Early-life stress at the time of the plague in Northern Italy (17th century): The case of two individuals with severe hypoplastic defects from Lazzaretto Vecchio (Venice).

OBJECTIVES: This study investigates two cases of severe enamel hypoplasia observed in individuals buried at Lazzaretto Vecchio (Venice, Italy) likely during the 1575-1630 plague outbreak. By integrating histological and geochemical evidence, we assess whether severe hypoplastic defects correspond to alterations in enamel formation dynamics or elemental composition and explore what such patterns may reveal about physiological disruption during crown development. DESIGN: A multi-method approach was applied, including morphological analysis for biological profile reconstruction, enamel histomorp

Melatonin Rescues Enamel Defects Induced by Maternal Circadian Disruption via Targeting the BMAL1-JNK3 Axis.

INTRODUCTION AND AIMS: Developmental defects of enamel (DDE) are highly prevalent, yet effective preventive measures remain elusive. The process of amelogenesis exhibits intrinsic circadian rhythmicity. While the photoperiod-regulated hormone melatonin is a recognized modulator of bone formation, its potential role in coordinating the circadian aspects of enamel biomineralization remains incompletely understood. This study aimed to decipher the impact of aberrant photoperiods on enamel development and to identify melatonin's regulatory targets. METHODS: We assessed the impact of maternal circ

Modified microabrasion protocol associated with resin infiltration. Two case reports.

Resin infiltration is an alternative for arresting and masking white spot lesions (WSLs). However, depending on their extent, depth, and activity, the voids within their body may not be completely filled by the infiltrant, leaving a certain degree of porosity inside the infiltrated lesion. The two case reports presented in this article aim to propose a modified microabrasion protocol for treating the enamel surface before resin infiltration to improve the esthetic outcome of teeth presenting inactive WSLs. The clinical procedures are described step by step based on data from the scientific lit

KMT2D Regulates Tooth Enamel Development.

Amelogenesis, the process of enamel formation, is tightly regulated and essential for producing the tooth enamel that protects teeth from decay and wear. Disruptions in amelogenesis can result in amelogenesis imperfecta, a group of genetic conditions characterized by defective enamel, including enamel hypoplasia, marked by thin or underdeveloped enamel. Mutations in the KMT2D (MLL4) gene, which encodes histone H3 lysine 4 methyltransferase, are associated with Kabuki syndrome, a developmental disorder that can involve dental anomalies such as enamel hypoplasia. However, the specific role of KM

Dental and Cranial Pathologies in Mice Lacking the Cl(-) /H(+) -Exchanger ClC-7.

ClC-7 is a 2Cl(-) /1H(+) -exchanger expressed at late endosomes and lysosomes, as well as the ruffled border of osteoclasts. ClC-7 deficiencies in mice and humans lead to impaired osteoclast function and therefore osteopetrosis. Failure of tooth eruption is also apparent in ClC-7 mutant animals, and this has been attributed to the osteoclast dysfunction and the subsequent defect in alveolar bone resorptive activity surrounding tooth roots. Ameloblasts also express ClC-7, and this study aims to determine the significance of ClC-7 in enamel formation by examining the dentitions of ClC-7 mutant m

Dental fluorosis in the European roe deer (Capreolus capreolus): A review of the pathological changes in the enamel of fluorotic cheek teeth and the abnormal pattern of dental wear in affected dentitions.

This article reviews the pathological changes in the enamel of permanent mandibular cheek teeth and their sequelae in European roe deer from regions polluted by anthropogenic fluoride emissions. The primary (developmental) changes of fluorotic roe deer enamel are hypomineralization and microstructural aberrations, including enamel hypoplasia. The hypomineralized fluorotic enamel is whitish-opaque on eruption and gets stained following tooth eruption. Moreover, it is prone to excessive wear and mechanical breakdown, resulting in posteruptive enamel lesions. These posteruptive lesions can morpho

Teeth as time-resolved perinatal archives: An integrated protocol for reconstructing early-life exposures associated with MIH.

INTRODUCTION/OBJECTIVES: Molar-incisor hypomineralization (MIH) is a highly prevalent developmental enamel defect with origins during perinatal life. Despite extensive research, its aetiology remains unclear, likely reflecting the interaction of multiple exposures during critical periods of enamel development. This study aims to establish an integrated exploratory framework for reconstructing biologically plausible early-life exposure profiles associated with MIH. METHODS: An exploratory observational case-control study will enrol 400 children aged 9-12 years, including 200 with MIH and 200 u

A probable genetic origin for pitting enamel hypoplasia on the molars of Paranthropus robustus.

We report the frequencies of linear enamel hypoplasia (LEH) and, specifically, pitting enamel hypoplasia (PEH) defects in the teeth of Paranthropus robustus, for comparison with four other South African hominin species and three extant nonhuman primate species. Unlike LEH, the lesser known PEH is characterized by multiple circular depression defects across a tooth crown and is often difficult to interpret in terms of developmental timing and etiology. Teeth in all samples were examined macroscopically with type, position and number of defects recorded. Frequencies of teeth with LEH vary among

Overexpression of ameloblastin in secretory ameloblasts results in demarcated, hypomineralized opacities in enamel.

Introduction: Developmental defects of the enamel manifest before tooth eruption and include amelogenesis imperfecta, a rare disease of underlying gene mutations, and molar-incisor hypomineralization (MIH), a prevalent disease in children originating from environmental and epigenetic factors. MIH enamel presents as the abnormal enamel marked by loss of translucency, demarcation between the healthy and affected enamel, and reduced mineral content. The pathophysiology of opaque, demarcated enamel lesions is not understood; however, the retention of enamel proteins in the matrix has been suggeste

Dental enamel defect diagnosis through different technology-based devices.

INTRODUCTION: Dental enamel defects (DEDs) are faulty or deficient enamel formations of primary and permanent teeth. Changes during tooth development result in hypoplasia (a quantitative defect) and/or hypomineralisation (a qualitative defect). OBJECTIVE: To compare technology-based diagnostic methods for detecting DEDs. MATERIAL AND METHODS: Two-hundred and nine dental surfaces of anterior permanent teeth were selected in patients, 6-11 years of age, with cleft lip with/without cleft palate. First, a conventional clinical examination was conducted according to the modified Developmental Def

Osseous repair of a lateral periodontal cyst.

THE LATERAL PERIODONTAL CYSTS is a slow-growing radiolucent, developmental lesion occurring most frequently in males during the sixth decade. As part of the differential diagnosis, it must be distinguished from the collateral keratocyst and the gingival cyst of adults as well as other entities. Speculation remains as to the lateral periodontal cyst's developmental origin. Whether it is from reduced enamel epithelium, remnants of dental lamina, or cell rests of Malassez remains to be determined. The following longitudinal case report describes the review of literature and clinical and histologi

The genetic basis of inherited anomalies of the teeth. Part 2: syndromes with significant dental involvement.

Teeth are specialized structural components of the craniofacial skeleton. Developmental defects occur either alone or in combination with other birth defects. In this paper, we review the dental anomalies in several multiple congenital anomaly (MCA) syndromes, in which the dental component is pivotal in the recognition of the phenotype and/or the molecular basis of the disorder is known. We will consider successively syndromic forms of amelogenesis imperfecta or enamel defects, dentinogenesis imperfecta (i.e. osteogenesis imperfecta) and other dentine anomalies. Focusing on dental aspects, we

Prenatal exposure to environmental toxins and comprehensive dental findings in a population cohort of children.

Environmental toxins are known to have many impacts on growth and development in humans, starting in utero. Alterations in amelogenesis, caused by chemical and physical trauma that occur during the antenatal, perinatal and postnatal time periods, may result in developmental defects in deciduous and permanent tooth enamel, as demonstrated in animal studies. These defects can be clinically visible and result in a variety of morphological and functional problems in the dentition. Since enamel does not remodel after formation, it may serve as a permanent record of insults during organ development.

Is Italian Dentists' Knowledge of Enamel Development Defects Adequate? A Nationwide Survey.

OBJECTIVES: Correct identification and management of Developmental Defects of Enamel (DDEs) are essential to provide the best possible treatment. The present survey aims to investigate Italian dentists' knowledge of DDEs, their ability to recognise the different clinical pictures, and to choose the most appropriate clinical approach. METHODS: A cross-sectional survey was planned based on a questionnaire including 27 closed-ended questions, and that proposed 4 clinical pictures, molar incisor hypomineralisation (MIH), amelogenesis imperfecta (AI), dental fluorosis (DF), and an initial caries l

Nonsyphilitic occurrence of mulberry molars: A rare case report.

Developmental defects of enamel may range from slight abnormalities of the tooth's color, shape and size to a complete absence of the enamel. Permanent first molar may show large variations in their morphological features and forms. Such changes may be in the form of anomalous cusps or variations in the occlusal surface showing globular shaped additional cusps and also increased in number of roots. However, to the best of our knowledge, this is the first nonsyphilitic case with the occurrence of multiple globular cusps. In the present case, occlusal anatomy is abnormal, with numerous disorgani

Essentiality of early diagnosis of molar incisor hypomineralization in children and review of its clinical presentation, etiology and management.

Molar incisor hypomineralization (MIH) is a common developmental condition resulting in enamel defects in first permanent molars and permanent incisors. It presents at eruption of these teeth. One to four molars, and often also the incisors, could be affected. Since first recognized, the condition has been puzzling and interpreted as a distinct phenomenon unlike other enamel disturbances. Early diagnosis is essential since, rapid breakdown of tooth structure may occur, giving rise to acute symptoms and complicated treatment. The purpose of this article is to review MIH and illustrate its diagn

Developmental defects of the dental crown in chimpanzees from the Taï National Park, Côte D'Ivoire: coronal waisting.

We describe a developmental defect that manifests as a mild constricted "waist" in anterior teeth from seven of nine chimpanzee individuals from Taï National Park, Côte D'Ivoire. The sample consists of 21 canine teeth and one incisor, imaged in profile with a digital microscope. Twelve teeth are affected. The waist develops during tooth formation as an external, encircling depression in the contour of the outer enamel surface, more easily seen labially. It is not a thinning of enamel per se, but rather a slight decrement in dentinal crown volume, shown in microCT scans as a change in contour o
