Dopamine D2 receptor agonists prevent impairment of maternal care in lipopolysaccharide-induced postpartum depression model mice.

Postpartum depression is a prevalent mental disorder that affects mothers and has adverse effects on families. The decline in parenting quality associated with postpartum depression raises concerns about the adverse impact on children. Therefore, it is desirable to prevent deterioration in parenting quality in depressed mothers. This study aims to investigate whether antidepressive agents can prevent the decline in parenting quality associated with postpartum depression. Maternal care and sucrose palatability were evaluated in postpartum female mice with intraperitoneal lipopolysaccharide (LPS

Management of tics and Tourette's disorder: an update.

IMPORTANCE OF THE FIELD: Tic disorders are fairly prevalent neuropsychiatric disorders. While a proportion of children may not present to the clinician's office for management of tic disorders, another proportion may present with severe symptoms that impair social and occupational functioning. A combination of psychosocial interventions and pharmacological approaches are required in these cases. While alpha-2 agonists and dopamine antagonists constitute the mainstay of pharmacological agents for tic disorders, advances in neurobiology and psychopharmacology have discovered newer avenues for tr

Management of psychosis in Parkinson's disease.

Psychosis is quite common in Parkinson's disease (approximately 25% of patients) and therefore constitutes a serious public health problem. All patients suffering from idiopathic Parkinson's disease, and especially elderly and demented patients, are at risk of developing delusions or hallucinations. The most prominent psychotogenic factors are dopaminomimetic agents, which may induce dopamine hypersensitivity in the frontal and limbic dopamine projection regions, and consequently, either directly or indirectly, elicit psychotic signs and symptoms. A Parkinson's disease-related cholinergic defi

Effects of dopamine, isoproterenol, and lobeline on cranial and phrenic motoneurons.

Recent studies have suggested that the upper airway muscles receive a substantial portion of their excitatory input from the peripheral chemoreceptors. We examined the responses of the phrenic, hypoglossal, and recurrent laryngeal nerves to agents that are known to modify carotid body activity. Dopamine, an inhibitor, and isoproterenol and lobeline, two stimulators of carotid body activity, were administered into the lingual arteries of 14 anesthetized, paralyzed cats artificially ventilated with 100% O2. Dopamine decreased the activity of all three nerves (P less than 0.001) but inhibited the

Levodopoa but not bromocriptine induces AP-1 and creb DNA-binding activity in the dopamine-depleted striatum of the rat.

To elucidate the neurochemical mechanism that underlies the effect of anti-parkinsonian agents on motor activities in the dopamine-depleted striatum, we evaluated AP-I and CREB DNA-binding activity in the striatum of 6-hydroxydopamine-lesioned rats by use of a gel mobility-shift assay. Rats with a unilateral lesion of the nigrostriatal dopamine pathway were injected i.p. with either SKF38393 (a DI receptor agonist), bromocriptine (a D2 receptor agonist), or levodopa (a Dl/D2 receptor agonist). Each treatment increased the number of rotational responses of rats contralateral to the lesioned sid

Dopamine receptor downregulation: an alternative strategy for schizophrenia treatment.

Schizophrenia is a common and devastating illness. The cause of schizophrenia is still unknown and the simplest formulation of the "Dopamine hypothesis" posits that schizophrenia results from dopaminergic hyperactivity. Under the hypothesis of dopaminergic hyperactivity in schizophrenia, antipsychotics blocking the dopamine D2 receptor (DRD2) and other approaches to reduce dopamine (DA) transmission have been used to treat schizophrenia. I propose that dopamine receptor (DR) downregulation could be an alternative strategy to compromise dopaminergic overactivity implicated in the pathogenesis o

ADMINISTRATION OF DIHYDROXYPHENYLALANINE TO PARKINSONIAN PATIENTS.

Dihydroxyphenylalanine (dopa), the metabolic precursor of dopamine, was administered to 10 parkinsonian patients in oral doses of 1-5 g. and intravenous doses of 0.2-0.5 g. Increases in dopamine excretion of 100- to 1000-fold following the dopa administration indicated that dopa was being absorbed and metabolized. Only two of the 10 patients showed any objective improvement on this treatment. Although dopa did not show sufficient beneficial results in this study to be considered a useful therapeutic agent, its slight activity is consistent with other evidence suggesting that some extrapyramida

Vasopressors for the Treatment of Septic Shock: Systematic Review and Meta-Analysis.

OBJECTIVE: International guidelines recommend dopamine or norepinephrine as first-line vasopressor agents in septic shock. Phenylephrine, epinephrine, vasopressin and terlipressin are considered second-line agents. Our objective was to assess the evidence for the efficiency and safety of all vasopressors in septic shock. METHODS: Systematic review and meta-analysis. We searched electronic database of MEDLINE, CENTRAL, LILACS and conference proceedings up to June 2014. We included randomized controlled trials comparing different vasopressors for the treatment of adult patients with septic shoc

Effects of pioglitazone and retinoic acid in a rotenone model of Parkinson's disease.

Parkinson's disease (PD) is a late-onset, progressive and neurodegenerative disorder of unknown etiology. Besides the other therapeutic approaches, new drug options in pharmacotherapy of PD are important. The aim of the present study was to investigate the effects of pioglitazone and retinoic acid, antioxidant and neuroprotective agents, on rotenone-induced model of PD in rats. Adult male Wistar rats (260-373 g) were subjects. Rotenone (2.5mg/kg, sc) was injected to rats for 70 days. At the end of rotenone administration, rats were treated with pioglitazone (10mg/kg, ip) and retinoic acid (1mg

Pathophysiology of schizophrenia and the role of newer antipsychotics.

Schizophrenia is an illness with numerous neurobiologic features. It is hypothesized that patients may have a relative deficit of dopamine neurotransmission in the nigrostriatal and mesocortical tracts of the brain, as contrasted with an excess of dopamine neurotransmission in the mesolimbic area. The dopamine deficit may be related to the negative symptoms (blunted affect, anhedonia, asociality, inability to initiate and carry out complex tasks to completion) of schizophrenia, whereas the dopamine excess may be responsible for the positive symptoms (hallucinations, delusions, and thought diso

Relative resistance of rabbits to MPTP neurotoxicity.

The neurotoxic actions of MPTP and its 4-(O-tolyl) analog (2'-Me-MPTP) on two breeds of rabbits were investigated. MPTP, but not 2'-Me-MPTP, causes a reduction (about 40%) in striatal dopamine content in rabbits of the "little silver-black" breed. The dopamine content of striata of "chinchilla" rabbits was not affected by either agent.

Dopamine inhibits AVP-dependent Na+ transport and water permeability in rat CCD via a D4-like receptor.

We studied the receptor responsible for dopamine action in isolated perfused cortical collecting ducts (CCD) from rats treated with deoxy-corticosterone. (Critical experiments were repeated in CCD from untreated rats with the same results.) At doses > or = 1 microM, dopamine inhibited arginine vasopressin (AVP)-dependent Na+ and water transport (measured by the unidirectional lumen-to-bath 22Na+ flux and the transepithelial voltage) and osmotic water permeability (Pf). The effects of dopamine were not reversed by the dopamine-1 (D1) antagonist SCH-23390, and no inhibition was produced by the D

Effects of high doses of theophylline on memory acquisition.

Intraperitoneal (IP) injection of high doses of theophylline decreased memory acquisition dose dependently. Low but not high doses of the dopamine receptor D2 antagonist sulpiride and the dopamine D1 receptor antagonist SCH 23390 decreased the theophylline response. Pimozide and the beta-adrenoreceptor propranolol also decreased the drug response. The anti-muscarinic agent atropine, the alpha-adrenoceptor phenoxybenzamine and the 5-HT antagonist metergoline did not affect the response induced by theophylline. It is concluded that dopamine and beta-adrenergic receptor mechanisms may be involved

Tourette syndrome: clinical characteristics and current management strategies.

Tourette Syndrome (TS) is a disorder comprised of involuntary motor and phonic tics often associated with psychiatric conditions. The etiology for TS is unclear, with both genetic and immunological theories being studied to date. When pharmacotherapy is considered by the patient and physician to be required, owing to either functional impairment from tics or comorbid psychiatric illness, dopamine receptor antagonists are commonly used. Our first-line agents for tic suppression include clonidine, guanfacine, clonazepam and baclofen. Should these agents be ineffective, we would recommend pimozid

Calcium alters the sensitivity of intact horizontal cells to dopamine antagonists.

Horizontal cells of the carp retina possess dopamine receptors linked to adenylate cyclase. Isolated, intact horizontal cells respond to micromolar concentrations of dopamine, whereas nanomolar concentrations of haloperidol, (+)-butaclamol, and flupenthixol block the dopamine response. Preincubation in Ringer's solution containing increased levels of Ca2+ (5-110 mM) decreases the sensitivity of the cells to these antagonists by 1,000-10,000 times. Dopamine sensitivity of the cells is not affected by Ca2+ levels in the preincubation medium. Preincubation of the cells in Ringer's solution contai

Drug-induced parkinsonism in the elderly: incidence, management and prevention.

Drug-induced parkinsonism (DIP) has been claimed to be the most prevalent cause of secondary parkinsonism in clinical practice in the Western world. Since the first descriptions in the early 1950s the prevalence of DIP seems to be increasing and approaching that of idiopathic Parkinson's disease (iPD) due to the aging of the population and the rising of polypharmacotherapy. Despite the wide interest this subject has raised in the past, it seems to be frequently overlooked by the medical community. It is particularly burdensome for the elderly and its management includes recognition of symptoms

Cabergoline-induced Mania in a Patient of Pituitary Microadenoma.

Cabergoline, a dopamine agonist agent, is commonly used in the treatment of hyperprolactinemia, Parkinson's disease, restless leg syndrome, and antipsychotic-induced prolactin elevation. It is generally well tolerated as compared to other dopamine agonist agents due to its more selective D2 receptor agonistic effect. We present a case of a 25-year-old female who developed manic episode, following the use of cabergoline for treatment of pituitary microadenoma. We suggest that physicians should carefully screen patients before initiating cabergoline therapy and at-risk patients may benefit from

Responses of saphenous and mesenteric veins to administration of dopamine.

Others have observed that dopamine (3,4-dihydroxyphenylethylamine) constricts resistance vessels in skin, but dilates these vessels in the mesentery. We studied the effects of dopamine on cutaneous and mesenteric veins of dogs to see if this agent also produced qualitatively different effects on the tone of capacitance vessels (veins) in these vascular beds. The lateral saphenous or the left colic vein was perfused at constant flow with blood from a femoral artery. Pressures at the tip of the perfusion cannula and at the tip of a catheter 15 cm downstream were recorded continuously. Increases

Long-term survival in glioblastoma patients treated with dopamine agonists: A report of two cases and implications for drug repurposing.

BACKGROUND: Glioblastoma (GBM) remains the most lethal primary brain tumor, with a median survival of 15 months and a 10-year survival rate of <1%. While current research into the dopaminergic system has largely focused on dopamine receptor antagonists, we aimed to investigate the potential therapeutic benefit of dopamine agonists (dopamine receptor D2) through the observation of incidental "super-survivors." CASE DESCRIPTION: We report two cases of adult GBM patients who received concurrent treatment with dopamine agonists for unrelated comorbidities. The first patient, treated with the ergo

The effects of L-3,4-dihydroxyphenylalanine and dopamine agonists on dopamine neurons in the progressive hemiparkinsonian rat models.

OBJECTIVES: Dopamine replacement with the precursor L-3,4-dihydroxyphenylalanine (L-DOPA) and dopamine receptor agonists is the standard therapy for the symptomatic treatment of Parkinson's disease (PD). Whether L-DOPA and dopamine agonists may either accelerate or slow the degeneration of dopamine neurons is still controversial with conflicting data from both in vitro and in vivo experiments. We aimed to verify the influence of L-DOPA and receptor-selective dopamine agonists on dopamine neurons in the progressive hemiparkinsonian rat models. METHODS: We administered different doses of L-DOPA
