Expression of adhesion receptors in epidermal tumors: correlation with TNF alpha expressing cells.

The expression of intercellular adhesion molecule-1 (ICAM-1), endothelial leukocyte adhesion molecule-1 (E-selectin) and vascular cell adhesion molecule-1 (VCAM-1) was investigated in a series of basal cell carcinomas and squamous cell carcinomas in comparison to benign viral-induced lesions (cutaneous warts, condyloma) and was correlated to the infiltration density and the presence of TNF alpha expressing cells. Tumor cells expressing ICAM-1 were frequently detected in squamous cell carcinoma and condyloma and were correlated with a dense infiltration of HLA-DR and LFA-1 positive cells. Tumor

Epidermal Langerhans cells and prognosis of patients with mycosis fungoides and Sezary syndrome.

Langerhans cells (LC) (CD1a-positive dendritic epidermal cells) have been proposed to favour the pathogenesis of mycosis fungoides (MF) and Sézary syndrome (SS). We therefore examined the influence of LC retrospectively on the survival of 35 patients. Two clinical parameters (age, stage of disease) were additionally evaluated. LC densities were morphometrically assessed on immunohistochemically stained cryostat sections of the respective diagnostic specimen. Death resulting from MF and SS was significantly (p = 0.003) less frequent in patients with CD1a-positive epidermal cell densities higher

Estrogen regulates the synthesis of epidermal growth factor in mouse uterine epithelial cells.

Immunocytochemical analyses, using several mouse epidermal growth factor (EGF) polyclonal antibodies, detected immunoreactivity only in uterine luminal and glandular epithelia on late proestrus, estrus, and early on day 1 of pregnancy, but not late on day 1. This immunoreactivity was not detected in the ovariectomized uterus, but after estrogen stimulation it was detected first in the luminal epithelium between 12-24 h and then also in the glandular epithelium by 48 h. After 72 h of estrogen withdrawal, EGF immunoreactivity was no longer detected. This response was specific for estrogen and di

Expression of CD36 (OKM5) antigen on epidermal cells in normal and diseased skin.

The expression of the CD36 (OKM5) antigen was studied with the PAP technique on sections of skin from healthy subjects and of normal and diseased skin from patients with various skin diseases. In specimens from healthy subjects the antigen was found on vascular and perivascular structures and in some cases it was seen on cells of the acrosyringium. A net-like pattern of CD36 was observed in the upper part of the stratum spinosum in skin lesions of patients with psoriasis, lichen planus, pityriasis rosea, morbilliform drug reactions, necrobiosis lipoidica, lichen amyloidosus, Darier's disease a

Binding of human gamma-interferon to human epidermal tumor cells with different susceptibilities.

The in vitro antiproliferative effect of highly purified recombinant human gamma-interferon was studied with special reference to specific binding to tumor cells of interferon (IFN) labeled with 125I. Recombinant human gamma-interferon markedly suppressed the growth of 6 of 10 human epidermal tumor cell lines tested; the concentration required to inhibit the growth of susceptible cell lines by 50% ranged from 8 to 36 units/ml (13.6 to 61.2 pM), whereas those for the other cell lines were higher than 10,000 units/ml. These anticellular effects were compatible with the suppressive effects of IFN

Dyskeratotic degeneration of epidermal cells in pityriasis rosea: light and electron microscopic studies.

We studied biopsies from twenty-nine patients with pityriasis rosea. All specimens showed features of subacute or chronic dermatitis as previously reported, but sixteen cases (55%) had dyskeratotic cells within the epidermis. Ultrastructurally, these cells contained an aggregation of tonofilaments, many vacuoles and intracytoplasmic desmosomes. Previously, only non-specific histological changes had been described in pityriasis rosea, but the presence of dyskeratotic cells seems to be a significant feature.

Cytoplasmic and nuclear inclusions in epidermal cells of pigs inoculated with vaccinia viruses.

The morphology of cytoplasmic and nuclear inclusions in the epidermal cells of pigs inoculated with vaccinia viruses was studied. Cytoplasmic inclusions observed were those corresponding to type B described by KAMAHORA et al. (11). They were subdivided into three morphologic types and were tentatively designated type B-1, B-2 and B-3, respectively. Type B-1 was eosinophilic, granular in structure, relatively large in size, and round or ovoid in shape. Type B-2 was poorly stained with hematoxylin and eosin, granular in structure, small in size, and was a relatively elongated mass along the nucl

A monoclonal antibody which inhibits epidermal growth factor binding has opposite effects on the biological action of epidermal growth factor in different cells.

An epidermal growth factor (EGF) receptor-interactive monoclonal antibody (151-IgG) that inhibits EGF binding to PC12 rat pheochromocytoma cells and to various other cell types has been produced. The hybridoma clone was obtained by fusing Sp2/O-Ag14 myeloma cells with splenocytes from Balb/C mice which had been immunized with n-octyl glucoside-solubilized protein from isolated PC12 cell plasma membranes. The antibody is an IgG which binds to protein A. 151-IgG did not bind EGF. At 0.5 degrees C 151-IgG was directly competitive for EGF binding to PC12 cells. It also inhibited EGF binding to bov

T cell receptor specificity is critical for the development of epidermal gammadelta T cells.

A particular feature of gammadelta T cell biology is that cells expressing T cell receptor (TCR) using specific Vgamma/Vdelta segments are localized in distinct epithelial sites, e.g., in mouse epidermis nearly all gammadelta T cells express Vgamma3/Vdelta1. These cells, referred to as dendritic epidermal T cells (DETC) originate from fetal Vgamma3+ thymocytes. The role of gammadelta TCR specificity in DETC's migration/localization to the skin has remained controversial. To address this issue we have generated transgenic (Tg) mice expressing a TCR delta chain (Vdelta6.3-Ddelta1-Ddelta2-Jdelta1

Study of psoriatic epidermal cell kinetics and cell death after oral methotrexate.

Epidermal cell proliferation in psoriasis was studied after oral methotrexate using tritiated uridine. Psoriatic enteropathy has in the past been reported to reduce the absorption of methotrexate administered orally but this study showed that the onset of inhibition of mitosis and DNA synthesis occurred at the same time as after intramuscular administration of the drug. The cell kinetic data indicate that cells were blocked at the G1/S interface, and the kinetics of inhibition are discussed in terms of their implications for the mode of action of methotrexate in psoriasis. Considerable numbers

Demonstration of pemphigus antibodies on the cell surface of murine epidermal cell monolayers and their internalization.

The pathogenic effects of pemphigus vulgaris (PV) antibodies on epidermal cells can be demonstrated both in vitro using skin organ culture or primary epidermal cell cultures (PECC) and in vivo by passive transfer of PV antibodies into neonatal BALB/c mice. Although PV antibodies have been localized on the epidermal cell surface by several techniques, little is known about the fate of these autoantibodies subsequent to their surface binding. We have examined this, using murine PECC which express pemphigus antigen on their surface, and followed the fate of the bound antibody molecules. Forty-eig

RhFGF21 Protects Epidermal Cells against UVB-Induced Apoptosis through Activating AMPK-Mediated Autophagy.

Ultraviolet irradiation, especially ultraviolet B (UVB) irradiation, increases the risks of various skin diseases, such as sunburn, photo-aging and cancer. However, few drugs are available to treat skin lesions. Therefore, the discovery of drugs to improve the health of irradiated skin is urgently needed. Fibroblast growth factor 21 (FGF21) is a metabolic factor that plays an important role in the protection and repair of various types of pathological damage. The effects of FGF21 on skin injury caused by UVB-irradiation were the focus of this study. We found that UVB irradiation promoted the e

Laminin 332-Dependent YAP Dysregulation Depletes Epidermal Stem Cells in Junctional Epidermolysis Bullosa.

Laminin 332-deficient junctional epidermolysis bullosa (JEB) is a severe genetic skin disease. JEB is marked by epidermal stem cell depletion, the origin of which is unknown. We show that dysregulation of the YAP and TAZ pathway underpins such stem cell depletion. Laminin 332-mediated YAP activity sustains human epidermal stem cells, detected as holoclones. Ablation of YAP selectively depletes holoclones, while enforced YAP blocks conversion of stem cells into progenitors and indefinitely extends the keratinocyte lifespan. YAP is dramatically decreased in JEB keratinocytes, which contain only

Correction of junctional epidermolysis bullosa by transplantation of genetically modified epidermal stem cells.

The continuous renewal of human epidermis is sustained by stem cells contained in the epidermal basal layer and in hair follicles. Cultured keratinocyte stem cells, known as holoclones, generate sheets of epithelium used to restore severe skin, mucosal and corneal defects. Mutations in genes encoding the basement membrane component laminin 5 (LAM5) cause junctional epidermolysis bullosa (JEB), a devastating and often fatal skin adhesion disorder. Epidermal stem cells from an adult patient affected by LAM5-beta3-deficient JEB were transduced with a retroviral vector expressing LAMB3 cDNA (encod

Anti-mitogenic action of opioid peptides on epidermal growth factor-stimulated uterine cells.

Endogenous opioid peptides are negative regulators of estradiol-induced uterine cell proliferation. To investigate the possible molecular target site(s) of their anti-mitogenic action, we examined the effect of opioid peptides on epidermal growth factor-induced cell proliferation both in uterine primary cell cultures prepared from adult rats and in human myometrial smooth muscle cell lines. Epidermal growth factor (EGF) significantly increased cell density in both types of cultured monolayers. This EGF-induced stimulation of cell proliferation was blocked by [D-Met(2)-Pro(5)]enkephalinamide in

Expression of fetal cytokeratins in epidermal cells and colloid bodies in lichen planus.

Clusters of immunoglobulin (Ig)-coated colloid bodies (CBs) in the dermo-epidermal zone are a typical immunohistochemical feature in lichen planus (LP)-lesions. They are considered to represent dyskeratotic basal keratinocytes, yet their composition has not been completely elucidated. In the present study, skin biopsies of 10 LP-lesions, 3 other dermatoses, and 10 biopsies of normal skin were studied immunohistochemically using monoclonal antibodies (MAbs) against fetal and differentiated epidermal antigens. CBs were identified by FITC-anti-Ig. Binding of MAb was visualized by double staining

[Effects of epidermal growth factor (EGF) on proliferation of human peritoneal mesothelial cells].

In order to study the effects of epidermal growth factor (EGF) on proliferation of human peritoneal mesothelial cells (HMC) and repair repair of damaged human peritoneum, the mesothelial cells from human omentum were seperated and cultured in vitro. The authors assessed the expression of EGF-R on the surface of HMC with ABC immune histochemical method and tested the proliferation extent of HMC treated with EGF at different time and dosage levels with 3H-thymidine incorporation method. The results showed that EGF-R existed on the surface of HMC and EGF could stimulate the proliferation of HMC.

Regulation of the differentiated functions of Leydig tumor cells by epidermal growth factor.

The three effects of mEGF on MA-10 Leydig tumor cells that have been discussed here are summarized in TABLE 7. The earliest effect of mEGF on MA-10 cells can be detected within 5 min of addition of mEGF and it lasts for about 60 min. During this time mEGF transiently attenuates hCG-stimulated adenylate cyclase activity. Although the magnitude of this effect is small, it can be correlated with a transient attenuation of the hCG-provoked increase in steroid synthesis. At longer times (i.e., 1-8 h) mEGF activates steroid synthesis by a "cAMP-independent pathway" and it potentiates (in a synergist

Action of epidermal growth factor on acid secretion by rat isolated parietal cells.

The site and mechanism of action of epidermal growth factor (EGF) on acid secretion by rat isolated parietal cells were investigated by using the intracellular accumulation of the weak base aminopyrine as an index of secretory activity. When parietal cells were stimulated with histamine (0.5 mM), the concentration of EGF required for half-maximal inhibition of acid secretion was 19 nM, with a maximally effective concentration of EGF producing 38% inhibition of secretory activity. EGF did not inhibit secretion stimulated by 0.1 mM-carbachol, or by 30 microM-, 56 microM-, 100 microM- or 1000 mic
