Cytokines produced in response to plaque bacteria clearly play a key role in the periodontal diseases. However, we know very little about the interactions between cytokines and periodontopathogenic bacteria. The aims of this study were to determine whether the key pro-inflammatory cytokines interleukin-1 beta (IL-1 beta) and IL-6 could affect the growth of Actinobacillus actinomycetemcomitans or Porphyromonas gingivalis and to determine whether these organisms could hydrolyse IL-1 beta, IL-6 or the anti-inflammatory IL-1 receptor antagonist (IL-1ra). Culture medium containing up to 100 ng/ml o

Patients with end-stage renal failure present with various debilitating forms of renal osteodystrophy characterized either by high bone turnover with or without mineralization defect or low bone turnover, that is, adynamic bone disease. Alterations in parathyroid hormone and calcitriol production do not completely account for the abnormalities in bone turnover. This suggest that other factors or mediators, or both, are involved in the regulation of bone turnover. Among them, the cytokine systems are of particular interest because of the following: cytokine production is altered in uremic patie

Allergic airway inflammation is characterized by peribronchial eosinophil accumulation within the submucosa surrounding the airways. The development of appropriate animal models to dissect the critical mechanisms involved in the deleterious responses is crucial for the development of proper therapeutic approaches. We have utilized a model of allergic airway inflammation induced by intratracheal challenge with parasite (Schistosoma mansoni) egg antigen in presensitized mice. The use of this model to examine the eosinophil recruitment has identified key cytokines involved in the responses. These

The role of cytokines was intensively discussed over the course of a two and a half day meeting sponsored by the US-JAPAN Cancer Cooperative Research Program of the Office of International Affairs, National Cancer Institute and held at The National Institutes of Health, Bethesda, Maryland on 15-17 January 1996. Most of the first day was devoted to a discussion of the role of cytokines in modulating angiogenesis and the consequent effect of this on tumor growth and metastases. This was followed by sessions on the effect of various cytokines in enhancing or suppressing immunological responses to

Cytokines, by definition, exert an effect on haematopoiesis. Diseases characterized by haematopoietic insufficiency, such as aplastic anaemia, should therefore be investigated for abnormal expression of these regulatory proteins. In studies on hairy cell leukaemia, a severe deficiency was found in the production of interleukin-3 (IL-3), granulocyte-macrophage colony stimulating factor (GM-CSF), granulocyte CSF, IL-6 and tumour necrosis factor alpha (TNF alpha). Further studies on IL-6 at the mRNA and protein levels revealed that peripheral blood mononuclear cells and even hairy cells could be

Virus vaccines prepared on the basis of cells of mammals (rabies, poliomyelitis, measles and hepatitis A vaccines) contain cytokines (IL-1 beta, IL-6, TNF-alpha), whose concentration depends on the kind of the vaccine. Cell lines (green monkey kidney cells, VERO, 4647), used for the preparation of commercial and experimental vaccines, do not produce spontaneously any of the above cytokines. Cell line L-68, used for the manufacture of experimental measles vaccine, is capable of the spontaneous synthesis of IL-6. In Russian and foreign preparations of interferon the presence of IL-1 beta and TNF

Studies that were published over the past year have added new knowledge to our understanding of cellular abnormalities in systemic lupus erythematosus (SLE). Antigen-specific and "pathogenic" T cells can be identified and characterized in SLE. Interleukin-10 has been added to the factors that may promote B cell overactivity and autoantibody production. Protein kinase isozyme I was shown to be deficient in patients with SLE, indicating defects in cell signaling events. Aberrant expression of adhesion molecules on the surface membrane of leukocytes and endothelial cells was shown, with important

The second international congress on cytokines, held in Florence (March 23-25, 1992), has been marked by the presentation of recent data about cytokine-deficient mice obtained by homologous recombination which, contrary to expectation, exhibit normal T and B cell development. The cloning of a few new cytokines and receptors were also reported as well as recent advances in the knowledge of the oldest members of this quickly growing family. The meeting has also been marked by a great number of reports providing insights into T cell-dependent immune response. Participants provided evidence, throu

Background: Neuroinflammation is increasingly recognized as an important contributor to Parkinson's disease (PD), yet perioperative immune responses in this population remain incompletely characterized. This study investigated perioperative cytokine dynamics in patients with PD compared with healthy controls (HCs) undergoing orthopedic surgery under general anesthesia. Methods: In this prospective pilot observational study, 50 patients scheduled for lower limb orthopedic surgery were enrolled (25 PD patients, 25 HCs). Serum cytokines (IL-6, IL-8, VEGF, MCP-1, HMGB1, S100B, and PARK7) were meas

PURPOSE OF REVIEW: Well tolerated, scalable strategies capable of targeting latent HIV reservoirs are essential for the development of cure approaches that can be implemented globally. In recent years, common γ-chain cytokines (γc-cytokines) have emerged as promising candidates as standalone interventions or in combination with complementary therapeutic strategies. γc-cytokines exhibit latency-reversing activity and immunomodulatory effects. In this review, we summarize recent in-vitro, ex-vivo, and in-vivo studies evaluating the use of γc-cytokines in HIV cure research.
RECENT FINDINGS: γc-c

Exploratory analyses have suggested associations between cytokine levels and symptom response in cancer patients receiving placebo; however, the inferential strength of these findings remains limited. This letter introduces the concept of "fragile results" to describe outcomes derived from underpowered models subject to extensive multiplicity and lacking internal validation. Key concerns include low events-per-predictor ratios, multiple hypothesis testing, and potential inflation of type I error in flexible modeling approaches. To guide future research, sample size requirements for confirmator

The aim of the present study was to evaluate the diagnostic significance of the dynamics of cytokines and growth factors during pregnancy with and without preeclampsia. The study included 168 pregnant women at risk of hypertensive disorders. The levels of biomarkers of all pregnant women were studied at 12-16 weeks, 28-30 weeks and 36-38 weeks. These included cytokines (tumour necrosis factor-α, interferon-, γinterleukin-4) and growth factors (placental growth factor, vascular endothelial growth factor). All pregnant women were divided into two groups: 124 patients with preeclampsia and 44 wit

Due to the close relationship between the vitreous and posterior eye layers, the microenvironment of these layers can affect the composition of the vitreous. Molecular analysis of the vitreous may therefore provide important insights into the pathogenesis of chorioretinal diseases. In this study, vitreous cytokines (n = 41) were evaluated to gain further insights into the tumor microenvironment in uveal melanoma (UM) arising from the choroid (CM). Cytokine levels were measured using a bead-based multiplex immunoassay panel in vitreous samples obtained from 32 eyes, including 18 with CM and 14

Meniere disease (MD) is a rare set of conditions associated with the accumulation of endolymph in the cochlear duct and the vestibular labyrinth with a decrease of endocochlear potential. It is considered a chronic inflammatory disorder of the inner ear with a multifactorial origin. The clinical syndrome includes several groups of patients with a core phenotype: sensorineural hearing loss, episodes of vertigo, and tinnitus with a non-predictable course. Genetic factors and the innate immune response seem to play a central role in the pathophysiology of the condition. Autoimmune MD should be di

During folliculogenesis, different proinflammatory cytokines have a physiological role in the weakening of the follicle wall and an eventual rupture at ovulation. Chronic inflammation is closely related to endothelial dysfunction, cardiovascular disease, coronary artery disease, and polycystic ovary syndrome. Polycystic ovary syndrome is characterized by androgen excess and ovarian dysfunction. Emerging evidence suggests that the long-term metabolic effects and cardiovascular complications observed in this syndrome may be related to the presence of a mild chronic inflammatory state. It is uncl

Fascioliasis is a foodborne trematode endemic worldwide. Children under 15 years have the highest prevalence of infection. We hypothesized that acute fascioliasis would be associated with more pronounced cytokine changes than in chronic disease or no helminth infections. To test this hypothesis, 33 children who lived in the Peruvian highlands were classified into 3 groups: acute fascioliasis, chronic fascioliasis, and no helminth infection. Type Th1, Th2, and Th17 cytokines were measured in plasma by cytometric bead array. Children with acute infection had higher levels of IL-5 and IL-17 compa

BACKGROUND: The role of cytokines in various disease states is a burgeoning field of academic study and clinical application, however there are no consensus documents on how certain cytokines should be stored prior to quantification. This information is especially of interest to researchers assembling a biobank or clinicians who have to transport specimens to a different location in order to be tested.
OBJECTIVE: To review the literature and synthesize prior findings on cytokine storage and freeze/thaw stability.
DESIGN: We searched PubMed for articles related to cytokine storage stability.

Epidemiological data demonstrate that patients with diabetes have an augmented risk of developing various types of cancers, accompanied by higher mortality. A number of mechanisms for this connection have been hypothesized, such as insulin resistance, hyperinsulinemia, hyperglycemia, and increased inflammatory processes. Apart from these potential mechanisms, several diabetes-associated dysregulated cytokines might be implicated in the link between diabetes and cancer. In fact, some inflammatory cytokines, e.g. TNF-α, IL-6 and leptin, have been revealed to play important roles in both initiati
