Executive processes in Parkinson's disease--random number generation and response suppression.

In producing random numbers, subjects typically deviate systematically from statistical randomness. It is considered that these biases reflect constraints imposed by underlying structures and processes, rather than a deficient concept of randomness. Random number generation (RNG) places considerable demands on executive processes, and provides a possibly useful tool for their investigation. A group of patients with Parkinson's disease (PD) and a group of controls were tested on a RNG task, both alone and with a concurrent attention-demanding task (manual tracking). Both groups showed the biase

Vascular endothelial cells generate peroxynitrite in response to carbon monoxide exposure.

Carbon monoxide causes a perivascular oxidative injury in animals, and we tested the hypothesis that endothelial cells could be a source of the injurious oxidants. Studies were undertaken to assess whether exposure to carbon monoxide would cause cultured bovine pulmonary artery endothelial cells to liberate reactive species. Concentrations of carbon monoxide between 11 and 110 nM caused progressively higher concentrations of nitric oxide to be released by endothelial cells based on measurements of nitrite and nitrate. Intracellular production of peroxynitrite was indicated by elevated concentr

Guinea-pig tracheal responsiveness in vitro following general anaesthesia with halothane.

Halothane and isoflurane induce potent bronchodilation during general anaesthesia and have been used successfully during status asthmaticus. The aim of this study was to determine whether airway hyporesponsiveness was prolonged after halothane administration. Sixteen guinea-pigs were submitted for 2 h to either 1.5% halothane in oxygen or 100% oxygen, and were killed 24 h later to elicit isometric tracheal contractions in organ baths with various agonists. Cumulative concentration-response curves to histamine or to KCl and contractions evoked with acetylcholine 1 mM (4.7 +/- 0.8 vs 4.6 +/- 0.5

Differences in eosinophil and neutrophil chemotactic responses in sowda and generalized form of onchocerciasis.

Extravasation of host's leukocytes from blood vessels into inflammatory tissues represents a prerequisite for a subsequent interaction with invaded parasites. The migratory responses of eosinophilic and neutrophilic granulocytes in the polar forms of the filarial infection onchocerciasis were investigated. The hyporeactive, generalized form, the chronic hyperreactive (sowda) form and persons without signs of onchocerciasis from a hypoendemic area for onchocerciasis were compared. Eosinophils from sowda patients responded more strongly to the inflammatory mediator platelet-activating factor (PA

Replacing maladaptive speech with verbal labeling responses: an analysis of generalized responding.

We taught three mentally handicapped students to answer questions with verbal labels and evaluated the generalized effects of this training on their maladaptive speech (e.g., echolalia) and correct responding to untrained questions. The students received cues-pause-point training on an initial question set followed by generalization assessments on a different set in another setting. Probes were conducted on novel questions in three other settings to determine the strength and spread of the generalization effect. A multiple baseline across subjects design revealed that maladaptive speech was re

Generation of procoagulant activity by hairy cells in response to endotoxin and phorbol esters.

Several investigators have suggested that hairy cells are neoplastic B lymphocytes. These cells, however, also share some biological properties with mononuclear phagocytes. A property of these cells is the capacity to generate procoagulant activity (PCA) in response to a variety of stimuli. In this study we investigated the PCA of peripheral blood hairy cells in 19 consecutive patients with hairy cell leukaemia (HCL). Monocyte-depleted blood mononuclear cells, tested immediately after isolation, expressed little, if any, activity. However, after exposure to endotoxin, a marked increase in PCA

Response to 30 generations of selection for open-field activity in laboratory mice.

High and low lines resulting from 30 generations of bidirectional selection for open-field activity have nonoverlapping distributions and more than a thirtyfold difference in mean activity. Open-field defecation scores of low-active lines are approximately 7 times higher than those of high-active lines, substantiating earlier reports of a large, negative genetic correlation between these characters. Since the selection experiment is replicated, other variables which are found to be reliably different among the high, control, and low lines are likely to be causally related to open-field activit

[The practice guideline 'Acne' (second revision) from the Dutch College of General Practitioners; a response from the perspective of general practice].

The strength of the second revision of the practice guideline 'Acne' from the Dutch College of General Practitioners (NHG) lies mainly in bringing its therapy up to date. The guideline also points out psychosocial problems that can arise with acne, but discounts stress and diet as the possible cause. Schedules showing a medical plan of action and information on medications complete this clear and practical guideline.

[The practice guideline 'Vaginal discharge' (first revision) from the Dutch College of General Practitioners; a response from the perspective of general practice].

The revised practice guideline by the Dutch College of General Practitioners (NHG) on vaginal discharge is intended to offer general practitioners a handle for management of vaginal complaints. Most general practitioners experience uncertainties in daily practice with respect to the diagnosis of vaginal discharge. In most cases history taking and physical examination fails to provide sufficient valuable information to arrive at a definite diagnosis. No further (microscopic) investigation is only permissible in a very obvious infection with Candida albicans. A recent onset of complaints of itch

[The practice guideline 'Children with fever' (second revision) from the Dutch College of General Practitioners; a response from the perspective of general practice].

The new version of the practice guideline 'Children with fever' has recently been published by the Dutch College of General Practitioners. There is no evidence to support antipyretic therapy in children with fever. Paracetamol (acetaminophen) is not advised for preventing seizures, but may be given to relieve other symptoms. Parents have a different attitude towards fever than do GPs. Parents are concerned about serious causes or complications of the fever. However, GPs are more interested in parameters other than fever. The practice guideline recommends that parents be advised and reassured,

[The practice guideline 'Otitis externa' (first revision) from the Dutch College of General Practitioners; a response from the perspective of general practice].

The first revision of the practice guideline 'Otitis externa' from the Dutch College of General Practitioners has been published. Data from a clinical trial in primary care clearly show that acid eardrops containing corticosteroids are superior to acid eardrops without corticosteroids. Other minor changes have been made. A KOH test is no longer necessary in general practice. Aluminium acetotartrate eardrops 1.2% are reserved for otitis externa with tympanic membrane perforation.

[The practice guideline 'Hormonal contraception' (second revision) from the Dutch College of General Practitioners; a response from the perspective of general practice].

The second revision of the Dutch College of General Practitioners' practice guideline on hormonal contraception discusses the various methods of hormonal birth control and can help the general practitioner to inform the patient when choosing the most desirable method. A thorough anamnesis is necessary with regard to the increased risks for breast cancer, uterine cancer and cardiovascular diseases in women using hormonal contraception, especially in those who smoke. The guideline also discusses non-hormonal methods briefly.

Patient motivational language in the prediction of symptom change, clinically significant response, and time to response in psychotherapy for generalized anxiety disorder.

Existing research demonstrates that patient change-talk (CT), or self-arguments for change, associates variably with favorable outcomes, whereas counter change-talk (CCT), or self-arguments against change, associates consistently with poorer outcomes. However, most studies on change language have focused on posttreatment versus more proximal outcomes. Addressing this gap, we examined Session 1 CT and CCT as predictors of during-treatment worry change, likelihood of clinically significant response during treatment, and time to response across cognitive-behavioral therapy (CBT; n = 43) and CBT i

Hydrogen Cyanide Generation by Pseudomonas aeruginosa Blunts the Host Innate Immune Response.

BACKGROUND: The biological mediator hydrogen cyanide (HCN) is produced by certain pathogenic bacteria. Hydrogen cyanide exerts multiple effects in mammalian cells: at low concentrations, it has cytoprotective regulatory effects, whereas at high concentrations, it blocks mitochondrial electron transport by inhibiting cytochrome c oxidase, thereby halting cellular metabolism. Here, we evaluated whether bacterial HCN confers resistance to the host immune response. We used Pseudomonas aeruginosa as a model organism, since it is a clinically significant HCN-generating bacterial species and a common

Glucocorticoid Receptor Translational Isoforms Generate Unique Glucocorticoid Responses in the Mouse Brain.

Glucocorticoids are primary stress hormones necessary for life that act on nearly every tissue in the body to maintain homeostasis. These hormones and their synthetic derivatives are widely used in the clinic to combat disease but are limited by serious adverse effects. The actions of glucocorticoids are mediated by the glucocorticoid receptor (GR). In addition to the classic full-length receptor (GR-A), seven highly conserved receptor isoforms with progressively shorter N-terminal transactivation domains (NTD) (GR-B, GR-C1, GR-C2, GR-C3, GR-D1, GR-D2, GR-D3) are produced from the single GR ge

The general practice workforce crisis in Europe: scale of the challenge and available policy responses.

General practice across the European region faces a systemic workforce crisis that threatens patient access to health services as well as the coordination, continuity, and equity of care within European health systems. In this Health Policy paper, we use an analysis of the WHO/Europe-Eurostat-Organisation for Economic Co-operation and Development (OECD) Joint Questionnaire on Non-Monetary Health Care Statistics dataset for 43 countries, evidence from a dedicated research collection, and a rapid review of policy interventions to assess the scale, drivers, and solutions to general practitioner (

Generation of transcriptomic resources in Oolichan to assess marine diesel gene expression responses.

Transcriptomic tools that quantify gene activity and expression offer a powerful approach to assess fish health response to environmental contaminants. Oolichan (Thaleichthys pacificus), a culturally and ecologically important and threatened anadromous fish, migrates through large river and estuarine waterways and may be exposed to oil products released from coastal industrial development and marine shipping activities. As part of the Conservation and Recovery Research on Oolichan in Haisla Territory (CAROOHT) program, we developed transcriptomic resources for Oolichan to assess gene expressio

Next-generation stimuli-responsive smart membranes: Developments in oil/water separation.

Effective treatment of oil-contaminated wastewater is essential due to its severe environmental and health impacts. The membrane-based separation is cost-effective, energy-efficient, and eco-friendly; however, fouling has remained a pressing issue. Stimuli-responsive membranes, which adjust their pore structure and surface properties in response to external triggers such as light, pH, and temperature, offer enhanced fouling resistance and improved separation performance. This review provides a comprehensive analysis of stimuli-responsive membranes for oil/water separation, emphasizing the role

Social well-being moderates behavioral therapy response for generalized anxiety disorder.

INTRODUCTION: Identifying moderators of treatment outcome for depression and anxiety offers promise for personalizing treatment or providing novel intervention targets. Indices of social well-being have emerged as candidate moderators due to their robust impact on mental health and quality of life. Here, we examine whether measures of social support, isolation, and loneliness moderate outcomes to behavioral activation (BA) and exposure-based therapy (EXP) for individuals with GAD. METHODS: This secondary analysis of a randomized clinical trial included 69 participants (31 EXP, 38 BA). Partici
