Joubert syndrome (JBTS) is a clinically and genetically heterogeneous group of ciliary diseases. To date, 34 subtypes of JBTS have been classified due to different causative genes or extra clinical features. Most of them are autosomal recessive, while only the subtype 10 (JBTS10) is a quite rare X-linked recessive disorder caused by OFD1 mutations with few reports. In this study, by using whole exome sequencing (WES), a novel OFD1 splicing mutation (c.2488+2T>C) was identified in a male fetus with suspected Dandy-Walker variant (DWV) and syndactyly, for whom abnormal karyotype and pathogenic C

A black and white female German Holstein calf showed a highly deformed cranium. The animal was not able to stand. Further findings were bilateral strabismus divergens and negative pupillary light reflexes. Magnetic resonance imaging and pathological-anatomical examination showed that the cerebrum was replaced to a high degree by the ventricle system filled with 1.5 liters of cerebrospinal fluid. The hemispheres of the cerebellum were ruptured by the dilated fourth ventricle. In addition, the vermis and pons were missing and fluid accumulation in the subarachnoidal space extending up to the fir

PURPOSE: The clinical characteristics and treatment of the rare methylmalonic academia (MMA) with Dandy-Walker syndrome (DWS) are still unclear. This study aims to provide insights into this rare pediatric condition.
METHODS: A retrospective analysis was conducted on MMA cases from our pediatric ICU database (2006-2021), identifying those with concurrent DWS. Onset age, clinical features, genetic types, treatment, and prognosis were analyzed. MMA diagnosis was confirmed via blood and urine tandem mass spectrometry, while DWS was diagnosed through cranial MRI. Follow-up was conducted via phone

BACKGROUND: Dandy-Walker syndrome (DWS) is an uncommon congenital abnormality characterised by agenesis or hypoplasia of the cerebellar vermis, cystic dilation of the fourth ventricle, and posterior fossa expansion. Neurological defects usually accompany the DWS; however, it has not been documented to coexist with amelia, genu recurvatum, complex congenital cardiac defects, or dyslipidaemia. CASE PRESENTATION: An 18-month-old girl who is a known case of DWS in association with multiple congenital anomalies, including bilateral amelia of the upper limbs, developmental dysplasia of the hip (DDH)

BACKGROUND: The Dandy-Walker malformation (DWM) is one of the commonest congenital cerebellar defects, and can be associated with multiple congenital anomalies and chromosomal syndromes. The occurrence of overlapping 3q deletions including the ZIC1 and ZIC4 genes in few patients, along with data from mouse models, have implicated both genes in the pathogenesis of DWM.
METHODS AND RESULTS: Using a SNP-array approach, we recently identified three novel patients carrying heterozygous 3q deletions encompassing ZIC1 and ZIC4. Magnetic resonance imaging showed that only two had a typical DWM, while

INTRODUCTION: Dandy-Walker syndrome (D-WS) is a rare congenital brain anomaly that primarily impacts the fourth ventricle and cerebellum. Its variant is even rarer and includes cerebellar dysgenesis, with possible posterior fossa enlargement and variable cerebellar vermis hypoplasia.
CASE REPORT: We present the case of a patient diagnosed late with the Dandy-Walker Syndrome variant, associated with Leigh Syndrome, at a tertiary hospital. The patient received an optimized, multidisciplinary treatment approach to improve prognosis.
DISCUSSION: Early intervention in pediatric neurodegenerative

OBJECTIVE: Treating Dandy-Walker syndrome-related hydrocephalus (DWSH) involves either a CSF shunt-based or endoscopic third ventriculostomy (ETV)-based procedure. However, comparative investigations are lacking. This study aimed to compare shunt-based and ETV-based treatment strategies utilizing archival data from the Hydrocephalus Clinical Research Network (HCRN) registry.
METHODS: A retrospective review of prospectively collected and maintained data on children with DWSH, available from the HCRN registry (14 sites, 2008-2018), was performed. The primary outcome was revision-free survival o

Dandy-Walker Syndrome (DWS) is a rare cerebellar malformation characterized by the underdevelopment of the cerebellar vermis and associated complications. This case report presents a 3-month-old female from Somalia who exhibited fever, loss of consciousness, vomiting, and developmental delays. Neurological examination suggested signs of meningitis, and MRI revealed an enlarged posterior fossa consistent with DWS. Despite initial antibiotic treatment for suspected bacterial meningitis, CSF cultures yielded no growth, raising concerns about prior antibiotic use. Unfortunately, the patient's cond

The Keratitis-Ichthyosis-Deafness syndrome (KIDS) is an autosomal dominant ectodermal dysplasia characterized by ocular, skin, and ear anomalies, including keratitis, palmoplantar keratoderma, and congenital hearing loss. Most cases are due to mutations in the GJB2 gene encoding connexin 26. The Dandy-Walker malformation (DWM) is a developmental anomaly of the midline of the cerebellum with complete or partial agenesis of the vermis and cystic dilatation of the fourth ventricle. The association of KID syndrome with DWM has been reported a few times, but thought to be coincidental. We report 4

We report on sisters with similar craniofacial anomalies, a brain malformation in the area of the posterior fossa, and a congenital heart defect. The craniofacial findings include macrocephaly, a prominent forehead and occiput, foramina parietalia, hypertelorism, downslanting palpebral fissures, a depressed nasal bridge, narrow palate, and apparently low-set ears. Patient 1 had a Dandy-Walker malformation with communicating hydrocephalus, aplasia of the posterior portion of the cerebellar vermis, and high insertion of the confluent sinus, while in patient 2, a Dandy-Walker variant was found wi

Cerebral salt wasting (CSW) is an uncommon cause of hyponatremia characterized by extracellular volume depletion, high urine sodium concentration and osmolality, and low serum uric acid concentration in association with central nervous system (CNS) disease. Distinguishing CSW from the syndrome of inappropriate secretion of antidiuretic hormone (SIADH), a much more common form of hyponatremia in this setting, can be challenging because both present with identical laboratory features. However, treatment of CSW and SIADH differs, making a correct diagnosis important. Here we present a case of CSW

Wisconsin syndrome is a very rare genetic condition characterized by coarse facies, prominent nasal tip, bushy high arched/upsweeping eyebrows, and a full/everted lower lip. Deletion of chromosome 3q24q25 region is considered critical for its manifestation. Deletion of the adjoining chromosome region 3q24 is associated with Dandy-Walker malformation, while 3q22.3 deletion is associated with blepharophimosis-ptosis-epicanthus inversus syndrome. Here, we report an individual with chromosome 3q22.2-q26.2 interstitial deletion presenting with Wisconsin syndrome, blepharophimosis-ptosis-epicanthus

UNLABELLED: For patients with comorbid complex posttraumatic stress disorder (PTSD) and psychotic disorder, trauma-focused therapy may be difficult to endure. Phase-based treatment including (a) stabilization, (b) trauma-focused therapy, and (c) integration of personality with recovery of connection appears to be the treatment of choice.
OBJECTIVE: The objective of this article is to describe and evaluate the therapeutic process of a single case from a holistic perspective.
METHOD: We present a case report of a 47-year-old woman treated for severe complex PTSD resulting from repeated sexual

Associations among Down syndrome (DS), Dandy-Walker variant (DWv), pulmonary hypertension (PH) and childhood interstitial lung disease (chILD) are extremely rare. Several cases of trisomy disorders with Dandy-Walker malformation (DWM) and neurodevelopmental outcomes have been reported. The extent to which moderate to severe pulmonary hypertension with complications of patent ductus arteriosus (PDA), PH and chILD leads to substantial morbidity and mortality in infants with DS and DWM should be studied. We report the case of an ex-premature 15-month-old girl with confirmed DS with DWv, who devel

We report on a female infant with postaxial polydactyly of the hands, preaxial polydactyly of the right foot, cleft palate, microglossia and tongue hamartomata consistent with an oral-facial-digital syndrome (OFDS). The patient also had optic nerve colobomata, a Dandy-Walker malformation, micrognathia and apneic spells. This combination of clinical features has not been previously reported. This patient either expands the clinical features of one of the existing OFDS or represents a new variant. A review of the literature highlights the difficulties in making a specific diagnosis because of th
