As part of the innate immune response, the host withholds metal micronutrients such as iron (Fe) from invading pathogens. To survive such Fe-limitation, the opportunistic fungal pathogen Candida albicans has evolved elaborate responses to Fe-starvation stress. One such adaptation involves the secretion of flavins, yellow isoalloxazine compounds that serve important redox roles in biology. Why the organism secretes flavins during Fe-starvation is not known. Moreover, the exact flavin secreted by the fungus or the effects of Fe-starvation on intracellular flavin pools have not been documented. M

Controllable regulatory elements, like inducible, titratable promoters, are highly desired in synthetic biology toolboxes. A set of previously developed erythritol-inducible promoters along with an engineered Yarrowia lipolytica host strain were shown to be a very potent expression platform. In this study, we push the previously encountered limits of the synthetic promoters' titratability (by the number of upstream motifs) by using a compatible transcription factor, Euf1, as the promoter titrator. Overexpression of spliced EUF1 turned out to be very efficient in promoting expression from the c

The filamentous ascomycete Trichoderma reesei, known for its prolific cellulolytic enzyme production, recently also gained attention for its secondary metabolite synthesis. Both processes are intricately influenced by environmental factors like carbon source availability and light exposure. Here, we explore the role of the transcription factor STE12 in regulating metabolic pathways in T. reesei in terms of gene regulation, carbon source utilization and biosynthesis of secondary metabolites. We show that STE12 is involved in regulating cellulase gene expression and growth on carbon sources asso

BACKGROUND: Adaptation to complex, rapidly changing environments is crucial for evolutionary success of fungi. The heterotrimeric G-protein pathway belongs to the most important signaling cascades applied for this task. In Trichoderma reesei, enzyme production, growth and secondary metabolism are among the physiological traits influenced by the G-protein pathway in a light dependent manner.
RESULTS: Here, we investigated the function of the SNX/H-type regulator of G-protein signaling (RGS) protein RGS4 of T. reesei. We show that RGS4 is involved in regulation of cellulase production, growth,

The polysaccharide chitosan is found in the cell wall of specific cell types in a variety of fungal species where it contributes to stress resistance, or in pathogenic fungi, virulence. Under certain growth conditions, the pathogenic yeast Candida dubliniensis forms a cell type termed a chlamydospore, which has an additional internal layer in its cell wall compared to hyphal or yeast cell types. We report that this internal layer of the chlamydospore wall is rich in chitosan. The ascospore wall of Saccharomyces cerevisiae also has a distinct chitosan layer. As in S. cerevisiae, formation of th

Aspergillus fumigatus is the most common cause of invasive aspergillosis, a life-threatening infection mainly affecting immunocompromised patients. The essential metals copper and iron play crucial roles in virulence of this mold. Recently, the copper-regulatory transcription factor Mac1 was reported to additionally be involved in the control of iron acquisition. However, in the current study, neither growth assays on solid and in liquid media, analysis of siderophore production nor expression analysis of genes involved in iron acquisition indicated the involvement of Mac1 in the regulation of

Paracoccidioidomycosis is a highly prevalent systemic mycosis in Latin America, caused by fungi of the genus Paracoccidioides. Copper is essential for eukaryotes and bacteria. This micronutrient is used in many vital biochemical processes, although metal excess levels can be toxic for organisms. Pathways underlying copper overload are poorly understood in members of the Paracoccidioides complex. The responses of Paracoccidioides lutzii yeast cells to copper overload were here evaluated. The results showed that under copper overload, cells presented a dark brown pigment, identified as melanin.

Candida albicans, a fungus that normally inhabits the digestive tract and other mucosal surfaces, can become a pathogen in immunocompromised individuals, causing severe or even fatal infection. Mechanisms by which C. albicans can evade commonly used antifungal agents are not fully understood. We are studying a model system involving growth of C. albicans on toxic sugar sorbose, which represses synthesis of cell wall glucan and, as a result, kills fungi in a manner similar to drugs from the echinocandins class. Adaptation to sorbose occurs predominantly due to reversible loss of one homolog of

As is often the case for microbial product formation, the penicillin production rate of Penicillium chrysogenum has been observed to be a function of the growth rate of the organism. The relation between the biomass specific rate of penicillin formation (q(p)) and growth rate (mu) has been measured under steady state conditions in carbon limited chemostats resulting in a steady state q(p)(mu) relation. Direct application of such a relation to predict the rate of product formation during dynamic conditions, as they occur, for example, in fed-batch experiments, leads to errors in the prediction,

We have previously shown that the beneficial filamentous fungus Trichoderma virens secretes the highly effective hydrophobin-like elicitor Sm1 that induces systemic disease resistance in the dicot cotton (Gossypium hirsutum). In this study we tested whether colonization of roots by T. virens can induce systemic protection against a foliar pathogen in the monocot maize (Zea mays), and we further demonstrated the importance of Sm1 during maize-fungal interactions using a functional genomics approach. Maize seedlings were inoculated with T. virens Gv29-8 wild type and transformants in which SM1 w

Peach shoot blight, caused by Diaporthe amygdali, is a serious challenge for China's peach industry, contributing to annual yield losses of 20-50%. Despite its economic significance, the molecular mechanisms underlying D. amygdali pathogenicity remain poorly characterized. Velvet proteins are fungal-specific regulatory factors that exhibit functional diversity across plant pathogenic fungi; however, their roles in D. amygdali remain uncharacterized. Here, we identified and systematically characterized four velvet proteins in D. amygdali, including DaVeA, DaVelB, DaVelC, and DaVosA. Expression

In a previous study on discovering new antimicrobial agents from microbial sources, nine bafilomycins were isolated from the fermentation broth of Streptomyces albolongus. Among them, bafilomycin C1 (Baf C1) showed strong antifungal activity against Candida albicans, with MIC value of 1.56 μg/mL. The aim of this study was to evaluate the action mechanism of Baf C1 against C. albicans. Quantitative PCR analysis revealed that ergosterol biosynthesis-related genes of C. albicans ACS1, HMG1, IDI1, ERG1, ERG2, ERG6, ERG7, ERG8, ERG9, ERG12, ERG13, ERG20, ERG24, ERG251, ERG252, ERG26, ERG27, and ERG

The Mkt1-Pbp1 complex promotes mating-type switching by regulating the translation of HO mRNA in Saccharomyces cerevisiae. Here, we performed in vivo immunoprecipitation assays and mass spectrometry analyses in the human fungal pathogen Cryptococcus neoformans to show that Pbp1, a poly(A)-binding protein-binding protein, interacts with Mkt1 containing a PIN like-domain. Association of Pbp1 with Mkt1 was confirmed by co-immunoprecipitation assays. Results of spot dilution growth assays showed that unlike pbp1 deletion mutant strains, mkt1 deletion mutant strains were not resistant to heat stres

The yeast pathogen Candidozyma (Candida) auris can form biofilms, which contribute to its virulence and nosocomial transmission. In this study, we identified the transcription factor Wor2 as a negative regulator of biofilm formation in C. auris. Wor2 hyperactivation in a strain of clade IV via the use of a protein tagging strategy resulted in downregulation of two important adhesins, SCF1 and ALS4112, and decreased biofilm-forming capacity. We showed that the impact on biofilm was predominantly mediated via decreased SCF1 expression in this strain. However, results of adhesion assays on inert

Biofilm formation is vital for the survival and pathogenicity of the fungus Candida albicans. Expression of biofilm-promoting genes is coordinated by a transcription factor network that governs the yeast-filament transition and other processes. A second cell type transition, the white-opaque transition, is coordinated by its own transcription factor network. Initial studies suggested that the 2 transcriptional networks have a mutually exclusive relationship, driven in part by reciprocal repression of biofilm regulator Efg1 and white-opaque regulator Wor1. However, recent studies have shown tha

During mixed-species invasive candidiasis-an emerging threat in healthcare-the presence of Candida albicans is nearly essential for host invasion by C. glabrata. However, the molecular communication underlying such intriguing inter-species fungal interactions is largely unexplored. Here, we show that C. glabrata secretes a unique small protein, Yhi1-encoded by a likely de novo gene-which induces hyphal growth via Tsa1 in C. albicans. Notably, this Yhi1-based inter-species interaction is specific to C. glabrata and C. albicans compared with other common species of Candida. Our structure-functio

BACKGROUND: Talaromyces sp., as powerful cellulolytic fungi, display a nearly complete enzymatic system, which exhibits a complex regulatory system involving multiple transcription factors to control the expression of cellulase genes. However, there are fewer studies on the transcriptional regulatory factors of cellulase expression in Talaromyces sp. This study provides a basis for in-depth analysis of the transcription regulatory mechanism of Talaromyces endophyticus for cellulase expression, offering new ways for the preparation of fermented sugars using straw resources to produce high value

Candida albicans is an opportunist pathogen responsible for a broad spectrum of infections, from superficial mycosis to the systemic disease candidiasis. C. albicans has various morphological forms, including unicellular budding yeasts, filamentous pseudohyphae and true hyphae, and the ability to switch from yeast to hyphal forms is a key survival mechanism underlying the adaptation of the pathogen to the microenvironments encountered within the host. Filamentation is regulated by multiple signalling pathways and its induction in different growth media in vitro has often led to conflicting res

Candida albicans is known for its pathogenicity, although it lives within the human body as a commensal member. The commensal nature of C. albicans is well controlled and regulated by the host's immune system as they live in the harmonized microenvironment. However, the development of certain unusual microhabitat conditions (change in pH, co-inhabiting microorganisms' population ratio, debilitated host-immune system) pokes this commensal fungus to transform into a pathogen in such a way that it starts to propagate very rapidly and tries to breach the epithelial barrier to enter the host's syst

Asparagus is a genus consisting of over two hundred species of perennial plants. Fusariumproliferatum is a major asparagus pathogen and it biosynthesizes a variety of mycotoxins, of which fumonisins B are prevalent. Our previous studies on F.proliferatum strains indicated that asparagus extract affects the expression of FUM1 gene, encoding polyketide synthase, a key enzyme of the FUM gene cluster governing the biosynthesis of fumonisins. An asparagus-derived F.proliferatum strain increased fumonisin B1 production after extract fractions' addition, reaching the maximum 2 or 24 h after treatment
