Win-Win Integration Strategy to Construct Multifunctional Fluorescence Hydrogels for On-Site Monitoring of Food Freshness Via Nucleophilic Substitution.

Fluorescence hydrogels have become attractive candidates for monitoring food freshness; however, the ability to improve anti-interference, broaden the color variation range, and introduce multifunctionality remains limited. Here, an ingenious fluorescence hydrogel design strategy for monitoring food freshness is proposed via integrating an OH--responsive dye into functional matrices. By adjusting the intermolecular charge transfer and π-π stacking effect, nitrobenzoxadiazaole-based dye BOD-R1 was prepared and responded to biogenic amines via nucleophilic substitution. Embedding BOD-R1 in benze

A Turn-On Fluorescent Chemical Sensor Based on Benzo[f]Chromen Structure for Cu2+-Sensitive Detection as Well as Applications.

A simple Schiff base probe (E)-N'-(1-(3-oxo-3H-benzo[f]chromen-2-yl)ethylidene) picolinohydrazide (CHH) was synthesized and characterized by Fourier transform infrared spectroscopy (FT-IR), 1H nuclear magnetic resonance spectra (1H NMR), 13C nuclear magnetic resonance spectra (13C NMR), and high-resolution mass spectrometry (HR-MS). The probe CHH shows high sensitivity and selectivity for the recognition of Cu2+ in EtOH-H2O (1:1, v/v), and exhibits an enhanced fluorescence phenomenon. The binding constant of CHH for Cu2+ is 2.74 × 105 M-1, and the binding stoichiometry of CHH and Cu2+ is 1:1.

Fabrication of Novel Yellow-Fluorescent Carbon Dots and Their Application for Fe³⁺ Detection.

Sensitive, rapid, and cost-effective detection of Fe³⁺ is urgently required for biological research, food safety, and environmental monitoring. Herein, novel yellow-fluorescent carbon dots (CDs) were successfully synthesized via a facile one-step solvothermal method using ursolic acid and o-phenylenediamine as green precursors. The as-prepared CDs, with an average particle size of 4.82 nm, exhibit bright yellow fluorescence (λem = 540 nm) and high selectivity toward Fe³⁺. The fluorescence intensity is linearly quenched by Fe³⁺ in the range of 0-150 µmol/L, with a low detection limit of 0.096 µ

Angelica dahurica-derived carbon dots as a sensitive fluorescent probe for Fe3+: investigation of quenching mechanism and ascorbic acid-induced fluorescence recovery.

Biomass carbon dots (CDs) have attracted more attention owing to their low cost, environmental sustainability, better biocompatibility, and low toxicity. However, it remains a major challenge to obtain CDs with high stability and superior luminescence properties. Herein, Angelica dahurica powder, a renewable and abundant Chinese herbal resource, was selected as a biomass carbon source, and high-performance blue fluorescence-emitting carbon dots (AD-CDs) were prepared by a one-step solvothermal method. Comprehensive characterizations of the composition and optical properties reveal that the AD-

Chemoselective and quantitative detection of acrolein based on pyrene excimer fluorescence via cycloaddition reaction of conjugated imine.

Acrolein is a highly reactive and toxic aldehyde generated in various biological and environmental contexts, including tobacco combustion and cellular oxidative stress. Herein, we present a novel quantitative method for acrolein detection using Probe A, which features an ethanolamine skeleton and a pyrene moiety. This method employs a 4 + 4 dimerization reaction, enabling selective interaction with acrolein, resulting in excimer fluorescence emission proportional to the acrolein concentration. This approach offers high sensitivity and selectivity across a broad dynamic range from micromolar to

Fluorescence-colorimetric dual-mode detection of cysteine in artificial urine with potential relevance to cervical cancer.

As cervical cancer poses a serious threat to the health of women, non-invasive and convenient diagnostic methods are still in urgent need. With cysteine (Cys) in urine as the disease marker, a xanthylium-based colorimetric and fluorescent probe SH-OH-NBD for Cys detection was constructed and its response to Cys was explored in artificial urine rather than common buffer solutions. Upon the action of Cys, the nitrobenzoxadiazole ether in SH-OH-NBD was cleaved, generating a new fluorescence peak at 567 nm. Thus, the concentration of Cys could be accurately determined based on the turn-on fluoresc

A cooperative sequential binding rhodamine-based fluorescent probe for total copper sensing: From environmental monitoring to cancer cell imaging.

BACKGROUND: Copper is an essential trace element involved in various biological processes. However, disruption of copper homeostasis can have adverse effects on human health. Selective and sensitive detection of copper ions in both oxidation state (Cu2+ or Cu+) is therefore important, as Cu2+ is toxic at elevated levels, while intracellular Cu+ levels are often elevated in cancer cells. Because Cu2+ and Cu+ predominate in extracellular (oxidizing) and intracellular (reducing) environments, respectively, a single probe capable of detecting both species would be particularly valuable. RESULTS:

A Fluorescent Probe for Carboxylesterases Detection in Estradiol Valerate-Treated Inflammation Alleviation.

In this work, a fluorescent probe PIPM-CE was developed for the carboxylesterases (CE) detection in the estradiol valerate (EV)-treated inflammation alleviation. Under the excitation at 368 nm, the notable fluorescence signal was found at 479 nm. The confirmation on the working conditions suggested the rapid recognition reaction process and stable reporting signal. The probe also showed relatively high sensitivity, high specificity, and low cytotoxicity. By the construction of the basic correlation between the CE level and the fluorescence signal intensity, PIPM-CE achieved imaging the EV-trea

Dissecting the pathological basis of brain disorders with genetically encoded fluorescent neurochemical sensors.

Brain disorders exhibit profound spatiotemporal complexity, involving intricate disruptions in synaptic transmission and neurochemical signaling. A precise understanding of these dynamic processes is crucial for developing effective therapeutics but remains technically challenging. Genetically encoded fluorescent sensors have emerged as transformative tools to bridge this gap by enabling the real-time monitoring of neurochemical events with high specificity. This review highlights how cutting-edge single-fluorophore biosensors for neurochemicals are being applied to dissect the pathological me

Protein Melting Biosensors for Label-Free and Self-Reporting of Iron-Binding Proteins and Iron Ion.

Protein fluorescence sensing holds great promise for food safety, disease diagnosis, and environmental monitoring. Nevertheless, the synthesis of specific fluorescent dye probes is usually necessary, which significantly increases the cost and complexity of the synthesis. Herein, we developed a label-free, melting-based protein sensing strategy. The melting biosensors monitor the change in fluorescence intensity produced by aromatic fluorescent amino acids during protein unfolding and determine the melting temperature (Tm) of the protein. Based on that, we developed a Fe3+-associated Tm-regulat

A near-infrared ratiometric fluorescent probe for visual sensing of H2S and monitoring its fluctuation in plant roots under drought and flooding stresses.

BACKGROUND: Hydrogen sulfide (H2S) is a key endogenous gasotransmitter involved in plant physiological regulation and stress responses. Monitoring its dynamic changes in plants is essential for understanding related signaling mechanisms. However, due to its chemical instability and the complexity of plant tissues, developing a reliable method for accurate quantification and real-time visualization of H2S in living plants remains challenging. Although fluorescent probes have been developed for H2S imaging, most still face critical limitations, such as emission in the visible region being suscep

Antioxidative and Robust Fluorescent Hydrogel for Deep Learning-Assisted Non-Destructive Detection of Hg2.

Flexible fluorescent hydrogels offer great potential for on-site heavy-metal sensing, but their practical use is often limited by a fundamental trade-off: materials that provide strong fluorescence responses typically suffer from poor stability in oxidative environments, while hydrogel matrices with good robustness or adhesion rarely support reliable fluorescence signaling. Here, we introduce a phycocyanin-polyacrylamide (PC-PAAM) hydrogel that resolves this conflict by integrating high mechanical flexibility and strong underwater adhesion with stable red fluorescence and intrinsic antioxidati

Recent Advances in the ESIPT-based Responsive Probes for the Detection of Bio-Active Molecules CO, NO and H₂S.

Excited-state intramolecular proton transfer (ESIPT)-based fluorescent probes have emerged as powerful tools for bioimaging and chemical sensing owing to their large Stokes shifts, dual-emission characteristics, high sensitivity, and reduced self-absorption effects. These unique photophysical properties have facilitated the development of advanced fluorescent probes for the detection of biologically important gasotransmitters, including carbon monoxide (CO), nitric oxide (NO), and hydrogen sulfide (H2S), which play crucial roles in numerous physiological and pathological processes. A Sensitive Green-Red FRET Sensor for Calpain-1 Utilizing Heterodimeric Fluorescent Proteins from Distinct Evolutionary Origins.

Calpain-1, a calcium-dependent cysteine protease, plays a pivotal role in various biological functions. However, real-time monitoring of its activity has been limited by the lack of high-performance calpain sensors. Here, we report the development of iFoCAL, a sensitive and specific fluorescence resonance energy transfer (FRET) sensor for calpain-1, which employs mClover3 and mScarletX, a newly engineered green-red FRET pair with weak interactions. Using iFoCAL, we revealed distinct spatiotemporal patterns of calpain-1 activation: gradual and global activation in whole cells during endoplasmic

A Double-Thiadiazole-Derived Fluorescent Probe for the Cyclooxygenase-2 Detection in Colorectal Tumor-Inflammation Interaction.

Cyclooxygenase-2 (COX-2) is an attractive biomarker for the early diagnosis in colorectal tumor. The previous fluorescent probes for the COX-2 detection relied on the inhibitors. The affection on the enzymatic activity may impact the accuracy. Herein, a double-thiadiazole-derived fluorescent probe BTD-COX2 was developed for the COX-2 detection in colorectal tumor-inflammation interaction. The design concept avoided the direct use of the moieties from the COX-2 inhibitors, and selected the thiadiazole subunit to construct the interaction. BTD-COX2 responded to COX-2 with the fluorescence enhanc

Interference-reduced detection of 1,2-dihydroxybenzene using an acid-tolerant luminescent probe.

BACKGROUND: Plant polyphenols, particularly flavonoids and phenolic acids containing 1,2-dihydroxybenzene structures, are bioactive compounds with health benefits, but their detection in acidic fruit matrices (pH 4-6) remains challenging. Conventional boronic acid-based probes are often limited by their instability in acidic media because ofthe susceptibility of ester linkages to hydrolysis. Meanwhile, although HPLC and LC‒MS techniques offer excellent sensitivity and selectivity, they remain labor-intensive and frequently encounter challenges in mitigating complex matrix interference. A Galactose-Modified Light-Activatable Persulfurated Arene Fluorescent Probe for Targeted Imaging of Polarity and Viscosity in Liver Cancer Cells.

Monitoring intracellular physiological changes, particularly the dysregulation of key microenvironmental parameters like viscosity and polarity, provides critical functional insights into diseased states. Herein, we report a fluorescent probe, HTB-βGal, based on persulfurated arenes and sensitive to both polarity and viscosity. Modified with galactose, the probe exhibits good water solubility and is initially non-emissive in aqueous solution. Upon light irradiation, its fluorescence gradually increases due to the photoexcitation-induced aggregation (PEIA) characteristic of the persulfurated ar

Ultrabright Carbon Dots as an Emerging Fluorescent Probe for Multivalent Labeling and Highly Sensitive Detection of Extracellular Vesicles.

Extracellular vesicles (EVs) play an important role in many physiological processes, and surface labeling is essential for the research and application of EVs. Fluorescence probes provide a sensitive and visual tool for EV labeling and imaging; however, most of them suffer from some native shortcomings, e.g., false positives caused by the aggregation of hydrophobic organic dyes and membrane destruction in inorganic phosphor labeling. To resolve these issues, in this work, we presented an emerging fluorescent probe, i.e., cholesteryl-modified ultrabright green emissive carbon dots (Chol-GCDs),
