Sludge anaerobic digestion with high concentrations of tetracyclines and sulfonamides: Dynamics of microbial communities and change of antibiotic resistance genes.

This study established two mesophilic anaerobic digesters to ascertain the microbial dynamics and variation characteristics of antibiotic resistance genes (ARGs) during sludge anaerobic digestion (AD) with high concentration of antibiotics. System parameters, microbial community, ARGs (tetA, tetM, tetW, sulI, sulII) and integrase gene of class 1 (intI1) were analyzed. General performance of AD showed methane production was inhibited by 17.1% under the pressure of antibiotics. Microbial 16S rRNA high-throughput sequencing results showed the richness of microbial community decreased, but a highe

Leveraging existing 16S rRNA gene surveys to decipher microbial signatures and dysbiosis in cervical carcinogenesis.

The presence of dysbiotic cervicovaginal microbiota has been observed to be linked to the persistent development of cervical carcinogenesis mediated by the human papillomavirus (HPV). Nevertheless, the characteristics of the cervical microbiome in individuals diagnosed with cervical cancer (CC) are still not well understood. Comprehensive analysis was conducted by re-analyzing the cervical 16S rRNA sequencing datasets of a total of 507 samples from six previously published studies. We observed significant alpha and beta diversity differences in between CC, cervical intraepithelial neoplasia (C

The association between microbial community and ileal gene expression on intestinal wall thickness alterations in chickens.

The dynamic development of the animal intestine with a concurrent succession of microbiota and changes in microbial community and metabolite spectrum can exert far-reaching effects on host physiology. However, the precise mechanism of mutual response between microbiota and the gut is yet to be fully elucidated. Broilers with varying developmental degrees of intestinal wall thickness were selected, and they were divided into the thick group (H type) and the thin group (B type), using multiomics data integration analysis to reveal the fundamental regulatory mechanisms of gut-microbiota interplay

Tagging the vanA gene in wastewater microbial communities for cell sorting and taxonomy of vanA carrying cells.

Failure to understand the microbial ecology driving the proliferation of antibiotic resistance in the environment prevents us from developing strategies to limit the spread of antibiotic resistant infectious disease. In this study, we developed for the first time a tyramide signal amplification-fluorescence in situ hybridization-fluorescence-activated cell sorting protocol (TSA-FISH-FACS) for the characterization of all vanA carrying bacteria in wastewater samples. Firstly, we validated the TSA-FISH protocol through microscopy in pure cultures and wastewater influent. Then, samples were sorted

Calcitonin gene-related peptide expression in sepsis: postulation of microbial infection-specific response elements within the calcitonin I gene promoter.

BACKGROUND: Recently, we reported an unexpected ubiquitous expression of calcitonin (CT)-mRNA in a hamster peritonitis model of sepsis. Using this animal model,we undertook a study to further investigate the pattern of expression of the calcitonin I (CALC-I) gene and CT gene-related peptide (CGRP)-mRNA in sepsis. METHODS: Live Escherichia coli impregnated in agar pellets were implanted in the peritoneal cavities of hamsters. Twelve hours after sepsis induction, the septic and healthy control animals were sacrificed and tissues and peritoneal macrophages were collected. CGRP-mRNA content was e

Biochar application alters soil properties and microbial gene profiles in a continuous cassava cropping system.

Continuous cassava cropping can lead to soil degradation and disturbances in soil microbial functions, threatening the sustainability of cassava production systems. To address these challenges, this study examined the effects of biochar on soil physicochemical properties and microbial functional profiles in a continuous cassava cropping system. Two biochar application rates (0 and 3 Mg ha-1) were applied, and metagenomic sequencing was conducted to evaluate microbial community composition and functional genes related to carbon and nitrogen cycling in rhizosphere and bulk soils. The results dem

Gene Editing-Driven Engineering of Microbial Systems and Metabolites for Precision Medicine and Sustainable Environmental Remediation.

The engineering of microorganisms is undergoing a fundamental paradigm shift, transitioning from the construction of static cell factories to the programming of dynamically responsive living materials. However, translating molecular interventions into robust macroscopic functions requires overcoming distinct microbial-specific barriers, including delivery bottlenecks and genetic stability. In this review, we establish a unified Edit-Reprogram-Functionalize conceptual framework that systematically delineates transient genetic regulation from permanent genomic engineering. We critically examine

From niche to niche: investigating microbial communities and their specialised metabolite gene clusters in human microbiomes.

Diverse microbial communities within the human microbiome perform vital functions which influence both health and disease in hosts. Specialized metabolites produced by microbes via biosynthetic gene clusters (BGCs) drive ecological interactions and offer possibilities for therapeutic application. The biosynthetic capabilities of microorganisms present in human microbiomes are still mostly unexplored despite metagenomics advancements. The study examines the variety of microbial communities and BGC locations through metagenomic data from 1,191 samples across eight human microbiomes taken from th

Host- and microbial-mediated mucin degradation differentially shape Pseudomonas aeruginosa physiology and gene expression.

Pseudomonas aeruginosa is a hallmark pathogen of cystic fibrosis (CF) airway infections, capable of reaching high cell densities despite its limited ability to directly utilize mucin glycoproteins as a nutrient source. In the CF lung, however, P. aeruginosa may access preferred carbon sources (e.g., amino acids and short-chain fatty acids) through metabolic cross-feeding with co-colonizing mucin-degrading microbes. Although host-derived enzymes such as neutrophil elastase can also degrade mucins, the extent to which host-mediated mucin breakdown supports P. aeruginosa growth remains unclear. T

Subsurface landfill leachate contamination affects microbial metabolic potential and gene expression in the Banisveld aquifer.

Microbial communities in groundwater ecosystems can develop the capacity to degrade complex mixtures of chemicals resulting from pollution by landfill leachate. Monitoring this natural attenuation requires insight into the metabolic potential and activity of microbial communities. We contrasted the metagenomes and metatranscriptomes from a leachate-polluted aquifer downstream of the Banisveld (the Netherlands) landfill with uncontaminated groundwater, which revealed changes in microbial genomic content and activity. Banisveld landfill leachate contains mono-aromatic hydrocarbons and the assess

Influence of secondary water supply systems on microbial community structure and opportunistic pathogen gene markers.

Secondary water supply systems (SWSSs) refer to the in-building infrastructures (e.g., water storage tanks) used to supply water pressure beyond the main distribution systems. The purpose of this study was to investigate the influence of SWSSs on microbial community structure and the occurrence of opportunistic pathogens, the latter of which are an emerging public health concern. Higher numbers of bacterial 16S rRNA genes, Legionella and mycobacterial gene markers were found in public building taps served by SWSSs relative to the mains, regardless of the flushing practice (P < 0.05). In reside

Antibiotic-driven mechanisms in endogenous partial denitrification (EPD): Nitrite accumulation, microbial adaptation, functional gene responses and resistance gene proliferation.

The mechanisms by which antibiotics affect Endogenous Partial Denitrification (EPD) systems remain unclear. This study investigated the relationships between antibiotic type and pollutant removal, along with the underlying microbial metabolic mechanisms in an EPD system exposed to three antibiotics. The results showed that sulfadiazine (SD) and sulfamethoxazole (SMZ) exhibited higher chemical oxygen demand removal efficiency than tetracycline (TC). In the SD system, NO₃⁻-N removal was 84.13%, whereas other systems achieved 97%, which was associated with a higher abundance of the n arG and nirS

Oxygen availability shapes microbial community composition and function to determine antibiotic resistance genes and expression during composting: A pilot-scale study.

Temperature was considered the primary factor reducing antibiotic resistance genes (ARGs) during composting, with oxygen availability serving as the fundamental driver of temperature differences. However, how different oxygen levels shaped the composition and function of microbial communities, and subsequently affected ARGs and resistance expression during composting, remained unclear. Here, a pilot-scale long-term thermophilic aerobic and mesophilic facultative composting were constructed with metagenomics and stress (Oxygen/Antibiotic) plate screening to reveal the resistance potential and d
