Heterozygotes for phenylketonuria and controls were given oral loads of 100 mg and 200 mg L-phenylalanine per kilogram body weight. The concentrations of urinary aromatic acids were determined by gas-chromatography after isolation by ion-exchange chromatography and ethylacetate extraction. On an intake of 100 mg L-phenylalanine per kilogram, controls and carriers of classical phenylketonuria excreted nearly the same amounts of aromatic acids (P greater than 0.05). However on an intake of 200 mg per kilogram L-phenylalanine they could be distinguished from one another (P less than 0.001).

Protein C activator from Agkistrodon halys halys venom has been isolated and purified by ion-exchange and gel-filtration chromatography. The purified enzyme consists of the single peptide-chain with molecular weight of 34 kDa. Its pH-optimum was in 7.5-8.0 range. The enzyme was inhibited by DFP, benzamidine, PMSF, EGTA. Protein C activator was as effective as "Protac" (Pentapharm AG, Switzerland).

OBJECTIVE: To determine the kinds of proteoglycan (PG) monochain and their distribution in the cell layer and cultural medium of bovine trabecular meshwork (BTM) culture.
METHODS: (3)H-glycosamine was incorporated in BTM cells in the third passage. PGs were extracted by 4 mol/L guanidine hydrochloride. The aliquots were chromatographed on Sephadex G-50 DEAE-Sephacel, Sepharose 4B and Sepharose CL-6B gel respectively. Automated cation exchange microcolumn chromatography of haemoglobins has been modified for the analysis of haemoglobin A2. It provides the quantitative data of sufficient precision and specificity for the investigation of potential heterozygotes for beta-thalassaemia. A monoclonal immunoglobulin, IgM-kappa, was detected in the serum of a patient with Burkitt's lymphoma. This rarely reported finding raises the possibility that undifferentiated neoplastic lymphoreticular cells in this disorder have the capacity to synthesize immunoglobulins. BACKGROUND: Patients with inherited disorders of amino acid metabolism including maple syrup urine disease, tyrosinaemia and phenylketonuria on dietary management require frequent monitoring of disease-relevant plasma amino acids in order to optimize therapeutic benefit.
