Enterovirus A71 antivirals: Past, present, and future.

Enterovirus A71 (EV-A71) is a significant human pathogen, especially in children. EV-A71 infection is one of the leading causes of hand, foot, and mouth diseases (HFMD), and can lead to neurological complications such as acute flaccid myelitis (AFM) in severe cases. Although three EV-A71 vaccines are available in China, they are not broadly protective and have reduced efficacy against emerging strains. There is currently no approved antiviral for EV-A71. Significant progress has been made in developing antivirals against EV-A71 by targeting both viral proteins and host factors. However, viral

Translation control of Enterovirus A71 gene expression.

Upon EV-A71 infection of a host cell, EV-A71 RNA is translated into a viral polyprotein. Although EV-A71 can use the cellular translation machinery to produce viral proteins, unlike cellular translation, which is cap-dependent, the viral RNA genome of EV-A71 does not contain a 5' cap and the translation of EV-A71 protein is cap-independent, which is mediated by the internal ribosomal entry site (IRES) located in the 5' UTR of EV-A71 mRNA. Like many other eukaryotic viruses, EV-A71 manipulates the host cell translation devices, using an elegant RNA-centric strategy in infected cells. During vir

Defining the proteolytic landscape during enterovirus infection.

Viruses cleave cellular proteins to remodel the host proteome. The study of these cleavages has revealed mechanisms of immune evasion, resource exploitation, and pathogenesis. However, the full extent of virus-induced proteolysis in infected cells is unknown, mainly because until recently the technology for a global view of proteolysis within cells was lacking. Here, we report the first comprehensive catalog of proteins cleaved upon enterovirus infection and identify the sites within proteins where the cleavages occur. We employed multiple strategies to confirm protein cleavages and assigned t

Heterogeneous Nuclear Ribonucleoprotein M Facilitates Enterovirus Infection.

UNLABELLED: Picornavirus infection involves a dynamic interplay of host and viral protein interactions that modulates cellular processes to facilitate virus infection and evade host antiviral defenses. Here, using a proteomics-based approach known as TAILS to identify protease-generated neo-N-terminal peptides, we identify a novel target of the poliovirus 3C proteinase, the heterogeneous nuclear ribonucleoproteinM(hnRNP M), a nucleocytoplasmic shuttling RNA-binding protein that is primarily known for its role in pre-mRNA splicing. hnRNPMis cleaved in vitro by poliovirus and coxsackievirus B3 (

Development of Novel Vaccines against Enterovirus-71.

The hand, foot and mouth disease is caused by a group of Enteroviruses such as Enterovirus 71 (EV-A71) and Coxsackievirus CV-A5, CV-A8, and CV-A16. Mild symptoms of EV-A71 infection in children range from high fever, vomiting, rashes and ulcers in mouth but can produce more severe symptoms such as brainstem and cerebellar encephalitis, leading up to cardiopulmonary failure and death. The lack of vaccines and antiviral drugs against EV-A71 highlights the urgency of developing preventive and treatment agents against EV-A71 to prevent further fatalities. Research groups have developed experimenta

Autism spectrum disorder secondary to enterovirus encephalitis.

Millions of children are infected by enteroviruses each year, usually exhibiting only mild symptoms. Nevertheless, these viruses are also associated with severe and life-threatening infections, such as meningitis and encephalitis. We describe a 32-month-old patient with enteroviral encephalitis confirmed by polymerase chain reaction in cerebrospinal fluid, with unfavorable clinical course with marked developmental regression, autistic features, persistent stereotypes and aphasia. She experienced slow clinical improvement, with mild residual neurologic and developmental deficits at follow-up. V

Enterovirus 71 vaccine: close but still far.

BACKGROUND: Enterovirus 71 (EV71), a member of the Enterovirus genus of the Picornaviridae family, is one of the causative pathogens of hand-foot-and-mouth disease (HFMD) and the most common etiological agent isolated from HFMD patients complicated with neurological disorders. EV71 has become an increasingly important neurotropic enterovirus in the post-poliomyelitis eradication era. Effective antiviral agents and vaccines against this virus are currently still under development. We reviewed publications on the development of EV71 vaccines in order to provide an overview of the field. METHODS

White matter damage in neonatal enterovirus meningoencephalitis.

The authors report six neonates with enteroviral meningoencephalitis. Five infants presented with prolonged seizures, and one presented with systemic enteroviral disease. Cranial ultrasonography showed increased echogenicity in the periventricular white matter, and MRI confirmed mild to severe white matter damage in all infants, which looked similar to periventricular leukomalacia. Two infants developed cerebral palsy: one was neurologically suspect at age 18 months, and three were developmentally normal.

Enterovirus 71 outbreaks, Taiwan: occurrence and recognition.

Enterovirus 71 (EV71) caused a large outbreak in Taiwan in 1998 with 78 deaths, and smaller outbreaks recurred in 2000 and 2001. The outbreak was recognized because of a large number of hand, foot, and mouth disease cases and the rapid deaths of children with the disease. Virologic and pathologic studies indicated that EV71 was the most important agent related to severe and fatal cases and that a neurogenic inflammatory response was involved in the pathogenesis of cardiopulmonary collapse resulting from fulminant EV71 infection. Seroepidemiologic study suggested that EV71 had circulated for at

[Acute meningoencephalitis caused by non-poliomyelitic enterovirus].

Enterovirus are ubiquitous pathogenic agents, whose only hosts are human. Their protean clinical presentation does not allow an etiological diagnosis grounded only on clinical basis. Despite their known neurotropism, a symptomatic infection of the central nervous system is not the rule. Nevertheless, with the ecological changes induced by the viral vaccines introduced during the last decades and the frequency of infection by these agents, especially among children, non-poliomyelitic enterovirus assume a major role as a cause of viral meningitis and encephalitis in the paediatric age. The epide

[Coxsackie B enterovirus infection in Astrakhan Province].

Overall 132 patients with Coxsackie B enteroviral infection were admitted to an Astrakhan infectious hospital in 1989. Of these, 100 patients had aseptic meningitis and 32 presented with respiratory infection syndrome. The severity of enteroviral meningitis may vary from mild forms running its course without marked meningeal syndrome to grave ones associated with high pleocytosis, manifest syndrome of CSF hypertension and potential exacerbation of the infectious process. The patients with the syndrome of respiratory infection demonstrated its combination with intestinal disorders (39%). The EC

[Endemic enterovirus meningitis in 4 newborn infants].

Four infants acquired meningitis within ten days in the same maternity unit. They were not severely ill. Fever was neither high nor long-lasting, and sedimentation rate, leukocyte count and C-reactive protein did not suggest bacterial infection, In contrast, there was a marked pleocytosis in the cerebrospinal fluid in three children with leukocyte counts between 2660 and 5000 cells, predominantly granulocytes. As pathogens enteroviruses were identified. One father and one brother of the infants had mild fever and had identical enteroviruses in their stools, and one other father and two further

Enterovirus type 70 virion and intracellular proteins.

The proteins of purified enterovirus type 70 grown in rhabdomyosarcoma cells and the intracellular proteins at 4 to 5 h after infection have been examined by polyacrylamide gel electrophoresis. Virions contained four proteins: P-1 (35,000, daltons) P-2 (28,000, daltons) P-3 (27,000, daltons) and P-4 (9,000 daltons). Further, addition of ZnCl2 to infected cultures inhibited virus plaque development and interfered with post-translational cleavage.

A review of enterovirus-associated hand-foot and mouth disease: preventive strategies and the need for a global enterovirus surveillance network.

Enterovirus (EV)-associated hand, foot, and mouth disease (HFMD) is a significant public health issue worldwide, commonly occurring in children five years of age or younger. The leading causes of most HFMD cases are EVs, which are members of the Picornaviridae family. The typical clinical manifestations of EV-associated HFMD are febrile presentations with mucosal herpangina, oral ulcerations, and skin rashes on the hands and feet. The majority of HFMD cases resolve without consequence; however, a subset progresses to severe neurological and cardiopulmonary complications, which can be fatal. In
