In this study, we describe the antimycobacterial activity of two pigments, violacein, a purple violet pigment from Janthinobacterium sp. Ant5-2 (J-PVP), and flexirubin, a yellow-orange pigment from Flavobacterium sp. Ant342 (F-YOP). These pigments were isolated from bacterial strains found in the land-locked freshwater lakes of Schirmacher Oasis, East Antarctica. The minimum inhibitory concentrations (MICs) of these pigments for avirulent and virulent mycobacteria were determined by the microplate Alamar Blue Assay (MABA) and Nitrate Reductase Assay (NRA). Results indicated that the MICs of J-

Approximately 4.2% of 4,000 Maryland-Virginia raw milk tanker samples developed ropiness when incubated at 10 degrees C. Of the 56 bacterial isolates 30 were identified by species. Klebsiella oxytoca and Pseudomonas aeruginosa were isolated most frequently. Other ropy isolates were identified as Pseudomonas spp., Chromobacterium, Flavobacterium multivorum, presumptive Yersinia pestis, Enterobacter agglomerans, Klebsiella pneumoniae, and Pasteurella-Actinobacter spp. Six of the Klebsiella oxytoca isolates were mesophilic (optimum temperatures of 32.0 to 37.8 degrees C) with two isolates having

Development of antimicrobial resistance in bacteria is a major concern. The study of quorum sensing (QS), the bacterial communication system that coordinates important events in an infection, opens a new avenue for treating infections. QS helps in biofilm formation which acts as a physical barrier and alters the microenvironment which traps the molecules or slows down the antibiotic diffusion making them less effective. Since the standard drugs are not very effective against bacteria anymore, demand for the discovery of other alternatives arises. Phytochemicals extracted from commonly used pla

Rapid technological advancement and relatively short life time of electronic goods have resulted in an alarming growth rate of electronic waste which often contains significant quantities of toxic and precious metals. Compared to conventional recovery methods, bioleaching is an environmentally friendly process for metal extraction. Gold was bioleached from electronic scrap materials (ESM) via gold-cyanide complexation using cyanide produced from pure and mixed cultures of cyanogenic bacteria Chromobacterium violaceum, Pseudomonas aeruginosa and Pseudomonas fluorescens. As ESM was toxic to the

Vibriosis is a major bacterial disease that causes significant mortality in farmed shrimp worldwide. Numerous studies have explored strategies to prevent mass mortalities associated with vibriosis outbreaks. This study aimed to identify microalgal species with immunostimulant and anti-Vibrio properties for shrimp culture. Microalgal screening began with centrifugal supernatant extraction, followed by metabolite extraction using ethyl acetate. The metabolites were subjected to an anti-quorum sensing (QS) bioassay via the disc diffusion method, using Chromobacterium violaceum as an indicator str

Spent automotive catalysts represent an important secondary resource for platinum group metals, offering environmental and economic advantages over primary mining. This review evaluates bioleaching-based recovery strategies of these metals as sustainable alternatives to conventional pyrometallurgical and hydrometallurgical processing. The cyanogenic bioleaching using Chromobacterium violaceum, Pseudomonas fluorescens, and Bacillus megaterium, and acidophilic bioleaching using Acidithiobacillus spp. for washcoat degradation and base-metal removal are discussed through the one-step, two-step, sp

A biosurfactant-producing fungal strain, Exophiala spinifera CSF123, was isolated from cashew oil-contaminated soil. Internal transcribed spacer sequencing followed by phylogenetic analysis revealed that the isolated fungus shared 98.42% similarity with E. spinifera CBS66.76. The biosurfactant molecule, named ES-414, produced by the fungus was purified to homogeneity by HPLC, and subsequent electrospray mass spectrometric analysis showed that its neutral mass is 414 Da. The molecule displayed remarkable antibiofilm activity. At concentrations of 15 and 80 μg/mL, ES-414 showed 62.17% biofilm in

The root of Platycodon grandiflorum has long been used as a vegetable and traditional medicine. Although the antibacterial activity of the plant's lipophilic (hexanes and dichloromethane) fractions has been reported, the specific antibacterial compounds have not been identified. In this study, chemical analysis of the lipophilic fractions of P. grandiflorum extracts led to the discovery of five new polyacetylenes (1-5) and nine known compounds (6-14). Their structures were elucidated and confirmed based on 1D and 2D NMR data together with mass spectra. In particular, the relative and absolute

INTRODUCTION: Burkholderia thailandensis is a study model for Burkholderia pseudomallei, a highly virulent pathogen, known to be the causative agent of melioidosis and a potential bioterrorism agent. These two bacteria use an (acyl-homoserine lactone) AHL-mediated quorum sensing (QS) system to regulate different behaviors including biofilm formation, secondary metabolite productions, and motility.
METHODS: Using an enzyme-based quorum quenching (QQ) strategy, with the lactonase SsoPox having the best activity on B. thailandensis AHLs, we evaluated the importance of QS in B. thailandensis by c

Lectins, also known as sugar binding proteins, play an essential role in the initiation of bacterial infections and biofilm production. To date, several lectins of Gram-negative bacteria such as Pseudomonas aeruginosa, Burkholderia cenocepacia, Ralstonia solanacearum and Chromobacterium violaceum have been identified. There are no published reports on the presence of lectins in Burkholderia pseudomallei, the causative agent of melioidosis. The aim of this study was to identify possible lectin genes of B. pseudomallei and generate recombinant proteins for assessment of hemagglutinating activity

The number of discarded electric devices containing traces of Au is currently increasing. It is desirable to recover this Au because of its valuable physicochemical properties. Au is usually dissolved with relatively high concentrations of cyanide, which is associated with environmental risk. Chromobacterium violaceum is able to produce and detoxify small amounts of cyanide, and may thus be able to recover Au from discarded electric devices. This study investigated the effects of cyanide and dissolved oxygen concentration on biological Au recovery. Cyanide production by C. violaceum was suffic

We have studied the intrinsic tryptophan fluorescence of the lipases from Chromobacterium viscosum (CVL), Pseudomonas species (PSL), and Rhizopus oryzae (ROL) in aqueous buffer, zwitterionic detergent micelles, and isopropanol-water mixtures. It was the purpose of this study to obtain information about biophysical properties of the respective enzymes under conditions that modulate enzyme activities and stereoselectivities to a significant extent. According to their decay-associated emission spectra, CVL tryptophans are located in the hydrophobic interior of the protein. In contrast, the PSL an

Quorum sensing (QS) is a critical cell-to-cell communication mechanism employed by microorganisms to coordinate their collective behaviors, including biofilm formation, virulence regulation, and development of antimicrobial resistance (AMR). Disruption of QS using quorum sensing inhibitors (QSIs) has emerged as a promising anti-virulence strategy for combating multidrug-resistant bacterial infections. This review focuses on new technologies in drug discovery applied to the identification and optimization of QSIs, with particular emphasis on computational and data-driven methods. The key method

Both the ability of bacteria to form biofilms and communicate through quorum sensing allows them to develop different survival or virulence traits that lead to increased bacterial resistance against conventional antibiotic therapy. Here, seventeen essential oils (EOs) were investigated for the antimicrobial, antibiofilm, and anti-quorum sensing activities on Escherichia. coli O157:H7, Escherichia coli O33, and Staphylococcus epidermidis ATCC 12228. All essential oils were isolated from plant material by using hydrodistillation and analyzed by GC-MS. The antimicrobial activity was performed by

In order to search for antifungal from biological origin, we performed a screening of marine microorganisms isolated from seawater, seaweed, sediment and marine invertebrates collected from different coastal areas of the Moroccan Atlantic Ocean. The antifungal activities of these isolates were investigated against the pathogenic yeasts involved in medical mycology. Whole cultures of 34 marine microorganisms were screened for antifungal activities using the method of agar diffusion against four yeasts. The results showed that among the 34 isolates studied, 13 (38%) strains have antifungal activ

Phenylketonuria (PKU) is a disease in which phenylalanine and phenylalanine-derived metabolites build up to neurotoxic levels due to mutations in the phenylalanine hydroxylase gene (PAH). Enzyme replacement therapy is a viable option to supply active PAH. However, the inherent protease sensitivity and potential immunogenicity of PAH have precluded adoption of this approach. In this report, we have used polyethylene glycol derivatization (PEGylation) to produce protected forms of PAH for potential therapeutic use. Three recombinantly produced PAH enzymes were reacted with activated PEG species,

Granulomas are defined by the presence of organized layers of immune cells that include macrophages. Granulomas are often characterized as a way for the immune system to contain an infection and prevent its dissemination. We recently established a mouse infection model where Chromobacterium violaceum induces the innate immune system to form granulomas in the liver. This response successfully eradicates the bacteria and returns the liver to homeostasis. Here, we sought to characterize the chemokines involved in directing immune cells to form the distinct layers of a granuloma. We use spatial tr

Quorum sensing (QS) is a cell-density-dependent communication system that coordinates bacterial virulence, biofilm formation, motility, and stress adaptation, making it a compelling target for antivirulence intervention. Due to the possibility of sulfonyl (-SO₂-) and sulfinyl (-S = O-) functional groups to impart specific electronic and steric properties that can modulate bacterial signaling and receptor interactions, organosulfur compounds are increasingly gaining attention. This review highlights recent advances in the development of sulfonyl- and sulfinyl-containing organosulfur compounds a
