Biochemical responses of cnidarian larvae to mercury and benzo(a)pyrene exposure.

The biochemical responses of planulae from the coral Porites astreoides exposed to 10 microg/L of benzo(a)pyrene (B(a)P) and to 10 microg/L of mercury (Hg) was evaluated. The survivorship of larvae only dropped significantly after 48 h of B(a)P exposure, whereas it remained at 98% for Hg exposure and up to 96 h. Exposure to B(a)P significantly increased free thiols, and the activity of glutathione-S-transferase and catalase were unaltered under exposure of any of the contaminants. This study is the first contribution of the biochemical effects in cnidarian larvae exposed to contaminants.

Painful toxins acting at TRPV1.

Many plant and animal toxins cause aversive behaviors in animals due to their pungent or unpleasant taste or because they cause other unpleasant senstations like pain. This article reviews the current state of knowledge of toxins that act at the TRPV1 ion channel, which is expressed in primary sensory neurons, is activated by multiple painful stimuli and is thought to be a key pain sensor and integrator. The recent finding that painful peptide "vanillotoxin" components of tarantula toxin activate the TRPV1 ion channel to cause pain led us to survey what is known about toxins that act at this r

Successive electrical responses to insemination and concurrent sperm entries in the polyspermic egg of the ctenophore Beroe ovata.

In the ctenophore Beroe ovata, action potentials and sperm-induced egg electrical responses were recorded, in unfertilized and in vitro inseminated eggs, respectively. The unfertilized egg had a resting membrane potential of -65.6 +/- 1.3 mV and a membrane resistance of 1.45 +/- 0.09 M omega. When depolarized beyond a threshold of about -20 mV, the membrane displayed an action potential with a fast phase and then a peak reaching 38 +/- 0.6 mV, followed by a positive declining plateau which lasted about 20 sec and ended by a biphasic repolarization of the membrane. The action potential was main

Fatal Portuguese man-o'-war (Physalia physalis) envenomation.

A fatal case of Physalia physalis (Portuguese man-o'-war) envenomation occurred on the Florida Atlantic coast in 1987. Despite appropriate beachside first aid, the patient was conscious only several minutes before having primary respiratory arrest and, later, cardiovascular collapse that resulted in death. Discharged nematocysts were still visible on the injured stratum corneum five days after envenomation. Additional treatment maneuvers suggested by this case include testing the tentacle fragments found on the victim's skin before their removal to ensure that nematocyst firing has been counte

Acute regional vascular insufficiency after jellyfish envenomation.

Three cases of jellyfish envenomation which occurred in the Indian Ocean and the Andaman Sea are reported. In each instance the stinging occurred on the upper extremities, and impaired the circulation to the stung limb(s), with absent distal arterial pulses, regional cyanosis, and the threat of distal gangrene. One of the patients is seriously and permanently handicapped, with bilateral upper-limb numbness and paresis; another patient has permanent sensory loss; the third patient, who also had brachial-artery narrowing that was demonstrated by angiography, made an uneventful recovery. The firs

Chilling Injury to Algal Symbionts Induces Host Starvation and Metabolic Reorganization in a Temperate Cnidarian.

Global temperature anomalies increasingly disrupt the cnidarian-algal symbiosis resulting in a phenomenon termed bleaching. In contrast to heat stress, the mechanisms underlying symbiotic breakdown under cold stress remain largely unknown. Combining physiological and metabolomic measurements, we investigated the response of the photosymbiotic sea anemone holobiont Aiptasia couchii to an experiment mimicking a cold spell in the Mediterranean Sea. Within 4 weeks, we observed the onset of symbiotic breakdown reflected in reduced algal endosymbiont density and chlorophyll a content. While photosyn

A novel, non-invasive cnidarian venom extraction device.

Cnidaria represent one of the most ancient venomous lineages with thousands of extant species and their toxins have long been known to signify a source of therapeutic potential. Despite this recognition, cnidarian toxin research has progressed relatively slowly when compared to other taxa. One of the major factors for this slow development pertains to the difficulties involved with obtaining samples, particularly from benthic species which are sessile, where dissected tissues have historically been required. Additionally, the instability of marine venoms has further hindered progression of cni

Equinins as Novel Broad-Spectrum Antimicrobial Peptides Isolated from the Cnidarian Actinia equina (Linnaeus, 1758).

Sea anemones are valuable for therapeutic research as a diversified source of bioactive molecules, due to their diverse bioactive molecules linked to predation and defence mechanisms involving toxins and antimicrobial peptides. Acid extracts from Actinia equina tentacles and body were examined for antibacterial activity against Gram-positive, Gram-negative bacteria, and fungi. The peptide fractions showed interesting minimum inhibitory concentration (MIC) values (up to 0.125 µg/mL) against the tested pathogens. Further investigation and characterization of tentacle acid extracts with significa

Multiplexed Spliced-Leader Sequencing: A high-throughput, selective method for RNA-seq in Trypanosomatids.

High throughput sequencing techniques are poorly adapted for in vivo studies of parasites, which require prior in vitro culturing and purification. Trypanosomatids, a group of kinetoplastid protozoans, possess a distinctive feature in their transcriptional mechanism whereby a specific Spliced Leader (SL) sequence is added to the 5'end of each mRNA by trans-splicing. This allows to discriminate Trypansomatid RNA from mammalian RNA and forms the basis of our new multiplexed protocol for high-throughput, selective RNA-sequencing called SL-seq. We provided a proof-of-concept of SL-seq in Leishmani

Occurrence and sequestration of toxins in food chains.

Animals may acquire toxicity by absorbing toxic compounds from their food, e.g. from plants or other animals. Sequestration and accumulation of toxins may provide protection from predators, which learn to avoid this prey because of unpleasant experiences such as bitter taste. This is a common phenomenon in marine as well as in terrestrial ecosystems. Moreover, toxins may enter food chains where they accumulate reaching high, often lethal concentrations. Palytoxin which had been primarily detected in marine zoanthids (Palythoa sp.), occurs also in a wide range of other animals, e.g. in sponges,

Environmental skin injuries in children.

Recently, interest in the potential impact of the environment on our general health has heightened. Particular focus has been directed toward environmental skin injuries because the integument, the only organ of the body that is constantly exposed to the surrounding environment, directly demonstrates the damaging effects of the environment and thereby allows for direct visual recognition by both physicians and laypersons. The characteristic cutaneous features, physiologic responses, therapeutic approaches to promote healing, and preventive measures that may be taken to avoid future environment

Changes in the distribution of filament-containing septate junctions as coelenterate myoepithelial cells change shape.

This paper describes the redistribution of septate junctions during an increase in diameter of myoepithelial cells from mesenteries of the sea anemone Metridium senile (L). Each septum was composed of a filament core, 9.5-10.2 nm in diameter, which had a double row of lateral projections from each side to the adjacent cell membrane. Septa were arranged in patches in which neighbouring septa lay parallel, 28-33 nm apart. When anaesthetized mesenteries were stretched, myoepithelial cell layers decreased from a mean of 32 to 8 micron thick; each cell shortened and its apical diameter increased. T

Oral inflammatory response to a sting from a Portuguese man-of-war.

A careful history was taken in an attempt to document an unusual delayed inflammatory reaction involving the ears and oral cavity. Severe oral and auricular inflammations have been described as apparent reactions to stings on the patient's foot by a Portuguese man-of-war (P physalia). Toxin was injected into the right foot and resulted in a delayed type of severe swellings and inflammation. The soft palate, cheek, periodontal tissues, and lower lip were primarily involved. Oral inflammations coincided with massive, acute, inflammatory lesions behind the ears. Delayed oral and ear inflammations

Placozoan fiber cells: mediators of innate immunity and participants in wound healing.

Placozoa is a phylum of non-bilaterian marine animals. These small, flat organisms adhere to the substrate via their densely ciliated ventral epithelium, which mediates mucociliary locomotion and nutrient uptake. They have only six morphological cell types, including one, fiber cells, for which functional data is lacking. Fiber cells are non-epithelial cells with multiple processes. We used electron and light microscopic approaches to unravel the roles of fiber cells in Trichoplax adhaerens, a representative member of the phylum. Three-dimensional reconstructions of serial sections of Trichopl

Neurotoxin localization to ectodermal gland cells uncovers an alternative mechanism of venom delivery in sea anemones.

Jellyfish, hydras, corals and sea anemones (phylum Cnidaria) are known for their venomous stinging cells, nematocytes, used for prey and defence. Here we show, however, that the potent Type I neurotoxin of the sea anemone Nematostella vectensis, Nv1, is confined to ectodermal gland cells rather than nematocytes. We demonstrate massive Nv1 secretion upon encounter with a crustacean prey. Concomitant discharge of nematocysts probably pierces the prey, expediting toxin penetration. Toxin efficiency in sea water is further demonstrated by the rapid paralysis of fish or crustacean larvae upon appli

Riding the Storm: Microeukaryote Community Responses to Hurricane Disturbance.

Sandy beaches are dynamic ecosystems where terrestrial and marine organisms coexist, resulting in diverse communities where microeukaryotes (protists, microalgae, and meiofauna) are key drivers. Understanding their diversity patterns and responses to stressors is essential for elucidating ecological processes, yet information on this subject remains limited. Herein, microeukaryotic diversity in a sandy beach in Acapulco, Guerrero, was analyzed via ITS1 metabarcoding assessing community structure changes following hurricane Otis. Overall, the number of ASVs decreased substantially from 260 befo

Proteomic Diversity of the Sea Anemone Actinia fragacea: Comparative Analysis of Nematocyst Venom, Mucus, and Tissue-Specific Profiles.

Sea anemones (Actiniaria, Cnidaria) are promising targets for biomedical research, as they produce unique bioactive compounds, including toxins and antimicrobial peptides (AMPs). However, the diversity and mechanisms underlying their chemical defenses remain poorly understood. In this study, we investigate the proteomic profiles of the unexplored sea anemone Actinia fragacea by analyzing its venom nematocyst extract, tissues, and mucus secretion. A total of 4011 different proteins were identified, clustered into 3383 protein groups. Among the 83 putative toxins detected, actinoporins, neurotox

Integrated multi-approaches reveal unique metabolic mechanisms of Vestimentifera to adapt to deep sea.

BACKGROUND: Vestimentiferan tubeworms are deep-sea colonizers, in which chemoautotrophic symbiosis was first observed. These animals are gutless and depend on endosymbiotic bacteria for organic compound synthesis and nutrition supply. Taxonomically, vestimentiferans belong to Siboglinidae and Annelida. Compared with other siboglinids, vestimentiferans are distinguished by high tolerance of the prevailing hydrogen sulfide in hydrothermal vents, rapid growth in local habitats, and a physical structure consisting of a thick chitinous tube. The metabolic mechanisms contributing to these features r

Thermal plasticity of a freshwater cnidarian holobiont: detection of trans-generational effects in asexually reproducing hosts and symbionts.

Understanding factors affecting the susceptibility of organisms to thermal stress is of enormous interest in light of our rapidly changing climate. When adaptation is limited, thermal acclimation and deacclimation abilities of organisms are critical for population persistence through a period of thermal stress. Holobionts (hosts plus associated symbionts) are key components of various ecosystems, such as coral reefs, yet the contributions of their two partners to holobiont thermal plasticity are poorly understood. Here, we tested thermal plasticity of the freshwater cnidarian Hydra viridissima
