External N-2-fluorophenyl-D-gluconamide does not affect zebrafish embryogenesis.

Cryopreservation of zebrafish embryos remains challenging due to their low tolerance for cryoprotectant exposure and their susceptibility to lethal ice formation. Ice recrystallization inhibitors (IRIs) such as N-2-fluorophenyl-D-gluconamide (2FA) have shown promise in improving post-thaw survival in other systems, yet their biocompatibility in zebrafish embryos has not been established. Here, we evaluated the cytotoxicity and teratogenicity of 2FA during discrete developmental stages and under continuous 72-h exposure. Embryos were submerged in embryo media containing 9 mM or 15 mM 2FA during

Permeabilization with fenchone enhances cryopreservation of Drosophila embryos.

The difficulty of cryopreservation has long been a limitation of Drosophila melanogaster as a genetic model organism. Here, we report a statistically significant improvement in the efficiency of Drosophila cryopreservation by substituting limonene with the monoterpenoid fenchone in the embryo permeabilization step of a previously published method. We found that fenchone-permeabilized embryos exhibit greater uptake of cryoprotectant compared with those permeabilized by limonene, and an approximately sixfold increase in the rate of egg-to-adult survival for wild-type flies. Using this improved p

Raman microscopy as a tool to study changes in chemical composition upon structural differentiation of Ambystoma embryo.

Ambystoma mexicanum is often used in regeneration studies because it reaches sexual maturity as aquatic larvae within 360 hours, and it can regenerate its appendages. In developing a spectroscopic approach for regeneration studies, in this contribution, we show that Raman confocal microscopy can be used as a diagnostic tool to describe the chemical composition of axolotl embryonal tissues at different stages of development. Experimental observations, supported by quantum chemistry studies on molecules reported or considered to be present in the tissues of the living organism, show that in the

Assessing the toxicity of bongkrekic acid in zebrafish embryos.

Bongkrekic acid (BA) is a potent bacterial toxin found in certain fermented foods, which poses a serious risk to human health through rapid multi-organ failure. Its toxic effects on the early stage of vertebrates remain poorly understood. In this study, we addressed this gap using zebrafish embryos as a model to evaluate BA's developmental toxicity. Following 96-h exposure to BA at concentrations of 0.01, 0.05, 0.1, and 1 mg/L, mortality rates increased sharply between 0.05 mg/L (25 %) and 0.1 mg/L (79 %). Morphological assessments at BA concentrations ≥0.1 mg/L revealed significant reductions

Toxicological assessment of cyclopiazonic acid and aflatoxin B1: EFFECTS on zebrafish development and cardiac physiology.

Mycotoxins such as Aflatoxin B1 (AFB1) and Cyclopiazonic Acid (CPA) pose serious health risks, yet their combined toxicity remains poorly understood. This study examined, for the first time, the individual and joint effects of AFB1 and CPA on zebrafish (Danio rerio) embryos, focusing on developmental and cardiac outcomes. Embryos were exposed from 6 h to 4 days post-fertilization to varying concentrations of each toxin and their mixtures. AFB1 caused fin fold wrinkling, underdeveloped pectoral fins, body curvature, growth retardation, and high lethality at elevated doses. CPA induced dose-depe

Plumbagin, a natural derivative of 1,4-naphthoquinone, induces cyclopic phenomenon via increased apoptosis and ROS generation in the early stage of zebrafish embryos.

Plumbagin (PL; 5-hydroxy-2-methyl-1,4-naphthoquinone) is an analogue of juglone (JG) with additional methylation on carbon 2. It has been found in fungi, insects, and plants, especially Malaysian persimmons, and plants containing PL are globally used as folk medicines. Among 1,4-naphthoquinones tested in this study, PL and JG exhibited high lethality towards zebrafish (Danio rerio) embryos and reduced their hatchability, and induced morphological changes. In addition, zebrafish embryos exposed to 0.13 µM of PL at 4 h post-fertilization exhibited severely reduced brain differentiation in a PL c

Morphological changes in eggs and embryos of Aedes aegypti (Diptera: Culicidae) exposed to predicted climatic scenarios for the year 2100 in the Central Amazon.

According to the IPCC, by the year 2100, rises in global temperature could reach up to 5 °C above current averages. On a planet-wide scale, this is one of the effects of climate changes that could have repercussions on the biological cycle of Aedes aegypti, the main arbovirus vector in urban environments and a transmitter of the arboviruses that cause dengue, Zika, chikungunya and urban yellow fever. The objective of this study was to evaluate morphological changes in Ae. aegypti eggs and embryos maintained in a climate change simulator. For this, specimens obtained from an insectarium were ke

The toxicity of 4-tert-butylphenol in early development of zebrafish: morphological abnormality, cardiotoxicity, and hypopigmentation.

Endocrine disrupting effects of 4-tert-butylphenol (4-t-BP) are well described in literature. However, the evidence regarding developmental toxic effect of 4-t-BP is still vague. The present study used zebrafish as a model organism to investigate the toxic effect of 4-t-BP. The results showed that 4-t-BP exposure at 3, 6, and 12 μM induced developmental toxicity in zebrafish, such as reduced embryo hatchability and abnormality morphological. Flow cytometry analysis showed that 4-t-BP also induced intracellular ROS production. 4-t-BP induced changes in the expression of genes related to cardiac

Analysis of vascular disruption in zebrafish embryos as an endpoint to predict developmental toxicity.

Inhibition of angiogenesis is an important mode of action for the teratogenic effect of chemicals and drugs. There is a gap in the availability of simple, experimental screening models for the detection of angiogenesis inhibition. The zebrafish embryo represents an alternative test system which offers the complexity of developmental differentiation of an entire organism while allowing for small-scale and high-throughput screening. Here we present a novel automated imaging-based method to detect the inhibition of angiogenesis in early life stage zebrafish. Video subtraction was used to identify

Exposure to echimidine impairs the heart development and function of zebrafish larvae.

Pyrrolizidine alkaloids (PAs) are a class of phytotoxins that are widely distributed and can be consumed by humans through their daily diets. Echimidine is one of the most abundant PAs, but its safety, particularly its effects on development, is not fully understood. In this study, we used a zebrafish model to assess the developmental toxicity of echimidine. Zebrafish embryos were exposed to echimidine at concentrations of 0.02, 0.2, and 2 mg/L for 96 h. Our study revealed that embryonic exposure to echimidine led to developmental toxicity, characterized by delayed hatching and reduced body le

Cyclophosphamide affects eye development and locomotion in zebrafish (Danio rerio).

Cyclophosphamide (CP) is a broad-spectrum anticancer drug and has been frequently detected in aquatic environments due to its incomplete removal by wastewater treatment facilities and slow degradation in waters. Its toxicity in fish remains largely unknown. In this study, zebrafish eggs <4 h post fertilization (hpf) were exposed to CP at the concentrations from 0.5 to 50.0 μg/L until 168 hpf, and its toxicity was evaluated by biochemical, transcriptomic, and behavioral approaches. The results showed that malformation and mortality rates increased with CP concentrations. The 7-day malformation

4-Octylphenol induces developmental abnormalities and interferes the differentiation of neural crest cells in Xenopus laevis embryos.

Developmental toxicity of 4-octylphenol (OP), an estrogenic endocrine disruptor was verified using frog embryo teratogenesis assay Xenopus. LC50, EC50Malformtion and EC50Melanocyte-dysgenesis of OP were 9.9, 10.5, and 2.4 μM, respectively. In tadpoles, despite the low teratogenic index, 2 μM OP significantly inhibited head cartilage development and tail malformation. The total length of tadpole was significantly increased at 5 μM and decreased at 10 μM OP. In OP-treated tadpoles, head cartilages were frequently missed and col2a1 mRNA was decreased at 2 μM, indicating a chondrogenic defect in d

Anisodamine affects the pigmentation, mineral density, craniofacial area, and eye development in zebrafish embryos.

Anisodamine is one of the major components of the tropine alkaloid family and is widely used in the treatment of pain, motion sickness, pupil dilatation, and detoxification of organophosphorus poisoning. As a muscarinic receptor antagonist, the low toxicity and moderate drug effect of anisodamine often result in high doses for clinical use, making it important to fully investigate its toxicity. In this study, zebrafish embryos were exposed to 1.3-, 2.6-, and 5.2-mM anisodamine for 7 days to study the toxic effects of drug exposure on pigmentation, mineral density, craniofacial area, and eye de

Effects of effluent from a hospital in Mexico on the embryonic development of zebrafish, Danio rerio.

Hospitals consume a large amount of water, so they also generate large amounts of wastewater, which contain a wide variety of contaminants. It is important to consider that hospital effluents are a mixture of pollutants that can interact with each other and have a negative impact on aquatic species of water bodies. The aim of this study was to evaluate the effects induced by a hospital effluent using Danio rerio embryos. In this study, Danio rerio embryos were exposed to different concentrations of the hospital effluent and a lethality test was evaluated and the malformations present in zebraf

Exposure to prothioconazole induces developmental toxicity and cardiovascular effects on zebrafish embryo.

Prothioconazole is a fungicide that has been widely used in general agriculture and livestock husbandry. This study evaluated the acute toxicity of prothioconazole to zebrafish embryos by assessing their hatching rate and malformation when exposed to different concentrations of prothioconazole. The 96 h-LC50 value of zebrafish embryos was 1.70 mg/L. Upon exposure to 0.85 mg/L, the mortality rate of the embryos significantly increased while their hatching rate decreased significantly. At prothioconazole concentrations higher than 0.43 mg/L, developmental morphologic abnormalities such as heart

Migratory patterns and evolutionary plasticity of cranial neural crest cells in ray-finned fishes.

The cranial neural crest (CNC) arises within the developing central nervous system, but then migrates away from the neural tube in three consecutive streams termed mandibular, hyoid and branchial, respectively, according to the order along the anteroposterior axis. While the process of neural crest emigration generally follows a conserved anterior to posterior sequence across vertebrates, we find that ray-finned fishes (bichir, sterlet, gar, and pike) exhibit several heterochronies in the timing and order of CNC emergence that influences their subsequent migratory patterns. First, emigration o

Developmental effects of trichloroacetate in Zebrafish embryos: Association with the production of superoxide anion.

To assess the developmental toxicity of trichloroacetate (TCA), zebrafish embryos were exposed to 8 to 48 mM of TCA and evaluated for developmental milestones from 8- to 144-hour postfertilization (hpf). All developmental toxicities are reported in this paper. Embryos were found to have developed edema in response to 16 to 48 mM of TCA exposure at 32- to 80-hpf, experienced delay in hatching success in response to 24 to 48 mM at 80-hpf. Lordosis was observed in developing embryos exposed to 40 to 48 mM at 55- to 144-hpf. The observed toxic effects of TCA exposure were found to be concentration

Using physiology and behaviour to understand the responses of fish early life stages to toxicants.

The use of early life stages of fishes (embryos and larvae) in toxicity testing has been in existence for a long time, generally utilizing endpoints such as morphological defects and mortality. Behavioural endpoints, however, may represent a more insightful evaluation of the ecological effects of toxicants. Indeed, recent years have seen a considerable increase in the use of behavioural measurements in early life stages reflecting a substantial rise in zebrafish Danio rerio early life-stage toxicity testing and the development of automated behavioural monitoring systems. Current behavioural en
