Analytical method for fast screening and confirmation of multi-class veterinary drug residues in fish and shrimp by LC-MS/MS.

A multi-class, multi-residue analytical method based on LC-MS/MS detection was developed for the screening and confirmation of 28 veterinary drug and metabolite residues in flatfish, shrimp and eel. The chosen veterinary drugs are prohibited or unauthorised compounds in Korea, which were categorised into various chemical classes including nitroimidazoles, benzimidazoles, sulfones, quinolones, macrolides, phenothiazines, pyrethroids and others. To achieve fast and simultaneous extraction of various analytes, a simple and generic liquid extraction procedure using EDTA-ammonium acetate buffer and

Involvement of growth hormone-insulin-like growth factor I system in cranial remodeling during halibut metamorphosis as indicated by tissue- and stage-specific receptor gene expression and the presence of growth hormone receptor protein.

The role of growth hormone (GH) and insulin-like growth factor-I (IGF-I) in the tissue remodeling associated with the transition of a symmetrical larva to an asymmetrical juvenile during flatfish metamorphosis is unknown. In order to investigate the potential role of these hormones in the remodeling of cranial bone and soft tissue that accompanies eye migration during metamorphosis of Atlantic halibut (Hippoglossus hippoglossus) larvae, tissue-specific gene expression was monitored by in situ hybridization for Atlantic halibut type I growth hormone receptor (hhGHR), type II hhGHR, and insulin-

Insemination and population density in Entobdella soleae, a monogenean skin parasite of the common sole, Solea solea.

Immature specimens of the monogenean skin parasite Entobdella soleae, experimentally isolated from other individuals, fail to self-inseminate when maturity is reached and self-insemination has not been recognized during many hours spent observing living parasites. E. soleae lives on the lower surface of its flatfish host, the common sole (Solea solea), in relatively small numbers, most commonly 1, 2 or 3 adults per fish, but in spite of the low density and extensive habitat, freshly collected wild parasites were found to be inseminated. If it is assumed that E. soleae is unable to self-insemin

Action on ileal smooth muscle of synthetic detergents and pardaxin.

Pardaxin (PX), a toxic and repellent substance isolated from the Red Sea flatfish, causes a sharp ball-like profile of drop of saline placed on a hydrophobic film to turn into a flattened one. This effect results with a decrease of the contact angle (theta) from 96 degrees to a maximum of 42 degrees at 10(-4) M of PX. The action of sodium dodecyl sulphate (SDS), a synthetic anionic detergent, benzalkonium chloride (BAC) cationic detergent and pardaxin (PX) a toxic protein with detergent properties, were studied in the ileal guinea-pig longitudinal smooth muscle preparation. SDS (4 X 10(-4) M)

Multifactorial neural disruption in the brain of the Senegalese sole (Solea senegalensis) under ocean acidification.

Global ocean acidification, driven by rising atmospheric CO2, threatens marine ecosystems and biodiversity, with increasing evidence of disruptive effects on fish neurobiology and behaviour. However, the precise mechanisms underlying these impacts remain largely unresolved. Here, we reveal how chronic exposure to future-predicted CO2 levels disrupts brain function in the marine teleost Solea senegalensis. Using an integrative approach combining electrophysiology, immunohistochemistry and transcriptomics, we demonstrate that elevated CO2 induce a complex multifaceted disruption in brain physiolo

Histopathological and functional evidence of intestinal mucosal disruption in emaciated olive flounder (Paralichthys olivaceus) infected with Enteromyxum leei.

Emaciation disease associated with Enteromyxum leei infection is a major health concern in olive flounder (Paralichthys olivaceus) aquaculture, but the structure-function basis underlying this condition remains incompletely understood. In this study, we investigated whether emaciation in E. leei-infected olive flounder is linked to intestinal mucosal disruption and impaired digestive and absorptive function. Fish were classified based on external appearance, relative condition factor, and polymerase chain reaction-based detection of E. leei. Infected fish exhibited markedly lower relative cond

Isolation and Characterisation of Serotype III in Miamiensis avidus (Ciliophora: Scuticociliatida) From Olive Flounder (Paralichthys olivaceus).

Miamiensis avidus (Ciliophora: Scuticociliatida, syn. Philasterides dicentrarchi) is a significant pathogen causing scuticociliatosis in olive flounder (Paralichthys olivaceus) in Korea and Japan. In this study, the BB19 strain, isolated from olive flounder, was identified as M. avidus through phylogenetic analysis of cytochrome c oxidase subunit 1 (cox1) sequences and classified as cox1 genotype IV. Serotype-specific PCR confirmed BB19 as serotype III. Agglutination assay and indirect ELISA also showed that BB19 exclusively reacted with anti-serotype III (Mie0301 and BB19) sera, with no cross

Scoliosis and kyphosis in blue-spotted and marbled flathead fish associated with a Myxobolus acanthogobii-like parasite.

Spinal deformities in finfish have the potential to impact aquaculture industries and wild populations by increasing morbidity, mortality, and reducing growth rates. Myxobolus acanthogobii has been implicated in causing scoliosis and lordosis in various aquatic species in Japan. We investigated 4 cases of spinal deformity in 2 flathead (Platycephalus) species that were submitted to the Elizabeth Macarthur Agricultural Institute (EMAI) in New South Wales (NSW), Australia, between 2015 and 2021. Flathead are commercially significant species that are popular among Australian consumers, and are al

Regulatory mechanism of miR-722 on C5aR1 and its functions against bacterial inflammation in half-smooth tongue sole (Cynoglossus semilaevis).

MicroRNAs (miRNAs) are small non-coding RNAs involved in various biological processes, including immunity. Previously, we investigated the miRNAs of half-smooth tongue sole (Cynoglossus semilaevis) and found that miR-722 (designated Cse-miR-722) was significantly differentially expressed after infection with Vibrio anguillarum, reflecting its importance in immune response. Our preliminary bioinformatic analysis suggested that Cse-miR-722 could target C5aR1 (designated CsC5aR1), which was known to play crucial roles in complement activation and inflammatory response, as a receptor of C5a. A new species of Neoheterobothrium Price, 1943 (Monogenea, Diclidophoridae) from Syacium papillosum (Linnaeus) (Pleuronectiformes, Paralichthyidae) in the Yucatan Shelf, with notes on the validity of the subfamilies in the Diclidophoridae.

Neoheterobothrium papillosum n. sp. (Monogenea: Diclidophoridae) is described based on specimens collected from the gills of the dusky flounder Syacium papillosum (Linnaeus) (Paralichthyidae) in the Yucatan Shelf. The new species can be differentiated from other congeneric species by the following characteristics: (1) number of hooks in the genital atrium (8 hooks), (2) number of testes (20-34), (3) pharynx size, (4) peduncle length, and (5) egg size. Neoheterobothrium papillosum n. sp. resembles Neoheterobothrium syacii; however, detailed morphological analysis allowed for their separation in

Effect of triploidy on digestive enzyme activity of early stages of turbot (Scophthalmus maximus).

In the present work, growth and digestive enzyme activities of total acid and alkaline proteases, pepsin, trypsin, lipase, and α-amylase, as well as partial characterization of enzyme activity, were studied in diploid and triploid turbot. Growth was similar between both groups. Acid protease activity increased consistently during the experiment, for both diploid (2n) and triploid (3n) fish. The alkaline protease activity was always higher for triploids throughout the experiment. Proteolytic acid activity (pH 2) was generally higher for diploids, at all temperatures tested. Higher activity was

Hematology-centered characterization of systemic physiological alterations during viral haemorrhagic septicemia virus (VHSV) infection in olive flounder (Paralichthys olivaceus).

Viral haemorrhagic septicemia virus (VHSV) is a major viral pathogen that causes severe haemorrhagic disease and hematological disturbances in olive flounder (Paralichthys olivaceus). However, integrated hematology-centered analyses of infection-associated blood alterations remain limited. In the present study, temporal changes in viral replication, hematological parameters, blood cell morphology, biochemical responses, and gene expression were analyzed following VHSV infection in olive flounder. VHSV infection caused dose-dependent mortality, reaching 100% cumulative mortality at 106-107 50%

Genome-wide characterization of integrin (ITG) gene family and their expression profiling in half-smooth tongue sole (Cynoglossus semilaevis) upon Vibrio anguillarum infection.

Integrins (ITGs) are transmembrane heterodimer receptors with ITGα subunit and ITGβ subunit, participating in various physiological processes, including immunity. At present, systematic research on ITGs in teleost is scarce, especially in half-smooth tongue sole (Cynoglossus semilaevis). In this study, a set of 28 ITG genes in half-smooth tongue sole have been identified and characterized. The phylogenetic analysis showed that ITGα and ITGβ subunits were respectively classified into five and two clusters, consistent with previous studies. The selection pressure analysis indicated that most of

Mucus piRNAs profiles of Vibrio harveyi-infected Cynoglossus semilaevis: A hint for fish disease monitoring.

The half-smooth tongue sole, Cynoglossus semilaevis, is an important cultured flatfish species. Vibrio harveyi is a common pathogen to this fish, which may result in great economic loss to C. semilaevis culture industry. piRNAs, a non-coding RNAs with 26-32 nt, have been regarded as promising biomarkers for cancer diagnosis and fish diseases. Here, we extracted the RNA from mucus of C. semilaevis and constructed the differential expression profiles of piRNAs between the sick fish (MS) and healthy fish (MC). We identified 45,696 differentially expressed piRNAs including 22,735 up-regulated piRN

Significant decrease in common dab (Limanda limanda) hepatic neoplasms revealed by long-term environmental monitoring.

The prevalence of hepatic neoplasms (carcinogenesis) in the common dab (Limanda limanda) flatfish was investigated using data collected between 2004 and 2020 from five biogeographic UK marine regions including the Northern North Sea, Southern North Sea, Irish Sea, Eastern Channel, and Western Channel and Celtic Sea. Our assessment considered age-related confounding effects, providing an improved estimation of how hepatic neoplasm prevalence differs between regions and over time. The data indicated a significant decline in prevalence in both the Irish Sea and Southern North Sea regions, with a

Determination of Flunixin and 5-Hydroxy Flunixin Residues in Livestock and Fishery Products Using Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS).

Flunixin is a veterinary nonsteroidal anti-inflammatory agent whose residues have been investigated in their original form within tissues such as muscle and liver. However, flunixin remains in milk as a metabolite, and 5-hydroxy flunixin has been used as the primary marker for its surveillance. This study aimed to develop a quantitative method for detecting flunixin and 5-hydroxy flunixin in milk and to strengthen the monitoring system by applying to other livestock and fishery products. Two different methods were compared, and the target compounds were extracted from milk using an organic sol

Characterization of pax3a and pax3b genes in artificially induced polyploid and gynogenetic olive flounder (Paralichthys olivaceus) during embryogenesis.

Although chromosome set manipulation techniques including polyploidy induction and gynogentic induction in flatfish are becoming increasingly mature, there exists a poor understanding of their effects on embryonic development. PAX3 plays crucial roles during embryonic myogenesis and neurogenesis. In olive flounder (Paralichthys olivaceus), there are two duplicated pax3 genes (pax3a, pax3b), and both of them are expressed in the brain and muscle regions with some subtle regional differences. We utilized pax3a and pax3b as indicators to preliminarily investigate whether chromosome set manipulati

A cytochrome P4501B gene from a fish, Pleuronectes platessa.

Tetrapod cytochrome P4501 family (CYP1A1, CYP1A2 and CYP1B1) enzymes are most active in hydroxylating a variety of environmental contaminants including polyaromatic hydrocarbons (PAH), planar polychlorinated biphenyls and arylamines and thus play a pivotal role in the toxicology of these compounds. Mammalian CYP1A1 and CYP1A2 genes appear to have diverged after the evolutionary emergence of mammals, whereas fish species apparently possess only one CYP1A family gene, and fish CYP1A enzymes exhibit properties of both of the mammalian isoforms. We have isolated a further CYP1 family gene from a m

Genomic selection accelerates genetic improvement of tolerance to visceral nodular disease in the half-smooth tongue sole (Cynoglossus semilaevis).

The half-smooth tongue sole (Cynoglossus semilaevis) is an economically important marine fish in China, but its vulnerability to visceral nodular disease (VND) caused by Mycobacterium marinum poses a significant threat to aquaculture. Based on traditional breeding methods, it is slowly to improve disease tolerance and then may lead to inbreeding depression. This study aimed to accelerate the development of VND-tolerant strains of C. semilaevis through genomic selection (GS). In this study, individuals from both reference and candidate populations were genotyped, and more than 7 million high-qu
