Analytical sensitivity of a multiplex quantitative PCR for Toxoplasma gondii and Neospora caninum.

Cyst-forming coccidia, Toxoplasma gondii and Neospora caninum, are recognised as important causes of animal disease. Molecular diagnostics based on the presence of DNA in animal tissue are required to specifically detect T. gondii and N. caninum while achieving high levels of analytical sensitivity. We optimised available single-plex probe base qPCR assays into a multiplexed qPCR panel to detect cyst-forming coccidia, i.e. T. gondii and N. caninum. The T. gondii assay is based on a 529-bp repetitive (REP) element and the N. caninum assay on the NC5 repetitive region. Using target sequence synt

A New Protocol for Molecular Detection of Cyclospora cayetanensis as Contaminants of Berry Fruits.

Cyclospora cayetanensis is a coccidian parasite that is associated with foodborne outbreaks of gastrointestinal illnesses. Raspberries have been implicated as a vehicle of infection in some of these outbreaks. Most of the molecular techniques used for the detection of parasites commonly use the 18s rRNA as a target gene, which is highly conserved. The conserved nature of the 18s rRNA gene among coccidia means that there is potential for cross-reactivity from primers intended to target this gene in C. cayetanensis with the same gene in related coccidia. This provides an additional challenge in

Integrative use of conventional and DNA metabarcoding methods reveals high diversity, prevalence and intensity of gastrointestinal parasites in pigs from Victoria, Australia.

This cross-sectional study provides the first integrated parasitological and molecular survey of gastrointestinal parasites in pigs from 69 commercial and backyard farms across Victoria, Australia. In total, 1222 faecal samples were tested using the modified McMaster technique to determine faecal egg and oocyst counts. Samples test-positive for strongylid nematode stages were characterised further using next-generation sequencing (NGS) targeting the second internal transcribed spacer (ITS-2) of ribosomal DNA as a species-specific genetic marker. Based on faecal egg/oocyst count, 39.1 % of farm

Eimeria in the Xochimilco axolotl Ambystoma mexicanum.

Coccidia are protozoal parasites that can cause coccidiosis, a disease linked to high mortality in young, stressed, and overcrowded animals. In the order Urodela, only 21 coccidia species have been registered, and none in the Critically Endangered Ambystoma mexicanum. The present study presents the detection and follow-up of Eimeria in A. mexicanum. The monitoring was conducted at CIBAC (Centro de Investigaciones Biológicas y Acuícolas de Cuemanco), analyzing fecal samples from 90 juveniles using sedimentation, Faust, and modified Zajíček McMaster techniques. An overall prevalence of 65.6% was

The evolutionary expansion of the Sporozoa.

The sporozoans comprise a coherent group of protozoans, with characteristic and complex life cycles, containing 4-5000 species parasitic in invertebrates, particularly annelids and arthropods, and vertebrates. The group is a very successful one but neither its origins nor evolution are well understood. Considerations of traditional life cycles combined with newer methodologies have thrown some light on the evolutionary expansions of the main groups of sporozoans, the gregarines, coccidia, haemosporidians and piroplasms. The sporozoans of economic importance such as the coccidia, malaria parasi

In vitro cultivation methods for coccidian parasite research.

The subclass Coccidia comprises a large group of protozoan parasites, including important pathogens of humans and animals such as Toxoplasma gondii, Neospora caninum, Eimeria spp., and Cystoisospora spp. Their life cycle includes a switch from asexual to sexual stages and is often restricted to a single host species. Current research on coccidian parasites focuses on cell biology and the underlying mechanisms of protein expression and trafficking in different life stages, host cell invasion and host-parasite interactions. Furthermore, novel anticoccidial drug targets are evaluated. Given the v

Influence of Dietary Zinc, Copper, and Manganese on the Intestinal Health of Broilers Under Eimeria Challenge.

The incidence of enteric infections in broiler chickens may increase worldwide due to mounting pressure to limit the use of sub-therapeutic antibiotics and ionophores for coccidia suppression/prevention in the diets of broilers. For this reason, we need expand our knowledge on the role that micro-minerals have in modulating the intestinal physiology, immunology, and microbiology of broiler chickens. There are issues associated with the use of high doses of some micro-minerals in the diets of animals, such as environmental contamination, bacterial resistance, and gizzard erosion. Therefore, the

Investigation of Intestinal Health in Broiler Chickens Following Salmonella Typhimurium and Coccidiosis Vaccination and Challenge with Salmonella Typhimurium.

Salmonella enterica and coccidia (Eimeria spp.) are important intestinal pathogens in broiler production. Salmonella has high zoonotic potential, and coccidia are responsible for large economic losses. Live vaccines reduce shedding of Salmonella and minimize the impact of coccidial infections on broiler performance. This study investigated the interaction between both vaccines on the intestinal health of broilers. The 2 × 2 experimental design included vaccination against Salmonella Typhimurium (ST) (no vaccination or vaccination on day 14) and vaccination against coccidiosis (no vaccination o

Comprehensive analysis of gut microbiome and host transcriptome in chickens after Eimeria tenella infection.

BACKGROUND: Coccidiosis is an intestinal parasitic disease caused by Eimeria protozoa, which endangers the health and growth of animals, and causes huge economic losses to the poultry industry worldwide every year. Studies have shown that poultry gut microbiota plays an important role in preventing the colonization of pathogens and maintaining the health of the host. Coccidia infection also affects host gene expression. However, the underlying potential relationship between gut microbiome and host transcriptome during E. tenella infection in chickens remain unclear. METHODS: In this study, me

First report of Caryospora sp. infection in free-living green turtles (Chelonia mydas) in Northeastern Brazil.

Infection by Caryospora cheloniae has been reported to be responsible for green turtle strandings with high morbidity and mortality rates worldwide. Although studies have already shown the pathogenesis of these infections, many aspects of this protozoan are still poorly understood, including their life cycle and infection dynamics in free-living sea turtle populations. Due to the lack of information about the infection by this protozoan in sea turtles in Northeastern Brazil, our study aims to describe Caryospora sp. infection and its pathological findings in free-living Chelonia mydas found on

Cryptosporidia in the cloacal coprodeum of red-lored parrots (Amazona autumnalis).

Cryptosporidiosis was diagnosed in the cloacal coprodeum of two red-lored parrots (Amazona autumnalis). The parasites were adhered to the microvillus border of epithelial cells and their presence was associated with loss and atrophy of microvilli. Merozoites, trophozoites, gametes, and oocytes were identified with light and transmission electron microscopy. The coprodeal epithelium was hyperplastic and the lamina propria was infiltrated with moderate numbers of heterophils and lymphocytes.
