Anaerobic growth of Corynebacterium glutamicum via mixed-acid fermentation.

Corynebacterium glutamicum, a model organism in microbial biotechnology, is known to metabolize glucose under oxygen-deprived conditions to l-lactate, succinate, and acetate without significant growth. This property is exploited for efficient production of lactate and succinate. Our detailed analysis revealed that marginal growth takes place under anaerobic conditions with glucose, fructose, sucrose, or ribose as a carbon and energy source but not with gluconate, pyruvate, lactate, propionate, or acetate. Supplementation of glucose minimal medium with tryptone strongly enhanced growth up to a

Pancreatic abscess caused by Corynebacterium coyleae mimicking malignant neoplasm.

A 50-year-old female was referred to our hospital because of postprandial epigastric pain and pancreatic head mass. On admission, an elastic hard mass with tenderness was palpable in the epigastric region. Laboratory findings showed no abnormalities, except for a slightly elevated C-reactive protein value and iron deficiency anemia. Serum levels of pancreatic enzymes and tumor markers were also within the reference range. Computed tomography (CT) demonstrated a 5-cm heterogenous mass at the head of the pancreas. Angiography showed that gastroduodenal artery was transformed and narrowed by the

Unusual Corynebacterium mucifaciens isolated from ear and nasal specimens.

OBJECTIVE: Corynebacterium mucifaciens was first proposed as a novel Corynebacterium species in 1997. This report describes our experience with the unexpectedly frequent isolation of C mucifaciens. STUDY DESIGN: By using agar supplemented with several antibiotics, microbiological studies were performed on specimens collected from the ear and nose. SETTING: A primary and secondary referral hospital. RESULTS: The organisms grew slowly. Five strains of C mucifaciens were found in specimens of middle ear effusion (MEE) from patients with otitis media with effusion (OME), and four strains were i

Comparative molecular characterization of Corynebacterium pseudotuberculosis of different origin.

Ribotyping and susceptibility to 17 antimicrobial agents were used to compare 37 isolates of Corynebacterium pseudotuberculosis (28 from horses, 1 from cattle, 3 from sheep and 5 from goats) derived from various types of lesions, and different geographic locations. According to the presence of nitrate reductase, all but one isolate from horses reduced nitrate (nitrate-positive), whereas all isolates from sheep and goats were unable to reduce nitrate (nitrate-negative). The ribotype of the nitrate-negative isolate from a horse with ulcerative lymphangitis was identical to all the other isolates

Corynebacterium JK: a cause of nosocomial device-related infection.

Ten cases of serious infection due to antibiotic-resistant Corynebacterium JK are reported. Five infections occurred in patients with hematologic malignancy and granulocytopenia, an association previously reported. Five additional nosocomial infections occurred in patients without underlying disease previously reported to be associated with JK. Nine of the 10 infections were nosocomial in origin and were associated with plastic devices that had been inserted into the patients. JK organisms were isolated from a variety of sources from an additional 40 uninfected patients. Although the majority

Citrate synthase condensation requires catalytic events in Corynebacterium glutamicum.

Protein condensation regulates cellular metabolism. Although many enzymes are highly structured proteins, the molecular mechanisms underlying their condensation remain poorly understood. Here, we report the discovery and characterization of Corynebacterium glutamicum citrate synthase (CgCS) condensates in relation to extracellular glutamate overproduction. CgCS, which does not possess typical intrinsically disordered sequences, formed droplet-like condensates in C. glutamicum. CgCS condensates were observed in growing cells and dispersed in stationary phase cells. Condensates were not formed f

Corynebacterium bovis Keratitis Mimicking Acanthamoeba Infection Following Ocular Trauma.

Corneal isolates of Corynebacterium species are frequently dismissed as contaminants, yet they are capable of causing severe, vision-threatening keratitis. A 45-year-old man with no history of contact lens use developed acute left eye pain and visual loss after a stone fragment struck his eye during work, with immediate contamination by garden hose water. Slit-lamp examination revealed a ring-shaped stromal infiltrate with visual acuity reduced to light perception, a presentation consistent with Acanthamoeba keratitis. Empiric therapy with fortified topical vancomycin and ceftazidime, antifung

Evaluation of omadacycline in vitro activity against Corynebacterium species.

Corynebacterium spp. are gram-positive bacteria increasingly recognized as pathogens. This study evaluates the MICs of omadacycline, a tetracycline, against 40 clinical Corynebacterium isolates using two methodologies: broth microdilution (BMD) and Liofilchem omadacycline MIC Test Strip (MTS). By BMD, the MIC50, MIC90, and MIC range were 0.5 µg/mL, 1 µg/mL, and 0.12-2.0 µg/mL, respectively. Comparing BMD to MTS, essential agreement (EA, within ±1 doubling dilution of the reference BMD MIC) was 87.5% (95% CI: 73.0%-95.4%).

Corynebacterium striatum-Induced Meningitis in a Non-immunocompromised Patient.

We present the case of a 46-year-old immunocompetent male diagnosed with meningitis caused by Corynebacterium striatum, a rare pathogen typically found in immunocompromised individuals. The diagnosis was confirmed by cerebrospinal fluid (CSF) culture, which grew C. striatum, and was supported by the exclusion of other common pathogens causing meningitis, including viral etiology, using polymerase chain reaction (PCR) testing. This case highlights the potential of C. striatum to cause central nervous system (CNS) infections in otherwise healthy individuals. The patient had a history of prosthet

A Rare Case of Locally Contracted Cutaneous Corynebacterium diphtheriae.

A 32-year-old man with a history of intravenous heroin use and housing instability presented with three years of worsening left forearm and wrist "infection," which had progressed over the past few months with worsening purulence, pain, and deformity. In the emergency department, he was afebrile with stable vitals. Superficial cultures drawn demonstrated polymicrobial growth, including heavy growth of Corynebacterium diphtheriae. He was treated with vancomycin and then IV penicillin to complete 10 days of therapy. Given the uncharacteristic appearance of the lesion, a biopsy was recommended, b

Identification and Evolutionary Relationship of Corynebacterium striatum Clinical Isolates.

Corynebacterium striatum has developed into a new community-acquired and hospital-acquired multi-drug resistance (MDR) bacterium, and is a potential target pathogen for infection control and antibacterial management projects. In this study, non-duplicate samples of inpatients were collected from a local central hospital. Mass spectrometry showed that 54 C. striatum isolates mainly appeared in secretion and sputum from 14 departments. Protein fingerprint cluster analysis showed that the isolates were divided into four groups, most of which appeared in summer. The drug resistance test showed tha

Impedance flow cytometry for viability analysis of Corynebacterium glutamicum.

Corynebacterium glutamicum efficiently produces glutamate when growth is inhibited. Analyses of viability in this non-growing state requires time consuming plating and determination of colony forming units. We here establish impedance flow cytometry measurements to assess the viability of non-growing, glutamate producing C. glutamicum cultures within minutes.

Late-onset native valve endocarditis caused by Corynebacterium kroppenstedtii.

Corynebacterium kroppenstedtii is an emerging cause of granulomatous mastitis and recurrent breast abscesses in women, but data on its clinical relevance in nongynecological disease conditions are limited. Here, we report the first case of a late-onset endocarditis of a native aortic valve in a 73-year-old male patient who presented with symptomatic aortic insufficiency. Echocardiography and cardiac computed tomography revealed the perforation of the noncoronary cusp and a large perivalvular abscess cavity. Hence, the surgical replacement of the aortic valve and aortic root were performed. Int
