HPCAL1 as a novel prognostic biomarker in acute myeloid leukemia and its correlation with immune infiltrates.

Acute myeloid leukemia (AML), a heterogeneous malignancy with complex molecular mechanisms, urgently requires improved prognostic biomarkers and therapeutic targets. This study investigated Hippocalcin-like 1(HPCAL1), a calcium-binding protein with dual functionality dependent on cancer type, as a potential prognostic marker and investigated its interaction with the tumor immune microenvironment in AML. Through analyses of multiple independent cohorts (GSE13159, GSE34184, GSE24395 and institutional data), we revealed that significant HPCAL1 overexpression in AML samples was correlated with poo

Highly aggressive lymphomas in adults.

Highly aggressive lymphomas are relatively uncommon in adults, comprising approximately 4% to 5% of all non-Hodgkin lymphomas in the United States and Western Europe. The designation of "highly aggressive" is generally restricted to precursor T-cell and B-cell lymphoblastic lymphoma/leukemia and Burkitt's lymphoma/leukemia. Treatment strategies for lymphoblastic lymphoma and Burkitt's lymphoma include complex, highly intensive combination chemotherapy regimens, which may be curative. As with other subtypes of NHL, emerging data from gene-expression profiling and related techniques are helping

In-Silico discovery of Pediatric Acute-Myeloid-Leukemia (pAML) causing druggable molecular signatures highlighting their pathogenetic processes and therapeutic agents through single-cell RNA-Seq profile analysis.

Pediatric acute-myeloid-leukemia (pAML) is an aggressive malignancy and the second most common blood cancer in children. In spite of significant advances in the frontline therapeutic approaches, approximately 50% of pAML patients show poor prognosis and relapse. Though drugs show positive response against the cancer cells initially, however, it becomes resistant in the long run of treatment, requiring the use of alternative drugs. Therefore, this study aimed to discover pAML-causing druggable molecular signatures highlighting their pathogenetic processes and alternative therapeutic agents. To

Heme synthesis inhibition induces the intrinsic apoptosis pathway in leukemia with OSGIN1 upregulation.

Acute myeloid leukemia (AML) is one of the hematologic malignancies with a poor outcome. AML has a unique metabolic status, and identifying its metabolic vulnerabilities is warranted. Recent genome-wide screenings suggest that heme synthesis might be such a vulnerability. Heme is required not only for hemoglobin synthesis but also for the proper function of hemoproteins. Cytochromes are such hemoproteins and are necessary for mitochondrial respiration. Therefore, heme synthesis inhibition can diminish AML by altering mitochondrial status and function. However, still little is known about the i

[Functional study of Esco2 in promoting proliferation and survival of acute myeloid leukemia cells].

The present study aimed to investigate the expression pattern of Esco2 in acute myeloid leukemia (AML) and its role in regulating the biological behavior of AML cells. The expression level of ESCO2 and its association with prognosis in patients with AML were analyzed using the TCGA-LAML and GTEx datasets. An sgRNA-mediated Esco2 depletion model was established in MLL-AF9-driven AML cells, and its function was validated through both in vitro and in vivo experiments. A murine bone marrow transplantation model was used to evaluate the effects of Esco2 depletion on leukemia progression. Colony for

BCR-ABL1 mediated miR-150 downregulation through MYC contributed to myeloid differentiation block and drug resistance in chronic myeloid leukemia.

The fusion oncoprotein BCR-ABL1 exhibits aberrant tyrosine kinase activity and it has been proposed that it deregulates signaling networks involving both transcription factors and non-coding microRNAs that result in chronic myeloid leukemia (CML). Previously, microRNA expression profiling showed deregulated expression of miR-150 and miR-155 in CML. In this study, we placed these findings into the broader context of the MYC/miR-150/MYB/miR-155/PU.1 oncogenic network. We propose that up-regulated MYC and miR-155 in CD34+ leukemic stem and progenitor cells, in concert with BCR-ABL1, impair the mo

Integrated computational biology analysis to evaluate target genes for chronic myelogenous leukemia.

Although hundreds of genes have been linked to chronic myelogenous leukemia (CML), many of the results lack reproducibility. In the present study, data across multiple modalities were integrated to evaluate 579 CML candidate genes, including literature‑based CML‑gene relation data, Gene Expression Omnibus RNA expression data and pathway‑based gene‑gene interaction data. The expression data included samples from 76 patients with CML and 73 healthy controls. For each target gene, four metrics were proposed and tested with case/control classification. The effectiveness of the four metrics present

Time-series analysis in imatinib-resistant chronic myeloid leukemia K562-cells under different drug treatments.

Chronic myeloid leukemia (CML) is characterized by the accumulation of active BCR-ABL protein. Imatinib is the first-line treatment of CML; however, many patients are resistant to this drug. In this study, we aimed to compare the differences in expression patterns and functions of time-series genes in imatinib-resistant CML cells under different drug treatments. GSE24946 was downloaded from the GEO database, which included 17 samples of K562-r cells with (n=12) or without drug administration (n=5). Three drug treatment groups were considered for this study: arsenic trioxide (ATO), AMN107, and

Expression of miR-196b is not exclusively MLL-driven but is especially linked to activation of HOXA genes in pediatric acute lymphoblastic leukemia.

BACKGROUND: Deregulation of microRNA may contribute to hematopoietic malignancies. MicroRNA-196b (miR-196b) is highly expressed in MLL-rearranged leukemia and has been shown to be activated by MLL and MLL-fusion genes. DESIGN AND METHODS: In order to determine whether high expression of miR-196b is restricted to MLL-rearranged leukemia, we used quantitative stem-loop reverse transcriptase polymerase chain reaction to measure the expression of this microRNA in 72 selected cases of pediatric acute lymphoblastic leukemia i.e. MLL-rearranged and non-MLL-rearranged precursor B-cell and T-cell acut

Gene expression profiling of minimally differentiated acute myeloid leukemia: M0 is a distinct entity subdivided by RUNX1 mutation status.

Minimally differentiated acute myeloid leukemia (AML-M0) is defined by immature morphology and expression of early hematologic markers. By gene expression profiling (GEP) and subsequent unsupervised analysis of 35 AML-M0 samples and 253 previously reported AML cases, we demonstrate that AML-M0 cases express a unique signature that is largely separated from other molecular subtypes. Hematologic transcription regulators such as CEBPA, CEBPD, and ETV6, and the differentiation associated gene MPO appeared strongly down-regulated, in line with the primitive state of this leukemia. AML-M0 frequently

Tumor suppressor effects of miR-143 and miR-199 on the K562 myelogenous leukemia cell line by targeting the RNA-binding protein Musashi2.

The RNA-binding protein Musashi2 plays a critical role in normal and malignant hematopoiesis. Aberrant expression of Musashi2 is linked to the inappropriate expression of several miRNAs. In this study, we observed downregulation of miR-143 and miR-199 in CML and AML cell lines. Bioinformatic analysis revealed potential interactions between these miRNAs and Musashi2 mRNA, which were also confirmed by our experimental data. In K562 cells, which showed the strongest negative correlation between Musashi2 and the candidate miRNAs, overexpression of miRNAs led to reduced cell proliferation and invas

miRNA expression based modulation: A new paradigm for the treatment of chronic myeloid leukemia.

MicroRNAs (miRNAs) are non-coding and internally derived small RNA molecules. They post-transcriptionally direct gene expression either by inhibiting translation or initiating degradation of mRNA. Conserved evolutionarily, these miRNAs have a significant role in several developmental and regulatory functions in organisms including mammalian cell growth, differentiation, and apoptosis. An individual miRNA can modulate a network of mRNA expression by binding to multiple mRNAs through imperfect sequence complementarity. In several types of cancer, dysregulation of miRNAs may contribute to cell in

Red cell antigen loss in a patient with chronic myeloid leukemia: a case of ABO discrepancy.

Change in ABO antigen expression on the surface of neoplastic cells have been seen for a variety of tumor types. This phenomenon has been linked with myeloid neoplasms as well but is reported infrequently. Here, we report a rare cause of ABO discrepancy in an elderly female having chronic myeloid leukemia.

Overexpression of delta-like (Dlk) in a subset of myelodysplastic syndrome bone marrow trephines.

The mRNA expression level of the gene delta-like (Dlk), coding for a signal transducer related to the Delta-Notch family, was measured using real-time PCR methodology in a large series of control biopsies (n = 61) and bone marrow trephines from patients with myelodysplastic syndrome (MDS) (n = 93) and related myeloid disorders (AML, n = 16 and chronic myeloproliferative disease (CMPD), n = 38). It turned out that dlk is strongly overexpressed in a subset of MDS (16%) cases, however with higher frequency in blast rich MDS cases. Therefore, the quantitative detection of dlk mRNA can support the

Regulation of the expression of annexin VIII in acute promyelocytic leukemia.

Annexin VIII is a calcium-dependent phospholipid-binding protein previously identified as a blood anticoagulant based on in vitro studies. However, the physiologic function of annexin VIII remains unknown. In acute promyelocytic leukemia (APL) the annexin VIII gene is highly expressed, but its expression is undetectable in the blasts of other acute leukemias. In the present investigation, we showed using the APL-derived NB4 cell line that expression of the annexin VIII gene is regulated at the transcription level during induced differentiation by all-trans retinoic acid (ATRA). The half-life o

Inhibition of the CtBP complex and FBXO11 enhances MHC class II expression and anti-cancer immune responses.

There is increasing recognition of the prognostic significance of tumor cell major histocompatibility complex (MHC) class II expression in anti-cancer immunity. Relapse of acute myeloid leukemia (AML) following allogeneic stem cell transplantation (alloSCT) has recently been linked to MHC class II silencing in leukemic blasts; however, the regulation of MHC class II expression remains incompletely understood. Utilizing unbiased CRISPR-Cas9 screens, we identify that the C-terminal binding protein (CtBP) complex transcriptionally represses MHC class II pathway genes, while the E3 ubiquitin ligas

Expression of HOXC4, HOXC5, and HOXC6 in human lymphoid cell lines, leukemias, and benign and malignant lymphoid tissue.

Besides their regulatory role in embryogenesis, homeobox (HOX) genes are expressed in a specific manner in hematopoietic cell lineages, implying a role in the molecular regulation of hematopoiesis. Some HOX C cluster genes are found to be expressed in lymphoid cells of mice and humans. Their function and expression in normal hematopoiesis are still largely unknown. We have studied the mRNA expression of HOXC4, HOXC5, and HOXC6 in several stages of lymphocyte maturation by reverse transcriptase-polymerase chain reaction (RT-PCR) and RNA in situ hybridization (RISH). We examined CD34+/CD38low an

ALPHA-TOCOPHEROL AND G-CSF CHANGE EXPRESSION OF GENES ASSOCIATED WITH DIFFERENTIATION OF K562 CHRONIC MYELOID LEUKEMIA CELLS DOWNREGULATING EMT-ASSOCIATED STEMNESS BIOMARKERS.

BACKGROUND: Chronic myeloid leukemia (CML) is a clonal myeloproliferative disorder characterized by a block of myeloid differentiation, finally resulting in the uncontrolled expansion of CML stem cells in a phase of blast crisis of the disease. Tyrosine kinase inhibitors (TKI) are effective in delaying CML progression for a long time. Nevertheless, CML cells become resistant to TKI over time. Therefore, the search for alternative and complementary therapies, including differentiation therapy, is currently in the limelight. The aim of the study was to explore the differentiation potential of al
