Short-time thermal inactivation of surrogates of the public transport microbiome with a low-cost thermoresistometer.

In this study, the thermal inactivation of the bacterial genera Staphylococcus, Enterococcus and Burkholderia, which can be found in public transportation environments, as well as the bacteriophage MS2 as a surrogate for potential viral pathogens are investigated. To quantify the thermal inactivation characteristic, an automated and inexpensive thermoresistometer is constructed and set up, which enables the microorganisms to be exposed to short-term thermal shocks. The time dependent temperature curves were measured to account for heat-up and cooling times. Afterwards, the microorganisms were

When "dead" isn't dead: VBNC entering and survival modeling of Salmonella Typhimurium ATCC 14028 in low-water-activity conditions.

Salmonella can persist in low water activity (aw) foods for years. This high resistance to conservation methods applied by the food industry makes this pathogen a public health concern. In this work, a predictive model to describe the survival of Salmonella enterica serovar Typhimurium ATCC 14028 under four different water activity (0.9, 0.8, 0.7 and 0.6 aw) conditions was determined, in parallel with the determination of cell viability throughout and after exposure to stress to monitor the entry and persistence of this pathogen in the VBNC state. In addition, morphological changes in the cell

In vitro susceptibility testing of amphotericin B for Cryptococcus neoformans variety grubii AFLP1/VNI and Cryptococcus gattii AFLP6/VGII by CLSI and flow cytometry.

Cryptococcus neoformans var. grubii AFLP1/VNI is the main causative agent of cryptococcosis associated with AIDS in the world. Cryptococcus gattii AFLP6/VGII causes mainly endemic primary infection in immunocompetent hosts. To determine the minimum inhibitory concentrations (MICs) of C. neoformans var. grubii AFLP1/VNI and C. gattii AFLP6/VGII against amphotericin B (AMB) in a short period of time, flow cytometry (FCM) with FUN-1 fluorochrome was used to compare with broth microdilution method (CLSI M27-A3). The minimum incubation period was evaluated by minimum fungicidal concentration proced

Enrichment of Yersinia pestis from blood cultures enables rapid antimicrobial susceptibility determination by flow cytometry.

Mortality from plague is high if not treated with the proper antibiotics within 18-24 hours after onset of symptoms. The process of antibiotic susceptibility determination of Yersinia pestis isolated from blood samples may extend from 4 to more than 7 days, since the in vitro growth is very slow. To accelerate this process, we developed an enrichment protocol as well as a non-standard yet reliable method for rapid antibiotic susceptibility analysis of Y. pestis from blood cultures using flow cytometry technology. This rapid method is applicable to blood cultures containing low levels of Y. pes

Use of the microbial growth curve in postantibiotic effect studies of Legionella pneumophila.

Using the standard Craig and Gudmundsson method (W. A. Craig and S. Gudmundsson, p. 296-329, in V. Lorian, ed., Antibiotics in Laboratory Medicine, 1996) as a guideline for determination of postantibiotic effects (PAE), we studied a large series of growth curves for two strains of Legionella pneumophila. We found that the intensity of the PAE was best determined by using a statistically fitted line over hours 3 to 9 following antibiotic removal. We further determined the PAE duration by using a series of observations of the assay interval from hours 3 to 24. We determined that inoculum reducti

The Association Between OR Traffic and Airborne Microbial Counts During Two Types of Abdominal Surgeries.

Airborne contamination via OR traffic (ie, number of personnel and door openings, personnel movement) can contribute to the deposit of microbes on surfaces and contribute to surgical site infections. Researchers at a midwestern health system used a mixed-methods approach to evaluate the association between OR traffic and microbial deposits during colon surgeries and abdominal hysterectomies using nonparticipant observations and agar settle plate testing. Door openings were at the low end of published rates, although some openings were for nonessential reasons. Door openings were strongly corre

Overview of endospore-forming bacteria in food: The road towards a harmonised method for the enumeration of their spores.

Endospore-forming bacteria are an important challenge for the food industry due to their ubiquitous nature, widespread presence in the food chain and sophisticated survival mechanisms. An accurate method is needed that can provide insight into the quality of raw materials, predict spoilage potential and ensure food safety. A plethora of methods exist for the enumeration of spore-forming bacteria which vary among countries, industries and food producers. These methods describe a wide range of values in the key method parameters, such as heat treatment, growth medium, incubation time, and temper

The disinfectant effects of portable ultraviolet light devices and their application to dentures.

Interest in ultraviolet (UV) light devices for disinfection has increased due to pandemic concerns, but their efficacy in denture disinfection is underexplored. This study evaluated seven-enclosed type portable UV irradiation devices (devices A-G). Among these, devices A, B, and C used bulb light sources, while devices D, E, F, and G utilized LED (light-emitting diode) light sources. All devices were assessed in agar plate assay, and Candida albicans and Staphylococcus aureus spread on agar plates and irradiated under two conditions: inoculated surface facing upward and downward. Colony counts

Two Methods of Measuring Cryptococcus neoformans Fungal Burden in Macrophages.

The ability of C. neoformans to survive and replicate within host phagocytes enables it to evade the immune system and allows for persistence of the infection. As such, measuring fungal burden of C. neoformans strains-and indeed how drug treatments can influence fungal burden-provides important information about C. neoformans pathogenesis. In this chapter, we describe two methods that may be used to appraise fungal burden: a standard end-point colony-formation assay for calculating the average number of yeast per host cell and a fluorescence microscopy-based method that may be used to measure

Fitness Differences Between Listeria monocytogenes Serotypes 1/2a, 4b, and 4bv-1 in Competition for Growth on Lettuce Leaf Sections.

Listeria monocytogenes is a foodborne pathogen that lives in nature as a saprophyte. Two of the three most common serotypes that cause foodborne listeriosis are 1/2a and 4b. Within serotype 4b, there is a variant called 4bv-1. In the last decade, several produce-related outbreaks (linked to leafy salad, caramel apples, and stone fruit) were linked to 4bv-1 strains, specifically those of Sequence Type 382. This study assessed the fitness of ST 382 strains on lettuce leaf sections to determine if they are more fit on produce than strains of other serotypes. Strains of serotypes 1/2a, 4b, and ST

Comparative evaluation of the effectiveness of alcohol-based sanitizers, UV-C radiation and hot air on three-age Salmonella biofilms.

Dry sanitation is recommended to control contamination and prevent microbial growth and biofilm formation in the low-moisture food manufacturing plants. The objective of this study was to evaluate the effectiveness of dry sanitation protocols on Salmonella three-age biofilms formed on stainless steel (SS) and polypropylene (PP). Biofilms were formed for 24, 48 and 96 h at 37 °C using a cocktail of six Salmonella strains (Muenster, Miami, Glostrup, Javiana, Oranienburg, Yoruba) isolated from the peanut supply chain. Then, the surfaces were exposed to UV-C radiation, hot air (90 °C), 70% ethanol

Quantitative modeling of the survival of Listeria monocytogenes in soy sauce-based acidified food products.

Primary and secondary models were developed for quantitatively characterizing the survival of Listeria monocytogenes in soy-sauce based acidified Asian style products that do not undergo a thermal treatment. The objective of this study was to quantify the effect of food matrix properties on L. monocytogenes' survival in soy sauce-based products. This quantification enables a product-specific estimation of 5-log reduction time to ensure a safe processing and management operation, to ultimately facilitate a science-based, safety-oriented product development process. A central composite design wi

Kitchen-Scale Treatments for Reduction of Listeria monocytogenes in Prepared Produce.

ABSTRACT: Listeriosis, a foodborne illness caused by Listeria monocytogenes, has relatively low incidence, but a substantial mortality rate, particularly in immunocompromised populations. Because of the known risk of L. monocytogenes and other pathogens in produce, immunocompromised individuals are often placed on neutropenic diets that exclude fresh produce. Therefore, this study aimed to evaluate several kitchen-scale treatments as potential interventions to reduce L. monocytogenes in prepared produce. Cucumbers, apples, and celery were dip inoculated with a three-strain cocktail of L. monoc

Light microscopy and scanning electron microscopy of bacterial colonies isolated from ras malai samples from different markets of Lahore, Pakistan.

The current study investigates the total bacterial contamination in various packed and unpacked ras malai samples of 14 different localities of Lahore, Pakistan. The bacterial colonies such as Bacillus sp. and Gamella sp. were isolated from ras malai samples and grown on agar-broth media under sterile environmental conditions. Serial dilution technique was used to compose the replicates to get a viable count of bacteria in the samples. Results indicated that in case of packed ras malai samples, maximum bacterial count was observed in Sample 1 (422 × 10-2 to 402 × 10-6 ) and minimum bacterial c

Estimating Listeria monocytogenes Growth in Ready-to-Eat Chicken Salad Using a Challenge Test for Quantitative Microbial Risk Assessment.

Currently, there is a growing preference for convenience food products, such as ready-to-eat (RTE) foods, associated with long refrigerated shelf-lives, not requiring a heat treatment prior to consumption. Because Listeria monocytogenes is able to grow at refrigeration temperatures, inconsistent temperatures during production, distribution, and at consumer's household may allow for the pathogen to thrive, reaching unsafe limits. L. monocytogenes is the causative agent of listeriosis, a rare but severe human illness, with high fatality rates, transmitted almost exclusively by food consumption.
