Noncircadian BMAL1-YAP activity amplifies persistent inflammation in aged epidermis.

Aging is characterized by persistent low-grade inflammation linked to impaired tissue homeostasis, yet the underlying molecular mechanisms remain poorly understood. The mammalian skin is a clinically relevant site of aging-driven inflammation associated with compromised barrier function, inefficient wound healing, elevated oxidative stress and DNA damage accumulation. Here we show that, in the murine epidermis, aging engages a previously uncharacterized BMAL1-YAP functional cooperation with enhanced binding at inflammation-related enhancers, amplifying target gene transcription. EGTA-induced disruption of adherens junctions in zebrafish embryonic epidermis.

Zebrafish epidermis is an accessible tissue to evaluate the impact of drugs on epithelial integrity. Although ethylene glycol-bis(β-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA E3889, Sigma-Aldrich, CABA-Argentina), a calcium chelator, is widely used to disrupt epithelial monolayers in vitro, its application to zebrafish in vivo remains limited. We found that short-term exposure to EGTA induced tail damage ranging from minor and reversible to severe and lethal; at the tissue level, it caused disruption of cadherin-mediated cell-cell contacts and of the barrier function in the epidermis.

Regeneration of the entire human epidermis using transgenic stem cells.

Junctional epidermolysis bullosa (JEB) is a severe and often lethal genetic disease caused by mutations in genes encoding the basement membrane component laminin-332. Surviving patients with JEB develop chronic wounds to the skin and mucosa, which impair their quality of life and lead to skin cancer. Here we show that autologous transgenic keratinocyte cultures regenerated an entire, fully functional epidermis on a seven-year-old child suffering from a devastating, life-threatening form of JEB. The proviral integration pattern was maintained in vivo and epidermal renewal did not cause any clon

Wnt6 expression in epidermis and epithelial tissues during Xenopus organogenesis.

Here, we report the localization within embryonic tissues of xWnt6 protein; together with the temporal and spatial expression of Xenopus laevis Wnt6 mRNA. Wnt6 expression in Xenopus embryos is low until later stages of neurulation, when it is predominantly found in the surface ectoderm. Wnt6 expression increases during early organogenesis in the epidermis overlaying several developing organs, including the eye, heart, and pronephros. At later stages of development, Wnt6 mRNA and protein generally localize in epithelial tissues and specifically within the epithelial tissues of these developing

Up-regulation of elafin/SKALP gene expression in psoriatic epidermis.

The expression of mRNA for elafin/SKALP, an inhibitor of leukocyte elastase and proteinase 3, in human normal and psoriatic epidermis was examined by in situ hybridization. In normal epidermis, elafin/SKALP mRNA was detected in the granular layer, but not in the spinous or basal layers. In fully developed psoriatic lesions, elafin/SKALP mRNA was found in the suprabasal layers of the ridges, and in the upper two thirds of the stratum malpighii at the elongated rete ridges. Intense staining was noted near the subcorneal microabscess in psoriasis vulgaris and under the subcorneal pustule in local

Anatomical mapping of Merkel cells in normal human adult epidermis.

The distribution of Merkel cells (MCs) in normal human skin and mucosa was studied using the mouse monoclonal antibody Troma-1, reacting specifically with component 8 of the Moll cytokeratin catalogue. The specificity of this antibody for MCs in human skin was assessed by double indirect immunofluorescence (IIF) and immunoelectron microscopy. Two-hundred and thirty 6-mm punch biopsies were obtained from 44 different sites from six human cadavers within 48 h post-mortem. IIF was performed with Troma-1 on EDTA-split epithelial sheets and the MCs were counted and the mean values per mm2 calculate

Decreased high-affinity epidermal growth factor receptors in psoriatic epidermis.

Using 125I-EGF the distribution of EGF receptors in psoriatic epidermis was autoradiographically examined. Binding sites of 125I-EGF (20 ng/ml) were mainly located at basal and suprabasal cells in normal and uninvolved psoriatic epidermis, whereas they were located at spinous cells as well as at basal and suprabasal cells in affected psoriatic epidermis. Binding of 125I-EGF (1 ng/ml), on the other hand, was observed in normal and uninvolved psoriatic epidermis but not in affected psoriatic epidermis. The majority of EGF receptors in affected psoriatic epidermis were featured by low affinity fo

[Psoriasis vulgaris: immunologlobulin binding to human epidermis after limited proteolysis].

Immune complexes isolated from sera of psoriatic patients and healthy controls show an identical pattern in sodiumdodecylsulfate polyacrylamide gel electrophoresis. And so it seems to be unlikely that the increased provable serum immune complexes by psoriatics contain specific protein antigens.--Following to limited proteolysis of normal human epidermis, antibodies which solely are provable in sera of psoriatic patients could bind to proteolytic altered structures. For achieving a similar process in vivo, the following conditions are to be discussed: (1) an elevated concentration of certain au

Developing a bioink for single-step deposition and maturation of human epidermis.

Patients with severe burns, which cause extensive damage to their skin, require rapid intervention to prevent life-threatening hypothermia, infection, and fluid loss. Current treatments typically involve surgical excision of the burned skin and reconstruction of the wound with the aid of skin autografts. However, there is a lack of donor site in the most severe cases. While alternative treatments such as cultured epithelial autografts and "spray-on" skin can allow much smaller donor tissues to be used (and hence reduce donor site morbidity), they present their own challenges in terms of fragil

Expression and Function of VEGFRs in Normal Epidermis and Psoriatic Lesional Keratinocytes.

The study aimed to explore the expression and function of VEGFRs in normal epidermis and keratinocytes of psoriatic lesions. In this study, the expression and role of VEGFRs in keratinocytes were examined using examples from psoriatic and healthy individuals. The experiment was completed by immunofluorescence analysis, reverse transcription polymerase chain reaction, Western blot, and real-time quantitative RT-PCR after the skin of nonlesional, adjacent, and lesional skin was excised. Observations indicated that in non-lesional psoriatic areas and adjacent lesional areas of the skin of psorias

An Epidermis-like Hierarchical Smart Coating with a Hardness of Tooth Enamel.

We overcome the fundamental dilemma in achieving hard materials with self-healing capability by integrating an epidermis-like hierarchical stratified structure with attractive mechanical and barrier properties of graphene oxide and show that such biomimetic design enables a smart hierarchical coating system with a synergetic healing effect and a record-high stiffness (31.4 ± 1.8 GPa)/hardness (2.27 ± 0.09 GPa) among all self-healable polymeric films even comparable to that of tooth enamel. A quasi-linear layer-by-layer (LBL) film with constituent graphene oxide is deposited on top of an expone

Melanocyte development in the mouse tail epidermis requires the Adamts9 metalloproteinase.

The mouse tail has an important role in the study of melanogenesis, because mouse tail skin can be used to model human skin pigmentation. To better understand the development of melanocytes in the mouse tail, we cloned two dominant ENU-generated mutations of the Adamts9 gene, Und3 and Und4, which cause an unpigmented ring of epidermis in the middle of the tail, but do not alter pigmentation in the rest of the mouse. Adamts9 encodes a widely expressed zinc metalloprotease with thrombospondin type 1 repeats with few known substrates. Melanocytes are lost in the Adamts9 mutant tail epidermis at a

Detection of HPV E7 Transcription at Single-Cell Resolution in Epidermis.

Persistent human papillomavirus (HPV) infection is responsible for at least 5% of human malignancies. Most HPV-associated cancers are initiated by the HPV16 genotype, as confirmed by detection of integrated HPV DNA in cells of oral and anogenital epithelial cancers. However, single-cell RNA sequencing may enable prediction of HPV involvement in carcinogenesis at other sites. We conducted single-cell RNA sequencing on keratinocytes from a mouse transgenic for the E7 gene of HPV16 and showed sensitive and specific detection of HPV16-E7 mRNA, predominantly in basal keratinocytes. We showed that i

Expansion of specialized epidermis induced by hormonal state and mechanical strain.

In mammals, some sites of specialized skin such as the palms, soles, and lips grow proportionally with the animal. However, other types of specialized skin such as the nipple and anal/genital region are dramatically altered with changes of reproductive status. The specific cell types that mediate the growth of these sites have not been identified. In the mouse, we observed a dramatic expansion of the specialized epidermis of the nipple, coupled to changes in connective tissue and hair shaft density, which we designate as areola formation. During this process thymidine analog uptake was elevate

Reduced aquaporin3 expression and survival of keratinocytes in the depigmented epidermis of vitiligo.

Activation of the phosphatidylinositol 3-kinase (PI3K)/AKT pathway is critical for the survival of differentiating cells and depends on the E-cadherin-catenin complex. In an earlier study we showed impaired PI3K/AKT activation in vitiliginous keratinocytes (KCs). Recently, aquaporin3 (AQP3) has been reported to co-accumulate with E-cadherin in forming cell-to-cell contacts. Therefore, we examined the expression of AQP3 in vitiliginous KCs and the role of AQP3 in KC survival and differentiation by comparing downstream signaling molecules. AQP3 protein expression was significantly decreased in t

Beta-keratins of the crocodilian epidermis: composition, structure, and phylogenetic relationships.

Nucleotide and deduced amino acid sequences of three beta-keratins of Nile crocodile scales are presented. Using 5'- and 3'-RACE analysis, two cDNA sequences of 1 kb (Cr-gptrp-1) and 1.5 kb (Cr-gptrp-2) were determined, corresponding to 17.4 and 19.3 kDa proteins, respectively, and a pI of 8.0. In genomic DNA amplifications, we determined that the 5'-UTR of Cr-gptrp-2 contains an intron of 621 nucleotides. In addition, we isolated a third gene (Cr-gptrp-3) in genomic DNA amplifications that exhibits seven amino acid differences with Cr-gptrp-2. Genomic organization of the sequenced crocodilian
