A causal mechanism for childhood acute lymphoblastic leukaemia.

In this Review, I present evidence supporting a multifactorial causation of childhood acute lymphoblastic leukaemia (ALL), a major subtype of paediatric cancer. ALL evolves in two discrete steps. First, in utero initiation by fusion gene formation or hyperdiploidy generates a covert, pre-leukaemic clone. Second, in a small fraction of these cases, the postnatal acquisition of secondary genetic changes (primarily V(D)J recombination-activating protein (RAG) and activation-induced cytidine deaminase (AID)-driven copy number alterations in the case of ETS translocation variant 6 (ETV6)-runt-relat

Deciphering functional landscapes of rumen microbiota unveils the role of Prevotella bryantii in milk fat synthesis in goats.

BACKGROUND: The rumen microbiome is critical for regulating milk synthesis in dairy livestock, yet the molecular mechanisms linking microbial functions to host lipid metabolism remain poorly understood. While host genetics and microbial composition have been studied, integrative analyses of the rumen-blood-mammary gland axis remain lacking. RESULTS: Here, we present the goat rumen microbial reference gene catalog and 5514 metagenome-assembled genomes (MAGs) from 160 multi-breed rumen samples. Integrating this resource with lactation data from 177 Saanen dairy goats, we identify Prevotella spp

Sex-specific gut microbiota and metabolite signatures in Parkinson's disease: implications for personalized therapeutics.

Parkinson's disease (PD) is characterized by dopaminergic neuron loss and α-synuclein aggregation in the substantia nigra pars compacta (SNpc). It is a multifactorial disorder with motor and non-motor manifestations and growing evidence suggests that gastrointestinal dysfunction may precede motor onset. Sex differences influence PD risk, onset and clinical features, with men exhibiting higher prevalence and earlier onset, driven by hormonal, genetic, and metabolic factors. The gut microbiota communicates bidirectionally with the central nervous system (CNS) via the gut-brain axis, modulating n

Engineered Biosynthesis of Pharmaceutically Important Compounds.

Natural products are an important source of medicinal seeds. The discovery of novel biosynthetic enzymes from nature is important for their use as biocatalysts for the enzymatic synthesis of useful natural products. In addition, genetics and structural biology developments have enabled the engineering of enzymes for the production of unnatural analogs of bioactive natural products. In this review, I describe the recent research on these two topics, the exploitation of a novel secondary metabolite enzyme involved in the biosynthesis of the sulfonamide natural product antibiotic SB-203208, and t

Advances in schistosomiasis research.

This review reports on recent developments in the areas of diagnosis, chemotherapy and pathology, where female genital schistosomiasis was recently identified as a common cause of morbidity in Schistosoma haematobium-endemic areas. As a profound understanding of processes underlying resistance and susceptibility to infection as well as pathology is central to schistosomiasis control, detailed immunological studies of immunity and granuloma formation dominate the discussion. Studies on host genetics, on T-helper cell type 1 or 2 cytokines as well as on cell types influencing immunity and granul

HLA-B27 modulates the composition of the gut microbiota and drives a proinflammatory intestinal microecology.

The pathogenesis of autoimmune diseases such as spondyloarthritis (SpA), rheumatoid arthritis (RA), and inflammatory bowel disease (IBD) involves genetic factors and gut microbiota dysbiosis, which have been widely reported in patients and animal models. Although genetic factors are known to reshape the gut microbiota, the mechanistic role of the host gene-reshaped gut microbiota in mediating inflammatory diseases remains poorly characterized. This study focused on HLA-B27 to investigate its impact on the gut microbial composition and its association with HLA-B27 related autoimmune inflammator

The gut microbiota and kawasaki disease: exploring the role of microbial dysbiosis and metabolites in pathogenesis and therapeutics.

Kawasaki disease (KD) is an acute, immune-mediated medium-vessel vasculitis and the leading cause of acquired heart disease in children, yet its underlying etiology remains only partially defined. Emerging evidence implicates the gut microbiota as a key modulator of KD susceptibility, immune dysregulation, and therapeutic response. This narrative review aims to synthesise current insights linking gut microbial dysbiosis and microbial metabolites to the pathogenesis, clinical expression, and treatment of KD, and to explore microbiome-informed strategies with diagnostic and therapeutic potential

Differences in rhizosphere microbial communities between Fusarium wilt-resistant and susceptible watermelon cultivars.

To elucidate genotype-associated differences in rhizosphere microbial community assembly, this study compared the microbiomes of three Fusarium wilt-resistant and three susceptible watermelon cultivars using amplicon sequencing. Results revealed distinct bacterial and fungal community structures between the two groups. Notably, resistant cultivars harbored a higher number of unique operational taxonomic units and displayed greater fungal richness compared to their susceptible counterparts. Beyond taxonomic composition, co-occurrence network analysis demonstrated that the fungal community withi

Food-chain spirochetes: a unified hypothesis for Parkinson's disease and dementia risk.

Parkinson's disease (PD) and dementia have risen markedly in many regions. While aging, genetics, toxicants, and protein misfolding explain key aspects of pathology, they do not fully account for synchronized temporal trends or pronounced regional heterogeneity in recent increases. This hypothesis and theory article proposes that persistent microorganisms in industrial food systems-particularly biofilm-forming spirochetes in high-throughput poultry processing-may represent an under-recognized upstream contributor via the gut-brain axis. Analysis of Global Burden of Disease and FAOSTAT data rev

Miscarriage and the Microbiome: Host Genetics, Immunity, and the Reproductive Tract Ecosystem.

Background/Objectives: Pregnancy loss is a common and multifactorial complication of human reproduction, traditionally attributed to fetal chromosomal abnormalities, maternal anatomical and endocrine disorders, and immune dysfunction. Growing evidence now indicates that the maternal microbiome, particularly within the reproductive tract, plays a critical role in implantation, placental development, and the maintenance of immune tolerance during early pregnancy. Importantly, the influence of the microbiome on miscarriage appears to be strongly modulated by host genetic background and immune reg

Microbiome and Development of Ovarian Cancer.

Cancer of the female reproductive system involves abnormal cell growth that can potentially invade the peritoneal cavity resulting in malignancy and disease severity. Ovarian cancer is the most common type of gynecological cancer, which often remains undiagnosed until the later stages of the disease or until cancer has metastasized towards the peritoneum and omentum, compelling it to be a deadly disease complicating the prognosis and therapeutics. Environmental, genetics and microbial factors are the common mainsprings to the disease. Moreover, human beings harbor rich microbial diversity in v

Microbial chassis selection for niche-specific engineered live biotherapeutics.

The human microbiota comprises a vast and diverse array of microorganisms that play critical roles in maintaining health and modulating diseases. Engineered live biotherapeutic products (eLBPs) harness genetically modified microbes to perform defined therapeutic functions within the host. A central challenge in developing effective eLBPs is the rational selection of an appropriate microbial chassis, which requires consideration of safety, genetic tractability, functional performance and the target host niche. As different body sites present distinct physiological and biochemical conditions tha

Microbial heterogeneity-mediated treatment evasion and the potential for engineered live biotherapeutic products.

Microbial populations exhibit both genetic and non-genetic heterogeneity. In the clinical context, this heterogeneity is of concern as it provides subsets of microbial populations with enhanced immune evasion abilities and antimicrobial resistance. Fungal pathogens are of special concern as fungal diseases and antifungal resistance are increasing worldwide and similarities between eukaryotic cells make it challenging to identify targets that are toxic to fungi without also harming the human host. Engineered live biotherapeutic products (eLBPs) could provide an alternative and complementary app

Metabolic engineering strategies: utilizing different microbial strains and advanced technologies for the synthesis of high-valued terpenoids.

Terpenoids represent the largest and most structurally diverse class of secondary metabolites, with extensive applications in the pharmaceutical, nutraceutical, cosmetic, agricultural, fragrance, and biofuel industries. The growing demand for these compounds has resulted in extensive exploitation of plant-derived terpenoids, raising concerns regarding resource availability and sustainability. Consequently, microbial production has emerged as a promising alternative because of the high genetic tractability, rapid growth, and ease of metabolic engineering offered by microbial hosts. Decoding gut microbiome alterations in celiac disease: Implications for pathogenesis and treatment.

Celiac disease (CD) is a chronic immune-mediated disorder triggered by dietary gluten in genetically predisposed individuals and characterized by intestinal inflammation, epithelial damage, and loss of immune tolerance. While a strict lifelong gluten-free diet (GFD) remains the cornerstone of treatment, accumulating evidence indicates that it does not consistently restore gut microbiome composition or function, and many patients experience persistent symptoms despite good dietary adherence. The gut microbiome has emerged as a key modulator of immune homeostasis, intestinal barrier integrity, a

Shotgun Metagenomics Reveals Skin Microbiome Composition and Function in Infant Atopic Disease.

BACKGROUND: Atopic dermatitis (AD), food sensitization (FS), and food allergy (FA) frequently co-occur in infancy, but the factors underlying distinct atopic phenotypes remain unclear. Although FLG null mutations are major genetic risk factors for AD, they explain only part of disease heritability, suggesting a potential role for the skin microbiome. This study examined how early-life skin microbiome composition and its interaction with host genetics contribute to distinct atopic phenotypes in infancy. METHODS: We analyzed > 1000 skin swabs from 429 infants in the VITALITY cohort using deep s
