Viral infections and risk of mental illness: A Mendelian randomization study.

BACKGROUND: Prior epidemiological evidence suggests associations between viral infections and psychiatric disorders, yet causal relationships remain insufficiently characterized. This study aims to investigate potential causal links using genetic instrumental variables. METHODS: A two-sample Mendelian randomization (MR) analysis was conducted using pooled European-ancestry genomic data. Exposures included hepatitis B virus (HBV), human immunodeficiency virus (HIV), severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), human papillomavirus (HPV), and Epstein-Barr virus (EBV) infections

Effects of nonalcoholic fatty liver disease on sarcopenia: evidence from genetic methods.

With the aging of the population, sarcopenia has become more common. Studies have shown a broad association between liver disease and sarcopenia. However, this link remains unclear. Our study explored the link between NAFLD and sarcopenia and predicting the pathogenesis. To begin, we investigated the causal relationship and genetic correlation between them using MR and LDSC. Second, each GWAS was annotated by MAGMA. The annotated genes were analyzed for pleiotropy using the PLACO approach. Finally, functional analysis was conducted on the identified pleiotropic genes. We observed a significant

Determinants of functional burden pleiotropy and gene dosage responses across human traits.

Pleiotropic and monotonic effects of gene dosage are central to understanding comorbidities in developmental pediatric and psychiatric disorders, yet the underlying biological processes are not well characterized. Here we develop a functional burden analysis to investigate the association of all protein-coding copy-number variants, genome-wide, with 43 complex traits in approximately 500,000 UK Biobank participants. We test variant associations disrupting 172 tissue or cell-type gene sets, finding associations for all traits, which we replicate in the All of Us cohort. Functional burden pleiot

Sexual antagonism for resistance and tolerance to infection in Drosophila melanogaster.

A critical task in evolutionary genetics is to explain the persistence of heritable variation in fitness-related traits such as immunity. Ecological factors can maintain genetic variation in immunity, but less is known about the role of other factors, such as antagonistic pleiotropy, on immunity. Sexually dimorphic immunity-with females often being more immune-competent-may maintain variation in immunity in dioecious populations. Most eco-immunological studies assess host resistance to parasites rather than the host's ability to maintain fitness during infection (tolerance). Distinguishing bet

Causal relationship between iron status and preeclampsia-eclampsia: a Mendelian randomization analysis.

BACKGROUND: Preeclampsia/eclampsia is a severe pregnancy-related disorder associated with hypertension and organ damage. While observational studies have suggested a link between maternal iron status and preeclampsia/eclampsia, the causal relationship remains unclear. The aim of this study was to investigate the genetic causality between iron status and preeclampsia/eclampsia using large-scale genome-wide association study (GWAS) summary data and Mendelian randomization (MR) analysis. METHODS: Summary data for the GWAS on preeclampsia/eclampsia and genetic markers related to iron status were

Infection Polygenic Factors Account for a Small Proportion of the Relationship Between Infections and Mental Disorders.

BACKGROUND: Several recent studies have suggested a role for infections in the development of mental disorders; however, the genetic contribution to this association is understudied. METHODS: We use the iPSYCH case-cohort genotyped sample (n = 65,534) and Danish health care registry data to study the genetic association between infections and mental disorders. To test the hypothesis that these associations are due to genetic pleiotropy, we estimated the genetic correlation between infection and mental disorders. Polygenic risk scores (PRSs) were used to assess whether genetic pleiotropy of in

Periodontitis and Sjogren's syndrome: a bidirectional two-sample mendelian randomization study.

OBJECTIVES: Observational studies indicated a controversial relationship between periodontitis (PD) and Sjogren's syndrome (SS). To overcome restrictions in conventional observational studies, we conducted a two-sample Mendelian randomization (MR) analysis to assess the potential bidirectional relationship between PD and SS. METHODS: We utilized the largest available genome-wide association study (GWAS) of European ancestry on both PD (17,353 cases-28,210 controls) and SS (2495 cases-365,533 controls) for MR genetic instrument selection. The random-effect inverse-variance weighted (IVW) metho

Causal relationship between psoriasis and sudden deafness: a Mendelian randomization study.

OBJECTIVE: This study aims to investigate the genetic link between psoriasis and sudden sensorineural hearing loss (SSNHL). METHODS: From a genetic standpoint, this study further highlighted the connection between psoriasis and SSNHL. Single nucleotide polymorphisms (SNPs) connected to SSNHL could be found using a genome-wide association study from the IEU OpenGWAS project website. In addition, the inverse variance weighted (IVW) technique was used to evaluate the connection between psoriasis and SSNHL. RESULTS: According to Mendelian randomization analysis, IVW supported the causal relation

Dissection of shared genetic architecture and biological association between psoriasis and cardiovascular disease based on genome-wide association studies.

BACKGROUND: The clinical link between psoriasis (PsO) and cardiovascular diseases (CVDs) is well-established, yet the genetic underpinnings of their comorbidity remain unclear. This study aimed to systematically map the shared genetic architecture between PsO and CVDs to identify key risk loci, effector genes, and biological pathways. METHODS: We analyzed large-scale genome-wide association study data for PsO and 11 CVDs to assess their genetic correlation. We then identified pleiotropic loci-variants associated with both PsO and CVDs-and applied colocalization analysis to test whether a sing

Drosophila Col4a1 Glycine mutations highlight allelic heterogeneity and mechanistic pleiotropy.

Collagen IV (Col4) is a heterotrimer containing a triple helical domain broken up by short interruptions. Mutation of Glycine residues within the Glycine-X-Y triple helical repeat leads to genetically dominant disease in humans that affects multiple organ systems. Mouse and cell culture-based models have revealed allelic heterogeneity, resulting in a range of Col4 secretion defects depending on the position of the mutation. However, genetic background also affects phenotypic severity, making it challenging to understand the precise underlying molecular mechanisms driving disease. Here, we char

Genomic Relationships, Novel Loci, and Pleiotropic Mechanisms across Eight Psychiatric Disorders.

Genetic influences on psychiatric disorders transcend diagnostic boundaries, suggesting substantial pleiotropy of contributing loci. However, the nature and mechanisms of these pleiotropic effects remain unclear. We performed analyses of 232,964 cases and 494,162 controls from genome-wide studies of anorexia nervosa, attention-deficit/hyperactivity disorder, autism spectrum disorder, bipolar disorder, major depression, obsessive-compulsive disorder, schizophrenia, and Tourette syndrome. Genetic correlation analyses revealed a meaningful structure within the eight disorders, identifying three g

Nicotine dependence and comorbid psychiatric disorders: examination of specific genetic variants in the CHRNA5-A3-B4 nicotinic receptor genes.

BACKGROUND: The associations between nicotine dependence and specific variants in the nicotinic receptor CHRNA5-A3-B4 subunit genes are irrefutable with replications in many studies. The relationship between the newly identified genetic risk variants for nicotine dependence and comorbid psychiatric disorders is unclear. We examined whether these genetic variants were associated with comorbid disorders and whether comorbid psychiatric disorders modified the genetic risk of nicotine dependence. METHODS: In a case control study of nicotine dependence with 2032 subjects of European descent, we us

Identifying shared polygenic risk across cancers.

BACKGROUND: Shared genetic susceptibility across cancers has been reported but is generally modest at the genome-wide level. Whether such shared polygenic risk exhibits structured convergence at regional or functional levels remains unclear. We investigated shared genetic risk across cancers by integrating local genetic correlation analyses with cross-cancer polygenic risk score (PRS) associations. METHODS: We estimated pairwise local genetic correlations across 16 specific cancers and one pan-cancer phenotype using SUPERGNOVA. Genome regions harboring multiple cancers with mutually correlate

From paradox to biology: shared genetic architecture underlies the inverse association between smoking and Sjögren's disease.

OBJECTIVES: Smoking has been consistently associated with a paradoxically lower risk of Sjögren's disease (SjD). We used genetic approaches to investigate whether this association reflects a causal effect of smoking or shared genetic architecture, and to identify shared loci that may help explain this paradox and inform disease biology. METHODS: We applied an integrated cross-trait genetic framework using genome-wide association data for smoking initiation and SjD. Conventional Mendelian randomization (MR) was used as an initial test of association, followed by genome-wide causal modelling (C

Causal association between vitamin D and gestational diabetes mellitus: a two-sample Mendelian randomization study.

BACKGROUND: Previous articles on the relationship between vitamin D and gestational diabetes mellitus (GDM) were inconsistent. Their relationship has been observed primarily through observational studies, and the causality of this association has not been established. METHODS: A two-sample Mendelian randomization (MR) research was conducted to test the causal association between vitamin D and GDM, utilizing publically available statistics from genome-wide association studies (GWAS). This study obtained genetic variants from GWAS including vitamin D (N = 373,045,10,783,672 Single Nucleotide Po

Genetic Causal Association between Iron Status and Osteoarthritis: A Two-Sample Mendelian Randomization.

Objective: Observational studies have shown the association between iron status and osteoarthritis (OA). However, due to difficulties of determining sequential temporality, their causal association is still elusive. Based on the summary data of genome-wide association studies (GWASs) of a large-scale population, this study explored the genetic causal association between iron status and OA. Methods: First, we took a series of quality control steps to select eligible instrumental SNPs which were strongly associated with exposure. The genetic causal association between iron status and OA was anal

Microevolutionary divergence and plasticity of laryngeal shape in the house mouse (Mus musculus).

The morphology of the larynx is essential for vocalization, respiration, and airway protection, yet the sources of phenotypic variation within species are not well understood. This study investigated whether genetic background exerts a stronger influence than environmental factors on laryngeal morphology in two house mouse strains. Using geometric morphometrics, we analyzed the size and shape of 79 laryngeal specimens and examined the effects of body size, genetics, obesity, exercise, and social housing. Our results demonstrate that genetic background significantly shapes laryngeal structure.

Investigating the common genetic architecture and causality of metabolic disorders with neurodegenerative diseases.

BACKGROUND: The co-occurrence of metabolic dysfunction and neurodegenerative diseases suggests a genetic link, yet the shared genetic architecture and causality remain unclear. We aimed to comprehensively characterise these genetic relationships. METHODS: We investigated genetic correlations among four neurodegenerative diseases and seven metabolic dysfunctions, followed by bidirectional Mendelian randomisation (MR) to assess potential causal relationships. Pleiotropy analysis (PLACO) was used to detect the pleiotropic effects of genetic variants. Significant pleiotropic loci were refined and
