Piroxicam-β-cyclodextrin: a GI safer piroxicam.

Although NSAIDs are very effective drugs, their use is associated with a broad spectrum of adverse reactions in the liver, kidney, cardiovascular (CV) system, skin and gut. Gastrointestinal (GI) side effects are the most common and constitute a wide clinical spectrum ranging from dyspepsia, heartburn and abdominal discomfort to more serious events such as peptic ulcer with life-threatening complications of bleeding and perforation. The appreciation that CV risk is also increased further complicates the choices of physicians prescribing anti-inflammatory therapy. Despite prevention strategies s

Osteotropic beta-cyclodextrin for local bone regeneration.

An osteotropic alendronate-beta-cyclodextrin conjugate (ALN-beta-CD) was developed as a bone-targeting delivery system for improved treatment of skeletal diseases. The conjugate shows very strong binding to hydroxyapatite (HA, main component of the skeleton). Its ability in forming molecular inclusion complex with prostaglandin E(1) (PGE(1), a potent bone anabolic agent) was confirmed by phase solubility experiments and differential scanning calorimetry (DSC). In a bilateral rat mandible model, ALN-beta-CD/PGE(1) molecular complex was shown to stimulate strong local bone anabolic reaction. In

Effects of dietary beta-cyclodextrin in hypercholesterolaemic rats.

Beta-cyclodextrin is a compound that forms inclusion complexes with a variety of molecules, specially bile acids and sterols. This study examines the effects of beta-cyclodextrin on cholesterol and bile acid metabolism in hypercholesterolaemic rats. Male Wistar rats were divided into 4 groups that received during 7 weeks: control diet, 2% cholesterol diet (A), A+2.5% beta-cyclodextrin (B) and A+5% beta-cyclodextrin (C). The cholesterol-rich diet induced hepatomegaly and fatty liver and significantly reduced cholesterol, bile acid and phospholipid secretion. Addition of beta-cyclodextrin normal

Cyclodextrin-based optosensor for the determination of warfarin in waters.

A flow-through optosensor for warfarin is described. The sensor is developed in conjunction with flow analysis systems and uses a commercial bound beta-cyclodextrin material as the sensing phase. A strong fluorescence signal was observed as a result of the formation of an inclusion complex between warfarin and beta-cyclodextrin. The analytical performance characteristics of the proposed sensor for analysis of low levels of warfarin were as follows: the detection limits for continuous and flow injection analysis systems were 2 and 19 ppb, respectively; the observed relative standard deviations

Cyclodextrins and Cyclodextrin-Based Polymers and Their Supramolecular Host-Guest Interactions for Sustainable Water Purification and Perovskite Solar Cell Devices: A Review.

Cyclodextrin-based polymers are a versatile class of emerging materials with unique supramolecular host-guest interaction features. Their diverse functional groups, hydrophobic cavities, and hydrophilic exteriors make them intriguing materials. These structural advantages promote molecular recognition, the absorption of guest molecules in their cavities, and the selective adsorption of guest molecules by the functional groups on their external surface. Recent research highlights that cyclodextrin polymers can simultaneously leverage supramolecular host-guest interactions and chelating ability

Dissipative particle dynamics simulation of Pickering emulsions co-stabilized by β-cyclodextrins and Tween 60.

In the field of food colloid systems, elucidating the synergistic stabilization mechanism of composite emulsifiers at the interface in Pickering emulsions is crucial for designing high-performance emulsions. This study investigates the synergistic stabilization mechanism of β-cyclodextrin (β-CD) and Tween 60 in Pickering emulsions using experiments and dissipative particle dynamics (DPD) simulations. The optimal mixing ratio (β-CD: Tween 60 = 1:3) achieves the lowest oil-water interfacial tension (5.38 ± 0.06 mN/m) and the smallest rearrangement rate constant (Kr). All systems exhibit contact

Formulation and stability study of an oral paediatric phenobarbital 1% solution containing hydroxypropyl-β-cyclodextrins.

OBJECTIVES: Phenobarbital is a barbiturate, used to treat focal and generalised epilepsy. Since the end of marketing of the oral solution KANEURON in 2017, phenobarbital tablets remain the only available dosage form. Development of an oral phenobarbital solution for paediatric use is therefore essential to fulfil clinical needs. A new formulation of phenobarbital with hydroxypropyl-β-cyclodextrins (HPBCD) was developed, and the physicochemical stability of the solution was evaluated. METHODS: Different excipients have been selected to formulate a solution of phenobarbital. Samples were dosed

Biopharmaceutical Assessment of Dexamethasone Acetate-Based Hydrogels Combining Hydroxypropyl Cyclodextrins and Polysaccharides for Ocular Delivery.

We previously developed two optimized formulations of dexamethasone acetate (DXMa) hydrogels by means of special cubic mixture designs for topical ocular administration. These gels were elaborated with hydroxypropyl-β-CD (HPβCD) and hydroxypropyl-γ-CD (HPγCD) and commercial hydrogels in order to enhance DXMa water solubility and finally DXMa's ocular bioavailability and transcorneal penetration. The main objective of this study was to characterize them and to evaluate in vitro, ex vivo, and in vivo their safety, biopermanence, and transcorneal permeation. Gels A and B are Newtonian fluids and

Cyclodextrins: Assessing the Impact of Cavity Size, Occupancy, and Substitutions on Cytotoxicity and Cholesterol Homeostasis.

Cyclodextrins (CDs) are cyclic oligosaccharides; the most common CDs contain six, seven, or eight glucose units called α-CDs, β-CDs, and γ-CDs, respectively. The use of CDs in biomedical research is increasing due to their ability to interact with membrane lipids as well as a wide variety of poorly water-soluble molecules. We assessed the impact of CD cavity size, occupancy, and substitutions on cytotoxicity and cholesterol homeostasis. The potency of CD-mediated cytotoxicity was in the order of β-CDs, α-CDs, and γ-CDs. Substitutions with hydroxypropyl or carboxymethyl group attenuated cytotox

Characterization of hydroxypropyl-beta-cyclodextrins used in the treatment of Niemann-Pick Disease type C1.

2-Hydroxypropyl-beta-cyclodextrin (HPβCD) has gained recent attention as a potential therapeutic intervention in the treatment of the rare autosomal-recessive, neurodegenerative lysosomal storage disorder Niemann-Pick Disease Type C1 (NPC1). Notably, HPβCD formulations are not comprised of a single molecular species, but instead are complex mixtures of species with differing degrees of hydroxypropylation of the cyclodextrin ring. The degree of substitution is a critical aspect of the complex mixture as it influences binding to other molecules and thus could potentially modulate biological effe

Curcumin complexation with cyclodextrins by the autoclave process: Method development and characterization of complex formation.

One approach to enhance curcumin (CUR) aqueous solubility is to use cyclodextrins (CDs) to form inclusion complexes where CUR is encapsulated as a guest molecule within the internal cavity of the water-soluble CD. Several methods have been reported for the complexation of CUR with CDs. Limited information, however, is available on the use of the autoclave process (AU) in complex formation. The aims of this work were therefore to (1) investigate and evaluate the AU cycle as a complex formation method to enhance CUR solubility; (2) compare the efficacy of the AU process with the freeze-drying (F

Mechanical capture, protection and interconversion of cyclodextrins in redox-responsive enzyme-catalysed dynamic combinatorial libraries.

Redox-controlled rotaxane formation can be employed as a novel strategy for cyclodextrin interconversion via a process of template-directed enzymatic synthesis, mechanical capture and release. Mechanical bonds are shown to protect cyclodextrins from hydolysis by α-amylase and cyclodextrin glucanotransferase, facilitating rotaxane isolation and enabling size-selective separation of cyclodextrins via reversible rotaxane formation.

Encapsulation of Polyhedral Borane Cluster trans-[B20H18]2- in Cyclodextrins: Stabilizing Effect in Aqueous Solution.

The preparation of polyhedral boranes/cyclodextrin (CD) inclusion complexes was studied to develop novel supramolecular assemblies that could be useful for biological applications. The encapsulation of the trans-[B20H18]2- anion in cyclodextrins was investigated using nuclear magnetic resonance (NMR) spectroscopy, isothermal titration calorimetry (ITC), and molecular dynamics (MD) combined with density functional theory (DFT) calculations. The results revealed the formation of strong inclusion complexes with γ- and β-CD, while weaker interactions were observed with the smaller α-CD. Very large

Establishment of a novel refinement workflow for oligopeptides containing branched-chain amino acids with cyclodextrins.

Peptides containing branched-chain amino acids (BCAAs) are recognized for their diverse physiological functions; however, effective methods for their target refinement remain underdeveloped. This study introduced a novel approach utilizing the cavity structure of cyclodextrins, along with specific adsorption (50% ethanol, 50°C) and crystallization conditions (75% ethanol, 4°C), to selectively isolate BCAA-containing oligopeptides from Antarctic krill. Following refinement, the relative BCAA content was elevated to 1.70-fold compared to the original peptide mixture, with the number and abundanc

Improved triamcinolone acetonide-eluting contact lenses based on cyclodextrins and high hydrostatic pressure assisted complexation.

Contact lenses (CLs) constitute an advantageous platform for the topical release of corticosteroids due to their prolonged contact with the eye. However, the lipophilic nature of corticosteroids hampers CLs' ability to release therapeutic amounts. Two approaches to improve loading and release of triamcinolone acetonide (TA) from poly(2-hydroxyethyl methacrylate)-based hydrogels were investigated: adding 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) to the monomers solution before polymerization (HEMA/i-CD) and an hydrogels' post-treatment with HP-β-CD (HEMA/p-CD). The effect of HP-β-CD and steriliz

Cyclodextrins reduce the ability of Pseudomonas aeruginosa outer-membrane vesicles to reduce CFTR Cl- secretion.

Pseudomonas aeruginosa secretes outer-membrane vesicles (OMVs) that fuse with cholesterol-rich lipid rafts in the apical membrane of airway epithelial cells and decrease wt-CFTR Cl- secretion. Herein, we tested the hypothesis that a reduction of the cholesterol content of CF human airway epithelial cells by cyclodextrins reduces the inhibitory effect of OMVs on VX-809 (lumacaftor)-stimulated Phe508del CFTR Cl- secretion. Primary CF bronchial epithelial cells and CFBE cells were treated with vehicle, hydroxypropyl-β-cyclodextrin (HPβCD), or methyl-β-cyclodextrin (MβCD), and the effects of OMVs
