[Establishment of an animal model with hypertrophic scar].

OBJECTIVE: To establish an animal model of hypertrophic scar on the rabbit ears and to observe the dynamic process of scar formation on the ventral side of the rabbit ears. METHODS: Full skin defect (2 cm x 5 cm in diameter) was created on the ventral side of 60 ears in 32 New Zealand white rabbits. 1% silver sulfadiazine cream was applied to the wounds, and the dressing was changed once a week till wound healing. Four ears without operation were taken as control. Wound healing under natural condition was observed continuously for 12 months after the operation. The scar proliferation on the r

Postburn galactorrhea with refractory hypertrophic scars: role of obesity under scrutiny.

Postburn galactorrhea, although relatively uncommon, is a complex problem to treat. Three of 25 female premenopausal patients who were admitted during the years 1995 to 2001 with more than 40% TBSA burns developed this problem. All three patients were obese according to body mass index and other clinical criteria. It was observed that the additional disturbance of equilibrium of hypothalamus because of burn injury, which is already disturbed as per se in obese patients, precipitates sustained release of prolactin, leading to galactorrhea. Hyperinsulinemia because of obesity and associated reac

Refractory Keloids and Hypertrophic Scars: Immune Dysregulation and Neuroimmune Mechanisms Underlying Treatment Failure.

Keloids and hypertrophic scars are fibroproliferative disorders of wound healing characterized by excessive extracellular matrix deposition, constant inflammation, and high recurrence rates despite appropriate management. Conventional therapies, including surgical excision, corticosteroid injections, laser therapy, and radiation, can provide temporary relief. However, treatment failure remains common, specifically in refractory keloids. Recent findings suggest these outcomes cannot be fully explained by technical or mechanical factors alone, and pathological scarring may reflect underlying imm

Fluid silicone gels in the prevention and treatment of hypertrophic scars: a systematic review and meta-analysis of randomised controlled trials.

Hypertrophic scars are a benign yet potentially debilitating cutaneous manifestation, characterised by elevated, pigmented scarring confined within the wound boundaries. They can affect the patients both physically, through pruritus, pain, stiffness and contractures, and also psychologically through stigmatization, anxiety, depression, diminished self-esteem and daily activity disruption, reducing the quality of life. There are currently various treatments used, and although silicone gel fluid is widely recognised as a leading non-invasive treatment, there is inconsistent evidence of superiori

Causal Association Between Inflammatory Factors and Hypertrophic Scar: A Two-Sample Mendelian Randomization Study.

BACKGROUND: Hypertrophic scars result from abnormal healing following skin injuries. AIM: To delve deeper into the causal association between inflammatory factors and hypertrophic scars. METHODS: This study utilized genetic data from the FINN cohort and pertinent literature to scrutinize the nexus between a spectrum of inflammatory factors-encompassing IL-1β, interleukin 1 receptor-like 1, MCP1, RANTES/CCL5, TNFα, IL-8, IL-18, and CTACK/CCL27-and the risk of hypertrophic scarring. Our analytical strategy was based on the inverse variance weighted (IVW) approach, further bolstered by MR-Egger

ACLP promotes hypertrophic scar pathogenesis by enhancing myofibroblast activation and cholesterol synthesis-related gene expression.

BACKGROUND: Hypertrophic scar (HS) is a severe fibrotic disease characterized by excessive fibroblast activation and extracellular matrix deposition. While previous studies have revealed the involvement of Aortic Carboxypeptidase Like Protein (ACLP) in other fibrotic diseases, the role of ACLP in HS has not been investigated. METHODS: Quantitative real-time PCR (qRT-PCR), Western blotting, and immunofluorescence were applied to investigate the expression and subcellular location of ACLP. Wound healing and Transwell assays were employed to evaluate the impact of ACLP on HSF migration after ACL

Inflammation in Wound Healing and Pathological Scarring.

Significance: The aberrant inflammation during wound healing results in pathological scarring, such as hypertrophic scars and keloids. This adversely affects the quality of life of patients due to the disfiguring appearance as well as the symptoms of itch and pain. This review summarizes the up-to-date knowledge of the immunopathogenesis and treatment options for pathological scars. Recent Advances: With the advent of new technologies, combined with in vitro and in vivo wound models, several inflammatory cells have been shown to have both direct and indirect effects on both wound healing and p

[New research advances in hypertrophic scar formation, prevention and treatment].

Scarring, naturally induced by fibroblasts(Fb) during wound healing, is an essential process in response to repair damaged tissue. Excessive Fb proliferation which produces the excessive collagen deposition, including increased extracellular matrix synthesis or insufficient decomposition, typically contributes to hypertrophic scar(HS) formation. Although exact mechanisms of HS are not yet fully understood, it is generally believed that dysfunction of Fb and regulation of signal pathways play an important role in HS formation. Biologically, Fb function is affected by various factors such as cyt

Immune cells and associated molecular markers in dermal fibrosis with focus on raised cutaneous scars.

Raised dermal scars including hypertrophic, and keloid scars as well as scalp-associated fibrosing Folliculitis Keloidalis Nuchae (FKN) are a group of fibrotic raised dermal lesions that mostly occur following cutaneous injury. They are characterized by increased extracellular matrix (ECM) deposition, primarily excessive collagen type 1 production by hyperproliferative fibroblasts. The extent of ECM deposition is thought to be proportional to the severity of local skin inflammation leading to excessive fibrosis of the dermis. Due to a lack of suitable study models, therapy for raised dermal sc

The Intra-rater reliability and validity of ultrasonography in the evaluation of hypertrophic scars caused by burns.

PURPOSE: Hypertrophic scars that occur after burns are less flexible and less elastic than normal skin. Objective measurement tools are required to assess hypertrophic scars after thermal injury. Cutometer® MPA 580 has been widely used for evaluating the properties of hypertrophic scars. Ultrasonography can evaluate elasticity, stiffness, and structure of tissues simultaneously using elastography and B-mode. This study aimed to investigate the intra-rater reliability and validity of elastography to visualize hypertrophic scars. METHODS: Sixteen participants with a total of 96 scars were evalu

Occlusion and hydration of scars: moisturizers versus silicone gels.

BACKGROUND: The mainstay of non-invasive scar management, consists of pressure therapy with customized pressure garments often combined with inlays, hydration by means of silicones and/or moisturizers as well as UV protection. It is generally accepted that scar dehydration resulting from impaired barrier function of the stratum corneum and expressed by raised trans epidermal water loss (TEWL) values, can lead to increased fibroblast activity and thereby hypertrophic scar formation. However, we have reached no consensus on exactly what optimal scar hydration is nor on barrier function repair: b

Establishment of a long-term hypertrophic scar model by injection of anhydrous alcohol: A rabbit model.

The processes of hypertrophic scar formation are extremely complex, and current animal models have limitations in terms of the complete characterization of lesions. An ideal animal model is indispensable for exploring the complex progression of scar formation to elucidate its pathophysiology and to perform therapeutic testing. This study aimed to establish a long-term, consistent and easily testable animal model by injecting anhydrous alcohol into the dorsal trunk dermis of rabbits. The rabbits were injected with different amounts of anhydrous alcohol. Anhydrous alcohol was infiltrated into th

[Research advances on the effect of mechanical tension in post-traumatic hypertrophic scar formation].

Traumatic scar can not only exert influence on appearance and function of patients, but also affect psychological health status and life quality of patients to varying degrees. At present, scholars have confirmed from basic research that mechanical tension promotes the proliferation of inflammatory cells, fibroblasts, and other cells, as well as angiogenesis and epithelialization through a variety of mechanical conduction pathways and plays an important role in the formation of hypertrophic scar. Clinical studies have confirmed that surgical methods and adjuvant treatment to reduce the mechani
