Sudden Death of a Toddler With Rubinstein-Taybi Syndrome: Forensic Investigation and Pathophysiological Correlation.

Sudden death in childhood is a particularly demanding subject of forensic investigation, especially when a known genetic syndrome coexists. We present the case of the sudden death of a three-year-old female toddler diagnosed with Rubinstein-Taybi syndrome, who was found pulseless and apneic at home and pronounced dead shortly after being transferred to a children's hospital. A comprehensive forensic investigation was conducted, including an autopsy, histological examination, and toxicological analysis. The histopathological findings revealed extensive lesions of severe bronchopneumonia with ne

A 30-InDel Assay for Genetic Variation and Population Structure Analysis of Chinese Tujia Group.

In the present study, thirty autosomal insertion and deletion polymorphic loci were simultaneously amplified and genotyped in a multiplex system, and their allelic frequencies as well as several forensic parameters were obtained in a sample of 236 unrelated healthy Tujia individuals. All the loci were in Hardy-Weinberg equilibrium after applying a Bonferroni correction and all pair-wise loci showed no significant linkage disequilibrium. These loci were observed to be relatively informative and discriminating, quite efficient for forensic applications. Allelic frequencies of 30 loci were compar

Psychiatry and the unknown future: the period of hope.

Modern psychiatry is transitioning from an entity-based taxonomy toward a systems-oriented science that integrates biological, psychological, social, and existential levels of analysis. We propose a conceptual framework that combines genetics and multi-omics approaches, synergetics, complex systems theory, and a neo-existentialist perspective. Within this framework, psychiatric illness is conceptualized as a crisis of regulation and meaning emerging across dynamically interacting multi-level networks. We discuss implications for clinical practice, including process-based psychotherapy, idiogra

"Shedding the status"-a meta-analysis on the individual propensity to shed skin material in the context of forensic trace analysis.

The so-called "shedder status", which can more precisely be referred to as "individual shedding propensity" (ISP), has been the subject of forensic genetic research for more than two decades. Numerous studies have been published on this topic many of which report contradictory and/or insufficiently documented results regarding the existence, influencing factors, classifications of and test methodologies for the ISP of skin material. To date, there is no scientific consensus on the best way to register and conceptualize this variable, that is essential for the assessment of DNA transfer events.

A comparison of genetic findings in sudden cardiac death victims and cardiac patients: the importance of phenotypic classification.

Sudden cardiac death (SCD) is responsible for a large proportion of non-traumatic, sudden and unexpected deaths in young individuals. Sudden cardiac death is a known manifestation of several inherited cardiac diseases. In post-mortem examinations, about two-thirds of the SCD cases show structural abnormalities at autopsy. The remaining cases stay unexplained after thorough investigations and are referred to as sudden unexplained deaths. A routine forensic investigation of the SCD victims in combination with genetic testing makes it possible to establish a likely diagnosis in some of the deaths

The forensic and clinicopathological spectrum of the vertebral artery.

We report the forensic and clinicopathological spectrum of 14 postmortem cases involving the vertebral artery. In all cases, there was either pontocerebellar infarction (n = 8) or subarachnoid hemorrhage (n = 6). The underlying pathology of the vertebral artery was segmental mediolytic arteriopathy (n = 5), traumatic rupture of the arterial wall (n = 3), arterial dissection (n = 2), or atherosclerosis (n = 4). Histopathologic changes were often present in both the intracranial and extracranial segments of the vertebral artery. In our case series, the most frequent disease in the vertebral arte

[Optimized approach to the study of the dentoalveolar apparatus in forensic identification].

The purpose of the work is to draw parallels between lifetime and posthumous studies of the dentoalveolar apparatus during the forensic examination of personal identification. The material obtained during life on the state of the dentoalveolar system, both in aggregate and separately, is valuable for comparative analysis with bone objects of the jaws delivered for medical and forensic examination. The relevance of the study is associated with increasing cases of terrorism, hostilities, mass graves, natural disasters, when conducting a genetic study of cadaveric material presents certain diffic

Forensic applications of rapidly mutating Y-STR markers: current status and future perspectives.

Over the past few years, the Y chromosome has emerged as a crucial investigative tool, gaining significant applications in multiple disciplines such as forensics, anthropology, and genetics. Standard Y-STRs are particularly useful for identifying male donors in mixed deoxyribonucleic acid (DNA) samples dominated by female DNA, in tracing male patrilineal lineage, and in disaster victim identification. Nonetheless, their limited ability to distinguish closely related males has constrained their forensic utility. Evidence from multiple studies shows that rapidly mutating (RM) Y-STR markers impro

Predicting Homo pigmentation phenotype through genomic data: from Neanderthal to James Watson.

BACKGROUND: Human pigmentation is regulated by several genes acting at different stages of melanin formation. Functional and association studies have elucidated the role of several of these genes in pigmentation phenotypes. Forensic and evolutionary studies can benefit from this knowledge. OBJECTIVES: To evaluate the reliability of the prediction of pigmentation phenotypes using a large database of genetic markers in individuals with known phenotypes; and from this try to predict the pigmentation phenotype of prehistoric Homo specimens and of contemporary individuals whose visible phenotypes

Medicolegal Death Investigation: Coroner and Forensic Pathology Functions and Processes in Victoria, Australia.

The medicolegal death investigation in Victoria, Australia is a traditional coroner system based on the model in England and Wales in the early 20th Century. In 1985, the first of a series of legislative amendments were made that proved the vanguard of reform of the coroners' jurisdictions in Australia. The Victorian Institute of Forensic Medicine (the Institute) was established by the Coroners Act 1985 (Vic.), now the Victorian Institute of Forensic Medicine Act 1985 (Vic.), to provide forensic pathology, medical, and related scientific services needed by the justice system. In addition to de

On the persistence of race: Unique skulls and average tissue depths in the practice of forensic craniofacial depiction.

The (re-)surfacing of race in forensic practices has received plenty of attention from STS scholars, especially in connection with modern forensic genetic technologies. In this article, I describe the making of facial depictions based on the skulls of unknown deceased individuals. Based on ethnographic research in the field of craniofacial identification and forensic art, I present a material-semiotic analysis of how race comes to matter in the face-making process. The analysis sheds light on how race as a translation device enables oscillation between the individual skull and population data,

Pitfalls in body fluid identification - age independent DNA methylation markers for vaginal secretions and menstrual blood in sexual assaults.

In recent years, determination of cellular origin of DNA has been acknowledged as an important tool in forensic genetics. It complements identification of the DNA donor by STR analysis, since it could provide further information about the origin of DNA found at a crime scene. A workflow (body fluid identification (BFI) workflow) developed in 2023 for determination of seven different body fluids in forensic genetic casework by DNA methylation analysis has been very helpful in context with sexual assaults, especially rapes. The verification of vaginal secretions and/or menstrual blood, for which

A melanoma patient with macrophage-cancer cell hybrids in the primary tumor, a lymph node metastasis and a brain metastasis.

In 1911 it was proposed that cancer might result from fusion and hybridization between macrophages and cancer cells. Using immunohistochemistry it was determined that essentially all solid tumors expressed macrophage-like molecules on their cell surface. More recently we have used forensic (STR) genetics that allows one to detect DNA from more than one individual in the same sample. By studying biopsies from individuals receiving allogeneic stem cell transplants and later developed solid tumor metastases, we were able to detect both donor and patient DNA sequences suggesting that hybrids were

From fathers to forebears: Benchmarking Y-chromosomal short tandem repeat haplogroup predictors for forensic paternal lineage inference.

The male-specific Y chromosome plays a pivotal role in population and forensic genetics. Its relatively conserved paternal inheritance and lack of recombination over 95% of its length provide valuable insights into paternal lineage ancestry and DNA kinship investigations. While slowly mutating Y-chromosomal single nucleotide polymorphisms (Y-SNPs) are traditionally used to assign evolutionary Y-(sub)haplogroups, the process is labour-intensive, time-consuming and costly. An efficient alternative approach involves the use of the more rapidly mutating familial Y-chromosomal short tandem repeats

Molecular autopsy and family screening in a young case of sudden cardiac death reveals an unusually severe case of FHL1 related hypertrophic cardiomyopathy.

BACKGROUND: Hypertrophic cardiomyopathy (HCM) is a genetic cardiomyopathy with a prevalence of about 1:200. It is characterized by left ventricular hypertrophy, diastolic dysfunction and interstitial fibrosis; HCM might lead to sudden cardiac death (SCD) especially in the young. Due to low autopsy frequencies of sudden unexplained deaths (SUD) the true prevalence of SCD and especially of HCM among SUD remains unclear. Even in cases of proven SCD genetic testing is not a routine procedure precluding appropriate risk stratification and counseling of relatives. METHODS: Here we report a case of

Recent advances in Forensic DNA Phenotyping of appearance, ancestry and age.

Forensic DNA Phenotyping (FDP) comprises the prediction of a person's externally visible characteristics regarding appearance, biogeographic ancestry and age from DNA of crime scene samples, to provide investigative leads to help find unknown perpetrators that cannot be identified with forensic STR-profiling. In recent years, FDP has advanced considerably in all of its three components, which we summarize in this review article. Appearance prediction from DNA has broadened beyond eye, hair and skin color to additionally comprise other traits such as eyebrow color, freckles, hair structure, hai

Human autosomal DNA and X chromosome STR profiles obtained from Chrysomya albiceps (Diptera: Calliphoridae) larvae used as a biological trace.

The use of insects to answer questions in criminal investigations, as well as a combination of forensic genetic techniques to obtain human DNA from the organisms, especially necrophagous dipterians, have gained ground in recent decades among researchers and professionals in this area. The objective of our study was to evaluate and compare two methods of human DNA extraction, commonly used for forensic samples, to obtain human autosomal DNA and X chromosome short tandem repeat profiles from the digestive tract of Chrysomya albiceps (Diptera: Calliphoridae) larvae. Immature specimens were collec
