Intracellular Vesicle Transport Impairment as a Candidate Systems-Level Bottleneck in Chronic Diabetic Foot Ulcers: Network Medicine Identifies KIF13A as a Potential Therapeutic Vulnerability.

Background: Growth factor therapy often fails in diabetic foot ulcers (DFUs). The reason remains unclear. Standard differential expression analysis may miss functionally critical genes with modest expression changes. Methods: We performed a secondary computational analysis of a longitudinal DFU transcriptomic dataset (Dryad; 17 patients, 117 serial biopsy samples, 12-week follow-up). Co-expression networks were built separately for healed (n = 37) and non-healed (n = 80) samples. Virtual gene knockout (VGK) was used to rank genes by topological impact on network cohesion. Single-cell analysis

Can local vibration alter the contribution of persistent inward currents to human motoneuron firing?

The response of spinal motoneurons to synaptic input greatly depends on the activation of persistent inward currents (PICs), which in turn are enhanced by the neuromodulators serotonin and noradrenaline. Local vibration (LV) induces excitatory Ia input onto motoneurons and may alter neuromodulatory inputs. Therefore, we investigated whether LV influences the contribution of PICs to motoneuron firing. This was assessed in voluntary contractions with concurrent, ongoing LV, as well as after a bout of prolonged LV. High-density surface electromyograms (HD-EMG) of the tibialis anterior were record

Visual Perception and Reading: New Clues to Patterns of Dysfunction Across Multiple Visual Channels in Developmental Dyslexia.

PURPOSE: The specificity of visual channel impairment in dyslexia has been the subject of much controversy. The purpose of this study was to determine if a differential pattern of impairment can be verified between visual channels in children with developmental dyslexia, and in particular, if the pattern of deficits is more conspicuous in tasks where the magnocellular-dorsal system recruitment prevails. Additionally, we also aimed at investigating the association between visual perception thresholds and reading. METHODS: In the present case-control study, we compared perception thresholds of

Modeling the suppression dynamics of Aedes mosquitoes with mating inhomogeneity.

A novel strategy for controlling mosquito-borne diseases, such as dengue, malaria and Zika, involves releases of Wolbachia-infected mosquitoes as Wolbachia cause early embryo death when an infected male mates with an uninfected female. In this work, we introduce a delay differential equation model with mating inhomogeneity to discuss mosquito population suppression based on Wolbachia. Our analyses show that the wild mosquitoes could be eliminated if either the adult mortality rate exceeds the threshold [Formula: see text] or the release amount exceeds the threshold [Formula: see text] uniforml

Models of bovine babesiosis including juvenile cattle.

Bovine Babesiosis in cattle is caused by the transmission of protozoa of Babesia spp. by ticks as vectors. Juvenile cattle (<9 months of age) have resistance to Bovine Babesiosis, rarely show symptoms, and acquire immunity upon recovery. Susceptibility to the disease varies between breeds of cattle. Models of the dynamics of Bovine Babesiosis transmitted by the cattle tick that include these factors are formulated as systems of ordinary differential equations. Basic reproduction numbers are calculated, and it is proved that if these numbers are below the threshold value of one, then Bovine Bab

Accuracy of the central vein sign in diagnosis of multiple sclerosis: a systematic review and meta-analysis.

BACKGROUND: The central vein sign (CVS) is a promising imaging marker of multiple sclerosis (MS). We performed a systematic review and meta-analysis to evaluate the diagnostic accuracy of CVS-based rules in the differential diagnosis of MS and to identify the best cutoffs for these rules. METHODS: PubMed, Embase, and Scopus were systematically searched for available evidence. Data extracted were entered into Bayesian models recommended by the Cochrane network. Summary sensitivity and specificity of CVS-based diagnostic rules across different positivity thresholds were calculated. A meta-regre

A mathematical model of Cheyne-Stokes or periodic breathing.

Cheyne-Stokes Breathing is a periodic cycle of apnea followed by hyperventilation. A theory of this phenomenon is developed based on a minimal set of physiological assumptions. The rate of loss of CO2 from venous blood is proportional to the CO2 concentration in the lungs times the respiration rate; the respiration rate is a linear function of arterial CO2 concentration above a threshold, and zero below that threshold. A time delay between blood in lungs and respiratory response allows the system to go into oscillation. These assumptions lead to a single nonanalytic delay-differential equation

Non-invasive testing to diagnose the underlying auditory pathology of hearing impairment.

Of the three major classifications of inner ear pathology, sensory, neural and metabolic, it is relatively easy to distinguish a sensory pathology from a neural or inner hair cell pathology using current clinical tools. Deficiencies in both distortion product otoacoustic emissions (DPOAE) and auditory brainstem responses (ABR) suggest a sensory pathology involving outer hair cells. Normal DPOAEs with deficiencies in ABRs suggest a neural pathology, involving inner hair cells, auditory nerve synapses or neural deficiencies. However, there are currently no non-invasive methods to distinguish a m

Different TMS patterns of intracortical inhibition in early onset Alzheimer dementia and frontotemporal dementia.

OBJECTIVE: To investigate putative changes in cortical excitability of patients affected by early-onset mild dementia by means of transcranial magnetic stimulation (TMS) and to verify whether a peculiar neurophysiological profile may contribute to characterise Alzheimer's disease (AD) vs frontotemporal dementia (FTD). METHODS: Motor threshold and intracortical inhibition (ICI) and facilitation (ICF) after paired-pulse TMS (inter-stimulus intervals from 1 to 20 ms) were studied in two groups of early-onset demented patients with a neuropsychological profile suggestive of AD (n = 12) and FTD (n

A Low-Complexity Hall-Based Measurement System Implementing a Dark/Illuminated Differential Estimator for Majority-Carrier Photoconductive Screening in Doped n-Type Silicon.

A low-complexity Hall-based measurement system implementing a dark/illuminated differential estimator of the majority-carrier photoconductive response is assessed on n-type crystalline silicon wafers with contrasting doping concentrations. The setup combines a Van der Pauw configuration, permanent neodymium magnets, a characterized white LED source, and a source-measure unit to extract an apparent Hall-derived differential indicator-not a calibrated majority-carrier density-under dark and illuminated steady-state conditions. At 1.81mWcm-2, the lightly doped wafer (ND≈1014cm-3) shows a resolved

The Prevalence of Microembolic Signals in Transcranial Doppler Sonography With Bubble Test in Acute Ischemic Stroke.

OBJECTIVES: Transcranial Doppler ultrasound (TCD) is noninvasive and highly sensitive and specific for the diagnosis of patent foramen ovale (PFO). We evaluated the diagnostic implications of the TCD with a saline agitation test as a routine work-up for ischemic stroke patients. METHODS: A TCD bubble study was performed in all consecutive ischemic stroke patients as a routine work-up. We evaluated the prevalence of microembolic signals (MES) for each stroke etiology and the optimal number of MES for predicting the PFO-attributable stroke. RESULTS: Subjects (N = 499) with acute ischemic strok

Population dynamics of live-attenuated virus vaccines.

Viruses contained in live-attenuated virus vaccines (LAVV) can be transmitted between individuals, resulting in secondary or contact vaccinations. This fact has been exploited successfully in the use of the Oral Polio Vaccine (OPV) to better control wild-type polio viruses. In this work we analyze general LAVV vaccination models for infections that confer lifelong immunity. We consider both standard (continuous) vaccination strategies and pulse vaccination programs (where mass vaccination is carried out at regular intervals). For continuous vaccination, we provide a complete global analysis of

[Fibromyalgia and Sjögren syndrome--clinical and methodological aspects].

The prevalence of fibromyalgia in primary (n = 18) or secondary (n = 20) Sjögren's syndrome was examined. In all patients with Sjögren's syndrome as well as in 31 fibromyalgia patients and 20 healthy individuals measurements of pressure pain threshold were done by palpation and dolorimetry. Widespread pain, functional complaints, as well as depression were determined by a questionnaire. The results show a high frequency of fibromyalgia in the group of patients with pSS (44%), 72% of pSS patients reported widespread pain. Only 5% of the sSS patients fulfilled the ACR criteria for fibromyalgia a

Tactile and proprioceptive dysfunction differentiates cervical dystonia with and without tremor.

OBJECTIVE: To determine whether different phenotypes of cervical dystonia (CD) express different types and levels of somatosensory impairment. METHODS: We assessed somatosensory function in patients with CD with and without tremor (n = 12 each) and in healthy age-matched controls (n = 22) by measuring tactile temporal discrimination thresholds of the nondystonic forearm and proprioceptive acuity in both the dystonic (head/neck) and nondystonic body segments (forearm/hand) using a joint position-matching task. The head or the wrist was passively displaced along different axes to distinct joint

Temporal predictions based on a gradual change in tempo.

Previous studies investigating sensitivity to step changes in tempo and prediction of tone onset time have generally utilized isochronous sequences. This study investigates subjects' ability to detect deviations from a gradual change in the tempo of a tone sequence (experiment 1) and their judgment of the perceptually optimal timing of this tone (experiment 2). In experiment 1, inter-onset-intervals within pairs of eight-tone sequences followed a geometric progression to create a gradual tempo change. In one sequence, the final tone was presented either earlier or later than specified by the p

The thresholds of change model: an approach to analyzing stages of change data.

Stage models are prominent in research describing health behavior change. Since stage models often propose that different factors have varying influences on membership in the different stage, statistical methods that can estimate the thresholds that separate the stages and the relative value of variables in influencing these thresholds are useful. This article describes use of a "thresholds of change" model for analyzing the thresholds separating stages and specifically for examining the effects of explanatory variables on these thresholds using a generalization of an ordinal logistic (or prob

Protein engineering reveals that gasdermin A preferentially targets mitochondrial membranes over the plasma membrane during pyroptosis.

When activated, gasdermin family members are thought to be pore-forming proteins that cause lytic cell death. Despite this, numerous studies have suggested that the threshold for lytic cell death is dependent on which gasdermin family member is activated. Determination of the propensity of various gasdermin family members to cause pyroptosis has been handicapped by the fact that for many of them, the mechanisms and timing of their activation are uncertain. In this article, we exploit the recently discovered exosite-mediated recognition of gasdermin D (GSDMD) by the inflammatory caspases to dev

Influence of pulse duration on ultrashort laser pulse ablation of biological tissues.

Ablation characteristics of ultrashort laser pulses were investigated for pulse durations in the range of 130 fs-10 ps. Tissue samples used in the study were dental hard tissue (dentin) and water. We observed differences in ablation crater morphology for craters generated with pulse durations in the 130 fs-1 ps and the 5 ps-10 ps range. For the water experiment, the surface ablation and subsequent propagation of stress waves were monitored using Mach-Zehnder interferometry. For 130 fs-1 ps, energy is deposited on the surface while for longer pulses the beam penetrates into the sample. Both stu
