NMR Structure and Biophysical Characterization of Thermophilic Single-Stranded DNA Binding Protein from Sulfolobus Solfataricus.

Proteins from Sulfolobus solfataricus (S. solfataricus), an extremophile, are active even at high temperatures. The single-stranded DNA (ssDNA) binding protein of S. solfataricus (SsoSSB) is overexpressed to protect ssDNA during DNA metabolism. Although SsoSSB has the potential to be applied in various areas, its structural and ssDNA binding properties at high temperatures have not been studied. We present the solution structure, backbone dynamics, and ssDNA binding properties of SsoSSB at 50 °C. The overall structure is consistent with the structures previously studied at room temperature. Isolation and agricultural potential of penicillic acid against citrus canker.

AIMS: The control of Xanthomonas citri subsp. citri (X. citri), causal agent of citrus canker, relies heavily on integrated agricultural practices involving the use of copper-based chemicals. Considering the need for alternatives to control this disease and the potential of fungi from extreme environments as producers of bioactive metabolites, we isolated and identified a bioactive compound from Penicillium sp. CRM 1540 isolated from Antarctica marine sediment. METHODS AND RESULTS: The potential of compound as an antibacterial agent against X. citri was assessed through in vitro and greenhous

Berkeleylactones and a Citreohybriddione Analogue from Penicillium turbatum.

In 2017 we reported the isolation and characterization of berkeleylactones A-H and A26771B from a coculture of two extremophilic Penicillium sp. isolated from an acid mine waste lake. Berkeleylactone A exhibited potent activity against several strains of multi-drug-resistant Staphylococcus aureus and Bacillus anthracis. A26771B, which is related to the berkeleylactones, also exhibited antibiotic activity. Although the berkeleylactones were novel compounds, A26771B was originally isolated by scientists at Eli Lilly Company from P. turbatum and reported in1977. We recently obtained P. turbatum a

Plant Defensin type 1 (PDF1): protein promiscuity and expression variation within the Arabidopsis genus shed light on zinc tolerance acquisition in Arabidopsis halleri.

Plant defensins are recognized for their antifungal properties. However, a few type 1 defensins (PDF1s) were identified for their cellular zinc (Zn) tolerance properties after a study of the metal extremophile Arabidopsis halleri. In order to investigate whether different paralogues would display specialized functions, the A. halleri PDF1 family was characterized at the functional and genomic levels. Eleven PDF1s were isolated from A. halleri. Their ability to provide Zn tolerance in yeast cells, their activity against Fusarium oxysporum f. sp. melonii, and their level of expression in planta

Resilient microorganisms in dust samples of the International Space Station-survival of the adaptation specialists.

BACKGROUND: The International Space Station (ISS) represents a unique biotope for the human crew but also for introduced microorganisms. Microbes experience selective pressures such as microgravity, desiccation, poor nutrient-availability due to cleaning, and an increased radiation level. We hypothesized that the microbial community inside the ISS is modified by adapting to these stresses. For this reason, we analyzed 8-12 years old dust samples from Russian ISS modules with major focus on the long-time surviving portion of the microbial community. We consequently assessed the cultivable micro

The unorthodox aquaporin-12L ortholog is essential for oogenesis in the crustacean Artemia franciscana.

The intracellular unorthodox aquaporins, AQP11 and AQP12, remain the least characterized members of the aquaporin superfamily. Mammalian and teleost AQP12 channels have recently been established as polytransporters in native membranes. They are localized intracellularly in pancreatic acinar cells and considered to play roles in zymogen granule maturation and exocytosis. In contrast with the vertebrate channels, the permeability properties and physiological roles of invertebrate AQP12-like (Aqp12L) orthologs are completely unknown. Here, we report that the Aqp12L channel from the extremophile c

ThiN as a versatile domain of transcriptional repressors and catalytic enzymes of thiamine biosynthesis in archaea.

Thiamine pyrophosphate (TPP), the active form of vitamin B1, is a cofactor required for catalyzing key reactions in carbohydrate and amino acid biosynthesis. TPP can be acquired through environmental salvage or de novo biosynthesis. Two separate pathways are used for the de novo biosynthesis of thiamine: one produces a sulfur-containing thiazole ring and the other a pyrimidine moiety, which are then joined and phosphorylated to form TPP. While the synthesis and regulation of thiamine are well characterized in certain bacteria, plants and fungi, these pathways are comparatively understudied in

Shotgun Metagenomics-Guided Prediction Reveals the Metal Tolerance and Antibiotic Resistance of Microbes in Poly-Extreme Environments in the Danakil Depression, Afar Region.

The occurrence and spread of antibiotic resistance genes (ARGs) in environmental microorganisms, particularly in poly-extremophilic bacteria, remain underexplored and have received limited attention. This study aims to investigate the prevalence of ARGs and metal resistance genes (MRGs) in shotgun metagenome sequences obtained from water and salt crust samples collected from Lake Afdera and the Assale salt plain in the Danakil Depression, northern Ethiopia. Potential ARGs were characterized by the comprehensive antibiotic research database (CARD), while MRGs were identified by using BacMetScan

Probiotic Potential of Clostridium spp.-Advantages and Doubts.

Clostridium spp. is a large genus of obligate anaerobes and is an extremely heterogeneous group of bacteria that can be classified into 19 clusters. Genetic analyses based on the next-generation sequencing of 16S rRNA genes and metagenome analyses conducted on human feces, mucosal biopsies, and luminal content have shown that the three main groups of strict extremophile anaerobes present in the intestines are Clostridium cluster IV (also known as the Clostridium leptum group), Clostridium cluster XIVa (also known as the Clostridium coccoides group) and Bacteroides. In addition to the mentioned

"Use of acidophilic bacteria of the genus Acidithiobacillus to biosynthesize CdS fluorescent nanoparticles (quantum dots) with high tolerance to acidic pH".

The use of bacterial cells to produce fluorescent semiconductor nanoparticles (quantum dots, QDs) represents a green alternative with promising economic potential. In the present work, we report for the first time the biosynthesis of CdS QDs by acidophilic bacteria of the Acidithiobacillus genus. CdS QDs were obtained by exposing A. ferrooxidans, A. thiooxidans and A. caldus cells to sublethal Cd2+ concentrations in the presence of cysteine and glutathione. The fluorescence of cadmium-exposed cells moves from green to red with incubation time, a characteristic property of QDs associated with n

Hydrated silica exterior produced by biomimetic silicification confers viral vaccine heat-resistance.

Heat-lability is a key roadblock that strangles the widespread applications of many biological products. In nature, archaeal and extremophilic organisms utilize amorphous silica as a protective biomineral and exhibit considerable thermal tolerance. Here we present a bioinspired approach to generate thermostable virus by introducing an artificial hydrated silica exterior on individual virion. Similar to thermophiles, silicified viruses can survive longer at high temperature than their wild-type relatives. Virus inactivation assays showed that silica hydration exterior of the modified virus effe

Characterization of a plant S-adenosylmethionine synthetase from Acacia koa.

The Acacia koa S-adenosylmethionine (SAM) synthetase was identified from transcriptome data and cloned into the T7-expression vector pEt14b. Assays indicate a thermoalkaliphic enzyme which tolerates conditions up to pH 10.5, 55 °C and 3 M KCl. In vitro examples of plant SAM-synthetase activity are scarce, however this study provides supporting evidence that these extremophilic properties may actually be typical for this plant enzyme. Enzyme kinetic constants (Km = 1.44 mM, Kcat = 1.29 s-1, Vmax 170 μM. min-1) are comparable to nonplant SAM-synthetases except that substrate inhibition was not a

Scalable and Consolidated Microbial Platform for Rare Earth Element Leaching and Recovery from Waste Sources.

Chemical methods for the extraction and refinement of technologically critical rare earth elements (REEs) are energy-intensive, hazardous, and environmentally destructive. Current biobased extraction systems rely on extremophilic organisms and generate many of the same detrimental effects as chemical methodologies. The mesophilic methylotrophic bacterium Methylobacterium extorquens AM1 was previously shown to grow using electronic waste by naturally acquiring REEs to power methanol metabolism. Here we show that growth using electronic waste as a sole REE source is scalable up to 10 L with cons

Thermostability Engineering in Therapeutic Antioxidant Enzymes: From Molecular Fundamentals to Oxidative Stress Applications.

The efficacy of enzyme therapy is limited by their poor stability under physiological conditions. Thermostable enzymes, derived from extremophilic organisms or generated by advanced protein engineering, offer a revolutionary solution to this long-standing challenge. They are widely used in industrial biocatalysis. Their therapeutic applications are poorly investigated and spread across diverse disciplines. While most applications are in the preclinical stages, emerging evidence from animal models demonstrates proof-of-concept for thermostable antioxidant enzymes in cardiovascular, neurodegener

A Tardigrade-Derived CAHS-Motif Enables Glycerol-Free Red Blood Cell Cryopreservation with Enhanced Low-Temperature Trehalose Loading and Synergistic Suppression of Ice Damage.

High-concentration glycerol, the clinical standard for red blood cell (RBC) cryopreservation, requires laborious deglycerolization and causes significant hemolysis, whereas trehalose is biocompatible but poorly permeable to the RBC membrane. Herein, we report a tardigrade-inspired, glycerol-free strategy for RBC cryopreservation based on a conserved CAHS-derived peptide motif. Systematic sequence analysis identified a minimal CAHS-motif that possesses an intrinsic helical propensity and adopts a stabilized amphipathic α-helical conformation under dehydration-mimicking conditions. Isolation and characterization of biosurfactant-producing bacteria from the Thar Desert.

The Thar Desert, one of the most extreme arid ecosystems, harbours a diverse and resilient microbial community adapted to high temperatures, low moisture, and osmotic stress. In this study, soil samples collected from sand dunes in Churu, Rajasthan, were explored for the isolation of biosurfactant-producing bacteria. A total of ten morphologically distinct bacterial isolates were obtained and screened for biosurfactant activity using drop collapse (DC), emulsification index percentage (EA24) %, surface tension (ST), and interfacial tension (IFT) assays. Among them, three isolates S4, S6, and S

Challenges, adaptations, and biotechnological potential of oligophilic bacteria.

The majority of the prokaryotic microorganisms, especially bacteria cannot be cultivated under laboratory conditions. Irrespective of environmental samples, direct count of prokaryotes always exceeds viable count, a condition popularly coined as "Great Plate count anomaly". Our inability to culture them in isolation under laboratory conditions is due to many reasons. One possible reason could be the existence of a special group of extremophilic bacteria, such as, oligophilic bacteria, a group, that loves to grow in nutrient-deficient conditions. They are slow growing, showing unique physiologi
