Cytokine signalling in rat pulp interstitial fluid and transcapillary fluid exchange during lipopolysaccharide-induced acute inflammation.

The dental pulp consists of loose connective tissue encased in rigid dentinal walls. Because of its topography the tissue has low interstitial compliance and limited capacity to expand during fluid volume changes. Due to limitations regarding access to interstitial fluid, basic knowledge on transcapillary fluid transport parameters is lacking for this organ. The scope of this project was dual: first we aimed at establishing a method for isolation of pulp interstitial fluid (IF), and second we applied the method in rats subjected to lipopolysaccharide (LPS)-induced endotoxaemia. The aim was to

Assessment of bond strength and bioactivity on a prototype resin-based luting agent containing a novel bioactive monomer.

A prototype luting agent based on a resin composite containing a novel bioactive monomer (a calcium salt of 4-methacryloyloxyethyl trimellitate; CMET) was developed, and its shear bond strength to tooth materials (dentin and enamel) and bioactivity was compared with those of commercially available resin-based luting systems. Extracted bovine incisors were embedded in a mold with a potting material. The labial bonding surface of the embedded tooth was wet-ground using #400 silicon carbide abrasive paper until sufficient superficial enamel or dentin was exposed. The bonding and luting agents ass

The in vitro assessment of the degree of monomer conversion, biaxial flexural strength, and mineral precipitation on demineralised dentine of novel resin composite containing monocalcium phosphate monohydrate and polylysine.

OBJECTIVE: The development of ion-releasing resin composites is expected to reduce the risk of secondary caries. This study compared the degree of monomer conversion, biaxial flexural strength/modulus, elemental release, and remineralisation potential of a novel ion-releasing dental composite (Renewal MI) containing monocalcium phosphate monohydrate and polylysine. MATERIALS AND METHODS: The degree of monomer conversion after light curing for 20 s was determined (n = 8). The biaxial flexural strength and modulus after immersion in water for 24 h (n = 8) were evaluated. Additionally, the relea

Topical application of tetracycline-HCl in human periodontitis.

Previous in vitro studies have suggested that tetracycline-HCl (TTC-HCl) is adsorbed and actively released from root dentin. The aim of the current study was to evaluate the binding to and release of TTC-HCl from human root dentin surfaces in vivo, and to evaluate the clinical utility of TTC-HCl irrigation as an adjunct to scaling and root planing. Experiment I utilized two contralateral mandibular single-rooted teeth which were examined in four adults with severe generalized periodontitis. One tooth in each patient was carefully scaled and root planed, under local anesthesia, and the other us

The role of magnesium in the aetiology and prevention of caries: some new findings and implications.

While some epidemiological studies seem to indicate that a high intake of magnesium should be associated with a low prevalence of dental caries, the results of experimental studies are mainly equivocal. Magnesium is probably not bound to the apatite lattice of dental enamel or dentine, or it is bound to a small degree only. It is mainly located in the hydration layer of the apatite crystallites. In the dental caries process it is preferentially dissolved together with the carbonate of the mineral phase. It is not known to what extent feasible dietary changes can modify tooth magnesium content

3D-printable bioactive glass-based polymer-infiltrated ceramic for biomimetic tooth root applications.

Titanium implants are widely utilized for tooth root reconstruction due to their excellent mechanical and biological properties. However, their mechanical properties differ from those of dentin. This study aims to develop a 3D-printable polymer-infiltrated ceramic network (PICN) as a tooth root restoration material that mimics the natural root shape, mechanical properties, and biocompatibility. A bioactive glass (BG)-based photocurable slurry was prepared for vat photopolymerization, printed, sintered, and subsequently polymer-infiltrated to form 3D-printable PICN (3D-PICN). For comparison, 3D

Calcium Silicate Particles: Biological and Mechanical Role in the Hybrid Layer.

The addition of ion-releasing particles to experimental adhesives has been evaluated as a strategy to increase the longevity of bonded interfaces. This study evaluated the effect of calcium silicate particles (CaSi) applied as a pretreatment to acid-etched dentin surfaces and/or added (5% by mass) to the bonding resin of a commercial 3-step adhesive system. The mechanical properties of the adhesive layer, hybrid layer, and underlying dentin were measured by nanoindentation after 24 h and 6 and 12 mo of storage in simulated body fluid (SBF) and observed by scanning electron microscopy. Effectiveness of dental resin composites containing calcium orthophosphate particles on dentin remineralization.

To assess the remineralization of artificial dentin lesions achieved with composites containing dicalcium phosphate dihydrate (DCPD) particles. Composites (42 vol% inorganic fraction) were tested: barium glass (BG; control), DCPD or 1DCPD:1BG. Degree of conversion (DC) was measured by FTIR. Ca2+ release was monitored using inductively coupled plasma optical emission spectrometry and scanning electrochemical microscopy (n=3). Artificial lesions were restored and kept in simulated body fluid for 28 days. Remineralization was assessed by mineral-to-matrix ratio (MMR; n=5) and nanoindentation (n=6

Diagnosis and Managment of Maxillary Incisor with Vertical Root Fracture: A Clinical Report with Three-Year Follow-Up.

According to the American Association of Endodontists, "a 'true' vertical root fracture is defined as a complete or incomplete fracture initiated from the root at any level, usually directed buccolingually." Vertical root fracture (VRF) usually starts from an internal dentinal crack and develops over time, due to masticatory forces and occlusal loads. When they occur in teeth, those types of fractures can present difficulties in diagnosis, and there are however many clinic and radiographical signs which can guide clinicians to the existence of the fracture. Prognosis, most often, is hopeless,

Pulpal hemodynamics and interstitial fluid pressure: balance of transmicrovascular fluid transport.

The net rate of fluid movement across the microvascular endothelium is governed by hydrostatic and protein osmotic pressures operating on each side of the vessel wall. These basic forces regulating the pulpal transmicrovascular fluid exchange are the same as in other tissues. However, the pulp is special in regard to its enclosement between rigid dentin walls, implying low interstitial compliance. In addition, the pulp has a relatively low perfusion pressure. In the low compliant pulpal interstitium, even a modest rise in net filtration will favor a relatively large increase in interstitial fl

Technical note: Micro-computed tomography calibration using dental tissue for bone mineral research.

Computed tomography (CT) and microcomputed tomography (μCT) require calibration against density phantoms scanned with specimens or during routine internal calibration for assessment of mineral concentration (MC) and density. In clinical studies involving bone, alternative calibration methods using bodily tissues and fluids ("phantomless" calibration) have been suggested. However, such tissues are seldom available in archeological and osteological research. This study investigates the potential of dental tissue as internal reference for calibration of μCT scans, facilitating the analysis of bon

Analytical methods for biomarker-based diagnosis of pulpitis in minimally invasive biological fluids: a systematic review.

INTRODUCTION: Inflammatory biomarkers measured in minimally invasive biological fluids are promising objective tools for diagnosing pulpitis. However, the analytical methods used to detect and quantify these biomarkers, and their diagnostic performance, have not been critically evaluated. This review aimed to identify and assess the analytical methods for detecting and quantifying inflammatory biomarkers in gingival crevicular fluid, dentinal fluid and pulpal blood to distinguish healthy pulp from different stages of pulpitis. METHODS: This systematic review followed PRISMA 2020 guidelines an

Preliminary Evidence That Bioactive Cements Occlude Artificial Marginal Gaps.

PURPOSE: The objective of this preliminary laboratory study was to explore whether bioactive dental cements have the ability to seal marginal gaps as compared with other classes of dental cements. MATERIALS AND METHODS: Tooth discs (dentin/enamel) were positioned adjacent to a second disc of one of five possible dental cements (two bioactive and three conventional) and separated by a 50-μm Mylar film, creating a controlled gap between the two discs. The edges of the tooth segments were abraded to expose dentin such that the dentin interface was positioned opposite the cement. The artificial g

The Role of Transient Receptor Potential (TRP) Channels in the Transduction of Dental Pain.

Dental pain is a common health problem that negatively impacts the activities of daily living. Dentine hypersensitivity and pulpitis-associated pain are among the most common types of dental pain. Patients with these conditions feel pain upon exposure of the affected tooth to various external stimuli. However, the molecular mechanisms underlying dental pain, especially the transduction of external stimuli to electrical signals in the nerve, remain unclear. Numerous ion channels and receptors localized in the dental primary afferent neurons (DPAs) and odontoblasts have been implicated in the tr

Two new in vitro calcification systems showing the higher calcifiability of enamel proteins than dentin and bone matrices.

One of the difficulties in simulating in vivo calcification by in vitro experiments is how to prepare and apply a suitable calcifying solution. We have previously developed an entirely new model system consisting of 40% acrylamide gel blocks that contains matrix proteins and is immersed in fetal calf serum at 37 degrees C. (40% gel system) for 18 hr (Connect. Tissue Res., 33, 185, 1995). The gels were analyzed for immobilized calcium. In this system bovine enamel proteins (0.1% in the gel) showed the highest calcifiability among the tested matrices, followed by insoluble bovine dentin, bone an

Clinical observations of noncoping overdenture abutments protected by tannin-fluoride preparation.

STATEMENT OF PROBLEM: The exposed dentin of overdenture abutments can lead to caries. Therefore chemical protection is needed to prevent caries. Considering that dentin contains both organic and inorganic components, reinforcement of both components should give reasonable results. Tannin-fluoride preparation strengthens both the organic and inorganic components of teeth. PURPOSE: This study examined whether the tannin-fluoride preparation protects noncoping abutments. MATERIAL AND METHODS: Thirty-one subjects aged 36 to 86 years participated in the clinical trial. A trial autopolymerizing re
