Microbial Fuel Cell Biosensor with Capillary Carbon Source Delivery for Real-Time Toxicity Detection.

A microbial fuel cell (MFC) biosensor with an anode as a sensing element is often unreliable at low or significantly fluctuating organic matter concentrations. To remove this limitation, this work demonstrates capillary action-aided carbon source delivery to an anode-sensing MFC biosensor for use in carbon-depleted environments, e.g., potable water. First, different carbon source delivery configurations using several thread types, silk, nylon, cotton, and polyester, are evaluated. Silk thread was determined to be the most suitable material for passive delivery of a 40 g L-1 acetate solution. T

Evaluation of NOx removal from flue gas and Fe(II)EDTA regeneration using a novel BTF-ABR integrated system.

A promising process is under development for the removal of NOx and regeneration of Fe(II)EDTA in a novel biotrickling filter-anaerobic baffled reactor (BTF-ABR) integrated system at 50 ± 0.5 ℃. In this work, we investigated the NOx removal capacity of a BTF under different O2 concentrations (7.0 vol%, 5.25 vol% and 3.5 vol%), and tested the effect of an ABR on NOx removal and regeneration of Fe(II)EDTA. The results showed that the NOx removal capacity was significantly increased with the O2 concentration reduced from 7.0% to 3.5%. The microoxygen environment produced by the BTF-ABR integrated

Electron donor-driven bacterial and archaeal community patterns along forest ring edges in Ontario, Canada.

Forest rings are 50-1600 m diameter circular structures found in boreal forests around the globe. They are believed to be chemically reducing chimney features, having an accumulation of reduced species in the middle of the ring and oxidation processes occurring at the ring's edges. It has been suggested that microorganisms could be responsible for charge transfer from the inside to the outside of the ring. To explore this, we focused on the changes in bacterial and archaeal communities in the ring edges of two forest rings, the 'Bean' and the 'Thorn North' ring, in proximity to each other in O

Fractionation of Mercury Stable Isotopes during Microbial Methylmercury Production by Iron- and Sulfate-Reducing Bacteria.

The biological production of monomethylmercury (MeHg) in soils and sediments is an important factor controlling mercury (Hg) accumulation in aquatic and terrestrial food webs. In this study we examined the fractionation of Hg stable isotopes during Hg methylation in nongrowing cultures of the anaerobic bacteria Geobacter sulfurreducens PCA and Desulfovibrio desulfuricans ND132. Both organisms showed mass-dependent, but no mass-independent fractionation of Hg stable isotopes during Hg methylation. Despite differences in methylation rates, the two bacteria had similar Hg fractionation factors (α

Substrate hydrophobicity and cell composition influence the extent of rate limitation and masking of isotope fractionation during microbial reductive dehalogenation of chlorinated ethenes.

This study investigated the effect of intracellular microscale mass transfer on microbial carbon isotope fractionation of tetrachloroethene (PCE) and trichloroethene (TCE). Significantly stronger isotope fractionation was observed for crude extracts vs intact cells of Sulfurospirillum multivorans, Geobacter lovleyi, Desulfuromonas michiganensis, Desulfitobacterium hafniense strain PCE-S, and Dehalobacter restrictus. Furthermore, carbon stable isotope fractionation was stronger for microorganisms with a Gram-positive cell envelope compared to those with a Gram-negative cell envelope. Significan

Cadmium solubility in paddy soil amended with organic matter, sulfate, and iron oxide in alternative watering conditions.

We successfully accounted for the dynamics of cadmium (Cd) solubility in paddy soil that contained a realistically low total Cd concentration (0.36 mg kg-1) after submergence and redrying by monitoring multiple chemical and microbial soil properties. The addition of rice straw promoted greater microbial iron reduction and induced a greater increase in soil pH after submergence, compared to the control and treatments with added gypsum or hematite. Consequently, dissolved Cd decreased to under the detection limit in one week in the straw-amended treatment but remained high even after eight weeks

Chlorine isotope effects from isotope ratio mass spectrometry suggest intramolecular C-Cl bond competition in trichloroethene (TCE) reductive dehalogenation.

Chlorinated ethenes are prevalent groundwater contaminants. To better constrain (bio)chemical reaction mechanisms of reductive dechlorination, the position-specificity of reductive trichloroethene (TCE) dehalogenation was investigated. Selective biotransformation reactions (i) of tetrachloroethene (PCE) to TCE in cultures of Desulfitobacterium sp. strain Viet1; and (ii) of TCE to cis-1,2-dichloroethene (cis-DCE) in cultures of Geobacter lovleyi strain SZ were investigated. Compound-average carbon isotope effects were -19.0‰ ± 0.9‰ (PCE) and -12.2‰ ± 1.0‰ (TCE) (95% confidence intervals). Using

Use of biogenic and abiotic elemental selenium nanospheres to sequester elemental mercury released from mercury contaminated museum specimens.

Mercuric chloride solutions have historically been used as pesticides to prevent bacterial, fungal and insect degradation of herbarium specimens. The University of Manchester museum herbarium contains over a million specimens from numerous collections, many preserved using HgCl(2) and its transformation to Hg(v)(0) represents a health risk to herbarium staff. Elevated mercury concentrations in work areas (∼ 1.7 μg m(-3)) are below advised safe levels (<25 μg m(-3)) but up to 90 μg m(-3) mercury vapour was measured in specimen boxes, representing a risk when accessing the samples. Mercury vapou

Water Activation on Sulfur Vacancies Enables Pyrite-Mediated Methylmercury Demethylation and Immobilization.

The accumulation of neurotoxic methylmercury (MeHg) in rice paddies presents a major global food safety challenge, yet effective in situ remediation strategies for flooded, anoxic soils are lacking. Here, we report that sulfur vacancy (SV)-rich pyrite efficiently degrades MeHg under dark, anoxic conditions via an innovative pathway involving vacancy-activated water (H2O). Experimental and theoretical evidence reveals that SVs serve a dual function: they enhance MeHg adsorption, weakening the carbon-mercury bond, while simultaneously activating ambient H2O to generate surface-bound hydroxyl rad

Insight into the enhancement effect of humic acid on microbial degradation of triclosan in anaerobic sediments.

Humic acid (HA) as one class of macromolecular substances plays important roles in mediating environmental behaviors of pollutants in sediments, but its effect on microbial degradation of triclosan (TCS), a common antibacterial drug, remains unclear. In this study, the effects of HA addition with different dosages (0-5%) on TCS degradation in anaerobic sediment slurries and the underlying microbial mechanisms were investigated. The results showed that HA addition significantly accelerated the TCS removal and the maximum removal percentage (30.2%) was observed in the sediment slurry with 5% HA

Development and characterization of DehaloR^2, a novel anaerobic microbial consortium performing rapid dechlorination of TCE to ethene.

A novel anaerobic consortium, named DehaloR^2, that performs rapid and complete reductive dechlorination of trichloroethene (TCE) to ethene is described. DehaloR^2 was developed from estuarine sediment from the Back River of the Chesapeake Bay and has been stably maintained in the laboratory for over 2 years. Initial sediment microcosms showed incomplete reduction of TCE to DCE with a ratio of trans- to cis- isomers of 1.67. However, complete reduction to ethene was achieved within 10 days after transfer of the consortium to sediment-free media and was accompanied by a shift to cis-DCE as the

Carbon tetrachloride transformation on the surface of nanoscale biogenic magnetite particles.

Iron-reducing conditions in subsurface environments promote dechlorination reactions via both biotic and abiotic pathways, the latter often mediated via biologically activated minerals formed by dissimilatory iron-reducing bacteria (DIRB). Here we report the major products and pathways associated with the abiotic transformation of carbon tetrachloride (CT) by nanoscale biogenic magnetite/maghemite particles produced by the DIRB Geobacter metallireducens. Product formation and free radical/carbene trapping studies indicate that CT transformation occurs via three parallel pathways. The first pat

Waste activated sludge stimulates in situ microbial reductive dehalogenation of organohalide-contaminated soil.

Due to its enriched organic matter, nutrients and growth cofactors, as well as a diverse range of microorganisms, waste activated sludge (WAS) might be an ideal additive to stimulate organohalide respiration for in situ bioremediation of organohalide-contaminated sites. In this study, we investigated the biostimulation and bioaugmentation impacts of WAS-amendment on the performance and microbiome in tetrachloroethene (PCE) and polychlorinated biphenyls (PCBs) dechlorinating microcosms. Results demonstrated that WAS-amendment increased PCE- and PCBs-dechlorination rate as much as 6.06 and 10.67

Effects of the addition of nitrogen and phosphorus on anaerobic ammonium oxidation coupled with iron reduction (Feammox) in the farmland soils.

Anaerobic ammonium oxidation coupled with iron reduction is termed as Feammox, and is a new nitrogen removal process. However, there is a paucity of studies on the response of nutrient additions on Feammox process in farmland ecosystems. In this study, we investigated the shifts of Feammox and iron-reducers under nitrogen (N) and phosphorus (P) applications via isotopic tracing and high-throughput sequencing technology. In the isotopic tracing experiment, Feammox rates was significantly greater in the N and/or P applications soil (0.184-0.239 μg N g-1 day-1) than in the no fertilizer soil (0.1

Performance of low temperature Microbial Fuel Cells (MFCs) catalyzed by mixed bacterial consortia.

Microbial Fuel Cells (MFCs) are a promising technology for treating wastewater in a sustainable manner. In potential applications, low temperatures substantially reduce MFC performance. To better understand the effect of temperature and particularly how bioanodes respond to changes in temperature, we investigated the current generation of mixed-culture and pure-culture MFCs at two low temperatures, 10°C and 5°C. The results implied that the mixed-culture MFC sustainably performed better than the pure-culture (Shewanella) MFC at 10°C, but the electrogenic activity of anodic bacteria was substan

One shuttle, two wins: AQDS cuts N2O while boosting denitrification in cold, carbon-limited biofilm systems.

Efficient biological nitrogen removal in biofilm systems is hampered under low-temperature and carbon-limited ("dual-low") conditions, where impaired microbial metabolism curtails denitrification and promotes nitrous oxide (N2O). Quinone moieties of humic substances can shuttle electrons between microorganisms, yet their role in sustaining denitrification under dual-low stress remains unknown. Here we show that supplementing biofilm microcosms with the model humic analogue anthraquinone-2,6-disulfonate (AQDS) raised nitrate (NO3--N) removal from below 60% to 81% at 4-6 °C and C/N = 4, while cu

Microbiological aspects of microbial electrolysis cells: Isolation, screening, and electron transfer mechanisms.

Biohydrogen is a leading candidate in the global shift toward renewable energy, offering a sustainable solution to reduce reliance on fossil fuels. This review underscores the use of microbial electrolysis cells (MECs) as a viable strategy to address increasing energy demands. In MECs, extracellular electron transfer (EET) is a vital mechanism that enables microorganisms to channel electrons (e⁻) from organic donors to external acceptors. Exo-electrogenic bacteria, viz., Geobacter, Shewanella, Pseudomonas, Nitrospira, and Desulfovibrio, are frequently reported for their ability to utilize Iron

Assembly Dynamics and Functional Divergence of Anaerobic Communities Driven by Iron Oxides.

Iron oxides play an important role in regulating global biogeochemical cycles, yet how their physicochemical properties influence community structure, function, and assembly remains poorly understood. Here, we investigated the effects of four representative iron oxides-ferrihydrite (Fh), goethite (Gt), haematite (Ht), and magnetite (Mt)-serving as terminal electron acceptors on microbial communities enriched from activated sludge. Metagenomic profiling revealed mineral-dependent divergence in community composition and functional potential. The communities developed on amorphous Fh demonstrated

Exploiting the intrinsic microbial degradative potential for field-based in situ dechlorination of trichloroethene contaminated groundwater.

Bioremediation of trichloroethene (TCE) polluted groundwater is challenging, with limited next generation sequencing (NGS) derived information available on microbial community dynamics associated with dechlorination. Understanding these dynamics is important for designing and improving TCE bioremediation. In this study, biostimulation (BS), biostimulation-bioaugmentation (BS-BA) and monitored natural attenuation (MNA) approaches were applied to contaminated groundwater wells resulted in ≥ 95% dechlorination within 7 months. Vinyl chloride's final concentrations in stimulated wells were between
