Analysis of Gambierdiscus transcriptome data supports ancient origins of mixotrophic pathways in dinoflagellates.

Toxic dinoflagellates pose serious threats to human health and to fisheries. The genus Gambierdiscus is significant in this respect because its members produce ciguatoxin that accumulates in predominantly tropical marine food webs and leads to ciguatera fish poisoning. Understanding the biology of toxic dinoflagellates is crucial to developing control strategies. To this end, we generated a de novo transcriptome library from G. caribaeus and studied its growth under different culture conditions to elucidate pathways of carbon (C) and nitrogen (N) utilization. We also gathered available dinofla

New insights into carbon acquisition and exchanges within the coral-dinoflagellate symbiosis under NH4+ and NO3- supply.

Anthropogenic nutrient enrichment affects the biogeochemical cycles and nutrient stoichiometry of coastal ecosystems and is often associated with coral reef decline. However, the mechanisms by which dissolved inorganic nutrients, and especially nitrogen forms (ammonium versus nitrate) can disturb the association between corals and their symbiotic algae are subject to controversial debate. Here, we investigated the coral response to varying N : P ratios, with nitrate or ammonium as a nitrogen source. We showed significant differences in the carbon acquisition by the symbionts and its allocation

Ecology of the ciguatera causing dinoflagellates from the Northern Great Barrier Reef: changes in community distribution and coastal eutrophication.

Ciguatera fish poisoning (CFP) is known to be caused by the ciguatoxins from the dinoflagellate genus Gambierdiscus, however, there is the potential for other toxins such as okadaic acid and dinophysistoxins from the genus Prorocentrum, and palytoxin from the genus Ostreopsis, to contaminate seafood. These genera may also be indicators of ecosystem health and potentially impact on coral reef ecosystems and the role they may play in the succession of coral to macroalgae dominated reefs has not been researched. Sixteen GBR field sites spanning inshore, mid-lagoon and outer lagoon (offshore) regi

Neurotoxic shellfish poisoning.

Neurotoxic shellfish poisoning (NSP) is caused by consumption of molluscan shellfish contaminated with brevetoxins primarily produced by the dinoflagellate, Karenia brevis. Blooms of K. brevis, called Florida red tide, occur frequently along the Gulf of Mexico. Many shellfish beds in the US (and other nations) are routinely monitored for presence of K. brevis and other brevetoxin-producing organisms. As a result, few NSP cases are reported annually from the US. However, infrequent larger outbreaks do occur. Cases are usually associated with recreationally-harvested shellfish collected during o

An overview of the marine food poisoning in Mexico.

In the course of the last decade, huge events related to harmful algal blooms (HAB) have severely affected the environment in Mexico, even causing several human casualties. The tally of the toxins known up to date in Mexican waters includes: neurotoxin shellfish poisoning (NSP), paralytic shellfish poisoning (PSP), amnesic shellfish poisoning (ASP), tetrodotoxin (TTX) or puffer fish poisoning, ciguatera fish poisoning (CFP) and diarrhetic shellfish poisoning (DSP). Actual epidemiological figures profoundly modified the trends manifested on previous decades. Notwithstanding that the red tides a

Characteristics of a Dinophysis cf acuminata Population from a Tidewater Glacier Lagoon in a Temperate Latitude: Applications to Dinophysis Studies.

Dinophysis acuminata, the main agent of diarrhetic shellfish poisoning (DSP) worldwide, shows a high variability in morphology and toxin content between strains from contrasting habitats. Most frequent uncertainties in morphological discrimination are within the "D. acuminata complex", but confusion with other species (e.g., D. norvegica, D. fortii) also occurs. Here we describe a unique PTX2-containing population of Dinophysis cf acuminata observed during opportunistic samplings in San Rafael Lagoon (Chilean Patagonia), the only tidewater glacier lagoon remaining in the glacier with the world

A novel glutathione S-transferase Gtt2 class from the marine toxic dinoflagellate Alexandrium pacificum and its roles in algicide exposures.

In the present study, we identified a novel cytosolic glutathione S-transferase (GST) gene, Gtt2, from the marine dinoflagellate Alexandrium pacificum and evaluated its transcriptional responses to six toxic chemicals. The full-length cDNA sequence of ApGtt2 was 1306 base pairs (bp), encoding 289 amino acids with a predicted molecular weight of 32.76 kDa. The genomic coding region of the gene contained a 785-bp intron. Bioinformatics analysis revealed that ApGtt2 has a thioredoxin fold domain at the N-terminus, showing a characteristic feature of cytoplasmic GSTs. Phylogenetic analysis reveale

A catastrophic marine mortality event caused by a complex algal bloom including the brevetoxin producer Karenia cristata.

Harmful algal blooms of Karenia brevis (Dinophyceae) are a global anomaly, occurring in one location worldwide, causing severe marine and acute human impacts via brevetoxins (BTXs). During 2025 an unprecedented mass marine mortality occurred in South Australia, across an area of ~20,000 km2, persisting for >12 months, resulting in the deaths of ~106 marine animals of >600 taxa, with human health impacts. Using custom metabarcoding, long-read sequencing and targeted qPCR, we characterized the microalgal assemblage. Karenia cristata dominated over the sampling area, in an assemblage with four ot

First record of Prorocentrum caipirignum in Rapa Nui (Easter Island) and its production of okadaic acid: Implications for DSP risk in the southeastern Pacific.

The genus Prorocentrum comprises several benthic dinoflagellate species known to produce diarrhetic shellfish poisoning (DSP) toxins. However, information on their distribution and toxic potential remains limited across large areas of the Polynesian region. In this study, a clonal strain D008-7, isolated from intertidal tide pools in Rapa Nui (Easter Island), was analyzed using an integrative approach combining morphological observations, molecular phylogenetic analyses, and toxicological characterization by LC-MS/MS. Morphological and molecular results conclusively identified the strain as P.

Saxitoxin: A Comprehensive Review of Its History, Structure, Toxicology, Biosynthesis, Detection, and Preventive Implications.

Saxitoxin (STX) is a potent toxin produced by marine dinoflagellates and freshwater or brackish water cyanobacteria, and is a member of the paralytic shellfish toxins (PSTs). As a highly specific blocker of voltage-gated sodium channels (NaVs), STX blocks sodium ion influx, thereby inhibiting nerve impulse transmission and leading to systemic physiological dysfunctions in the nervous, respiratory, cardiovascular, and digestive systems. Severe exposure can lead to paralysis, respiratory failure, and mortality. STX primarily enters the human body through the consumption of contaminated shellfish

Ultrastructural characterization of Amyloodinium ocellatum (Dinoflagellata): Insights into trophont morphology and parasitic mode.

Amyloodinium ocellatum is a parasitic dinoflagellate that infects marine fish in tropical and subtropical regions, causing significant economic losses in aquaculture. This study used light microscopy (LM), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and electron tomography (ET) to comprehensively characterize the morphology, ultrastructure, and specialized attachment apparatus of A. ocellatum trophonts. The trophonts primarily parasitized the gill filaments of infected fish, reaching 89.8 ± 10.3 μm after 48 h of infection. Under LM, trophonts appeared as dark, s

Deeper insight into Gambierdiscus polynesiensis toxin production relies on specific optimization of high-performance liquid chromatography-high resolution mass spectrometry.

Ciguatera food poisoning affects consumer health and fisheries' economies worldwide in tropical zones, and specifically in the Pacific area. The wide variety of ciguatoxins bio-accumulated in fish or shellfish responsible for this neurological illness are produced by marine dinoflagellates of the genus Gambierdiscus and bio-transformed through the food web. The evaluation of the contents of ciguatoxins in strains of Gambierdiscus relies on the availability of standards and on the development of sensitive and specific tools to detect them. There is a need for sensitive methods for the analysis

Ciguatera Fish Poisoning: The Risk from an Aotearoa/New Zealand Perspective.

Gambierdiscus and Fukuyoa species have been identified in Aotearoa/New Zealand's coastal waters and G.polynesiensis, a known producer of ciguatoxins, has been isolated from Rangitāhua/Kermadec Islands (a New Zealand territory). The warming of the Tasman Sea and the waters around New Zealand's northern subtropical coastline heighten the risk of Gambierdiscus proliferating in New Zealand. If this occurs, the risk of ciguatera fish poisoning due to consumption of locally caught fish will increase. Research, including the development and testing of sampling methods, molecular assays, and chemical

First Report of Paralytic Shellfish Toxins in Marine Invertebrates and Fish in Spain.

A paralytic shellfish poisoning (PSP) episode developed in summer 2018 in the Rías Baixas (Galicia, NW Spain). The outbreak was associated with an unprecedentedly intense and long-lasting harmful algal bloom (HAB) (~one month) caused by the dinoflagellate Alexandrium minutum. Paralytic shellfish toxins (PSTs) were analyzed in extracts of 45 A. minutum strains isolated from the bloom by high-performance liquid chromatography with post-column oxidation and fluorescence detection (HPLC-PCOX-FLD). PSTs were also evaluated in tissues from marine fauna (invertebrates and fish) collected during the e

Grazer-induced bioluminescence gives dinoflagellates a competitive edge.

Bioluminescent dinoflagellates grow at one third the rate of their competitors of equivalent size, such as diatoms [1]. Despite this disadvantage, dinoflagellates successfully persist within phytoplankton communities and even form large blooms during favourable conditions. One explanation for this paradox is that bioluminescence acts as a defence that reduces losses to zooplankton grazers, such as copepods [2,3]. Lindström et al.[4] found that the dinoflagellate Lingulodinium polyedra (F.Stein) J.D.Dodge increase their bioluminescence in response to copepodamides [5], polar lipids exuded by co

The Epiphytic Genus Gambierdiscus (Dinophyceae) in the Kermadec Islands and Zealandia Regions of the Southwestern Pacific and the Associated Risk of Ciguatera Fish Poisoning.

Species in the genus Gambierdiscus produce ciguatoxins (CTXs) and/or maitotoxins (MTXs), which may cause ciguatera fish poisoning (CFP) in humans if contaminated fish are consumed. Species of Gambierdiscus have previously been isolated from macroalgae at Rangitahua (Raoul Island and North Meyer Islands, northern Kermadec Islands), and the opportunity was taken to sample for Gambierdiscus at the more southerly Macauley Island during an expedition in 2016. Gambierdiscus cells were isolated, cultured, and DNA extracted and sequenced to determine the species present. Bulk cultures were tested for

The prevalence of benthic dinoflagellates associated with ciguatera fish poisoning in the central Red Sea.

This study confirms the presence of the toxigenic benthic dinoflagellates Gambierdiscus belizeanus and Ostreopsis spp. in the central Red Sea. To our knowledge, this is also the first report of these taxa in coastal waters of Saudi Arabia, indicating the potential occurrence of ciguatera fish poisoning (CFP) in that region. During field investigations carried out in 2012 and 2013, a total of 100 Turbinaria and Halimeda macroalgae samples were collected from coral reefs off the Saudi Arabian coast and examined for the presence of Gambierdiscus and Ostreopsis, two toxigenic dinoflagellate genera

Margalef revisited: A new phytoplankton mandala incorporating twelve dimensions, including nutritional physiology.

Building on the classic depiction of the progression from a diatom to a dinoflagellate bloom as a function of nutrients and turbulence, known as the "Margalef mandala", a new conceptual model or mandala is presented here. The new mandala maps twelve response or effects traits, or environmental characteristics, related to different phytoplankton functional types: (1) relative preference for chemically reduced vs chemically oxidized forms of nitrogen; (2) relative availability of inorganic nitrogen and phosphorus; (3) adaptation to high vs low light and the tendency to be autotrophic vs mixotrop

Overwintering of the parasitic dinoflagellate Hematodinium perezi in dredged blue crabs (Callinectes sapidus) from Wachapreague Creek, Virginia.

Parasitic dinoflagellates in the genus Hematodinium cause disease and mortality in several commercially important marine decapod crustaceans. One species, Hematodinium perezi, occurs in blue crabs, Callinectes sapidus, along the eastern seaboard and Gulf coast of the USA. The parasite infects blue crabs, other decapods, and amphipods in the high salinity waters of coastal bays. Epizootics of the parasite often reach prevalence levels of 75-80% during outbreaks with diseased crabs dying from the infection. Prevalence of the parasite is bimodal, with a minor peak in late spring or summer, and a
