The enzyme was purified to homogeneity by ion-exchange and size-exclusion chromatography. BACKGROUND: Photoallergic contact dermatitis from ketoprofen has been recognized since the mid-1980s. Skin reactions have been reported to continue weeks after discontinuation of ketoprofen. One reason for this could be residual ketoprofen in the skin, which has been shown in a skin biopsy specimen.

A glutathione peroxidase was purified from bovine ciliary body by ammonium sulfate fractionation. Tonin was isolated from rat submandibular glands by a very convenient procedure consisting of sequential anion-exchange, hydrophobic interaction and gel filtration chromatographies. The method is superior to earlier purifications as it consists of fewer stages, resulting in a much higher recovery (41%) of tonin. The final preparation was seen to be pure on SDS gel electrophoresis (M(r) 32,800) and on gel isoelectric focusing (pI 6.15). The stability of tonin and its interaction with various inhibitors were investigated, and compared with the corresponding behavior of rat tissue kallikrein.

An allergenic protein fraction was isolated from wheat gluten by ion exchange and gel filtration chromatography. On sodium dodecyl sulphate gel electrophoresis the molecular mass of the protein was found to be 65 kDa. By Western blotting it was confirmed that the 65 kDa protein was the major allergenic fraction causing dermatitis herpetiformis (DH) and it contained glutamic acid and proline as the major amino acids.
