Steroids with anti-inflammatory, anti-allergic, endocrine-regulating, and other pharmaceutical activities represent the second most widely used class of drugs worldwide, following antibiotics. Their industrial production primarily relies on Mycobacteria-mediated biotransformation of sterols into key intermediates, followed by chemical or enzymatic modifications. While the sterol metabolic pathways in Mycobacteria have been intensively studied, the identification and functional characterization of key enzymes, particularly cytochrome P450 enzymes (CYPs or P450s) and their cognate redox partners

Hydrogenases are central to energy conservation and CO2 fixation in acetogenic bacteria during H2-driven autotrophic growth. The model acetogenic bacterium Clostridium ljungdahlii encodes multiple hydrogenases, with the NADP+- and ferredoxin-dependent electron-bifurcating [FeFe]-hydrogenase HytCBDE1AE2 presumed to be primary. In this study, the hydrogenase catalytic subunit of the HytCBDE1AE2 complex, HytA was knocked out by the CRISPR/Cas9-mediated mutagenesis. Unexpectedly, the ΔhytA mutant exhibited significantly enhanced autotrophic growth on H2/CO2, achieving a ∼ 3-fold higher cell densit

Osteoporosis, a common orthopedic disease predominantly caused by estrogen deficiency in postmenopausal women, continues to pose a significant public health challenge due to the poorly understood molecular mechanisms. While cuproptosis has been implicated in various pathological conditions, its concrete role in the pathogenesis of osteoporosis remains unknown. Equally ambiguous remains the functional role of Sirtuin 5 (SIRT5), a mitochondrial deacylase with well-characterized involvement in aging and bone formation, in estrogen deficiency-associated osteoporosis. In the present study, we ident

Cytochrome P450 CYP152s catalyze decarboxylation of fatty acids to generate terminal alkenes, valuable compounds for various industries. Here, we identified, overexpressed, and characterized a new CYP152 enzyme from Lacicoccus alkaliphilus (OleTLA) and compared its biophysical and biochemical properties with the well-studied OleTJE from Jeotgalicoccus sp. 8456. Improved expression protocols gave the highest yields of CYP152 holoenzymes reported to date. OleTLA exhibits twice the catalytic turnover number of OleTJE when using hexadecanoic acid and H2O2 as substrates in 10% (v/v) ethanol (EtOH).

An analysis of the paramagnetic components present in mitochondria isolated from the poky mutant of Neurospora crassa is described. The study was undertaken with a view to shedding light on the nature of the cyanide- and antimycin A-resistant alternative terminal oxidase which is present in these preparations. Of the ferredoxin-type iron-sulfure centers, only Centers S-1 and S-2 of succinate dehydrogenase could be detected in significant quantities. Paramagnetic centers attributable to Site I were virtually absent. In the oxidized state, at least two 'high potential iron sulfur' centers could

CYP106A2 is one of only a few known steroid hydroxylases of bacterial origin, which might be interesting for biotechnological applications. Despite the enzyme having been studied for more than 30 years, its physiological function remains elusive. To date, there have been no reports of the redox potential of CYP106A2, which was supposed to be unusually low for a cytochrome P450. In this work we show that cyclic voltammetry is not only suitable to determine the redox potential of challenging proteins such as CYP106A2, measured at -128 mV vs. NHE, but also to study molecular interactions of the e

Most disorders of steroidogenesis, such as forms of congenital adrenal hyperplasia (CAH) are caused by mutations in genes encoding the steroidogenic enzymes and are often recognized clinically by cortisol deficiency, hyper- or hypo-androgenism, and/or altered mineralocorticoid function. Most steroidogenic enzymes are forms of cytochrome P450. Most P450s, including several steroidogenic enzymes, are microsomal, requiring electron donation by P450 oxidoreductase (POR); however, several steroidogenic enzymes are mitochondrial P450s, requiring electron donation via ferredoxin reductase (FDXR) and

Fhit protein is lost in cancers of most, perhaps all, cancer types; when restored, it can induce apoptosis and suppress tumorigenicity, as shown in vitro and in mouse tumor models in vivo. Following protein cross-linking and proteomics analyses, we characterized a Fhit protein complex involved in triggering Fhit-mediated apoptosis. The complex includes the heat-shock chaperonin pair, HSP60/10, which is likely involved in importing Fhit into the mitochondria, where it interacts with ferredoxin reductase, responsible for transferring electrons from NADPH to cytochrome P450 via ferredoxin, in ele

Biofuel production from lignocellulosic biomass offers a transformative solution to reduce global fossil fuel dependency. Certain thermophilic anaerobes, including Clostridium thermocellum, show promise for renewable ethanol production due to their ability to break down plant material at high temperatures. However, achieving commercially viable ethanol yields has proven challenging despite extensive engineering efforts. Here, we characterized 27 ferredoxin:NADP+ oxidoreductase (Fnor) enzymes for their enzyme activity, nicotinamide cofactor specificity, thermotolerance, and functional expressio

Iron is a highly abundant element, essential for life on earth and integral to various biological processes. It originated from stellar nucleosynthesis, particularly in supernovae type Ia, which produced iron used in planetary formation. Life's emergence on earth, about 3.5 billion years ago, occurred in an oxygen-free atmosphere, with iron playing a pivotal role in early biochemical reactions. Hydrothermal vents, rich in iron-sulfur (Fe-S) minerals, provided the conditions for prebiotic chemistry and CO2 fixation, potentially driving the synthesis of key organic compounds. Iron-sulfur cluster

Nitroanisoles are used widely as synthetic intermediates and explosives. Although bacteria have been reported to degrade 4-nitroanisole (4NA) under aerobic conditions, the key enzymes and the catalytic mechanism have remained elusive. Rhodococcus sp. strain JS3073 was isolated for its ability to grow on 4NA as the sole carbon and energy source. In this study, whole cell biotransformation experiments indicated that 4NA degradation is initiated by O-demethylation to form 4-nitrophenol (PNP), which undergoes subsequent degradation by a previously established pathway involving formation of 1,2,4-b

CONTEXT: The cholesterol side-chain cleavage enzyme P450scc, encoded by CYP11A1, converts cholesterol to pregnenolone to initiate steroidogenesis. P450scc deficiency can disrupt adrenal and gonadal steroidogenesis, resembling congenital lipoid adrenal hyperplasia clinically and hormonally; only 12 such patients have been reported previously.
OBJECTIVE: We sought to expand clinical and genetic experience with P450scc deficiency.
PATIENTS AND METHODS: We sequenced candidate genes in 7 children with adrenal insufficiency who lacked disordered sexual development. P450scc missense mutations were

Rat alveolar macrophages incubated with soybean trypsin inhibitor and beta-amylase produced nitrite in a dose- and time-dependent manner. This production depends on the presence of L-arginine (L-arg) in the culture medium. The precursor of this nitrite was demonstrated as being nitric oxide by bleaching ferredoxin at 410 nm when added to the culture medium. NG-Monomethyl-L-arginine and the tetrahydrobiopterin biosynthesis inhibitor 2,4-diamino-6-hydroxypyrimidine inhibited the release of nitrite in a dose-dependent manner. Dexamethasone was able to modulate this release. These data indicate th

Microbial reduction of soluble selenium oxyanions is a sustainable strategy for remediating selenium-contaminated environments, yet the molecular mechanisms underlying selenite tolerance in the genus Rhodococcus remain poorly understood. In this study, we investigated the proteomic adaptation of the highly tolerant strain Rhodococcus qingshengii PM1 under high-concentration selenite stress (50 mM Na2SeO3) using a data-independent acquisition (DIA)-based quantitative proteomics approach. A total of 3335 proteins were identified, and 3310 proteins were retained for downstream analysis. Comparati

Defects in iron-sulphur [Fe-S] cluster biogenesis are increasingly recognized as causing neurological disease. Mutations in a number of genes that encode proteins involved in mitochondrial [Fe-S] protein assembly lead to complex neurological phenotypes. One class of proteins essential in the early cluster assembly are ferredoxins. FDX2 is ubiquitously expressed and is essential in the de novo formation of [2Fe-2S] clusters in humans. We describe and genetically define a novel complex neurological syndrome identified in two Brazilian families, with a novel homozygous mutation in FDX2. Patients

BACKGROUND: The spread of artemisinin (ART)-resistant Plasmodium falciparum threatens the control of malaria. Mutations in the propeller domains of P. falciparum Kelch13 (k13) are strongly associated with ART resistance. Ferredoxin (Fd), a component of the ferredoxin/NADP+ reductase (Fd/FNR) redox system, is essential for isoprenoid precursor synthesis in the plasmodial apicoplast, which is important for K13-dependent hemoglobin trafficking and ART activation. Therefore, Fd is an antimalarial drug target and fd mutations may modulate ART sensitivity. We hypothesized that loss of Fd/FNR functio

Pseudomonas aeruginosa JB2 can use 2-chlorobenzoate (2-CBa), 3-CBa, 2,3-dichlorobenzoate (2,3-DCBa), and 2,5-DCBa as sole carbon and energy sources, whereas strain 142 can only grow on 2-CBa and 2,4-DCBa. Both strains, however, harbor the same halobenzoate 1,2-dioxygenase (ohbAB) and chlorocatechol (clcABD) degradation genes necessary for the metabolism of ortho-CBas. In addition, the hybABCD operon, encoding a salicylate 5-hydroxylase, is also found in both strains. The expression of ohbAB, hybABCD, and clcABD operons was measured in cultures grown on different CBas as the sole carbon source

Biological conversion of one-carbon (C1) feedstocks frequently requires coupling oxidative and reductive reactions through an appropriate electron-transfer pathway. The combination of carbon monoxide dehydrogenase (CODH), which oxidizes CO to CO2, and formate dehydrogenase (FDH), which reduces CO2 to formate, serves as a representative model for examining this requirement. CODH-FDH coupling has been investigated in cell-free systems, whereas intracellular and distributed configurations provide emerging or prospective routes for extending this reaction across different biological environments.

Ferredoxin 1 (FDX1) emerges as a crucial regulator of autophagy and copper metabolism in ovarian cancer (OC), as revealed by this investigation. Predominantly localized to the cytoplasm and mitochondria, FDX1 coordinates autophagic activity by modulating the AMPK and mTOR signaling pathways. Its role extends to preserving mitochondrial integrity and facilitating sulfation of DLAT/DLST, ensuring effective autophagic flux. Knockdown of FDX1 disrupts these processes, exacerbating mitochondrial dysfunction. In vivo studies further demonstrate that overexpressing FDX1, combined with Compound C trea
