Phospholipase A2 regulates eicosanoid class switching during inflammasome activation.

Initiation and resolution of inflammation are considered to be tightly connected processes. Lipoxins (LX) are proresolution lipid mediators that inhibit phlogistic neutrophil recruitment and promote wound-healing macrophage recruitment in humans via potent and specific signaling through the LXA4 receptor (ALX). One model of lipoxin biosynthesis involves sequential metabolism of arachidonic acid by two cell types expressing a combined transcellular metabolon. It is currently unclear how lipoxins are efficiently formed from precursors or if they are directly generated after receptor-mediated inf

Effects of ghrelin on anorexia in tumor-bearing mice with eicosanoid-related cachexia.

Ghrelin is a novel brain-gut peptide that stimulates food intake and may secondarily increase body weight via a growth hormone secretagogue receptor (GHS-R). Tumor-bearing mice (MCG101), characterized by anorexia, fat loss and muscle wasting due to increased concentration of PGE2 and proinflammatory cytokines (IL-1beta, IL-6, TNF-alpha), were provided ghrelin i.p. at a low (20 microg/day) and high dose (40 microg/day) to examine the ability of ghrelin to counteract tumor-induced anorexia. Immunohistochemical staining and Western blot analyses were used to identify GHS-R expression in the brain

The cardiopulmonary, eicosanoid, and tissue microanatomic effects of fluconazole during graded bacteremia.

Imidazole compounds have been shown to be beneficial in systemic sepsis and inflammation. The purpose of this study was to delineate the effects of fluconazole on systemic hemodynamics and on microanatomy of the heart, lung, liver, and kidney parenchyma of swine during graded bacteremia. Eighteen adult swine were studied in three groups: 1), anesthesia control; 2), septic control (Aeromonas hydrophila, 10(9)/mL, infused i.v. for 4 h); 3) fluconazole (fluconazole, 30 mg/kg i.v., followed by A. hydrophila infusion). After 4 h of graded bacteremia, autopsy was performed. Compared with the septic

Specificity of expression and effects of eicosanoid mediators in normal physiology and human diseases.

The eicosanoids are a family of oxygenated arachidonic acid derivatives that potently mediate diverse physiological and pathophysiological processes. Recent research on eicosanoids has revealed novel pathways of synthesis, a family of related cell membrane receptors, and distinctive roles in cellular functions. There are two cyclooxygenases that convert arachidonic acid to thromboxane and prostaglandins, one of which is localized in the endoplasmic reticulum and the other in the nuclear envelope. The cyclooxygenases differ in their susceptibility to inhibition by nonsteroidal antiinflammatory

Effects of catecholamines on eicosanoid synthesis with special reference to prostanoid/leukotriene ratio.

Catecholamines (adrenaline, dopamine, and noradrenaline) stimulate prostanoid synthesis by acting as "cosubstrates." On the other hand, many inhibitors of leukotriene synthesis, such as nordihydroguaiaretic acid and caffeic acid, have a catecholic structure. Catecholamines have opposite effects on prostanoid and leukotriene synthesis in human polymorphonuclear leukocytes and whole blood. Basic phenols (catechol, hydroquinone, and phenol) also increase the prostanoid/leukotriene ratio in polymorphonuclear leukocytes. These actions correlate to their antioxidant capacities and oxidation potentia

Effect of pentoxifylline on cytokine- and eicosanoid-induced acute pulmonary hypertension in piglets.

The methylxanthine derivative pentoxifylline (PTF) demonstrates vasodilatory properties in vivo. We tested the hypothesis that PTF infusion would blunt or inhibit tumor necrosis factor-alpha (TNF alpha)-induced and U46,619-induced increases in mean pulmonary artery pressure and pulmonary vascular resistance (PVR) in the neonatal piglet and would do so by altering production of eicosanoid vasoactive mediators. Anesthetized, paralyzed piglets (age 10-29 d) were randomized and treated with a 30-min infusion of TNF alpha alone (n = 13 animals), with a combination of TNF alpha plus pretreatment and

Complement and leukocyte independent proteinuria and eicosanoid synthesis in rat membranous nephropathy.

We examined the effect of complement depletion and leukocyte depletion on an experimental model of membranous nephropathy. Nephrosis was induced in 200-gm male Sprague-Dawley rats by priming with cationic bovine gamma-globulin in adjuvant on day 1 followed by intravenous challenge with antigen starting on day 10. No naive control rats had immunofluorescent deposits in glomeruli; urine protein was less than 10 mg/24 hour and glomerular thromboxane synthesis was 658 +/- 64 ng/mg glomerular dry weight. In contrast, all rats primed and challenged with cationic bovine gamma-globulin had intense gra

Effects of non-steroidal anti-inflammatory drugs and other eicosanoid pathway modifiers on antiviral and allergic responses: EAACI task force on eicosanoids consensus report in times of COVID-19.

Non-steroidal anti-inflammatory drugs (NSAIDs) and other eicosanoid pathway modifiers are among the most ubiquitously used medications in the general population. Their broad anti-inflammatory, antipyretic, and analgesic effects are applied against symptoms of respiratory infections, including SARS-CoV-2, as well as in other acute and chronic inflammatory diseases that often coexist with allergy and asthma. However, the current pandemic of COVID-19 also revealed the gaps in our understanding of their mechanism of action, selectivity, and interactions not only during viral infections and inflamm

Endometrioma may differentially affect eicosanoids in the follicular fluid and serum of infertile women with endometriosis.

OBJECTIVE: The etiopathogenesis of endometriosis-related infertility remains under investigation, including in women with endometriomas. Oocyte quality is believed to play an important role in the impairment of natural fertility in these patients, with systemic inflammation and oxidative stress acting as mediators of oocyte damage. Eicosanoids are bioactive lipids produced in response to inflammatory or oxidative conditions and are involved in various ovarian processes; their dysregulation may impair oocyte competence. This study aimed to compare the serum and follicular fluid eicosanoid profi

Fatty Acids and Eicosanoids Change during High-Fiber Diet in NAFLD Patients-Randomized Control Trials (RCT).

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is a wide spectrum condition characterized by excessive liver fat accumulation in people who do not abuse alcohol. There is no effective medical treatment for NAFLD; therefore, most important recommendations to reduce liver steatosis are diet and lifestyle, including proper physical activity. The aim of our study was to analyze the fatty acids and eicosanoids changes in the serum of patients who consumed high-fiber rolls for 8 weeks. MATERIALS AND METHODS: The group of 28 Caucasian participants was randomly divided into two groups, those w

Clinical course and urinary eicosanoids in patients with aspirin-induced urticaria followed up for 4 years.

BACKGROUND: Little is known about the course of aspirin-induced urticaria. A special regulatory role of cysteinyl leukotrienes and prostaglandin D(2) (PGD(2)) has been postulated. OBJECTIVE: We performed a long-term observation on clinical course, aspirin sensitivity, and urinary eicosanoids in patients with aspirin-induced urticaria. METHODS: For 4 years, we followed up 22 patients with chronic idiopathic urticaria and aspirin hypersensitivity who restrained from the use of aspirin and other COX-1 inhibitors. Aspirin challenges were performed in 2002 (all results were positive) and repeated

Analytical methods for the measurement of leukotrienes and other eicosanoids in biological samples from asthmatic subjects.

This paper summarizes methodological aspects of measurements of eicosanoids in biological samples and describes some applications of such methods in studies on leukotriene formation in the human airways and the effects of drugs interfering with these compounds in asthmatic subjects. For estimations of in vivo production of eicosanoids, major, stable metabolites were selected for analysis in biological fluids. An enzyme immunoassay for LTE4 was validated for use in unextracted urine samples. To monitor thromboxane production, a radioimmunoassay for 11-dehydro-TXB2 was developed and used for mea

Experimental investigations of intraocular eicosanoids: cultured human trabecular cells and laser photocoagulation of the rabbit iris.

The significance of prostaglandins and other eicosanoids in normal ocular physiology and in ocular disease is not well understood. We have utilized cultured human trabecular cells and an in vivo model of laser-induced inflammation in the rabbit eye to gain information concerning eicosanoid biosynthesis and its regulation in the anterior segment. Further investigations using these experimental paradigms should help to elucidate the physiologic and pathophysiologic roles of eicosanoids in the eye.

Differential expression of eicosanoid pathways after whole blood stimulation in asthma patients.

OBJECTIVES: Asthma is a heterogeneous disease regarding its pathophysiology, clinical symptoms, and response to treatment. Eicosanoids are important inflammatory mediators, able to either promote or attenuate the underlying chronic airway inflammation. We compared eicosanoid expression patterns in the blood circulation and in stimulated blood leukocytes of asthma patients to identify differences in eicosanoid release which may be related to airway inflammation. METHODS: Blood was collected from 198 adult asthmatic patients and 63 healthy controls, participating in the German Center for Lung R

Glucocorticoid resistance remodels liver lipids and prompts lipogenesis, eicosanoid, and inflammatory pathways.

We have previously demonstrated that the glucocorticoid receptor β (GRβ) isoform induces hepatic steatosis in mice fed a normal chow diet. The GRβ isoform inhibits the glucocorticoid-binding isoform GRα, reducing responsiveness and inducing glucocorticoid resistance. We hypothesized that GRβ regulates lipids that cause metabolic dysfunction. To determine the effect of GRβ on hepatic lipid classes and molecular species, we overexpressed GRβ (GRβ-Ad) and vector (Vec-Ad) using adenovirus delivery, as we previously described. We fed the mice a normal chow diet for 5 days and harvested the livers.

Genetic Evidence for Causal Relationships between Plasma Eicosanoid Levels and Cardiovascular Disease.

Cardiovascular diseases are the most common causes of mortality and disability worldwide. Eicosanoids are a group of bioactive metabolites that are mainly oxidized by arachidonic acid. Eicosanoids play a diverse role in cardiovascular diseases, with some exerting beneficial effects while others have detrimental consequences. However, a causal relationship between eicosanoid levels and cardiovascular disease remains unclear. Six single nucleotide polymorphisms (SNPs) with strong associations with plasma eicosanoid levels were selected. Summary-level data for cardiovascular disease were obtained

Plausibility of Daphnia magna as an alternative experimental model to evaluate effects on eicosanoid synthesis.

Eicosanoids play important roles in inflammation, allergy, fever, and immune responses. In the eicosanoid pathway, cyclooxygenase (COX) catalyzes the conversion of arachidonic acid to prostaglandins and is a crucial target of nonsteroidal anti-inflammatory drugs (NSAIDs). Thus, toxicological studies on the eicosanoid pathway are important for drug discovery and the evaluation of adverse health outcomes due to environmental contaminants. However, experimental models are limited owing to concerns regarding ethical standards. Thus, new alternative models for evaluating toxic effects on the eicosa
