NtZFP66L1 Regulates Cold Tolerance in Tobacco by Modulating the ABA Signaling Pathway and the GLK1-Mediated Adaptive Pathway.

Cold stress is a major environmental challenge that affects tobacco (Nicotiana tabacum L.) growth and yield potential. However, the precise mechanisms underlying the tobacco plant's response remain largely unclear. In this study, we identified a cold-induced CCCH-type zinc finger protein, NtZFP66L1, as an important positive regulator of cold tolerance in tobacco. Overexpressing NtZFP66L1 improved cold resistance, while knocking it out made plants more sensitive to cold. NtZFP66L1 enhances cold tolerance by targeting NtPP2C-37 and NtGLK1; knocking out NtPP2C-37 increased cold tolerance, while l

Melatonin enhances the cold tolerance of tomatoes by promoting ABA accumulation.

SlSNAT1-dependent melatonin synthesis enhances tomato cold tolerance by promoting SlNCED1-mediated ABA accumulation, thereby strengthening antioxidant capacity, maintaining photosynthesis, and reducing ROS-induced membrane damage under cold stress. Cold stress limits greenhouse tomato growth by suppressing photosynthesis, promoting reactive oxygen species (ROS) accumulation, and disrupting membrane integrity, which ultimately reduces yield. Melatonin (MT) enhances plant stress tolerance, but its role in regulating cold adaptation in tomato remains unclear. Here, we used CRISPR/Cas9-generated S

A Comprehensive Analysis of Transcriptomics and Proteomics Elucidates the Cold-Adaptive Ovarian Development of Eriocheir sinensis Farmed in High-Altitude Karst Landform.

Background: In high-altitude regions, sporadic two-year-old immature Chinese mitten crabs (Eriocheir sinensis) would overwinter and mature in their third year, developing into three-year-old crabs (THCs) with a cold-adaptive strategy. Compared to two-year-old crabs (TWCs) from low-altitude Jiangsu, THCs from Karst landform and high-altitude Guizhou exhibit significantly larger final size but lower gonadosomatic index (GSI) (p < 0.01). Methods: To elucidate the molecular mechanisms underlying this delayed ovarian development, integrated transcriptomic and proteomic analyses were conducted. Resu

The AtHDA6-AtSK2 module promotes cold tolerance by enhancing shikimate metabolism and antioxidant activity.

Low temperature is an environmental factor that significantly impairs the normal development of plants by limiting yield and quality. Although histone deacetylase HDA6 is involved in various biological processes, the specific molecular mechanisms underlying its response to low temperatures remain unexplored in Arabidopsis. In this study, we investigated the HDA6 expression pattern at low temperatures and discovered that cold stress-induced transcriptional activity increased the HDA6 protein level. Freezing experiments demonstrated that HDA6 functions as a positive regulator in response to low

Molecular Mechanisms of Lipid-Based Metabolic Adaptation Strategies in Response to Cold.

Temperature changes and periods of detrimental cold occur frequently for many organisms in their natural habitats. Homeothermic animals have evolved metabolic adaptation strategies to increase mitochondrial-based energy expenditure and heat production, largely relying on fat as a fuel source. Alternatively, certain species are able to repress their metabolism during cold periods and enter a state of decreased physiological activity known as torpor. By contrast, poikilotherms, which are unable to maintain their internal temperature, predominantly increase membrane fluidity to diminish cold-rela

An integrative multi-project transcriptomic and structural prediction framework identifies candidate cold-responsive transcription factors in Medicago sativa.

Cold stress limits alfalfa (Medicago sativa) growth and persistence, but public transcriptomic datasets differ widely in genotype, tissue, treatment duration, and experimental design. We integrated RNA-seq data from ten independent BioProjects using a common processing workflow while retaining project-specific structures. A recurrent contrast-level DEG-derived pool of 4,354 genes was ranked by random forest using expression profiles from 240 samples. The original model showed strong internal discrimination (OOB ROC-AUC = 0.937), whereas fully nested leave-one-BioProject-out validation yielded

Heavy Metal Scavenger Metallothionein Rescues Against Cold Stress-Evoked Myocardial Contractile Anomalies Through Regulation of Mitophagy.

Cold stress prompts an increased prevalence of cardiovascular morbidity yet the underneath machinery remains unclear. Oxidative stress and autophagy appear to contribute to cold stress-induced cardiac anomalies. Our present study evaluated the effect of heavy metal antioxidant metallothionein on cold stress (4 °C)-induced in cardiac remodeling and contractile anomalies and cell signaling involved including regulation of autophagy and mitophagy. Cold stress (3 weeks) prompted interstitial fibrosis, mitochondrial damage (mitochondrial membrane potential and TEM ultrastructure), oxidative stress

Diurnal physiological and immunological responses to a 10-km run in highly trained athletes in an environmentally controlled condition of 6 °C.

PURPOSE: The Clara cell protein CC16, secreted from Clara cells in the lung, is discussed as a potential biomarker for toxic effects on the airways. An increased concentration of CC16 in serum may be caused by increased permeability of the lungs. To investigate the changes in P-CC16 in response to an intense exercise bout performed at different times of day (9 am and 4 pm) of highly trained individuals. METHOD: Using a crossover randomized design, 8 runners (mean VO2max 71 ml kg-1 min-1, SD 6) performed a 10-km time trial run, at 9 am and 4 pm, in an environmental chamber set at 6 °C. Lung fu

Optimal HSF1 activation in response to acute cold stress in BAT requires nuclear TXNIP.

While TXNIP (thioredoxin interacting protein) in the plasma membrane and vesicular location is known to negatively regulate cellular glucose uptake by facilitating glucose transporter endocytosis, the function of TXNIP in the nucleus is far less understood. Herein, we sought to determine the function of nuclear TXNIP in vivo, using a new HA-tagged TXNIP knock-in mouse model. We observed that TXNIP can be found in the nucleus of a variety of cells from different tissues including hepatocytes (liver), enterocytes (small intestine), exocrine cells (pancreas), and brown adipocytes (BAT). Neonatal sepsis and septic shock: concepts update and review.

The nonspecific presentation of neonatal sepsis and systemic inflammatory response syndrome preceding septic shock delay the early diagnosis of septic shock and increase its mortality rate. Early diagnosis involves suspecting septic shock in every newborn with tachycardia, respiratory distress, difficult feeding, altered tonus and skin coloration, tachypnea and reduced perfusion, specially in case of maternal peripartum infection, chorioamnionitis or long-term membranes rupture. This article aims to review current knowledge on neonatal period peculiarities, fetal circulation dynamics, and the

The transcriptional cascade CiERF023-CiERF041 of Citrus ichangensis regulates small heat shock protein gene CiHSP26.5 to modulate cold tolerance.

Ichang papeda (Citrus ichangensis), a wild primitive citrus species, exhibits remarkable cold tolerance, yet the transcriptional regulation of cold-responsive genes in C. ichangensis remains poorly explored. In this study, we identified the small heat shock protein (sHSP) CiHSP26.5 as a crucial member that was substantially induced under cold stress and played a positive role in cold tolerance. Furthermore, two APETALA2/Ethylene Responsive Factor (AP2/ERF) family transcription factors, CiERF023 and CiERF041, were found to directly bind to the dehydration-responsive element (DRE) motifs in the

Transcriptome analysis revealed lncRNA-mRNA modules responsive to low temperature stress in Qingke.

BACKGROUND: Qingke (hull-less barley), a staple crop on the Qinghai-Xizang Plateau, has evolved unique stress tolerance mechanisms. Cold stress damages plants through ice formation and oxidative stress, triggering antioxidant systems and stress-related gene expression. Long non-coding RNAs (lncRNAs) are known to regulate these responses. RESULTS: Physiological analysis revealed a dynamic defense strategy: antioxidant systems (SOD/POD) activated early (12 h), shifting to osmotic regulation (soluble protein) later (48 h). Membrane damage (MDA/conductivity) increased under stress but showed repa

Genetic dissection of inherited cold-stress reveals resilience hotspots in barley.

Cold episodes during early crop development threaten cereal establishment and yield, but it remains unclear whether ancestral cold exposure can generate measurable inherited resilience in barley. We tested this question in a three-generation field experiment using 138 barley accessions and eight third-generation lineage-treatment combinations that separated no-memory, transgenerational, intergenerational and repeated cold-memory classes under control and recurrent cold-stress conditions. Cold stress was imposed by delayed sowing, and establishment, physiological, biochemical, growth, reproduct

Review on bacterial stress topics.

A complex network of regulatory systems ensures a coordinated and effective response to different types of stress that can act on a bacterium. Bacterial stress response generates changes that influence efflux system and virulence factor expression. Thus, partial or total loss of pathogenicity islands in uropathogenic Escherichia coli can be induced by SOS-dependent or SOS-independent pathways related to selection of quinolone-resistant mutants. Likewise, hyperosmolarity and some chemicals, including fluoroquinolones, salicylate, nonantimicrobial medicaments like diazepam and anti-inflammatory

Temporal plasticity of thermal tolerance and cellular responses in Atlantic salmon (Salmo salar) during warm acclimation and subsequent cold acclimation to baseline temperature.

Global warming presents a substantial threat to aquatic ectotherms, including the economically significant Atlantic salmon (Salmo salar). Although warm acclimation can increase heat tolerance, little is known about the dynamic processes and cellular responses associated with the acquisition and loss of this increased tolerance. This study investigated changes in critical thermal maximum (CTmax), liver structure, hepatic antioxidant activity, and heat shock protein (HSP) expression in Atlantic salmon during prolonged warm acclimation followed by cold acclimation back to baseline temperature. Overexpression of AtCSP4 affects late stages of embryo development in Arabidopsis.

Eukaryotic cold shock domain proteins are nucleic acid-binding proteins that are involved in transcription, translation via RNA chaperone activity, RNA editing, and DNA repair during tissue developmental processes and stress responses. Cold shock domain proteins have been functionally implicated in important developmental transitions, including embryogenesis, in both animals and plants. Arabidopsis thaliana cold shock domain protein 4 (AtCSP4) contains a well conserved cold shock domain (CSD) and glycine-rich motifs interspersed by two retroviral-like CCHC zinc fingers. AtCSP4 is expressed in

Integrative physiological and behavioural responses to sudden cold-water immersion are similar in skilled and less-skilled swimmers.

We examined the initial physiological responses and subsequent capacity to swim following cold-water immersion. An ecologically-valid model was used whereby immersion was sudden (<2s) and participants had to actively remain afloat. Participants (15 skilled swimmers, 17 less-skilled swimmers) undertook four experimental test sessions: a physiological test and a swimming test in both cold (10°C) water and temperate (27°C) water in a swimming flume (temperature order counter-balanced). For physiological testing, measures of brain perfusion [flow velocity (MCAv, Doppler) and oxygenation (NIRS)] an

QTL Mapping and RNA-Seq Revealed Loci and Candidate Genes Associated with Cold Tolerance of Wheat Root at Seedling Stage.

OBJECTIVES: Low-temperature stress has become a key factor severely restricting seedling growth in wheat, highlighting the growing importance of research on low-temperature tolerance in wheat. Most previous studies focused on the aboveground organs, and only a minority specifically targeted the roots. METHODS: In this study, a recombinant inbred line population derived from a cross between DH118 and Jinmai 919 was used to dissect the genetic regulatory mechanism underlying root growth and development in wheat seedlings under low-temperature conditions. Genome-Wide Identification and Functional Characterization of CaGRAS43 in Enhancing Cold Stress Tolerance in Pepper.

Cold stress severely limits plant growth and crop productivity, necessitating the identification of key regulatory genes that enhance tolerance. In this study, we performed a genome-wide identification and expression analysis of the GRAS transcription factor family in pepper under cold stress. In total, 64 GRAS genes (CaGRAS1-CaGRAS64) were identified in the pepper genome and distributed across 10 different chromosomes. Additional in silico analysis provided key insights on the identified CaGRAS genes, including their conserved domains, exon-intron organization, evolutionary relationships, gen

Transcription and phosphorylation of CATALASE2 contribute to cold stress tolerance in Hevea brasiliensis.

Despite the economic importance of the rubber tree (Hevea brasiliensis) and its sensitivity to cold stress, the molecular mechanisms that govern its cold response are still largely unknown. Here, we identified H. brasiliensis CATALASE2 (HbCAT2) as a factor associated with cold tolerance in this species, likely through reducing excess H2O2. Cold stress significantly induced HbCAT2 transcription and catalase activity in H. brasiliensis, and overexpression of HbCAT2 in both H. brasiliensis and Arabidopsis enhanced cold tolerance and reduced H2O2 accumulation. Furthermore, we found that HbCBF1, a
