Structure and action of the binary C2 toxin from Clostridium botulinum.

C2 toxin from Clostridium botulinum is composed of the enzyme component C2-I, which ADP-ribosylates actin, and the binding and translocation component C2-II, responsible for the interaction with eukaryotic cell receptors and the following endocytosis. Three C2-I crystal structures at resolutions of up to 1.75 A are presented together with a crystal structure of C2-II at an appreciably lower resolution and a model of the prepore formed by fragment C2-IIa. The C2-I structure was determined at pH 3.0 and at pH 6.1. The structural differences are small, indicating that C2-I does not unfold, even a

Natural Clostridium botulinum type C toxicosis in a group of cats.

Clinical signs of botulism were observed in a group of eight cats, four of which died, after being fed pelican carrion. Clostridium botulinum type C was isolated from one cat. The microorganism and its toxin were found in the pelican. This is apparently the first report of natural botulism in cats.

Molecular properties of a hemagglutinin purified from type A Clostridium botulinum.

Clostridium botulinum causes the food poisoning disease botulism by producing botulinum neurotoxin, the most potent toxin known. The neurotoxin is produced along with a group of neurotoxin-associated proteins, or NAPs, which protect it from the low pH and proteases of the gastrointestinal tract. Recently, we isolated one of the major components of NAPs, a 33-kDa hemagglutinin (Hn-33) [Fu et al. (1998), J. Protein Chem. 17, 53-60]. In this study, we present molecular properties of Hn-33 derived from several biochemical and biophysical techniques. Hn-33 in pure form requires a 66-fold lower conc

[3 cases of Clostridium botulinum type C intoxication in the dog].

Three cases of dogs showing symptoms of acute lower motor neurone disease are reported, in which a diagnosis of type C botulism was established. In one of the dogs, botulism was believed to be due to type C toxin-containing carcases in poultry droppings; in the other two, causative relationship with poultry was extremely suggestive. The toxin was shown to be present in the serum on days one and three. Clostridium botulinum was still present in cultures of the faeces within sixteen days after onset of the symptoms.

Management of facial spasm with Clostridium botulinum toxin, type A (Oculinum).

One hundred five patients received 391 graded injections of Clostridium botulinum type A toxin (Oculinum) to treat uncontrollable facial muscle spasm. Patients had essential blepharospasm (n = 61), hemifacial spasm (n = 24), or aberrant regeneration of the seventh cranial nerve (n = 20). Muscle spasms were reduced within two days of the first injection of toxin and, in most cases, the drug effect lasted three to four months. Control of facial muscle spasm was achieved in all patients. Complications related to treatment included transient blepharoptosis (n = 7), diplopia (n = 2), and altered fa

Kinetics of growth and toxigenicity of Clostridium botulinum in experimental wound botulism.

An animal model of wound botulism was developed in mice using an inoculum of Clostridium botulinum type A spores. The number of C. botulinum in infected wounds was quantitated by culturing on egg yolk agar, and the level of C. botulinum toxin in infected wound tissue was measured by a bioassay in mice and by an enzyme-linked immunosorbent assay. All infected mice receiving no further treatment developed neuroparalytic symptoms consistent with botulism after an incubation period of ca. 48 h, and all of these animals died. Serotherapy with C. botulinum type A antitoxin initiated 24 h postchallen

Nitrite, nitrite alternatives, and the control of Clostridium botulinum in cured meats.

Historically, nitrite has been a component of meat-curing additives for several centuries. In recent years the safety of nitrite as an additive in cured meats has been questioned mainly because of the possible formation of carcinogenic nitrosamines. Nitrite has many important functions in meat curing including its role in color development, flavor, antioxidant properties, and antimicrobial activity. The inhibition of Clostridium botulinum growth and toxin production is an especially important antimicrobial property of nitrite. This review discusses the effects of processing, curing ingredients

Quantitation of Clostridium botulinum organisms and toxin in the feces of an infant with botulism.

A 4-month-old boy presented with symptoms and signs characteristic of infant botulism. Examination of feces revealed Clostridium botulinum type B and type B toxin. The numbers of C. botulinum and the amount of toxin in feces were measured throughout the 4-week period in hospital. The maximum numbers and amounts were detected in a fecal specimen collected 16 days after admission: this contained 8.4 X 10(6) C. botulinum type B colony-forming units and 61,440 mouse 100% lethal doses of type B toxin per g (wet weight) of feces. This latter figure is the highest fecal toxin titer reported yet for a

[Studies on the persistence of Clostridium botulinum on a cattle farm (author's transl)].

In the winter of 1978-1979, the presence of Clostridium botulinum was studied on a cattle farm, on which botulism caused by feeding the animals contaminated brewers' grains occurred in 1977. Cl. botulinum type B, the cause of mortality among cattle at the time, was detected in grass silage prepared in 1978. This organism was not detectable in a grass silage pit dating from 1977 and made prior to the outbreak of botulism. Investigations showed that proteolytic types of C. botulinum having grass as the substrate may produce large amounts of toxin. The production of toxin in grass silage pits may

Susceptibility of foxes to Clostridium botulinum type C and E toxins.

Investigations were performed to determine the exact susceptibility of foxes to Clostridium botulinum type C and E toxins. Doses of 5 mill. MLD type C toxin mixed with the feed did not cause symptoms of botulism in either cubs or adult foxes. Subcutaneous injections of 300,100(0 MLD or more were fatal to cubs, while 750,000 MLD caused the death of all adults. Regarding type E toxin, doses of 1 mill. MLD affected neither cubs nor adults on oral administration. Subcutaneously injected doses of 5,000 MLD or more killed all cubs, while 10,000 MLD was required to produce lethal effect on adult anim

Oral toxicities of Clostridium botulinum toxins in response to molecular size.

Clostridium botulinum type A, B, and F toxins of different molecular sizes were fed to mice to compare the oral toxicities. The progenitor toxin, a complex of a toxic and nontoxic component, of any type was higher in oral toxicity to mice than the dissociated toxic component or the derivative toxin. The former may no doubt play a more important role in the pathogenesis of food-borne botulism. The higher oral toxicity possessed by the progenitor toxin, including the exceptionally high one found with type B-L toxin, can be explained solely by the protection afforded by the nontoxic component att

Observations on the distribution and ecology of Clostridium botulinum type E in Alaska.

Environmental samples collected along the coastline and from the interior of Alaska were examined for the presence of Clostridium botulinum. Clostridium botulinum type E was detected in soils from 5 of 12 beaches; in 7 of 115 non-coastal soil samples; in sediments from six of eight locales; in gills of salmon from two fishing areas; and in the feces of 1 of 44 colonic samples from marine mammals. The basic biochemical characteristics of the isolates were determined. Tube tests for demonstrating gelatin liquefaction proved insensitive with these strains, whereas a plate test detected gelatinase

Detection and characterization of Clostridium botulinum isolated from powdered infant formula.

INTRODUCTION: As a part of an ongoing investigation of the 2025 multistate outbreak of infant botulism linked to consumption of powdered infant formula (PIF), various samples - including unopened containers of PIF and base powder (bulk PIF before packaging) - were analyzed for the presence of Clostridium botulinum. METHODS: Samples were screened using a tiered analytical approach: anaerobic enrichment followed by real-time PCR targeting botulinum neurotoxin-associated genes, confirmatory PCR, amplicon sequencing, colony isolation, and whole-genome sequencing (WGS). C. botulinum was detected i

Opportunistic Features of Non-Clostridium botulinum Strains Containing bont Gene Cluster.

The cluster of genes determining the production of botulinum toxins is an attribute of not only the Clostridium botulinum species. This cluster is also found in other members of the Clostridium genus, such as C. baratii, C. butyricum, and C. sporogenes. The occurrence of a botulinum-like cluster has also been recorded in strains of other genera, i.e., Enterococcus faecium, as well as in a Gram-negative species isolated from freshwater sediments; however, the biological activity of bont-related genes has not been noted. It can be said that the mentioned species have a dual nature. Another speci

Structural Features of Clostridium botulinum Neurotoxin Subtype A2 Cell Binding Domain.

Botulinum neurotoxins (BoNT) are a group of clostridial toxins that cause the potentially fatal neuroparalytic disease botulism. Although highly toxic, BoNTs are utilized as therapeutics to treat a range of neuromuscular conditions. Several serotypes (BoNT/A-/G, /X) have been identified with vastly differing toxicological profiles. Each serotype can be further sub-categorised into subtypes due to subtle variations in their protein sequence. These minor changes have been attributed to differences in both the duration of action and potency for BoNT/A subtypes. BoNTs are composed of three domains

Distinguishing highly-related outbreak-associated Clostridium botulinum type A(B) strains.

BACKGROUND: In the United States, most Clostridium botulinum type A strains isolated during laboratory investigations of human botulism demonstrate the presence of an expressed type A botulinum neurotoxin (BoNT/A) gene and an unexpressed BoNT/B gene. These strains are designated type A(B). The most common pulsed-field gel electrophoresis (PFGE) pattern in the C. botulinum PulseNet database is composed of A(B) strains. The purpose of this study was to evaluate the ability of genome sequencing and multi-loci variable number of tandem repeat analysis (MLVA) to differentiate such strains. RESULTS

Invasive slug populations (Arion vulgaris) as potential vectors for Clostridium botulinum.

BACKGROUND: Norwegian meadows, including those for silage production, are recently found heavily invaded by the slug Arion vulgaris in exposed areas. As a consequence, large numbers of slugs might contaminate grass silage and cause a possible threat to animal feed quality and safety. It is well known that silage contaminated by mammalian or avian carcasses can lead to severe outbreaks of botulism among livestock. Invertebrates, especially fly-larvae (Diptera), are considered important in the transfer of Clostridium botulinum type C and its toxins among birds in wetlands. C. botulinum form high
