Visuocortical changes during a freezing-like state in humans.

An adaptive response to threat requires optimized detection of critical sensory cues. This optimization is thought to be aided by freezing - an evolutionarily preserved defensive state of immobility characterized by parasympathetically mediated fear bradycardia and regulated by the amygdala-periaqueductal grey (PAG) circuit. Behavioral observations in humans and animals have suggested that freezing is also a state of enhanced visual sensitivity, particularly for coarse visual information, but the underlying neural mechanisms remain unclear. We induced a freezing-like state in healthy volunteer

Increased freezing and decreased positive affect in postinstitutionalized children.

BACKGROUND: Early neglect is associated with increased risk of internalizing disorders in humans and with increased fear behavior in animals. When children are adopted out of orphanages in which they experienced institutional neglect, anxiety and depressive disorders often are not seen until adolescence. What has not been examined is whether even young children adopted from institutional care exhibit heightened fear or behavioral inhibition. METHOD: Children adopted between 15 and 35 months from institutional care were examined twice during their first year postadoption and compared with chil

Heating-induced freezing and melting transitions in charged colloids.

We examine influence of temperature on the phase behavior of dilute aqueous dispersions of charged colloidal silica and polystyrene particles. They undergo either freezing or melting transitions with increasing temperature. Freezing occurs in the case of low-charge, low-salt colloids, and melting is observed in the case of high-charge, high-salt colloids. All of these phase transitions are thermoreversible. These intriguing behaviors can be qualitatively explained in terms of the decrease in the permittivity of water at elevated temperatures.

Effect of the "cough technique" on cryotherapy freezing temperature.

OBJECTIVE: To assess the impact of the cough technique on the tip temperature reached in a cryotherapy device commonly used to treat precancerous cervical lesions. METHODS: During February to March 2009, bench testing was conducted in Peru on 3 Wallach LL100 cryotherapy units using locally obtained carbon dioxide as the refrigerant. The temperature at the cryo tip was measured continuously with a thermocouple. Mean temperatures recorded with and without the cough technique were compared across the 3 cryotherapy devices. RESULTS: Higher mean temperatures were observed with the cough technique

In vivo detection of membrane injury at freezing temperatures.

The release of hydrogen cyanide from Amelanchier alnifolia was monitored at 30 C and -10 C following lethal freezing at both slow and fast rates. Assuming that hydrogen cyanide release indicates membrane damage, it was concluded that during a fatal freeze-thaw cycle membrane damage occurred during cell contraction and, therefore, was not dependent upon membrane area expansion during thawing.

Sample preparation of some plant tissues for transmission electron microscopy using high pressure freezing and freeze substitution.

For sample preparation of biological block sectioning, high pressure freezing and freeze substitution (HPF-FS) is a better choice than chemical fixation. Freezing fixation can retain the original structures and preserve the soluble materials, and avoid the disadvantage of chemical fixation. Using high-pressure freezing, plant tissues are more difficult to freeze successfully than animal tissues and microbes because they have cell walls and varying degrees of vacuolation. For plant tissues, including the roots of Arabidopsis thaliana, 1-hexadecene is an ideal cryoprotectant/filler. The ability

Electron microscopic analysis of the mouse and human alveolar epithelial glycocalyx using high-pressure freezing and freeze substitution.

Alveolar epithelial cells (AECs) are covered apically by the alveolar lining layer (ALL). The alveolar epithelial glycocalyx (AEGCX), which is part of the ALL, consists of a mixture of glycoproteins, proteoglycans, and glycosaminoglycans. In comparison with the endothelial glycocalyx (GCX), much less is known about the composition and structure of the AEGCX, although it is presumed to be involved in many cellular processes. The challenges of in vivo visualization of the AEGCX, coupled with the distorting effects of aldehyde fixation, make it difficult to accurately determine its structural org

Immunocytochemical localization of amylase in gerbil salivary gland acinar cells processed by rapid freezing and freeze-substitution fixation.

We examined the immunocytochemical localization of amylase in cryofixed serous acinar cells of gerbil major salivary glands by indirect immunostaining, using anti-gerbil parotid amylase antibody and protein A-gold complex. Fresh tissue blocks were quickly frozen by the metal-contact method, using liquid helium, and were freeze-substituted with either osmium-acetone solution or glutaraldehyde-containing acetone. They were then embedded in an epoxy resin mixture which was polymerized at 60 degrees C. Some tissue blocks substituted with aldehyde-acetone solution were embedded in Lowicryl K4M, pol

Effects of supercooling, sub-freezing, and deep-freezing pre-cooling on freshness, flavor, water-holding capacity, and microstructure of beef during EPS-based distribution.

This study examined how pre-cooling temperature affects beef quality and temperature fluctuations during expanded polystyrene (EPS)-based distribution, simulated as 48 h at 25 °C followed by chilled storage at 4 °C. Beef steaks were pre-cooled to -0.5 °C (supercooling), -5 °C (sub-freezing), or -18 °C (deep-freezing) and compared with a 4 °C refrigerated control. Quality was evaluated by freshness indicators (total volatile basic nitrogen (TVB-N), thiobarbituric acid reactive substances (TBARS), total viable count (TVC)), water-holding capacity, flavor (electronic tongue, electronic nose, gas

Effect of sample volume on the variation of freezing characteristics, mechanical properties, and microstructure of gelatin gel with depth after liquid nitrogen spray freezing.

Liquid nitrogen spray freezing (LNSF, -20, -60, and - 100 °C) was adopted to evaluate the effect of four cubic sample (with side lengths of 25, 50, 75, and 100 mm) volumes on the freezing curves, microstructure, texture, and gel strength of gelatin gels (16 % with 4 % sucrose, w/w), serving as a food model, along the depth in gels. The total freezing time significantly increased (P < 0.05) with increasing freezing temperature and depth, leading to larger ice crystals and more severe damage to the gel network. The gel strength significantly decreased from 3480 g at a depth of 5 mm to 2570 g at

Defensive Freezing and the Evolutionary Origins of Meditation in Humans.

The biological origins of meditation in humans remain underexplored, despite extensive scholarship on its cultural history and health effects. We present a theoretical account that traces the origins of meditation to the evolutionarily conserved repertoire of defensive freezing. We propose that this ancient survival response-characterized by motoric immobility, heightened vigilance with narrowed attentional focus, and bradycardia-provided a behavioral, neural, and physiological substrate upon which operant and social reinforcement could act. Over evolutionary time, these response components ma

Towards the Prevention of Freezing of Gait in Parkinson's Disease.

Despite optimal management, freezing of gait (FOG) remains a disabling complication of Parkinson's disease (PD), contributing substantially to falls, loss of independence and reduced quality of life. This highlights the pressing need for a shift towards preventive approaches, aiming to postpone or even prevent its development. In this narrative review, we examine potential avenues for the secondary prevention of FOG. We discuss how FOG might be prevented specifically, by targeting modifiable risk factors that seem associated with the development of FOG. Examples of potentially effective strate

Effect of freezing on mechanical properties of ovine brain tissue.

Freezing at -20 °C impacts the mechanical properties of post mortem brain tissue, which has implications for forensic science, particularly for estimating the post mortem interval (PMI). Freezing is examined as a method that may offer an extended window for forensic analysis by halting tissue degradation processes or allow tissue transfer to specialized departments. A rheological examination of samples from various brain regions of 21 ovine brains (frontal lobe, anterior deep brain, posterior deep brain, parietal lobe, superior colliculus, medulla, pons, cerebellum) was conducted. The brain ti

Functional connectivity compensation in Parkinson's disease with freezing of gait.

Freezing of gait (FOG) is a disabling motor symptom prevalent in patients with Parkinson's disease (PD); however, its pathophysiological mechanisms are poorly understood. This study aimed to investigate whole-brain functional connectivity (FC) pattern alterations in PD patients with FOG. A total of 18 PD patients, 10 with FOG (PD-FOG) and 8 without FOG (PD-nFOG), and 10 healthy controls were enrolled. High-resolution 3D T1-weighted and resting-state functional MRI (rs-fMRI) data were obtained from all participants. The groups' internetwork connectivity differences were explored with rs-fMRI FC

Use of electrolyte leakage to assess floral damage after freezing.

PREMISE: With growing interest in the impact of false springs on plant reproduction, there is the need to develop reliable, high-throughput methods for assessing floral freezing damage. Here we present a method for use with floral tissue that will facilitate more comparative work on floral freezing tolerance in the future. METHODS AND RESULTS: We examined the effectiveness of a modified electrolyte leakage protocol to assess floral freezing damage. By comparing data from temperature response curves to an estimate of visual tissue damage, we optimized the protocol for different floral types an

Cortical mechanisms of movement recovery after freezing in Parkinson's disease.

Involuntary interruptions of upper limb movements, referred to as "upper limb freezing" (ULF) belong to the most disabling symptoms of Parkinson's disease (PD). Our study aimed to explore the cortical neuronal mechanisms underlying the reinstation of regular movement after a freezing episode and to control them by voluntary stops. We hypothesized that this movement recovery after a freeze would be accompanied by a decrease of beta power (13-30 Hz) over the primary sensorimotor cortex (electrode "C3"). We recorded a 62-channel surface EEG in 14 PD patients during a repetitive finger tapping tas
