Unrecognized platelet physiology is the cause of rewarming deaths in accidental hypothermia and neonatal cold injury.

BACKGROUND: The lack of improvement in prognosis of accidental hypothermia and neonatal cold injury suggests that a major cause of mortality has not been appreciated. AIM OF THE ARTICLE: To show that thrombocytopenia that deepens on rewarming under certain conditions is that missing factor. SCIENTIFIC BASIS: Below 34 °C the first stage of aggregation is accentuated, the platelets are more sensitive to ADP and aggregation studies show an increased response "first stage hyper aggregation". We have confirmed that the irreversible second stage of platelet aggregation does not occur below 34 °C.

Cold ischemia-reperfusion injury of the liver. Role of the liver donor nutritional status in rats.

To verify the role of donor nutritional status on the quality of liver preservation after cold storage, we assessed hepatocyte and liver endothelial cell viabilities and functions in an isolated perfused rat liver model. Livers from fed and fasted Wistar rats were isolated and perfused either immediately after liver harvesting or after a 24-hr cold (4 degrees C) preservation in University of Wisconsin solution. Hyaluronic acid (150 ng/ml) and taurocholate (11.5 micrograms/ml) were infused into the reservoir, and their eliminations were assessed to evaluate liver endothelial cell function and h

Nature and mechanism of peripheral nerve damage in an experimental model of non-freezing cold injury.

Non-freezing cold injury (NFCI), so called trench foot, is a condition characterised by a peripheral neuropathy, developing when the extremities are exposed for prolonged periods to wet conditions at temperatures just above freezing. Classically, military personnel are affected, with 14% of casualties in the Falklands conflict afflicted. Clinically, NFCI is characterised by a well-defined acute clinical picture and chronic sequelae. Little is known regarding the pathophysiology and treatment of this condition. Opinions vary as to the type of nerve fibres most susceptible to damage and proposed

Developing and Consumer Testing Fire and Burn and Cold Injury Prevention Education With People Who Are Unhoused: Equity in Injury Prevention Toolkit.

Fifteen percent of people receiving care for major burn injuries in urban burn centers across North America were homeless pre-injury. The number and risk of such injuries are increasing due to greater numbers of people experiencing homelessness (PEH) and the frequency of extreme climate events. Prevention education, along with passive and active fire and cold weather protections, is critical for preventing these injuries. To increase acceptability, understandability, and actionability, prevention education needs to be in plain language, contextualized, and consumer-tested. We aimed to test new

Galangin alleviates cold stress-induced splenic injury in Danzhou chickens by inhibiting NLRP3 inflammasome-mediated pyroptosis.

Cold stress poses a significant challenge to the poultry industry, leading to impaired health and economic losses. However, the specific mechanisms by which cold stress induces injury in the chicken spleen, a critical immune organ, remain poorly defined. This study aimed to investigate the protective effects of the natural flavonoid galangin against cold stress-induced splenic injury and to elucidate the underlying mechanisms, with a focus on the NLRP3 inflammasome-mediated pyroptosis pathway. Using an in vivo chicken model, we demonstrated that acute cold exposure (10±1°C for 48 h) significan

The Optimization of Renal Graft Preservation Temperature to Mitigate Cold Ischemia-Reperfusion Injury in Kidney Transplantation.

Renal transplantation is the preferred treatment for patients with end-stage renal disease. The current gold standard of kidney preservation for transplantation is static cold storage (SCS) at 4 °C. However, SCS contributes to renal ischemia-reperfusion injury (IRI), a pathological process that negatively impacts graft survival and function. Recent efforts to mitigate cold renal IRI involve preserving renal grafts at higher or subnormothermic temperatures. These temperatures may be beneficial in reducing the risk of cold renal IRI, while also maintaining active biological processes such as inc

Cold Stress Induced Liver Injury of Mice through Activated NLRP3/Caspase-1/GSDMD Pyroptosis Signaling Pathway.

The body needs to generate heat to ensure basic life activities when exposed to cold temperatures. The liver, as the largest glycogen storage organ in the body and main heat-producing organ at rest, may play a role in chronic cold exposure. Recent studies suggested that pyroptosis plays a crucial role in liver diseases. However, the role of pyroptosis in cold stress-induced liver injury is not clear. Hence, in this study, we attempted to investigate the effects of chronic cold exposure on liver function, apoptosis, oxidative stress and inflammation in mice by establishing a mouse model of chro

Developing a novel rabbit model of atherosclerotic plaque rupture and thrombosis by cold-induced endothelial injury.

BACKGROUND: It is widely believed that atherosclerotic plaque rupture and subsequent thrombosis leads to acute coronary events and stroke. However, study of the mechanism and treatment of human plaque rupture is hampered by lack of a suitable animal model. Our aim was to develop a novel animal model of atherosclerotic plaque rupture to facilitate the study of human plaque disruption and thrombosis. METHODS: 28 healthy male New Zealand white rabbits were randomly divided into two groups: rabbits in group A (n = 12) were only fed a high-fat diet for eight weeks; rabbits in group B (n = 16) unde

Fibronectin-alpha4beta1 integrin interactions modulate p42/44 MAPK phosphorylation in steatotic liver cold ischemia-reperfusion injury.

We investigated the effects of the connecting segment-1 (CS1) peptide, which blocks fibronectin (FN)-alpha4beta1 integrin interactions upon cell signaling, leukocyte migration, and secretion of proinflammatory cytokines, in a well-established steatotic rat liver model using ex vivo cold ischemia followed by isotransplantation. In this model, CS1 peptides were administered through the portal vein of steatotic Zucker rat livers prior and after cold ischemic storage. Lean Zucker recipients of fatty orthotopic liver transplantation (OLT) received an additional 3-day course of CS1 peptides post-OLT

Nerve conduction studies in experimental non-freezing cold injury: II. Generalized nerve cooling by limb immersion.

After immersion of the hind limb of the rabbit, up to the lower thigh, in a waterbath, at 1 degree C for 10 to 14 hours under light anesthesia, there was evidence of persistent nerve damage to the tibial nerve, which varied in severity in different animals. Nerve conduction studies, carried out within 24 hours of removal from the bath, showed that in a proportion of the motor and/or afferent fibers, there was conduction failure between the knee and ankle. This was followed, over the next 48 hours, by distal degeneration of the affected fibers. No persistent conduction block was seen. After dis

Oxidative Stress and Apoptosis Inhibition Mitigate Static Cold Storage-Induced Injury in Liver Sinusoidal Endothelial Cells.

Donor liver scarcity is exacerbated by preservation-related injury, particularly ischemia-reperfusion injury (IRI), which reduces organ utilization and increases discard rates. Liver sinusoidal endothelial cells (LSECs) play a critical role in mediating IRI, yet their response to static cold storage (SCS) remains poorly understood. Here, we investigate the impact of SCS on isolated rat LSECs. Isolated rat hepatocytes, stellate cells, Kupffer cells, and LSECs were subjected to up to 3 days of SCS followed by up to 2 days of recovery, with LSECs also receiving apoptosis and/or oxidative stress i

Acute cold stress induces intestinal injury via CIRP-TLR4-IRE1 signaling pathway in pre-starter broilers.

BACKGROUND: Cold stress is a common environmental stress in broiler chicks. Cold-inducible RNA-binding protein (CIRP) is a conserved cold shock protein that can regulate inflammatory response through Toll-like receptor 4 (TLR4). The mechanism that how CIRP involves in the regulation of cold stress in broilers remains unclear. METHODS AND RESULTS: In this study, 360 7-day-old healthy male SZ901 chicks were selected and randomly allocated to four groups, and then subjected to acute cold exposure at the ambient temperature of 12 ± 1 °C for 0 h, 4 h, 8 h, and 12 h, respectively. Bile salt toxicity aggravates cold ischemic injury of bile ducts after liver transplantation in Mdr2+/- mice.

Intrahepatic bile duct strictures are a serious complication after orthotopic liver transplantation (OLT). We examined the role of endogenous bile salt toxicity in the pathogenesis of bile duct injury after OLT. Livers from wild-type mice and mice heterozygous for disruption of the multidrug resistance 2 Mdr2 gene (Mdr2+/-) were transplanted into wild-type recipient mice. Mdr2+/- mice secrete only 50% of the normal amount of phospholipids into their bile, leading to an abnormally high bile salt/phospholipid ratio. In contrast to homozygous Mdr2-/- mice, the Mdr2+/- mice have normal liver histo

Lumbar sympathectomy attenuates cold allodynia but not mechanical allodynia and hyperalgesia in rats with spared nerve injury.

UNLABELLED: In certain patients with neuropathic pain, the pain is dependent on activity in the sympathetic nervous system. To investigate whether the spared nerve injury model (SNI) produced by injury to the tibial and common peroneal nerves and leaving the sural nerve intact is a model for sympathetically maintained pain, we measured the effects of surgical sympathectomy on the resulting mechanical allodynia, mechanical hyperalgesia, and cold allodynia. Military cold injury during the war in the Falkland Islands 1982: an evaluation of possible risk factors.

Throughout the history of war, there have been many instances when the cold has ravaged armies more effectively than their enemies. Delineated risk factors are restricted to negro origins, previous cold injury, moderate but not heavy smoking and the possession of blood group O. No attention has been directed to the possibility that abnormal blood constituents could feasibly predispose to the development of local cold injury. This study considers this possibility and investigates the potential contribution of certain components of the circulating blood which might do so. Three groups of soldier
