Collagen XVII/BP180: a collagenous transmembrane protein and component of the dermoepidermal anchoring complex.

Collagen XVII, or BP180, is a collagenous transmembrane protein and a structural component of the dermoepidermal anchoring complex. Molecular studies reveal that it has a globular cytosolic amino-terminal domain and flexible-rod extracellular carboxyterminal domain. The extracellular portion of collagen XVII is constitutively shed from the cell surface by ADAMs (proteinases that contain adhesive and metalloprotease domains). Cell biological analyses suggest that collagen XVII functions as a cell-matrix adhesion molecule through stabilization of the hemidesmosome complex. This concept is suppor

Type III collagen is crucial for collagen I fibrillogenesis and for normal cardiovascular development.

Type III collagen is a fibrillar forming collagen comprising three alpha1(III) chains and is expressed in early embryos and throughout embryogenesis. In the adult, type III collagen is a major component of the extracellular matrix in a variety of internal organs and skin. Mutations in the COL3A1 gene have been implicated as a cause of type IV Ehlers-Danlos syndrome, a disease leading to aortic rupture in early adult life. To directly study the role of Col3a1 in development and disease, we have inactivated the Col3a1 gene in embryonic stem cells by homologous recombination. The mutated allele w

Collagen and Collagen Peptide From the Alosa Sapidissima Protect Against UVB-Induced Skin Damage.

Ultraviolet-B (UVB) radiation exacerbates oxidative stress, accelerating collagen degradation and skin aging. While collagen and its hydrolysates offer therapeutic potential, medical apparatus and instruments call for high quality and traceability. Here we investigated the synergistic effects of collagen and collagen peptides derived from Shad (Alosa sapidissima) scales against UVB-induced skin damage. Collagen was extracted and peptides were obtained through enzymatic hydrolysis. Structural characterization confirmed collagen's triple helix and peptide fragments. In vitro assays demonstrated

Injection of recombinant human type VII collagen restores collagen function in dystrophic epidermolysis bullosa.

Dystrophic epidermolysis bullosa (DEB) is a family of inherited mechano-bullous disorders that are caused by mutations in the type VII collagen gene and for which ex vivo gene therapy has been considered. To develop a simpler approach for treating DEB, we evaluated the feasibility of protein-based therapy by intradermally injecting human recombinant type VII collagen into mouse skin and a DEB human skin equivalent transplanted onto mice. The injected collagen localized to the basement membrane zone of both types of tissues, was organized into human anchoring fibril structures and reversed the

Collagen-based nanomaterials for tissue engineering: A review from nanocollagen to collagen-based nanocomposites.

Collagen is widely used as a scaffold material and drug delivery vehicle in tissue engineering due to its excellent biocompatibility and low immunogenicity. However, conventional collagen-based biomaterials suffer from inadequate mechanical strength, poor thermal stability, and excessively rapid degradation. Nanotechnology provides effective strategies to overcome these limitations. Collagen-based nanomaterials generally exhibit significantly enhanced mechanical properties and thermal stability, prolonged drug half-life, and optimized drug release behavior, showing great promise for repairing

Collagen Gly missense mutations: Effect of residue identity on collagen structure and integrin binding.

Gly missense mutations in type I collagen, which replace a conserved Gly in the repeating (Gly-Xaa-Yaa)n sequence with a larger residue, are known to cause Osteogenesis Imperfecta (OI). The clinical consequences of such mutations range from mild to lethal, with more serious clinical severity associated with larger Gly replacement residues. Here, we investigate the influence of the identity of the residue replacing Gly within and adjacent to the integrin binding 502GFPGER507 sequence on triple-helix structure, stability and integrin binding using a recombinant bacterial collagen system. Loss of collagen-receptor DDR1 delays renal fibrosis in hereditary type IV collagen disease.

Alport syndrome is a hereditary type IV collagen disease leading to progressive renal fibrosis, hearing loss and ocular changes. End stage renal failure usually develops during adolescence. COL4A3-/- mice serve as an animal model for progressive renal scarring in Alport syndrome. The present study evaluates the role of Discoidin Domain Receptor 1 (DDR1) in cell-matrix interaction involved in pathogenesis of Alport syndrome including renal inflammation and fibrosis. DDR1/COL4A3 Double-knockouts were compared to COL4A3-/- mice with 50% or 100% expression of DDR1, wildtype controls and to DDR1-/-

Type III collagen can be present on banded collagen fibrils regardless of fibril diameter.

Monoclonal antibodies that recognize an epitope within the triple helix of type III collagen have been used to examine the distribution of that collagen type in human skin, cornea, amnion, aorta, and tendon. Ultrastructural examination of those tissues indicates antibody binding to collagen fibrils in skin, amnion, aorta, and tendon regardless of the diameter of the fibril. The antibody distribution is unchanged with donor age, site of biopsy, or region of tissue examined. In contrast, antibody applied to adult human cornea localizes to isolated fibrils, which appear randomly throughout the ma

Urinary Collagen Peptides Predict Mortality.

Organ fibrosis caused by the presence of excessive extracellular matrix (ECM) is related to mortality. Urinary peptide signatures were reported to be predictive of death in SARS-CoV-2 and chronic kidney disease. Such signatures were composed for 68% of collagen fragments. In this study, we examined whether urinary collagen peptides, potentially representing organ fibrosis, could predict mortality in patients with critical and non-critical conditions. Urinary proteomic data from 1012 patients with follow-up information from the CRIT-COV-U study were investigated for the association of collagen

Collagen crosslinking in thin corneas.

BACKGROUND: Collagen cross linking (CXL) is the only modality that can arrest or mitigate the progression of keratoconus. However, conventional CXL using the Dresden protocol cannot be used in corneas less than 400 µm in thickness due to the risk of endothelial damage. A number of modifications have been implemented to solve this problem. PURPOSE: To highlight the parameters used in the Dresden protocol, its pitfalls in thin corneas and elucidate the modifications that allow cross-linking in thin corneas. SYNOPSIS: This video serves to present the Dresden protocol, the ensuing problem in thi

Type VII Collagen Disorders Simplified.

Epidermolysis bullosa acquisita (EBA) and bullous systemic lupus erythematosus (BSLE) are autoimmune mechanobullous diseases that are caused by autoantibodies directed against type VII collagen. The functionality of type VII collagen is vital to the skin and mucous membranes because it makes up the anchoring fibrils that adhere the epithelium to the underlying connective tissue. Dystrophic epidermolysis bullosa (DEB), which bears some clinical similarities to EBA and BSLE, is caused by mutations in the type VII collagen gene (COL7A1) that may be dominant or recessive, leading to partial or tot

Metabolic control of collagen synthesis.

The extracellular matrix (ECM) is present in all tissues and crucial in maintaining normal tissue homeostasis and function. Defects in ECM synthesis and remodeling can lead to various diseases, while overproduction of ECM components can cause severe conditions like organ fibrosis and influence cancer progression and therapy resistance. Collagens are the most abundant core ECM proteins in physiological and pathological conditions and are predominantly synthesized by fibroblasts. Previous efforts to target aberrant collagen synthesis in fibroblasts by inhibiting pro-fibrotic signaling cascades h

Collagen fleece in orthopaedic infections.

Collagen fleece is a relatively new development. It represents another option in the battle against infection. It is a cheap, biocompatible, and resorbable local antibiotic delivery mechanism with favorable drug release kinetics and low risk of adverse effects or toxicity. Benefit may be conferred when used in contaminated cases. Significantly more research is still needed before the adoption of collagen fleece as the standard of care. However, we can likely conclude that there are no major adverse effects and it can be safely used as an adjunct in addition to conventional therapies for the pr

Authentication of collagen VI antibodies.

BACKGROUND: Collagen VI is a ubiquitously-expressed macromolecule that forms unique microfibrillar assemblies in the extracellular matrix. Mutations in the COL6A1, COL6A2 and COL6A3 genes result in congenital muscular dystrophy, arguing that collagen is critical for skeletal muscle development and function. Antibodies against collagen VI are important clinical and diagnostic tools in muscular dystrophy. They are used to confirm genetic findings by detecting abnormalities in the distribution, organization and overall levels of collagen VI in cells and tissues isolated from patients. METHODS: M

Collagen cross linking: current perspectives.

Keratoconus is a common ectatic disorder occurring in more than 1 in 1,000 individuals. The condition typically starts in adolescence and early adulthood. It is a disease with an uncertain cause and its progression is unpredictable, but in extreme cases, vision deteriorates and can require corneal transplant surgery. Corneal collagen cross-linking (CCL) with riboflavin (C3R) is a recent treatment option that can enhance the rigidity of the cornea and prevent disease progression. Since its inception, the procedure has evolved with newer instrumentation, surgical techniques, and is also now perf

[Collagen VI-related muscle disorders].

Collagen VI-related muscle disorders include severe Ullrich's disease (Ullrich congenital muscular dystrophy:UCMD) and milder Bethlem myopathy. Mutations in the 3 collagen VI genes, namely, COL6A1, COL6A2, and COL6A3, cause both diseases. UCMD is inherited in an autosomal recessive manner, and de novo dominant mutations are also reported. Bethlem myopathy is usually inherited in an autosomal dominant manner, but a rare autosomal recessive inheritance has recently been reported. Patients with UCMD have generalized muscle weakness, multiple contractures of the proximal joints, and hyperextensibi
