In silico analysis of luteolin derivatives as antibacterial agents targeting DNA gyrase and CTX-M-15 extended-spectrum β-lactamase of Escherichia coli.

Luteolin exhibited antibacterial activity against Escherichia coli and its chemical structure similar to that of ciprofloxacin (CPF) which works by inhibiting DNA gyrase. Filtrate from passion fruit extract containing luteolin and its derivatives could inhibit extended-spectrum β-lactamase (ESBL)-producing E. coli. Antibacterial compounds that can also inhibit ESBL will be valuable compounds to overcome the problem of resistant bacteria. This study aimed to ensure the potency of luteolin and luteolin derivatives targeting DNA gyrase and ESBL by in silico approach. Docking simulation of ligands

Water network chemistry to exploit the nature of catalytic water molecules in Mtb DNA gyrase: a computational study to understand the binding mechanism of fluoroquinolones.

The dynamics of DNA gyrase and mutants of DNA gyrA such as G88A, A90V, S91P, D94A, D94G, D94N, D94Y; and double-point mutant (S91P-D94G), are meticulously investigated using computational approaches. Molecular dynamics (MD) and hydration thermodynamics have shed light on the fundamental, mechanistic basis of mutations on the conformational stability of Quinolone Binding Pocket (QBP) of DNA gyrase. Analysis of MD results revealed the displacement of a single crystal water molecule (HOH201) from the catalytic site of wild-type (WT) and mutants of DNA gyrA. This prompted our research group to pro

Epigenetic Activation of Silent Pathways in Endophytic Aspergillus sp. EGP214 Reveals Porphyrin-Derived Antimicrobial Metabolites Targeting DNA Gyrase: In Vitro and In Silico Evaluation.

Endophytic fungi are a reservoir for cryptic metabolites to discover new bioactive compounds through their research on inactive biosynthetic gene clusters. In this study, Aspergillus sp. EGP214 was used to identify Aspergillus sp. EGP214, which they isolated from Hyoscyamus muticus during their study. The expression of cryptic genes needed stimulation through the use of two epigenetic modifiers, which included trichostatin A as a histone deacetylase inhibitor and 5-aza-2'-deoxycytidine as a DNA methyltransferase inhibitor, during fermentation. GC-MS analysis revealed that both treatments signi
