Spider toxin (JSTX) on the glutamate synapse.

A new neurotoxin (JSTX) was separated from spider (Nephila clavata, Joro spider) venom. JSTX irreversibly suppressed the excitatory postsynaptic potential (EPSP) and the glutamate potential in the lobster neuromuscular junction with high degree of specificity. The threshold concentration for suppressing EPSPs corresponds to a small fraction of the toxin in a venom gland, roughly estimated as low as 10(-10) M/l. 10(-10) M/l. In the giant synapse of squid stellate ganglion JSTX suppressed EPSPs without affecting the antidromic response. Glutamate-induced membrane depolarization was blocked by JS

Inactivation of the Na permeability in squid giant axons.

Inactivation of the Na permeability has been studied in intact and perfused squid giant axons with the voltage clamp method. The main results are: 1. Upon depolarization inactivation develops along an exponential time course; the upper limit for an initial delay in the development of inactivation is 50-100 musec. 2. Adding 20-40 mM KCl to K-free external solution accelerates the development of inactivation and slows its removal. 3. Scorpion venoms increase the maintained conductance, i.e. make inactivation less complete; the voltage dependence of the maintained conductance is different from th

Cuttlefish Ink-Derived Melanin/MXene Composites: Boosting Stability and Unleashing Synergistic Photothermal-Mechanical Antimicrobial Effects Against Biofilms.

The advancement of MXene-based antibacterial platforms has been impeded by two critical limitations. Specifically, these include rapid oxidative degradation in physiological environments and potential cytotoxicity derived from their metallic nature. To address these challenges, we develop a core-shell structured nanikohybrid (CI@MXene) through the integration of natural cuttlefish ink melanin (CI) with MXene nanosheets. This biomimetic design establishes a protective barrier that effectively prevents MXene oxidation, thereby preserving its structural integrity and granting it durable antibacte

Screening of Potential Angiotensin-Converting Enzyme-Inhibitory Peptides in Squid (Todarodes pacificus) Skin Hydrolysates: Preliminary Study of Its Mechanism of Inhibition.

BACKGROUND: Hypertension has been identified as a significant risk factor for cardiovascular disease. Given the prevalence of the adverse effects of angiotensin-converting enzyme-inhibitory (ACEI) drugs, natural and effective alternatives to these medications need to be identified. METHODS: An investigative study was conducted to assess the ACEI capacity and structural characteristics of enzymatic hydrolysates with varying molecular weights derived from squid skin. The amino acid sequences of the enzymatic digests were analyzed via Nano LC-MS/MS and screened for peptides with ACEI activity us

Superb bio-effectiveness of Cobalt (II) phthalocyanine and Ag NPs adorned Sm-doped ZnO nanorods/cuttlefish bone to annihilate Trichinella spiralis muscle larvae and adult worms: In-vitro evaluation.

Herein, innovative biocides are designed for the treatment of Trichinella spiralis muscle larvae (ML) and adult worms. Samarium-doped ZnO nanorods (Sm-doped ZnO) are stabilized onto the laminar structure of cuttlefish bone (CB) matrix and adorned by either Ag NPs or cobalt phthalocyanine (CoPc) species. Physicochemical characteristics of such nanocomposites are scrutinised. Adorning of Sm-doped ZnO/CB with Ag NPs shortens rod-like shaped Sm-doped ZnO nanoparticles and accrues them, developing large-sized detached patches over CB moiety. Meanwhile, adorning of Sm-doped ZnO/CB by CoPc species de

Proprioceptive hair cells on the neck of the squid Lolliguncula brevis: a sense organ in cephalopods for the control of head-to-body position.

Decapod cephalopods, such as cuttlefishes and squids, have a distinct neck region that allows movements (roll, pitch and yaw) of the head relative to the body. This paper describes the structure, innervation and central pathways of proprioceptive hair cells on the neck of the squid Lolliguncula brevis that sense such movements and control head-to-body position. These hair cells exist on the dorsal side of the neck underneath the nuchal cartilage, close to the animal's midline on either side of the nuchal crest. On each side, the hair cells can be divided into an anterior and a posterior group

Abundant mRNAs in the squid light organ encode proteins with a high similarity to mammalian peroxidases.

A library derived from mRNA in the bacterial light organ of the squid, Euprymna scolopes, contained an unexpectedly high proportion of cDNAs that encode proteins with approximately 30% similarity to a family of mammalian peroxidases (PO) including myelo-PO, eosinophil PO, and thyroid PO (donor:hydrogen-peroxide oxidoreductase; EC 1.11.1.7). Two nearly full-length cDNAs were determined to encode putative PO of nearly 93 kDa each that are 97% identical in amino acid sequence to each other. Each contains four potential glycosylation sites, and His416, believed to be within the active site of the

Selective modification of the squid axon Na currents by Centruroides noxius toxin II-10.

We have studied the effects of the purified toxin II-10, from the venom of the scorpion Centruroides noxius Hoffmann, on the Na and K currents of voltage clamped squid giant axons. Extracellular applications of 10 microM of toxin II-10 produced a selective depression of peak Na currents, with no significant effects on the time course of K currents. On pharmacologically separated Na currents, low concentrations of toxin II-10 (0.28-1 microM) caused a reversible decrease in inward and outward peak INa, with little effect on either the maintained level of the currents or their turning-off. At hig

Salting-induced lipid hydrolysis and oxidation in dried squid fillets: A mechanistic link to formaldehyde formation, color browning, and flavor alteration.

Salt-dried squid fillets (SDSF) is a popular seafood product. In this study, the effect of different NaCl concentrations (0 %, 1 %, 5 % and 10 %) on the color, formaldehyde (FA) and volatile compounds (VOCs) of SDSF was investigated by determining the acid value (AV), thiobarbituric acid reactive substances (TBARS) value, lipid-related enzyme activities and lipidomics. These findings showed that the AV and TBARS value initially increased and subsequently decreased with increasing NaCl concentrations. Lipidomics indicated salting facilitated the hydrolysis of triglycerides and phospholipids by

Isolation and structural characterisation of acid- and pepsin-soluble collagen from the skin of squid Sepioteuthis lessoniana (Lesson, 1830).

Acid-solubilised collagen (ASC) and pepsin-solubilised collagen (PSC) were effectively isolated from squid skin with good yield and total protein content. ASC and PSC consist of two α-chains with an imino acid content of 182.6 and 184 imino acid residues/1000 residues. The molecular weight was determined to be between 73 and 107 kDa by using SDS-PAGE. For peptide mapping, collagens were digested with achromo endopeptidase, and all components, including α, β-chains, were markedly hydrolysed. Degradation peptides with molecular weights between 106.9 and 15.47 kDa were obtained. UV-vis absorption

A peroxidase related to the mammalian antimicrobial protein myeloperoxidase in the Euprymna-Vibrio mutualism.

Many animal-bacteria cooperative associations occur in highly modified host organs that create a unique environment for housing and maintaining the symbionts. It has been assumed that these specialized organs develop through a program of symbiosis-specific or -enhanced gene expression in one or both partners, but a clear example of this process has been lacking. In this study, we provide evidence for the enhanced production of an enzyme in the symbiotic organ of the squid Euprymna scolopes, which harbors a culture of the luminous bacterium Vibrio fischeri. Our data show that this enzyme has a

[Antioxidant effect of a phospholipid complex isolated from marine organisms].

Antioxidative effect of phospholipid complex from the sea organisms (preparation "Kalmofil") is observed comparatively with other phospholipid fractions of animal and plant origin. Most antioxidative action of the phospholipid complex from Mollusca (preparation "Kalmofil") on the nonenzymatic LPO reactions in vitro is established. Analogous fractions from different functional animal tissues have not such influence. Application of "Kalmofil" in vivo under experimental rat hepatitis provoked by CCl4 has a corrective effect on both nonenzymatic and Fe(2+)- and NADP.H-depending LPO reactions. This

Potential-dependent variations of the intracellular pressure in the intracellularly perfused squid giant axon.

Intracellular pressure responses were recorded from squid giant axons after the axoplasm was removed by the intracellular perfusion technique. A glass tube was inserted into the axon and the movement of the air-water interface formed on the end of the tube was observed with a Y-shaped fibrescope. The intracellular pressure increased and decreased rapidly when an action potential was induced by electrical stimulation. The amplitude of the response was about 10 mPa (or 1 X 10(-3) mmH2O), which was very large in comparison with that observed in unperfused axons. It was sensitive to extracellular

Modification of sodium channel inactivation in single myelinated nerve fibers by methionine-reactive chemicals.

Several methionine-reactive reagents, including N-bromoacetamide, N-bromosuccinimide, chloramine-T, and N-chlorosuccinimide, irreversibly slowed and prevented Na channel inactivation in single myelinated nerve fibers, whereas sulfhydryl- or tyrosine-modifying reagents had little effect. The activation process was not modified by the reagents that altered inactivation and could be modulated normally by Ca++ ions and Centruroides scorpion toxin II alpha. These results suggest that externally applied N-bromoacetamide and its related compounds specifically affect Na channel inactivation by modifyi

The effect of local anaesthetics on the components of the asymmetry current in the squid giant axon.

The effects of local anaesthetics and holding potential on sodium and asymmetry currents were studied in intracellularly dialysed squid giant axons. The asymmetry currents were fractionated into their inactivating and non-inactivating components, and the charge displacements Qi and Qn of the two components were determined for pulse potentials between -20 and +40 mV. The charged local anaesthetic RAD 366, a quaternary derivative of lidocaine, applied internally at a concentration in the dialysis solution of 1 mM, did not change Qn, but reduced Qi about 3-fold. The neutral local anaesthetic benz

Active control of intracellular pH.

When the interior of a squid giant axon is loaded with acid, the cell responds by extruding that acid in an energy-requiring process. Our results indicate that this acid extrusion is accomplished by exchanging external HCO3- for internal Cl- at the expense of ATP.

Mode of action of trichloroethylene on squid axon membranes.

The mode of action of trichloroethylene on electrical properties of squid giant axons has been studied by means of voltage clamp techniques. Trichloroethylene decreased the resting membrane potential in a manner dependent upon the concentration, the depolarization by 50% saturated trichloroethylene attaining 28.4 and 32.7% of the initial value at 20 and 10 degrees C, respectively. Leakage conductance was decreased to 34.6% of the control by 30% saturated trichloroethylene at 10-12 degrees C. It appears that the trichloroethylene-induced depolarization is at least in part due to a decrease in r

Health risks associated with the consumption of shrimp and squid contaminated with heavy metals in Poland.

Seafood may accumulate heavy metals (HMs) from environmental sources, posing potential health risks through dietary intake. This study aimed to assess the health risk associated with dietary intake of Hg, Cd, As and Zn from selected seafood available in Poland. In this study, a total 51 samples of shrimp and squid were analysed. The metals content was determined using such analytical methods as ET-AAS with a graphite furnace in the case of Cd, ICP-OES in the case of As and Zn, and CV-AFS in the case of Hg. The mean content of Cd and Zn were found to be 0.25 and 16.94 mg/kg f. w., respectively.
