Emphysematous pyelonephritis (EPN) is a rare, life-threatening complication of upper urinary tract infections that is characterized by the presence of gas in the renal parenchyma and perirenal space. It commonly occurs in diabetic patients. Escherichia coli are the most common causative organisms, with few reports implicating Citrobacter freundii as the etiologic agent in EPN. A 57-year-old woman with diabetes and neurogenic bladder visited at our department with confused mentality, myalgia, and general weakness. Further investigation revealed that the patient suffered from unilateral EPN with

Citrobacter freundii biotype 4280 produces attaching and effacing (AE) lesions in the large intestine of laboratory mice and is the causative agent of transmissible murine colonic hyperplasia. AE lesions are also produced by enteropathogenic Escherichia coli in humans. Southern analysis revealed that biotype 4280, but not 20 other strains of C. freundii, contained DNA homologous to the eae (E. coli attaching and effacing) gene which is necessary for AE activity by enteropathogenic E. coli in vitro. We have cloned and determined the nucleotide sequence of the C. freundii eae homolog. Our findin

BACKGROUND: Shiga toxin (Stx)-producing Escherichia coli (STEC) colonizes the gut and causes enteritis through bacterial attachment-effacement (A/E), leading to mucosal damage. This facilitates systemic uptake of Stx, which can often lead to hemolytic-uremic syndrome (HUS) and acute renal failure, particularly in children <5 years of age. STEC infections expressing Stx2 variants are a major risk factor for HUS. Beyond patients receiving supportive care, there is no treatment for STEC-mediated HUS, and antibiotics are contraindicated. The 3- to 7-day prodromal interval between onset of STEC dia

Citrobacter rodentium (CR) is a murine-specific enteric pathogen widely used to model infections of the attaching and effacing pathogens enteropathogenic and enterohaemorrhagic Escherichia coli. Infection induces colonic epithelial damage and inflammation that are resolved in resistant mice but result in severe disease and mortality in mice lacking interleukin-22 (IL-22). While IL-22-deficient (Il22-/-) mice succumb to CR infection due to dehydration, in part mediated by disruption of the barrier integrity by the type III secretion system (T3SS) effector EspF, the contribution of immunopatholo

Pancreatic cancer is highly malignant, and while fluoropyrimidines (5-fluorouracil [5-FU] and capecitabine) are critical first-line treatments for metastatic cases, drug resistance remains a major challenge. In this study, we identified an association between Citrobacter enrichment in pancreatic tumors and poor overall survival in patients with pancreatic cancer, particularly in patients receiving fluoropyrimidine-based treatment. Co-culture of 5-FU with the Citrobacter freundii strain isolated from pancreatic cancer and intratumoral injection of this strain in xenograft models significantly r

Colonic goblet cells play a crucial role in mucosal defense by secreting Muc2 mucin and other proteins that entrap and expel enteropathogens. However, the role of innate effectors in the gut like cathelicidin peptides in regulating the mucus barrier during infections remains unclear. In this study, we used cathelicidin-deficient (Camp-/-) littermates, colonoids, and human LS174T goblet-like cells to investigate how cathelicidin modulates goblet cell function and mucosal defense against attaching/effacing enteropathogen Citrobacter rodentium. We showed that increased fecal shedding and epitheli

The emergence of carbapenemase-producing organisms poses an escalating public health risk. In this case study, we used whole-genome sequencing (WGS) to characterize the carbapenem resistance mechanisms of a multidrug-resistant Citrobacter amalonaticus (C. amalonaticus) strain recovered from a surgical site infection. The isolate was initially identified using a MALDI-TOF mass spectrometry system. Phenotypic antibiotic susceptibility testing revealed the presence of carbapenem-resistant C. amalonaticus isolate with high-level resistance to both imipenem and meropenem (imipenem minimum inhibitor

BACKGROUND: Citrobacter braakii (C. braakii) is a gram-negative bacterium associated with hospital-acquired infections such as respiratory tract infections and bacteremia. There has been a gradual increase in the number of C. braakii infection cases in recent years. The antimicrobial resistance level of C. braakii has been steadily increasing, and the coexistence of multiple resistance genes further complicates the selection of appropriate clinical antibiotic therapies.
RESULTS: we reported a multidrug-resistant C. braakii W221 co-harboring blaNDM-1, blaIMP-4, and blaOXA-181 with four key res

The emergence of carbapenem-resistant Acinetobacter baumannii (A. baumannii) has severely limited available therapeutic options, posing a significant clinical challenge. This study aimed to evaluate the antibacterial and antibiofilm efficacy of biosynthesized silver nanoparticles (AgNPs) against clinical multi-drug resistant (MDR) A. baumannii isolates. Silver nanoparticles were biosynthesized using Citrobacter farmeri (C. farmeri) (A121) and subsequently characterized for their physicochemical properties. Antibacterial activity against A. baumannii was assessed using the disc diffusion assay,

Gut microbiota and human physiology are closely linked, and our microbiota can influence disease through immunological and metabolic activity. Quaternary amines, such as choline and carnitine, are abundant in the human gut and are found in red meat, beef, eggs, seafood, wheat, and beets. Canonically, choline and carnitine are broken down into the pro-atherogenic metabolite, trimethylamine (TMA), by various microbes in the human gut. A glycyl radical enzyme, CutC, is involved in the breakdown of choline to TMA. Carnitine is broken down to TMA via a gamma-butyrobetaine intermediate. TMA from the

Extended-spectrum beta-lactamase (ESBL) and ampicillin resistance gene type C (ampC) in foodborne Citrobacter freundii (C. freundii) pose a severe public health risk, especially when they combine with other critical resistance genes to produce a reservoir of resistance. Therefore, the current study aimed to estimate the prevalence, phenotypic, and genotypic profiling of antimicrobial resistance in C. freundii isolates obtained from diseased broiler chickens and native Egyptian buffaloes in Kafr El-Sheikh and Dakahlia governorates, Egypt. Also, the investigation of the antibacterial activity of

BACKGROUND: Aminoglycoside-modifying enzymes (AMEs) play an essential role in bacterial resistance to aminoglycoside antimicrobials. With the development of sequencing techniques, more bacterial genomes have been sequenced, which has aided in the discovery of an increasing number of novel resistance mechanisms.
METHODS: The bacterial species was identified by 16S rRNA gene homology and average nucleotide identity (ANI) analyses. The minimum inhibitory concentration (MIC) of each antimicrobial was determined by the agar dilution method. The protein was expressed with the pCold I vector in E. c

Enteropathogenic E. coli (EPEC) is a causal agent for diarrheal diseases and contributes to morbidity and mortality in children under the age of five years. The emergence and rapid spread of antibiotic resistant EPEC strains necessitate the search for novel alternatives to antibiotics. In this study, we used Citrobacter rodentium, a natural mouse pathogen that mimics many aspects of human EPEC infections, to investigate the antimicrobial properties of the blueberry anthocyanin malvidin 3-glucoside (MG) using a multi-omics approach. MG supplementation reversed the bodyweight loss induced by C.

This study aimed to characterize the molecular basis of beta-lactamase induction in Citrobacter diversus. The chromosomal beta-lactamase encoding region from C. diversus, strain NF85, was cloned and expressed in Escherichia coli. The cloned region was sequenced and open-reading frames encoding a class A beta-lactamase, designated cdiA, and a putative LysR-type transcriptional regulator protein, divergently transcribed from the beta-lactamase gene and designated cdiR, were identified. The nucleotide sequence of the NF85 cdiA was identical to that of the published C. diversus ULA27 ampC sequence

Citrobacter freundii, a common zoonotic pathogen affecting humans, livestock and fish, is recognized for its substantial impact on largemouth bass (Micropterus salmoides) mortality. However, the mechanisms of C. freundii infection in largemouth bass remain poorly understood. Based on the results of extracellular enzyme activity detection, it was speculated that the production of extracellular enzymes may be related to bacterial pathogenicity. To elucidate the immune response mechanism, transcriptomic analysis was conducted on spleen tissues from largemouth bass infected with C. freundii. Follo

We used the mouse attaching and effacing (A/E) pathogen Citrobacter rodentium, which models the human A/E pathogens enteropathogenic Escherichia coli and enterohemorrhagic E. coli (EPEC and EHEC), to temporally resolve intestinal epithelial cell (IEC) responses and changes to the microbiome during in vivo infection. We found the host to be unresponsive during the first 3 days postinfection (DPI), when C. rodentium resides in the caecum. In contrast, at 4 DPI, the day of colonic colonization, despite only sporadic adhesion to the apex of the crypt, we observed robust upregulation of cell cycle

OBJECTIVES: This study aimed to develop a novel model for detection of extended spectrum beta-lactamase (ESBL), AmpC and metallo-beta-lactamase (MBL) producing Serratia and Citrobacter species using cefoperazone sulbactam as well as other inducer-substrate combinations in a disc approximation assay. In the absence of molecular tools in developing countries, we attempted to standardize simple phenotypic techniques for detection of beta-lactamases to allow effective patient care in our countries. These techniques have been scarcely used in Serratia and Citrobacter spp., which are emerging as sig
