A twenty-week double-blind study was conducted to compare the efficacy and side-effect profile of haloperidol decanoate and fluphenazine decanoate, both given four-weekly, in fifty-one chronic schizophrenic patients. A comparison of fluphenazine decanoate depot injection used alone and fluphenazine decanoate with fluphenazine HCl i.m. was undertaken in South African Xhosa patients with schizophrenia diagnosed according to DSM-III-R. The combined treatment significantly shortened the length of hospital stay to a mean of 30.5 days from a mean of 40.7 days for the single therapy group. The combined treatment group also obtained significantly higher improvement ratings one week after the start of treatment than the single treatment patients.

A sensitive radioimmunoassay was developed to determine the steady-state, fixed-dose pharmacokinetic profile of fluphenazine decanoate in nine outpatients. To see if neuroleptic blood levels measured by this assay correlated with a clinical event, we performed a cross-sectional study of fluphenazine blood levels in 11 patients with and 17 patients without tardive dyskinesia. The results do not support the hypothesis that fluphenazine decanoate serum levels correlate with TD occurrence.

We evaluated the effectiveness and the side effects of what we defined as low (5-mg) and conventional (25-mg) doses of fluphenazine decanoate administered every two weeks in a double-blind comparison. Subjects were 66 patients who fulfilled DSM-III criteria for schizophrenic disorder. Serum neuroleptic levels were measured by radioreceptor assay in 24 schizophrenic patients maintained on fluphenazine decanoate. Clinical state was assessed at the time of neuroleptic level measurement and during the subsequent 6 months.
