Inversion 2La is associated with enhanced desiccation resistance in Anopheles gambiae.

BACKGROUND: Anopheles gambiae, the principal vector of malignant malaria in Africa, occupies a wide range of habitats. Environmental flexibility may be conferred by a number of chromosomal inversions non-randomly associated with aridity, including 2La. The purpose of this study was to determine the physiological mechanisms associated with the 2La inversion that may result in the preferential survival of its carriers in hygrically-stressful environments. METHODS: Two homokaryotypic populations of A. gambiae (inverted 2La and standard 2L+(a)) were created from a parental laboratory colony polym

Spatial and sex-specific dissection of the Anopheles gambiae midgut transcriptome.

BACKGROUND: The midgut of hematophagous insects, such as disease transmitting mosquitoes, carries out a variety of essential functions that mostly relate to blood feeding. The midgut of the female malaria vector mosquito Anopheles gambiae is a major site of interactions between the parasite and the vector. Distinct compartments and cell types of the midgut tissue carry out specific functions and vector borne pathogens interact and infect different parts of the midgut. RESULTS: A microarray based global gene expression approach was used to compare transcript abundance in the four major female

Unexpected high losses of Anopheles gambiae larvae due to rainfall.

BACKGROUND: Immature stages of the malaria mosquito Anopheles gambiae experience high mortality, but its cause is poorly understood. Here we study the impact of rainfall, one of the abiotic factors to which the immatures are frequently exposed, on their mortality. METHODOLOGY/PRINCIPAL FINDINGS: We show that rainfall significantly affected larval mosquitoes by flushing them out of their aquatic habitat and killing them. Outdoor experiments under natural conditions in Kenya revealed that the additional nightly loss of larvae caused by rainfall was on average 17.5% for the youngest (L1) larvae

Short-range attractiveness of pregnant women to Anopheles gambiae mosquitoes.

Malaria is a major cause of illness and an indirect cause of mortality in pregnant women. It can also cause stillbirths and low-birthweight babies. We have shown previously that pregnant women attracted twice as many Anopheles gambiae mosquitoes, the principal African malaria vector, as their non-pregnant counterparts over distances of about 15 m. In the current study (in 1998/99) we compared the short-range attractiveness of both pregnant and non-pregnant women sleeping under untreated bednets in Gambian villages. First, we measured the rate of mosquito entry under bednets and, second, we cal

Pallor as a clinical sign of severe anaemia in children: an investigation in the Gambia.

Anaemia associated with malaria is a major public health problem in African countries. Since most primary health facilities have to rely on physical signs and not laboratory tests to detect anaemic patients who need referral for blood transfusion, we have assessed the reliability of simple clinical signs to predict severe anaemia. A trained field assistant examined 368 children admitted to a tertiary care hospital, assessing the pallor of their eyelids (conjunctiva), palms and nailbeds, counting the respiratory rate, and looking for signs of respiratory distress. After the children's admission

Evaluation of the PlenOptika QuickSee Free Portable Autorefractor in Schools in The Gambia.

PURPOSE: To evaluate the ability of the monocular QuickSee Free (QSF) portable autorefractor (PlenOptika) to detect and measure refractive error relative to gold standard cycloplegic retinoscopy in a population of school-aged children in a low-resource setting in The Gambia. METHODS: A total of 101 children, aged between 3 and 17 years (10.2 ± 3.45 years) underwent visual acuity screening, autorefraction by the QSF, and complete ophthalmic examination including cycloplegic retinoscopy. The agreement of the results was evaluated by Bland-Altman plots. The sensitivity and specificity of detecti

The circadian clock modulates Anopheles gambiae infection with Plasmodium falciparum.

Key behaviours, physiologies and gene expressions in Anopheles mosquitoes impact the transmission of Plasmodium. Such mosquito factors are rhythmic to closely follow diel rhythms. Here, we set to explore the impact of the mosquito circadian rhythm on the tripartite interaction between the vector, the parasite and the midgut microbiota, and investigate how this may affect the parasite infection outcomes. We assess Plasmodium falciparum infection prevalence and intensity, as a proxy for gametocyte infectivity, in Anopheles gambiae mosquitoes that received a gametocyte-containing bloodfeed and me

Epizootiology and Molecular Identification of Trypanosome Species in Livestock Ruminants in the Gambia.

INTRODUCTION: African Animal Trypanosomiasis (AAT) or nagana in animals, is caused by the blood-borne parasitic protozoa called trypanosomes, and is potentially fatal. It is estimated that Africa loses $4‒5 billion annually due to the death of livestock to nagana in the tsetse belt. PURPOSE: Although The Gambia lies within this belt, there is scanty data regarding the epizootiology of nagana in The Gambia. Here, records of reported cases of nagana for the period 2010-2019 at the International Trypanotolerance Centre (ITC) in The Gambia were analyzed retrospectively. METHODS: For insights int

Molecular evolution of immune genes in the malaria mosquito Anopheles gambiae.

BACKGROUND: As pathogens that circumvent the host immune response are favoured by selection, so are host alleles that reduce parasite load. Such evolutionary processes leave their signature on the genes involved. Deciphering modes of selection operating on immune genes might reveal the nature of host-pathogen interactions and factors that govern susceptibility in host populations. Such understanding would have important public health implications. METHODOLOGY/FINDINGS: We analyzed polymorphisms in four mosquito immune genes (SP14D1, GNBP, defensin, and gambicin) to decipher selection effects,

Virological and immunological correlates of mother-to-child transmission of cytomegalovirus in The Gambia.

BACKGROUND: Cytomegalovirus (CMV) is the most common congenital infection and can follow primary and recurrent maternal infection. We studied correlates of vertical transmission of CMV in The Gambia, where most children acquire CMV during the first year of life. METHODS: A cohort of 281 mothers and infants was recruited at birth. Infants were prospectively followed up for CMV infection during the first year of life. Excretion of CMV and antiviral immune response were studied at birth in mothers of children infected in utero, early during infancy, or late during infancy or not infected at 1 ye

Molecular characterization of the malaria vector Anopheles gambiae s.s. in Madagascar.

Anopheles gambiae s.s. Giles (Diptera: Culicidae), the primary African malaria vector, has been characterized at the subspecies level in Madagascar, where only the molecular form S and haplotype gIA occur. The haplotype gIC proposed by other authors was not observed amongst the 35 mosquito genomes sequenced. These S/gIA characteristics are also found on the Comoros archipelago and in continental Africa.

Identification and biochemical characterization of the Anopheles gambiae 3-hydroxykynurenine transaminase.

Spontaneous oxidation of 3-hydroxykynureine (3-HK), a metabolic intermediate of the tryptophan degradation pathway, elicits a remarkable oxidative stress response in animal tissues. In the yellow fever mosquito Aedes aegypti the excess of this toxic metabolic intermediate is efficiently removed by a specific 3-HK transaminase, which converts 3-HK into the more stable compound xanthurenic acid. In anopheline mosquitoes transmitting malaria, xanthurenic acid plays an important role in Plasmodium gametocyte maturation and fertility. Using the sequence information provided by the Anopheles gambiae

Causes of mortality in twins in a rural region of The Gambia, West Africa.

Little is known about birth or mortality rates of twins in The Gambia. There are no reports of the causes of death among twins in sub-Saharan Africa. We identified twin births and deaths from a community-based study which had been carried out in a large rural region of The Gambia over a 5-year period from 1989 to 1993. The overall twinning rate excluding stillbirths was 14.3 (95% CI 12.9, 15.8) per 1000 live deliveries. This was significantly lower among the Mandinka ethnic group (8.8 per 1000) than among the Serahulis (15.1 per 1000) or Fulas (18.3 per 1000). The early-neonatal, late-neonatal

Strengthening healthcare through family medicine training in The Gambia: The journey so far.

According to the World Health Organizations (WHO) family medicine forms the bedrock upon for accessible, affordable and equitable healthcare for any country. The need for family doctors is more acute for low income countries like The Gambia. More so that The Gambian health infrastructure is suboptimal and appropriate health personnel is low. This is worsened by brain drain leading to poor health indices. Despite these challenges and more, the department of Family Medicine was accredited for training in the Gambia with improved infrastructure (at the training centre), with 7 residents. Though t

An exotic abscess within the United Kingdom from The Gambia: a case report.

BACKGROUND: Furuncular myiasis is a parasitic infection of a live mammal by fly larvae commonly seen in Africa. However, with an increase in international tourism, there is a significant rise in exotic infection in non-endemic areas which can pose a diagnostic challenge to doctors and potentially lead to delay in treatment. From the current literature, only 12 cases were reported in the UK. CASE PRESENTATION: We report an unusual case of multiple abscesses in a 32-year-old white British woman presenting to our Emergency department in the UK after returning from a holiday in The Gambia, West A
