Determination of haloacetic acids in municipal tap water and swimming pool water using portable capillary liquid chromatography - mass spectrometry.

In this study, a compact capillary liquid chromatography (capLC) system (<10 Kg) has been coupled with two small-footprint and portable single quadrupole mass spectrometers (<35 Kg) for the analysis of haloacetic acids (HAAs) in water samples. The portable capillary liquid chromatograph was equipped with a polar-modified C18 column (100 × 0.5 mm, particle size: 3 μm) and utilised gradient elution with 0.05 % formic acid and methanol to selectively separate and quantify the 9 HAAs (HAA9) recommended for monitoring by United States Environmental Protection Agency (USEPA). The successful quantifi

Formation of disinfection by-products during chlorination of organic matter from phoenix tree leaves and Chlorella vulgaris.

To better understand the precursor of disinfection by-products (DBPs) and provide useful information for water utilities to manage the drinking water, a study of DBP formation was conducted through chlorination of leaf organic matter (OM) from phoenix tree and algal OM from Chlorella vulgaris. DBPs investigated include trichloromethane (TCM), trichloroacetic acid (TCAA), dichloroacetic acid (DCAA), chloroacetic acid (CAA), dichloroacetonitrile (DCAN) and trichloroacetonitrile (TCNM). Results show that the specific yields (μg/mg C) of C-DBPs (TCM, CAA, DCAA and TCAA) from leaf OM were higher bu

[Formation of disinfection by-products: temperature effect and kinetic modeling].

Water temperature has significant effects on the disinfection by-product (DBP) formation and concentration in many water utilities and distribution systems. To study the temperature effect on the DBP concentration, the uniform formation condition (UFC) test was referred in testing the formation concentration of DBPs [including (trihalomethanes) THMs and (haloacetic acids) HAAs] at different temperatures during chlorination of the humic acid (HA) solution. A kinetic model was consequently proposed to predict DBP concentration during chlorination. Results show that for the three detected DBPs, i

Species differences in response to trichloroethylene. II. Biotransformation in rats and mice.

Detailed analysis of urine from two strains of rats and mice dosed po with trichloroethylene at four doses from 10 to 2000 mg/kg failed to detect any major species or strain differences in the metabolism of trichloroethylene. Although a greater proportion of the dose was metabolized in mice than in rats, the relative proportions of the major metabolites were very similar in both strains and were unaffected by the dose amount. Analysis of the same urine samples for minor metabolites failed to establish a major species difference. Small amounts of dichloroacetic acid (less than 1% of the dose) w

Blocking Lysine Crotonylation and Aerobic Glycolysis as Targeting Strategy Against mpox Virus Replication.

The global outbreak of mpox caused by the mpox virus (MPXV) in 2022 and 2024 underscores the urgent need to elucidate mechanisms governing viral replication during pathogenesis. Metabolic reprogramming is a conserved hallmark of viral infections, however, the precise mechanisms by which MPXV manipulates host cell metabolism remain unknown. Here, it is demonstrated that MPXV hijacks aerobic glycolysis via lysine crotonylation of its I3 protein, which is essential for MPXV replication. Mechanistically, MYST histone acetyltransferase 1 (MYST1), an acetyltransferase upregulated by MPXV, binds to a

Size-exclusion chromatography of poly(ethylene 2,6-naphthalate).

A solvent mixture of 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and dichloroacetic acid (DCAA) is used to dissolve difficultly soluble poly(ethylene 2,6-naphthalate) (PEN). Solutions can be diluted and analyzed in a common size-exclusion chromatography (SEC) eluent, HFIP. The HFIP/DCAA mixture is better at dissolving PEN than either solvent individually and it is easier and safer to work with than phenolic and strongly acidic eluents. Dissolution temperatures between 50 and 60 °C are sufficiently low to minimize hydrolytic degradation of the polyester. PEN does not dissolve in the solvent mixtur

Formation and interdependence of disinfection byproducts during chlorination of natural organic matter in a conventional drinking water treatment plant.

It is crucial to explore the source, formation process and interdependence of disinfection byproducts (DBPs) to reduce their risk on public health. In this investigation, a source water was chlorinated to evaluate the initial formation rates and the maximum yields of trichloromethane (TCM), dichloroacetic acid (DCAA), and trichloroacetic acid (TCAA) based on a hyperbola model. The results showed that TCM achieved the highest initial formation rate and maximum theoretical concentration compared with DCAA and TCAA. The TCM yield can be used to forecast the yields of DCAA and TCAA throughout the

A subchronic exposure to trichloroethylene causes lipid peroxidation and hepatocellular proliferation in male B6C3F1 mouse liver.

The common groundwater contaminant trichloroethylene (TCE), when given by oral gavage, can produce free radical species during metabolism. Furthermore, TCE end-stage metabolites, trichloroacetic acid and dichloroacetic acid, cause lipid peroxidation in mouse liver. The time courses of lipid peroxidation, free radical generation, and 8-hydroxydeoxyguanosine (8OHdG) formation were used to assess the level of oxidative stress in the liver of B6C3F1 mice dosed orally once daily, 5 days a week for 8 weeks at 0, 400, 800, and 1200 mg/kg TCE in corn oil. Peroxisomal proliferation, cell proliferation,

Trichloroethylene oxidation by purified toluene 2-monooxygenase: products, kinetics, and turnover-dependent inactivation.

Trichloroethylene is oxidized by several types of nonspecific bacterial oxygenases. Toluene 2-monooxygenase from Burkholderia cepacia G4 is implicated in trichloroethylene oxidation and is uniquely suggested to be resistant to turnover-dependent inactivation in vivo. In this work, the oxidation of trichloroethylene was studied with purified toluene 2-monooxygenase. All three purified toluene 2-monooxygenase protein components and NADH were required to reconstitute full trichloroethylene oxidation activity in vitro. The apparent Km and Vmax were 12 microM and 37 nmol per min per mg of hydroxyla

Functional diagnostics in mitochondrial diseases.

Mitochondrial diseases (MD) with respiratory chain defects are caused by genetic mutations that determine an impairment of the electron transport chain functioning. Diagnosis often requires a complex approach with measurements of serum lactate, magnetic resonance spectroscopy (MRS), muscle histology and ultrastructure, enzymology, genetic analysis, and exercise testing. The ubiquitous distribution of the mitochondria in the human body explains the multiple organ involvement. Exercise intolerance is a common symptom of MD, due to increased dependence of skeletal muscle on anaerobic metabolism,

Bioactivation of tetrachloroethylene. Role of glutathione S-transferase-catalyzed conjugation versus cytochrome P-450-dependent phospholipid alkylation.

The metabolism of [14C]tetrachloroethylene (Tetra) and its metabolite S-(1,2,2-trichlorovinyl)-L-cysteine (TCVC) was investigated with in vitro systems to substantiate metabolic pathways of Tetra deduced from in vivo experiments. In the presence of NADPH, rat hepatic microsomal fractions metabolized Tetra to soluble metabolites, which were identified as trichloroacetic acid and oxalic acid by gas chromatography/mass spectroscopy and a metabolite largely bound to microsomal macromolecules. The majority of the alkylated macromolecules were identified as N-trichloroacetylated phospholipids by hig

Platelet mitochondrial evaluation during cytochrome c and dichloroacetate treatments of MELAS.

We hypothesized that serial changes in platelet (PLT) mitochondrial enzyme (ME) activities might correspond to the effects of medications for mitochondrial encephalomyopathy and stroke-like episodes (MELAS). Cytochrome c and sodium dichloroacetate (DCA) were given to a 7-year-old girl with MELAS who had an A3243G mitochondrial DNA mutation. The effects were evaluated with whole PLT-ME assays, developed by our group, using a microplate-reader. During cytochrome c treatment, complex II+III (II+III), complex IV (IV) and citrate synthase (CS) activities showed gradual but statistically significant

Combined aerobic training and dichloroacetate improve exercise capacity and indices of aerobic metabolism in muscle cytochrome oxidase deficiency.

There is no generally effective therapy for mitochondrial myopathies. In this study, we measured responses to combined aerobic training and oral dichloroacetate (DCA) therapy in a 25-year-old woman with a mitochondrial myopathy caused by cytochrome oxidase deficiency. The patient trained for 14 weeks, and DCA therapy was begun after 8 weeks. Independent indices of aerobic capacity and oxidative metabolism showed substantial improvement. Venous lactate concentrations at rest, and after a constant amount of work, decreased by approximately 50% after 8 weeks of aerobic training, and by more than

Have animal data been used inappropriately to estimate risks to humans from environmental trichloroethylene?

Trichloroethylene (TCE) is widely viewed as an environmental hazard. Its major metabolite, chloral hydrate, is a currently used medicine. Regulation of TCE is based on a linear extrapolation from effects of high doses in rodents to risks for humans at low doses. However, metabolic, toxicologic, and epidemiologic data on trichloroethylene and chloral hydrate as well as water chlorination studies call this approach into question. The mechanism of carcinogenesis of TCE and chloral hydrate in rodents is nonlinear: very high doses, sufficient to cause cellular necrosis, are necessary. Malignancy ar

Effect of dichloroacetate on PaCO2 responses to hypoxia in awake goats.

To gain insight into the role of cerebral lactic acidosis in the hypoxic ventilatory response, we administered dichloroacetate (DCA) intravenously to inhibit lactic acid production in 7 awake goats (40-70 kg) during 0.5 h of normoxia (inspired O2 fraction = 0.209) and 5 h of poikilocapnic hypoxia (inspired O2 fraction = 0.125). On separate days, these goats were also studied with a continuous saline infusion (18 ml/h iv) during 5 h of normoxia and hypoxia. Arterial PCO2 (PaCO2) did not change during the 5-h normoxic period. During hypoxia, arterial PO2 fell significantly (P < 0.05) with both s

A partition coefficient determination method for nonvolatile chemicals in biological tissues.

Partition or distribution coefficients are critical elements in efforts designed to describe the uptake, distribution, biotransformation, and excretion of organic chemicals in biological systems. In order to estimate the partition coefficients needed to describe the biological distribution of low-volatility compounds, an experimental method was developed to measure partitioning of nonvolatile compounds into biological tissues. Blood, fat, muscle, liver, and skin were individually incubated in a saline solution containing the chemical of interest. Each sample was centrifuged and 2.0 ml of the s

Assessment of genotoxic chemicals using chemogenomic profiling based on gene-knockout library in Saccharomyces cerevisiae.

Understanding the adverse effects of genotoxic chemicals and identifying them effectively from non-genotoxic chemicals are of great worldwide concerns. Here, Saccharomyces cerevisiae (yeast) genome-wide single-gene knockout screening approach was conducted to assess two genotoxic chemicals (4-nitroquinoline-1-oxide (4-NQO) and formaldehyde (FA)) and environmental pollutant dichloroacetic acid (DCA, genotoxicity is controversial). DNA repair was significant enriched in the gene ontology (GO) biology process (BP) terms and KEGG pathways when exposed to low concentrations of 4-NQO and FA. Higher

Chlorination of oxybenzone: Kinetics, transformation, disinfection byproducts formation, and genotoxicity changes.

UV filters are a kind of emerging contaminant, and their transformation behavior in water treatment processes has aroused great concern. In particular, toxic products might be produced during reaction with disinfectants during the disinfection process. As one of the most widely used UV filters, oxybenzone has received significant attention, because its transformation and toxicity changes during chlorine oxidation are a concern. In our study, the reaction between oxybenzone and chlorine followed pseudo-first-order and second-order kinetics. Three transformation products were detected by LC-MS/M

Mice deficient in glutathione transferase zeta/maleylacetoacetate isomerase exhibit a range of pathological changes and elevated expression of alpha, mu, and pi class glutathione transferases.

Glutathione transferase zeta (GSTZ1-1) is the major enzyme that catalyzes the metabolism of alpha-halo acids such as dichloroacetic acid, a carcinogenic contaminant of chlorinated water. GSTZ1-1 is identical with maleylacetoacetate isomerase, which catalyzes the penultimate step in the catabolic pathways for phenylalanine and tyrosine. In this study we have deleted the Gstz1 gene in BALB/c mice and characterized their phenotype. Gstz1(-/-) mice do not have demonstrable activity with maleylacetone and alpha-halo acid substrates, and other GSTs do not compensate for the loss of this enzyme. When
