Clonorchis sinensis and Opisthorchis spp. in Vietnam: current status and prospects.

Clonorchis sinensis and Opisthorchis viverrini are clinically important small liver flukes because of their known association with development of cholangiocarcinoma. In Vietnam, high prevalence of C. sinensis infection in humans was previously reported in northern provinces, and O. viverrini infection has been detected in several central provinces. However, diagnosis of C. sinensis and O. viverrini infections in the past was merely based on faecal egg examination. This method alone can lead to misidentification at the species level because of morphological similarity between the eggs of these

[From recognition to practice: The 140th anniversary of the discovery of Clonorchis sinensis].

It has been the 140th anniversary since the discovery of Clonorchis sinensis, of which adult worms were found by McConnell in an oversea Chinese in Calcutta, India, September of 1874. Then, Japanese scholar Kobayashi proved that freshwater fish served as the second intermediate hosts in 1910, while another Japanese parasitologist Muto found that the first intermediate hosts were freshwater snails in 1918. However, the perniciousness has not been recognized until recently. C. sinensis infection was classified as definite carcinogen (group 1) in cholangiocarcinoma in 2009 by the International Ag

Molecular cloning, expression, and characterization of cyclophilin A from Clonorchis sinensis.

This study described the recognization, cloning, and recombinant expression of cyclophilin A-like gene from Clonorchis sinensis adult complementary DNA library (CsCyPA) and its expression and secretion in adult. Western blotting demonstrated the recombinant CsCyPA could be recognized by sera of clonorchiasis patients and a sole protein of the same size in the excretory-secretory antigens of in vitro cultured adult could be recognized by antiserum raised against the recombinant CsCyPA. Immunohistochemistry demonstrated that the CsCyPA was secreted in scattered vesicles from subtegumental parenc

The draft genome of the carcinogenic human liver fluke Clonorchis sinensis.

BACKGROUND: Clonorchis sinensis is a carcinogenic human liver fluke that is widespread in Asian countries. Increasing infection rates of this neglected tropical disease are leading to negative economic and public health consequences in affected regions. Experimental and epidemiological studies have shown a strong association between the incidence of cholangiocarcinoma and the infection rate of C. sinensis. To aid research into this organism, we have sequenced its genome. RESULTS: We combined de novo sequencing with computational techniques to provide new information about the biology of this

Clonorchis sinensis infection is positively associated with atopy in endemic area.

BACKGROUND: Epidemiologic studies have suggested that helminth infections play a protective role against allergy; this inverse association, however, has not been consistent. Clonorchis sinensis, the liver fluke of human, is prevalent in the Far East. The association between C. sinensis infection and allergy has not yet been reported. OBJECTIVE: We evaluated the association between clonorchiasis and atopy or allergic diseases in adults in endemic areas of clonorchiasis. METHODS: A total of 1116 subjects (males 419, females 697; age range, 30-86; mean age=61 years) were recruited from two ende

Identification and characterization of microRNAs in Clonorchis sinensis of human health significance.

BACKGROUND: Clonorchis sinensis is a zoonotic parasite causing clonorchiasis-associated human disease such as biliary calculi, cholecystitis, liver cirrhosis, and it is currently classified as carcinogenic to humans for cholangiocarcinoma. MicroRNAs (miRNAs) are non-coding, regulating small RNA molecules which are essential for the complex life cycles of parasites and are involved in parasitic infections. To identify and characterize miRNAs expressed in adult C. sinensis residing chronically in the biliary tract, we developed an integrative approach combining deep sequencing and bioinformatic

Infection status of freshwater fish with metacercariae of Clonorchis sinensis in Korea.

This study investigated freshwater fish for their current infection status with metacercariae of Clonorchis sinensis in Korea. Twenty-one species of freshwater fish (n = 677) were collected from 34 regions nationwidely from February 2007 to June 2008. They were individually examined by digestion technique. Eight species of freshwater fish from 17 different regions were recognized positive for the metacercariae of C. sinensis. The positive rates (range of metacercariae number per fish) of fish by the species were as follows: 48% (1-1,142) in Pseudorasbora parva, 60% (1-412) in Pungtungia herzi,

Clonorchis sinensis infection and increasing risk of cholangiocarcinoma in the Republic of Korea.

To evaluate the prevalence and risk factors of Clonorchis sinensis infection, an epidemiologic survey was performed (3,169 residents; age range: 30-87 years; mean age = 59; 1,973 women) in three areas with different mortality rates of cholangiocarcinoma. C. sinensis prevalence was 2.1% in Chuncheon, 7.8% in Chungju, and 31.3% in Haman, where cholangiocarcinoma incidence rate was 0.3, 1.8, and 5.5 per 100,000 persons, respectively. Of the four major risk factors for C. sinensis (male sex, alcohol consumption, raw freshwater fish intake, and area of residence), area was the strongest. Efforts to

Antigenic profile and localization of Clonorchis sinensis proteins in the course of infection.

In the course of Clonorchis sinensis infection, antigens presented to the hosts may be in a close relation to growth of the fluke. The antigenic proteins stimulating IgG antibody production were chronologically identified by immunoblot and localized by immunohistochemical staining. In the early stage of infection until 12 weeks post-infection (PI), antigens were proteins with molecular mass larger than 34 kDa which were derived from the tegument, testes and intrauterine eggs. After 20 weeks PI, antigens recognized were 29, 27 and 26 kDa proteins from the intestine, excretory bladder and reprod

Image analytical observation on the growth and development of Clonorchis sinensis in rats.

For the two-dimensional observation on the growth and development of Clonorchis sinensis, an image analysis system (IBAS 2000, Kontron, Germany) was used in this study. On 3, 6, 9, 12, 15, 21, 30 and 90 days of infection, 474 worms were collected from rats infected with 50 metacercariae each. The overall recovery rate was 31.6%. The worms grew rapidly in their length and width up to 30 days of infection and then, did slowly to 90 days of infection. However, the growth pattern of body area was not similar to that of body length and width, because the body area increased continuously even after

Intraductal variant of peripheral cholangiocarcinoma of the liver with Clonorchis sinensis infection.

Peripheral cholangiocarcinoma arises from the small bile ducts and presents with an infiltrative growth, but intraductal growth in the peripheral large duct tributaries is an extremely unusual manifestation, especially in cases with Clonorchis sinensis infection. A case of surgically resected intraductal variant of peripheral cholangiocarcinoma of the liver in a 46-year-old Korean man who had eaten raw fresh-water fish is described. The tumors were entirely confined within the dilated peripheral tributaries of the left intrahepatic bile duct, and no tumorous extension beyond the bile duct wall

The effects of gamma irradiation on the survival and development of Clonorchis sinensis metacercariae.

The effects of gamma irradiation on the survival and development of C. sinensis metacercariae were studied to evaluate the feasibility of irradiation as a control measure for clonorchiasis. Pseudorasbora parva were collected at an endemic river of clonorchiasis and were used for irradiation of the fluke in three schemes. The first (Scheme 1) was irradiation of the isolated metacercariae from the fish followed by infection to experimental rats. The second (Scheme 2) was irradiation of the fish, and then the metacercariae were isolated and infected to rats. The third (Scheme 3) was irradiation o

Helminths in the induction of cancer: Opisthorchis viverrini, Clonorchis sinensis and cholangiocarcinoma.

Opisthorchis and Clonorchis parasitize the bile ducts of millions of persons in the Far East. The most important aspect of infection with these flukes is their ability to initiate cancer. Numerous studies have shown these flukes to occur in association with cholangiocarcinoma far more frequently than can be explained by chance. Experimental studies in animals have confirmed the carcinogenic potential of these parasites. Although the pathogenesis remains unclear, the initial carcinogenic event is probably a function of the length and severity of infection, the host's immune response, and such v
