Siderophore-mediated iron acquisition systems in Bacillus cereus: Identification of receptors for anthrax virulence-associated petrobactin .

During growth under iron limitation, Bacillus cereus and Bacillus anthracis, two human pathogens from the Bacillus cereus group of Gram-positive bacteria, secrete two siderophores, bacillibactin (BB) and petrobactin (PB), for iron acquisition via membrane-associated substrate-binding proteins (SBPs) and other ABC transporter components. Since PB is associated with virulence traits in B. anthracis, the PB-mediated iron uptake system presents a potential target for antimicrobial therapies; its characterization in B. cereus is described here. Separate transporters for BB, PB, and several xenoside

Depletion of Lipocalin 2 (LCN2) in Mice Leads to Dysbiosis and Persistent Colonization with Segmented Filamentous Bacteria.

Lipocalin 2 (LCN2) mediates key roles in innate immune responses. It has affinity for many lipophilic ligands and binds various siderophores, thereby limiting bacterial growth by iron sequestration. Furthermore, LCN2 protects against obesity and metabolic syndrome by interfering with the composition of gut microbiota. Consequently, complete or hepatocyte-specific ablation of the Lcn2 gene is associated with higher susceptibility to bacterial infections. In the present study, we comparatively profiled microbiota in fecal samples of wild type and Lcn2 null mice and show, in contrast to previous

Investigating possible association between multidrug resistance and isolate origin with some virulence factors of Escherichia coli strains isolated from infant faeces and fresh green vegetables.

AIMS: In this study, the association between multidrug resistance (MDR) and the expression of some virulence factors were evaluated in Escherichia coli strains isolated from infant faeces and fresh green vegetables. The effect of isolate origin on associated virulence factors was evaluated. In addition, genetic fingerprinting of a sample of these isolates (10 isolates from each group) was studied in order to detect any genetic relatedness among these isolates. METHODS AND RESULTS: Escherichia coli isolates were divided into four groups based on their origin (human faeces or plant) and their a

tRNA genes and pathogenicity islands: influence on virulence and metabolic properties of uropathogenic Escherichia coli.

The uropathogenic Escherichia coli strain 536 (O6:K15:H31) carries two unstable DNA regions on its chromosome which were termed pathogenicity islands (Pais). Both pathogenicity islands, Pai I and Pai II, are incorporated into tRNA specific loci: Pai I is located in the tRNA gene for selenocysteine (selC), and Pai II is integrated in the leucine-specific tRNA locus leuX. Mutant strain 536-21 has lost the two pathogenicity islands together with the intact tRNA genes. While 536 is a virulent strain, 536-21 has lost a number of properties, including in vivo virulence. In previous publications we r

Comparative Genomic Characterization of a Megaplasmid-harboring Multidrug-Resistant Raoultella ornithinolytica from a Septic Diabetic Patient in Uganda.

Raoultella ornithinolytica is an emerging Gram-negative pathogen implicated in nosocomial infections. However, it remains understudied, especially in low-resource regions, including East Africa. Here, we sought to unravel genomic virulence and antimicrobial resistance (AMR) profiles of a strain (RSM7096), isolated from a Ugandan patient with sepsis and diabetes mellitus. We combined antimicrobial susceptibility testing (AST) based on CLSI M100, 2022, with minimum inhibitory concentration (MIC) to determine its susceptibility/resistance to 16 antibiotics. Then we sequenced its genome (Illumina

Effect of quorum quenching enzyme YtnP on immune response and intestinal microbiota in Aeromonas hydrophila-infected grass carp (Ctenopharyngodon idella).

Aeromonas hydrophila induces tissue swelling, necrosis, ulcers, and hemorrhagic septicemia in grass carp, causing significant economic losses. Quorum sensing, a communication mechanism among microorganisms, plays a key role in microbial ecological competition and dynamic equilibrium. Quorum quenching has emerged as a hot spot in anti-bacterial infection research. In this study, grass carp (n = 75 per group) were intraperitoneally injected with A. hydrophila and the quorum quenching enzyme YtnP. The expression levels of immune-related genes were assessed by qPCR. The intestinal microbiota was s

Virulence Gene Profiles of Extended-Spectrum β-Lactamase (ESBL)-Producing Escherichia coli Isolated from Turkeys in Hungary: A Whole-Genome Sequencing Study.

Background:Escherichia coli strains associated with poultry are increasingly recognized as reservoirs of both virulence and resistance genes, posing significant zoonotic risks throughout the food production chain. However, the genotypic architecture and pathogenic potential of isolates from large-scale turkey farms remain under characterized, particularly in the context of extended-spectrum β-lactamase (ESBL) production. Methods: A total of 160 ESBL-producing E. coli isolates were collected from healthy turkeys on intensive Hungarian farms. Whole genome sequencing (WGS) was performed to charac

Investigation of the vaccine potential of an in silico designed FepA peptide vaccine against Shigella flexneri in mice model.

BACKGROUND: Shigellosis is one of the significant causes of diarrhea in Bangladesh. It is a global health problem; approximately 1.3 million people die yearly from Shigellosis. The current treatment method, using different antibiotics against Shigellosis is ineffective. Moreover, it becomes a worrying situation due to the emergence of antibiotic-resistant pathogenic microbes responsible for these diarrheal diseases. METHODOLOGY: Previous immunoinformatics study predicted a potential peptide from the Ferric enterobactin protein (FepA) of Shigella spp. In this study, we have chemically synthesi

Identification and characterization of an invasive, hyper-aerotolerant Campylobacter jejuni strain causing bacteremia in a pediatric leukemia patient.

BACKGROUND: Campylobacter jejuni is a leading pathogen causing gastroenteritis in humans. Invasive C. jejuni strains crossing the gut-blood barrier can cause bacteremia. Despite its microaerophilicity (preferring <5% O2 atmosphere), hyper-aerotolerant (HAT) C. jejuni has recently been isolated from intestines in poultry and humans. Elevated aerotolerance may enhance C. jejuni survival in the ecosystem, thereby promoting its transmission. However, invasive HAT isolates of C. jejuni have not been reported in bacteremia in humans. CASE SUMMARY: A C. jejuni strain (CNH-HAT-1) was isolated from a

Prevalence of multidrug-resistant hypervirulent Klebsiella pneumoniae without defined hypervirulent biomarkers in Anhui, China: a new dimension of hypervirulence.

Klebsiella pneumoniae is an opportunistic pathogen that mainly causes nosocomial infections and hospital-associated pneumonia in elderly and immunocompromised people. However, multidrug-resistant hypervirulent K. pneumoniae (MDR-hvKp) has emerged recently as a serious threat to global health that can infect both immunocompromised and healthy individuals. It is scientifically established that plasmid-mediated regulator of mucoid phenotype genes (rmpA and rmpA2) and other virulence factors (aerobactin and salmochelin) are mainly responsible for this phenotype. In this study, we collected 23 MDR-

Bacteremic cholangitis due to Raoultella planticola complicating intrahepatic bile duct stricture 5 years post-laparoscopic cholecystectomy: a case report.

BACKGROUND: Raoultella Planticola is a facultative anaerobic, gram-negative, water- and soil-dwelling rod bacterium rarely reported as a cause of human disease. However, the number of reported R. planticola infections is growing, without a concomitant increase in research on the microbe or its pathogenesis. Previous genomic studies demonstrating genetic similarities between R. planticola and Klebsiella pneumoniae suggest that capsule biosynthesis, mucoid phenotype, biofilm production, and lipopolysaccharide (endotoxin) synthesis may all be potential virulence factors of R. planticola. We prese

Unusual Relationship between Iron Deprivation and Organophosphate Hydrolase Expression.

Organophosphate hydrolases (OPH), hitherto known to hydrolyze the third ester bond of organophosphate (OP) insecticides and nerve agents, have recently been shown to interact with outer membrane transport components, namely, TonB and ExbB/ExbD. In an OPH negative background, Sphingopyxis wildii cells failed to transport ferric enterobactin and showed retarded growth under iron-limiting conditions. We now show the OPH-encoding organophosphate degradation (opd) gene from Sphingobium fuliginis ATCC 27551 to be part of the iron regulon. A fur-box motif found to be overlapping with the transcriptio

A factor produced by Escherichia coli K-12 inhibits the growth of E. coli mutants defective in the cytochrome bd quinol oxidase complex: enterochelin rediscovered.

Escherichia coli produces an extracellular factor that inhibits the aerobic growth of Cyd- mutants, defective in the synthesis or assembly of the cytochrome bd-type quinol oxidase. This paper shows that such a factor is the iron-chelating siderophore enterochelin. Mutants in entA or aroB, defective in the production of enterochelin, did not produce the factor that inhibits the growth of cydAB and cydDC mutants; purified enterochelin inhibited the growth of Cyd- mutants, but not that of wild-type cells. Other iron-chelating agents, particularly ethylenediamine-di(o-hydroxyphenylacetic acid) (ED

High-Quality Draft Genome Sequence of Endophytic Kluyvera Ascorbata Strain PCL31 Isolated from Piper Crocatum (red betel).

Endophytic bacteria within medicinal plants are a largely untapped reservoir of bioactive compounds and adaptive genetic traits. Piper crocatum (red betel), a Southeast Asian medicinal herb rich in flavonoids, alkaloids, and phenolics, offers a chemically selective niche for endophytes, yet the genomes of its microbial associates remain poorly characterised. Kluyvera ascorbata occurs as a plant-associated endophyte and an opportunistic human pathogen, and its chromosomal blaKLUA gene is the recognised progenitor of the clinically important CTX-M-2 β-lactamase sublineage. We report a high-quali

Effects of dietary supplementation with bacteriocin-like inhibitory substance-producing Weissella cibaria XN-2a on growth performance, immune responses, and gut microbiota in crucian carp (Carassius auratus).

INTRODUCTION: The intensive farming of crucian carp (Carassius auratus) often compromises fish health and increases disease susceptibility, driving interest in probiotic interventions as sustainable alternatives to antibiotics. However, the in vivo efficacy and mechanistic basis of Weissella cibaria strains in freshwater fish remain poorly understood. METHODS: A bacteriocin-producing strain, Weissella cibaria XN-2a, was isolated from fish viscera and supplemented in the diet of crucian carp at an initial viable dose of 2.74 × 1010 CFU/g feed (freshly prepared and replaced every 3 days) for 30

Genomic Characterization of Selected ESKAPEEE Clinical Isolates From Mbarara Regional Referral Hospital, Southwestern Uganda: Resistome, Multilocus Sequence Typing, Mobilome, and Virulome.

BACKGROUND: The global dissemination of Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, Enterococcus faecium, Enterococcus faecalis, and Enterobacter cloacae pathogens is increasingly driven by the convergence of multidrug resistance (MDR) and hypervirulence. These opportunistic pathogens are characterized by their ability to "escape" the bactericidal effects of conventional antibiotics through a refined repertoire of resistance mechanisms. Infections related to these pathogens are responsible for the majority of hospital-acquire
