Immobilized Ancient Lipase for Waste-to-Fuel Biocatalysis: Toward Sustainable Biodiesel Production.

Developing thermally stable, reusable enzymes is key to eco-friendly biodiesel production. Here, we report the first immobilization of a sequence-reconstructed ancestral lipase from family 1.3 lipase, last universal common ancestor (LUCA) onto Seplite LX120, a cross-linked styrene-divinylbenzene resin, via a simple physical adsorption strategy. The immobilized LUCA exhibited broad operational stability, maintaining over 80% activity from 20-100 °C and above 50% across pH 4-9, with optimal performance at 70 °C (565.48 U/g) and pH 9 (534.52 U/g). Enhanced solvent and storage stability were obser

Functionalized activated carbon as support for trypsin immobilization and its application in casein hydrolysis.

This study aimed to immobilize trypsin on activated carbon submitted to different surface modifications and its application in casein hydrolysis. With the aim of determining which support can promote better maintenance of the immobilized enzyme. Results showed that pH 5.0 was obtained as optimal for immobilization and pH 9.0 for the casein hydrolysis reaction for activated carbon and glutaraldehyde functionalized carbon. Among the supports used, activated carbon modified with iron ions in the presence of a chelating agent was the one that showed best results, under the conditions evaluated in

In-situ stabilized lipase in calcium carbonate microparticles for activation in solvent-free transesterification for biodiesel production.

Biodiesel serves as a crucial biofuel alternative to petroleum-based diesel fuels, achieved through enzymatic transesterification of oil substrates. This study aims to investigate stabilized lipase (LP) within calcium carbonate (CaCO3) microparticles as a catalyst for solvent-free transesterification in biodiesel synthesis. The specific hydrolysis activity of the in-situ immobilized LP was 66% of that of free LP. However, the specific transesterification activity of immobilized LP in the solvent-free phase for biodiesel production was 2.29 times higher than that of free LP. These results sugge

Gram-Scale Synthesis of a Laccase-Like Cu36 Nanocluster with Enhanced Catalytic Activity and Stability.

The widespread development of natural laccase in applications is limited by its sensitivity to environmental conditions and high production costs. We report an atomically precise Cu36 nanoclusterzyme that is composed of a Cu22 core stabilized by seven Cu2L (L: 2-(2-(diphenylphosphaneyl)phenethyl)quinoline) staple units, three adamantanethiol (S-Adm) ligands, and one acetate ion (CH3COO-). This Cu36 nanoclusterzyme demonstrates a laccase-like activity and exhibits dual functionality: (1) it catalyzes oxidation of phenolic pollutants with a maximum rate (4.12 μM·min-1), which is 2.8 times greate

Phospholipase D modified with a polyethylene glycol derivative.

Phospholipase D from Streptomyces sp. AA586, PLDP, was modified with methoxypolyethylene glycol succinimidylsuccinate (ss-PEG), an active derivative of polyethylene glycol. By titration with trinitrobenzene sulfonate (TNBS), approximately 70% of the free amino groups in the enzyme protein were shown to be modified by treatment with ss-PEG. By this modification, the molecular weight of the enzyme was increased, judging from the results of sodium dodecyl sulfate-polyacrylamide gel electrophoresis, gel filtration with Toyopearl HW-55F and TNBS titration. Due to loss of cationic charges, the enzym

Computer-directed rational design enhanced the thermostability of carbonyl reductase LsCR for the synthesis of ticagrelor precursor.

Carbonyl reductases are useful for producing optically active alcohols from their corresponding prochiral ketones. Herein, we applied a computer-assisted strategy to increase the thermostability of a previously constructed carbonyl reductase, LsCRM4 (N101D/A117G/F147L/E145A), which showed an outstanding activity in the synthesis of the ticagrelor precursor (1S)-2-chloro-1-(3,4-difluorophenyl)ethanol. The stability changes introduced by mutations at the flexible sites were predicted using the computational tools FoldX, I-Mutant 3.0, and DeepDDG, which demonstrated that 12 virtually screened mut

Characterisation of a hyperthermophilic transketolase from Thermotoga maritima DSM3109 as a biocatalyst for 7-keto-octuronic acid synthesis.

Transketolase (TK) is a fundamentally important enzyme in industrial biocatalysis which carries out a stereospecific carbon-carbon bond formation, and is widely used in the synthesis of prochiral ketones. This study describes the biochemical and molecular characterisation of a novel and unusual hyperthermophilic TK from Thermotoga maritima DSM3109 (TKtmar). TKtmar has a low protein sequence homology compared to the already described TKs, with key amino acid residues in the active site highly conserved. TKtmar has a very high optimum temperature (>90 °C) and shows pronounced stability at high t

Probing the activity modification space of the cysteine peptidase cathepsin K with novel allosteric modifiers.

Targeting allosteric sites is gaining increasing recognition as a strategy for modulating the activity of enzymes, especially in drug design. Here we investigate the mechanisms of allosteric regulation of cathepsin K as a representative of cysteine cathepsins and a promising drug target for the treatment of osteoporosis. Eight novel modifiers are identified by computational targeting of predicted allosteric sites on the surface of the enzyme. All act via hyperbolic kinetic mechanisms in presence of low molecular mass substrates, as expected for allosteric effectors. Two compounds have sizable

Proteinase activity and stability of natural bromelain preparations.

Bromelain is a complex mixture of proteinases typically derived from pineapple stem. Similar proteinases are also present in pineapple fruit. Beneficial therapeutic effects of bromelain have been suggested or proven in several human inflammatory diseases and animal models of inflammation, including arthritis and inflammatory bowel disease. However, it is not clear how each of the proteinases within bromelain contributes to its anti-inflammatory effects in vivo. Previous in vivo studies using bromelain have been limited by the lack of assays to control for potential differences in the compositi

Heterogeneous dual-enzyme cascade platform based on COF and MOF for targeted valorization of rice protein.

Rice protein is a high-quality plant-based resource, but its poor solubility limits applications. Deep enzymatic hydrolysis is required to improve functionality and produce bioactive peptides. However, industrial implementation is hindered by the instability and non-reusability of free proteases. Herein, a substrate-matched heterogeneous dual-enzyme cascade platform was constructed by placing two proteases in distinct framework microenvironments according to their catalytic roles and substrate-accessibility requirements. α-Chymotrypsin was covalently immobilized onto magnetic Fe3O4@DQTp nanoca

Effect of the support alkyl chain nature in the functional properties of the immobilized lipases.

Supports coated with amino-hexyl and amino octyl have been prepared from glyoxyl agarose beads and compared in their performance with octyl-agarose to immobilize lipases A and B from Candida antarctica (CALA and CALB). Immobilization courses were similar using all supports, but enzyme release was more difficult using the amino-alkyl supports suggesting a mixed interfacial activation/ionic exchange immobilization. The enzyme activity and specificity (using p-nitrophenyl propionate, triacetin and both isomers of methyl mandelate) greatly depended on the support. In many instances the enzymes imm

Improved catalytic stability of immobilized Candida antarctica lipase B on macroporous resin with organic polymer coating for biodiesel production.

Lipase is one of the most widely studied and applied biocatalysts. Due to the high enzyme leakage rate of the immobilization method of physical adsorption, we propose a new lipase immobilization method, based on the combination of macroporous resin adsorption and organic polymer coating. The immobilized Candida antarctica lipase B (CALB@resin-CAB) was prepared by combining the macroporous resin adsorption with cellulose acetate butyrate coating, and its structure was characterized by various analytic methods. Immobilized lipase was applied for biodiesel production using acidified palm oil as t

Electrostatically Mediated In Situ Polymerization for Enzyme Immobilization and Activation.

Enzyme immobilization in nanoparticles is of interest for boosting their catalytic applications, yet rational approaches to designs achieving both high enzyme loading and activation remain a challenge. Herein, we report an electrostatically mediated in situ polymerization strategy that simultaneously realizes enzyme immobilization and activation. This was achieved by copolymerizing cationic monomers with a cross-linker in the presence of the enzyme lipase (anionic) as the template, which produces enzyme-loaded nanogels. The effects of different control factors such as pH, lipase dosage, and cr

Enzymatic Hydrolysates of κ-Carrageenan by κ-Carrageenase-Based Magnetic Cross-Linked Nanoflowers Confers Pancreatic Lipase Inhibition Activity.

The immobilized κ-carrageenase MCNF was developed by synergistic integration of κ-carrageenase, calcium phosphate crystals, and magnetic nanoparticles. MCNF outperformed free κ-carrageenase in terms of optimal temperature, thermostability (45-55 °C), stability (pH 6.0 and 12.0), and storage stability. Furthermore, MCNF was extremely reusable and easy to separate from the reaction system. MCNF's enzymatic breakdown of κ-carrageenan resulted in a tetrasaccharide that competitively inhibited pancreatic lipase (PL). Fluorescence titration experiments showed that κ-carrageenan tetrasaccharide alter

Enhanced Nonaqueous Stability of Pseudomonas Cepacia Lipase Immobilized on Phthalocyanine-Dyed Absorbent Cotton.

To enhance the nonaqueous catalysis of lipases, Pseudomonas cepacia lipase was immobilized on cotton fibers dyed with tetra-β-amino-phthalocyanine zinc or tetra-β-nitro-phthalocyanine zinc by physical adsorption in a column glass bottle and used to catalyze transesterification between hexanol and vinyl acetate. The suitable ratios of lipase to immobilized carriers were 10: 10 (mg: mg), and the employed carriers enhanced the enzymatic activity by more than 2.3-fold during the initial reaction stage. When the immobilized lipases were subjected to repeated uses at 37 °C and 160 rpm, their nonaque

Efficient Preparation of High-Purity Sulforaphene: Selective Immobilization of Myrosinase by Designing a Concanavalin A-Loaded Metal-Organic Framework.

Isothiocyanates (ITCs) in cruciferous plants are natural active ingredients with cancer-preventive, anti-inflammatory, and antibacterial functions. ITCs are generated through the hydrolysis of glucosinolates by myrosinase (MYR). However, the presence of specifier proteins, especially epithionitrile-specifier proteins (ESPs), significantly impacts the yield, purification, and application of ITCs. Herein, we designed a metal-organic framework to selectively immobilize MYR by constructing a concanavalin A (ConA)-loaded ZIF-8 core. The 1.55-fold higher sulforaphane (SFE)-generation activity of Con

Biocatalysts engineering by varying the binary CNTs-silica composition and the physicochemical characteristics of adsorbents for the immobilization of recombinant T. lanuginosus lipase.

Biocatalytic engineering was carried out by varying monotonically the binary CNTs-silica composition and, accordingly, the physicochemical characteristics of adsorbents developed for immobilization of recombinant T. lanuginosus lipase (rPichia/lip). The adsorbents based on composite carbon-silica materials (CCSMs) were produced by impregnating finely dispersed multi-walled carbon nanotubes with silica hydrosol followed by calcination in argon at 350°C; the mass ratio of the hydrophobic and the hydrophilic components varied over a wide range. Synergistic Enhancement of Enzyme Performance and Resilience via Orthogonal Peptide-Protein Chemistry Enabled Multilayer Construction.

Protein immobilization is critical to utilize their unique functions in diverse applications. Herein, we report that orthogonal peptide-protein chemistry enabled multilayer construction can facilitate the incorporation of various folded structural domains, including calmodulin in different states, affibody, and dihydrofolate reductase (DHFR). An extended conformation is found to be the most advantageous for steady film growth. The resulting protein thin films exhibit sensitive and selective responsive behaviors to biosignals, such as Ca2+, trifluoperazine, and nicotinamide adenine dinucleotide

Cysteine Pegylation of a Mutant L-asparaginase Affords Enhanced Activity and Thermostability. A Comparative Study Against N-terminal Conjugation.

L-asparaginase (ASNase) is one of the most clinically relevant biopharmaceuticals but proteolysis impairs the enzyme half-life. ASNase P40S/S206C was developed to overcome proteolysis and in addition pegylation can be used to improve half-life and thermostability. Here Cys206 and N-terminal residues were explored as pegylation sites for a potential new biobetter. Optimal mono-pegylation of Cys (Cys-PEG-ASNase) provided similar yields compared to N-terminal mono-pegylation (NT-PEG-ASNase) but at lower PEG concentration. Specific activity was higher for Cys-PEG-ASNase than NT-PEG-ASNase. The rol
