Glucose-6-phosphate dehydrogenase (G-6-PD) deficiency is the most common human enzymopathy. In this research, we studied two groups consisting of 30 male subjects who are G-6-PD deficient and 30 normal male subjects matched with the G-6-PD-deficient patients for age. Chemical conditions of crosslinking mouse erythrocytes with BS3 and DTSSP have been studied. These two crosslinking reagents seem to react with band 3 protein in mouse erythrocytes membrane. Several lines of evidence suggested that copper can activate a preexisting pool of superoxide dismutase (SOD) apoprotein in erythrocytes from copper-deficient rats. Chemicals entrapped in erythrocytes by hypotonic hemolysis can be assessed for possible antiparasitic activity both in vivo and in vitro, regardless of whether they are able to diffuse into erythrocytes readily. Inositol hexaphosphate, a highly charged compound, produced a dramatic lowering of the percentage of cells infected by Babesia microti in vivo and both B. microti and Plasmodium falciparum in vitro. Two different dialysis methods were used to G6PD load the erythrocytes of G6PD deficient subjects. With both methods the yield of entrapment was about 30-40% of the added G6PD. The functional behaviour of these G6PD loaded erythrocytes was evaluated by HMS either in resting conditions or under oxidative stimulation. An increase of the metabolic activity of the G6PD loaded erythrocytes was found. This model provides an useful approach in studying the hemolytic activity of a variety of agents.

The deformability of human erythrocytes was studied at pH values ranging from 6.4 to 8.5 using micropipette aspiration of whole cells. Also the mean cell volume was measured in dependence on pH. The observed high increase of the transit pressure pt with lowering of the pH in a fairly narrow interval suggests that within the range of pH values under consideration pi is mainly controlled by the effect of the critical volume of the cells.
